Comparison of preseasonal and coseasonal Allpyral with Depo-Medrone in summer hay-fever.

Three hundred patients with grass pollen hay-fever, with or without pollen asthma, were given one of three forms of treatment: preseasonal or coseasonal alum-precipitated pyridine extracted grass pollen (Allpyral) or methylprednisolone acetate in slow-release form (Depo-Medrone). Significant improvement was obtained with preseasonal Allpyral and with Depo-Medrone, but the degree of improvement obtained with coseasonal Allpyral fell within the limits of placebo response.Nevertheless, it is considered that the definite suppression of the pituitary-adrenal function which results from the use of a long-term steroid is not justified in a benign condition such as hay-fever.

Evidence for circulating IgE complexes in food allergy.

Antigen entry and the formation of immune complexes occur in atopic subjects after food ingestion. We have studied patients with asthma, eczema and arthralgia and we were able to detect the presence of IgE immune complexes and the antigen entry from the gut before and after food challenge. Food allergic subjects showed, after food challenge, the presence of IgE and IgG immune complexes, which correlates with the subsequent occurrence of symptoms. Both the symptoms and the appearance of IgE immune complexes could be prevented by pretreatment with oral sodium cromoglycate (SCG). IgE immune complexes were isolated by an isokinetic ultracentrifugation gradient and the components were analyzed for antigen-specific IgE and IgG as well as free antigen levels. The observation that SCG reduces antigen entry, the appearance of IgE immune complexes and the occurrence of symptoms point to the central role of IgE in the intestinal mucosa.

Critical evaluation of an indirect enzyme-linked immunosorbent assay for IgG antibodies to Dactylis glomerata.

Experiments are reported for the evaluation of an indirect ELISA using peroxidase conjugate for the detection of IgG antibodies to Dactylis glomerata (DG), a grass pollen antigen frequently responsible for allergic rhinitis and asthma. When optimized, the test proved to be useful in clinical practice for a semiquantitative, cheap and simply-performed determination of IgG antibodies to DG. The advantages and disadvantages of this method, compared with those of elaborate antigen-binding assays, are discussed.

Stimulation of human peripheral lymphocytes by methionine enkephalin and delta-selective opioid analogues.

The opioid peptide methionine enkephalin was shown to stimulate human peripheral lymphocyte proliferation in vitro in the absence of mitogen. A study of the time course of stimulation revealed a maximum proliferative response, measured by [3H]thymidine incorporation, after 4-5 days incubation. The kinetics of the response are similar to those for in vitro T cell responses to antigen rather than via polyclonal activation through lectin or anti-CD3 triggering, suggesting a physiological basis for the phenomenon. The stimulatory influence was blocked by the delta-selective antagonist ICI-174864, suggesting the mediation of classical opioid delta receptors. The opioid receptor specificity is further demonstrated using delta- and mu-selective agonists. The delta-selective agonists DSLET and DPDPE stimulate proliferation, whereas the mu-selective agonist DAGON was without effect.

Immunogenetic studies of multiple sclerosis patients in Caithness County, Scotland.

The present report indicates that there is an increased frequency of HLA-B7 in multiple sclerosis patients compared to unrelated controls and unaffected siblings. HLA-A3 was not significantly increased. The increased frequency of certain HLA antigens was not reflected in increased titres of serum measles antibodies in this group. In contrast to our previous studies, there were only minimal differences in the cellular response to measles antigen in multiple sclerosis patients as compared to siblings and unrelated controls.
