HLA antigens and infertility with sperm autoantibodies in man.

Vasectomized men with sperm auto-antibodies (S.A.A.) and high frequency of HLA-A20 antigen (Ag) were observed by Law et al. (1979); Hancock et al. (1983) observed the prevalence of HLA-A28 in infertile men with S.A.A. HLA-A, B and DR Ag were determined in a population of 80 Caucasian infertile men with (n = 22) or without (n = 58) agglutinating and/or cytotoxic S.A.A. in serum (S) and/or seminal plasma (SP), and the association between HLA-determinants and infertility with S.A.A. was investigated. The frequence of HLA-A28 Ag was found higher in the group with S.A.A. than without (18.2% vs 0.0%) but not statistically significant (chi 2 = 8.87; pc less than 0.15). The same result was obtained with HLA-B13 Ag (chi 2 = 8.87; pc less than 0.32). Lastly, there was no prevalence of HLA-DR Ag in men with S.A.A. In conclusion, no association was found between HLA-A, B and DR Ag and infertility with S.A.A. Nevertheless, two HLA-Ag were more frequently observed in the group with S.A.A.

Stimulation of motility and energy metabolism of spermatozoa from asthenozoospermic patients by 17 beta-estradiol.

Spermatozoa from nine asthenozoospermic patients were incubated for 3 h in Krebs Phosphate Ringer (KRP), supplemented or not with 17 beta-estradiol. 17 beta-estradiol increased the mean velocity and maintained the percentage of motility during the first 2 h of incubation. Oxydative metabolism and intracellular ATP concentrations were enhanced, too, whereas glycolysis remained unchanged.

Effects of estradiol and its antagonist tamoxifen on motility and metabolism of human spermatozoa.

An investigation was conducted to evaluate the motility and metabolism of human spermatozoa incubated at 37 degrees Celsius for 3 hours in a krebs phosphate Ringer solution containing different concentrations of 17beta-estradiol and tamoxifen. 17beta-estradiol (17beta E2) stimulated significantly the motility of human spermatozoa. The oxidative metabolism and the intracellular ATP concentrations are enhanced with 10-20 mcg/ml of 17beta E2; the lactate production remains unchanged however. Tamoxifen, an antiestrogen, inhibited the motility of spermatozoa, and was toxic at high concentrations. Its effects on the energetic metabolism is dose-dependent; it increases the oxidative metabolism and decreases the lactate production and intracellular ATP at 3 mcg/ml.

Objective evaluation of fibrosis in human testicular biopsies by analysis based on optical diffractometry.

Bilateral testicular biopsies were obtained from 27 patients and submitted to two different treatments for each of them: firstly, a classical fixation and colouring, permitting the histopathological diagnosis and subjective appreciation of the degree of fibrosis; secondly, a new objective technique based on optical diffractometry: this method is based on the analysis of light intensity distribution in the diffraction pattern of an original image. Using two different methods of discriminant analysis, we observed some errors due to the subjective examination; in particular, several fibrosed samples had been judged as 'normal' by the histopathologist. Moreover, we showed the existence of some heterogeneity between different slides from the same original biopsy.

Glycolytic activity of human spermatozoa in normospermic men and in men with abnormal spermograms.

The glycolytic activity of human spermatozoa was studied in samples from supposed fertile and infertile subjects. The quantity of L-lactate produced during the first hour of incubation in an enriched medium was measured. There was no difference between the oligozoospermic subjects and the volunteer donors constituting the control group. Lactate production was considerably decreased in men consulting for sterility but with a normal sperm examination, as well as in asthenozoospermic men. In oligoasthenozoospermic men abnormally high quantities of lactate were present in samples from certain individuals or it was unexpected, reflecting extensive metabolic disturbances. Positive correlations were noted between motility and glycolysis only in the controls. A negative correlation was noted between the concentration of spermatozoa and their metabolic activity, especially evident in oligoasthenozoospermic men.

[Liquefaction of human sperm by a nuclear magnetic resonance technic. Effect of temperature].

An investigation of the liquefaction of human semen was performed using the technique of Nuclear Magnetic Resonance, by measuring the time of spin-spin proton relaxation. Measurements which were made at 24 degrees C and 37 degrees C over a 15-day period showed that significant degradation processes and protein transformations occurred during the first 3-5 days. Different phases of these processes were identified.

Influence of cations and albumin on human spermatozoa.

The effects of potassium, calcium and magnesium cations, and albumin on motility of spermatozoa, were tested after 15 min, 4 hr, and 24 hr of incubation using laser Doppler velocimetry. The optimal concentration of K+ was found to be 11 mM; higher values decreased motility more severely than did lower ones, even when K+ was completely removed. Motility was increased in the presence of small amounts of calcium (from 0.8-1.7 mM) but was depressed by magnesium concentrations above 0.8 mM. Moderate concentrations of albumin (0.5-1%) improved motility, but the protecting effect of this compound--leading to a best sperm survival--appeared with higher concentrations (4%). In conclusion we observed that optimal ionic conditions presently found were corresponding well to the tubal environment in preovulatory phase.

Application of the optical Fourier transform for analysis of the spatial distribution of collagen fibers in normal and osteopetrotic bone tissue.

Optical Fourier analysis was applied for evaluation of the differences between normal and pathologically changed bone tissue. Collagen fibers were used as markers of bone structure. To prove the usefulness of this technique for objective mathematical analysis of these differences the spatial spatial distribution of collagen fiber bundles was evaluated in normal and osteopetrotic bone. The variation in the spatial distribution of collagen fiber bundles in cross sections of femur diaphyses was evaluated quantitatively by optical diffraction three groups of Fatty Orl-op strain rats, i.e. phenotypically normal animals, osteopetrotic (op/op) mutants and op/op-mutants cured by transplantation of normal syngenic bone marrow. The histological sections of decalcified bone were stained with Sirus-Red and then photographed under polarizing microscope. The Sirus-Red staining was used to enhance the natural birefringency of collagen fibers. Diffractograms obtained from microphotographs of selected bone section areas, i.e. outer and inner circumferential lamellae and haversian bone of normal and cured op/op animals as well as whole cortical bone and woven bone filling the medullary cavities in op/op mutants were analysed separately. Diffractograms contain summarized information on the size and relative position of these structures in histological sections. The radial and angular distribution of light energy were evaluated for each diffractogram with an electronic detector. The obtained distributions were described by several sets of parameters concerning the position, level and shape of local maxima and minima. Out of these parameters five with the highest discriminant power were chosen for further mathematical analysis. This analysis was based on the calculation of the position of centroids in the multidimensional space described by the linear functions of the chosen parameters for each of the evaluated bone section areas. The centroids (mean values of discriminant scores of each group) represent the centers of gravity of the analysed groups, while the separation of the centroids tested by the F-test illustrates the differences between the respective groups of selected bone section areas. A high level of separation of centroids was found when osteopetreotic bone was compared with normal one, what means that the spatial distribution, size and interstructural distances between the collagen fiber bundles in bone tissue in these two groups of animals differ markedly. A similar situation was observed when osteopetrotic bone was compared with bone tissue obtained from op/op mutants cured by transplantation of normal syngenic bone marrow. On the other hand, the level of separation of centroids was low when bone tissue of cured op/op mutants was compared with the control one, a finding which corresponds to the less pronounced histological differences between these two groups of animals. Polarizing microscopy of Picrosirius stained bone sections as a method for analysis of spatial distribution of collagen fibers by optical diffractometry.

Cross sections of femur diaphysis obtained from control and osteopetrotic rats were stained with hematoxylin-eosin (HE) and Picrosirius (SR). Analogous selected areas of bone sections photographed under a polarizing microscope were analysed by optical diffractometry. Since the collagen fibers are a good marker for the structure of bone tissue, their spatial distribution evaluated by optical diffractometry provides information on the tissue architecture. The Picrosirius staining technique enhances the natural birefringency of collagen fibers. Therefore, in the polarizing microscope, pictures of high contrast are obtained. This procedure, by increasing the amount of information in the image, increases the quantity of the data obtained by optical diffractometry in comparison with the HE staining method. The results obtained prove that SR staining combined with polarizing microscopy might be useful for optical diffractometry in analysis of the spatial distribution of collagen fibers in all connective tissues, where they could serve as markers of tissue architecture.

Enzymes in the seminal plasma from azoospermic men: correlation with the origin of their azoospermia.

Semi-quantitative estimations of the activities of 65 enzymes were studied for cases of azoospermia in an attempt to correlate the seminal enzyme profile with the origin of the azoospermia. One single enzyme--alpha-glucosidase--showed variation. All other specificities tested gave identical results for all the azoospermic samples and for normal controls. The alpha-glucosidase activity that is present in normal semen was absent from samples from patients with azoospermia associated with a complete obstruction of the genital tract, and also from all semen samples from vasectomized subjects. This enzyme appears to be secreted by the epididymis, and the activity present in azoospermic semen can provide information concerning the origin of the azoospermia and the functional state of the epididymis. Positive but reduced alpha-glucosidase activity appears to be frequently associated with inflammatory disease. The consequences of these findings are discussed.

[Congenital or acquired obstructions of the human epididymis: study of the motility of the spermatozoa above the obstruction].

Azoospermia is frequently due to an obstruction of genital pathways. In this work, concerning 22 patients submitted to surgical investigation for sterility, we made a systematic histological investigation of the lesions, and we studied the motility of epididymal spermatozoa. We observed with an unexpected frequency abnormalities of junctions either between seminiferous tubules and efferents ducts - leading to disappearance of rete testis - or between efferent ducts and epididymis. In each case a fibrosis of the epididymis was discovered, the consequence was either an interruption or, more frequently a simple narrowing of the lumen of the epididymal duct. The presence of spermiophages which destroyed all spermatozoa, was also systematically observed. The origin of the lesions (congenital, inflammatory, ischemic... ) could not be determined by simple histological study. Motility of spermatozoa was generally observed in the initial portion of the epididymal duct, but motile spermatozoa were sometimes still observed in the efferent tubules. Motility could be initiated by caffeine while other compounds as albumin, seminal plasma and different organic substrates, had no effects.

[Pharmacological effects of veralipride on the mammary gland of female rats: a complementary study (author's transl)].

Administration of 2 mg/kg/day of veralipride to intact female rats for 3 weeks provokes mammary gland development and, in some animals, the onset of secretory activity. These modifications do not appear after veralipride administration, under the same conditions, to castrated female rats. In castrated female rats previously conditioned by estradiol benzoate administration, some development of several acinous islets is observed in about half of the animals treated or not with veralipride. This suggests the development of a sensitivity of the mammary gland to the action of pituitary hormones, the secretion of which is stimulated by either castration or veralipride. It would seem that the absence of corpora lutea secretions prevented as great a mammary gland development as that observed in intact animals treated with veralipride.

Individual characteristics of human spermatozoa.

We have investigated the variability of some characteristics of a sperm sample in 18 young volunteers in good health who were examined twice per month over a period of two years, and in about one hundred men undergoing treatment for sterility and for whom more than 4 spermiograms had been established during the past three years. The results of statistical analysis indicate that the most characteristic individual feature is the concentration of spermatozoa.

Morphometric studies of cell nuclei by means of image analysis and computer data processing.

A review of the recent results of the authors in the field of morphometric studies of Feulgen-stained cell nuclei is given. The data acquisition with Quantimet 720 and data processing with computer were used in the study. The discrimination between male and female cell nuclei from buccal smears on the basis of morphometric data is described. The morphometry-based classification of cells from vaginal smears according to the computer procedure is presented. The results are discussed in the context of the general strategy of morphometric work.
