Missing evidence for HLA antigen association with Eales' disease, chorioretinitis, central serous retinopathy, and malignant choroidal melanoma.

Patients with Eales' disease, chorioretinitis, central serous retinopathy, or malignant choroidal melanoma were tested for HLA antigen deviation. When corrected p values (pc) are used, the first three disorders did not show any significant deviation, whereas a significant increase of HLA-Aw32 (pc = 0.026) was found in the malignant melanoma group. For conclusive evidence the latter finding needs confirmation by analysis of a greater number of patients with this disorder.

[HLA antigens in patients with rhegmatogenous retinal detachment (author's transl)].

An analysis of HLA-antigens was carried out in 132 patients with rhegmatogenous retinal detachment. While emmetropic and hyperopic patients showed no deviation from the normal HLA distribution pattern, HLA-B 12 was found to be significantly increased (30,3% versus a control frequency of 21,8%) at the Pc less than 0,05 level in myopic patients. This deviation frequency was identical in patients with and without a positive family history and was not related to the degree of myopia.

Association between HLA-B7 and allergic reactions to insulin in insulin-dependent diabetes mellitus.

Forty-four cases of insulin-dependent diabetes mellitus with cutaneous allergic reactions to insulin were typed for HLA-A and -B antigens. HLA-B7, which commonly is negatively associated with insulin-dependent diabetes mellitus, showed a positive association with insulin allergy. A second antigen, HLA-Bw21 may also be positively associated with local insulin allergy. These data were discussed in relation to heterogeneity of insulin-dependent diabetes mellitus and to the existence of B7-associated immune response genes for allergic-immunoreactivity to insulin.

The HLA linkage group and disease susceptibility.

The HLA linkage group, located on chromosome 6, is composed of serologically defined (SD-) loci (HLA-A, -B and -C) and of a MLC (= mixed lymphocyte culture-) or lymphocyte defined (LD-) locus (HLA-D). Different alleles of these loci are linked together as well as with loci for the synthesis of some complement components and the properdin factor B (= Bf-system) by strong positive linkage disequilibria (gametic association). The indication of a HLA linked genetic control of immunoresponsiveness makes the HLA system one of the biologically most important immunogenetic systems in man. The associations, found between single HLA determinants and diseases, can be explained in different ways, but may contribute to a better understanding of the etiopathogenesis of diseases and of the biological role of the HLA linkage group.

[Autolymphocytotoxins (CoCoCy) in different forms of hepatitis and cirrhosis (author's transl)].

Sera of altogether 282 patients with different forms of hepatitis and cirrhosis were screened for cold reacting complement dependant auto-lympho-cytotoxins (CoCoCy). These antibodies are 19S-IgM-immunoglobins and have no HLA-antigen-specificity. CoCoCy occurred in 48% of the patients with chronic aggressive hepatitis (CAH), in 14% of the patients with chronic persistent hepatitis (CPH) and in intermediate rates in the sera of patients with acute hepatitis. No correlations was found between CoCoCy and hepatitis B-antigen (HB-Ag). CoCoCy could be demonstrated also in 20% of the sera of a HB-Ag-positive and in 6% of a HB-Ag-negative control group. The serum concentration of CoCoCy is low. CoCoCy seems to be of T-cell-specificity and to reflect the overall-immunoreactivity without relation to the specificity of the antigenic stimulus. Thus demonstration of CoCoCy may be of pathogeneic and pathodynamic rather than of diagnostic interest.

HLA antigens and immunoresponsiveness to insulin in insulin-dependent diabetes mellitus.

HLA-A and B antigens were determined in 112 patients with insulin-dependent juvenile onset diabetes mellitus, who could be subdivided into "non" and "high responder" to insulin. The data revealed a trend of an association of these diabetes subgroups with only one of the diabetes-associated antigens HLA-B8 and HLA-BW15 and indicated the existence of at least two different genetic constellations for susceptibility to juvenile diabetes mellitus. One form with a strong immune-response to insulin seemed to be associated with HLA-BW 15 and the other form without humoral immunoreactivity to insulin seemed to be associated with the presence of HLA-B8 and the absence of HLA-B7.

HLA antigen segregation analysis in multiple sclerosis (MS) families.

HLA antigen segregation analysis was performed in 38 families with a total of 52 multiple sclerosis (MS) patients including 14 families with 2 affected sibs. HLA-A3 and especially B7 occurred more frequently in familial MS cases (propositi) (A3: 42.9%, B7: 64.3%) than in non-familial cases (A3: 35.8%, B7: 35.3%). As could be expected from the increased phenotype frequencies of A3 and B7, the haplotype A3-B7 showed a disturbed segregation among the MS patients but not among their healthy sibs. A second haplotype, A1-B8, was found more frequently than expected among the MS patients but also among their unaffected sibs. These data may suggest increased MS susceptibility associated with the haplotype A3-B7 and, conversely autoimmune or protective properties associated with A1-B8 in MS patients and their healthy sibs, respectively.

HL-A antigens in carcinoma of the breast, ovarium, cervix and endometrium: possible association of haplotype HL-A10-W18 with carcinoma of the breast.

24 HL-A antigens were determined in a total of 163 Caucasien cancer patients (60 breast cancers, 64 cancers of the cervix uteri, 19 cancers of the endometrium and 20 cancers of the ovary). The comparison of the antigen frequencies with the analogous values from 1000 healthy controls yielded no outstandingly significant deviations for cancer of the cervix, endometrium and ovary. In breast cancer patients, however, there might be an association with the antigens HL-A10 and W18 or the haplotype HL-A10-W18, as indicated by the results of a combined evaluation of our data together with the results of four different HL-A studies in breast cancer patients.
