Three acidic polysaccharide (AP) fractions and the prostaglandin-like substances (PLS) isolated from P. acnes were investigated regarding their chemotactic activities on polymorphonuclear leukocytes. Both AP's and PLS induced a significant chemotatic response, which suggests their involvement in inflammatory acne vulgaris.
Co-trimazine, a new preparation of sulphadiazine and trimethoprim, was compared with nitrofurantoin in a double-blind trial on patients with acute urinary tract infections. Co-trimazine had a bacteriological cure rate of 95% as compared to 88% for nitrofurantoin. There were no statistically significant differences in laboratory findings or in clinical side-effects between the two laboratory findings or in clinical side-effects between the two preparations. It was concluded that co-trimazine might constitute a valuable addition amongst drugs suitable for treatment of acute urinary tract infections.
The influence of 2 etracycline preparations, lymecycline and doxycycline, on leukocyte chemotaxis, was investigated in vitro and in vivo. Doxycycline in high concentrations caused a severe depression of both spontaneous and induced migration, an effect which was more pronounced than that caused by similar concentration of lymecycline. Corresponding results were obtained both in experiments on healthy volunteers given recommended dosages of the antibiotics, and in patients treated with doxycycline for postoperative infections.
Doxycycline was given as preventive preoperative treatment to 64 patients with fractured collum femoris. 46 patients, not administered doxycycline, served as controls. The overall incidence of postoperative infection was 6.3 and 6.5% in treated and control groups respectively. It is concluded that doxycycline preventive treatment offers no advantage in this type of operative procedure.
The influence of doxycycline on granulocyte migration was investigated on patients undergoing fracture surgery. The granulocyte migration of the patients was depressed when doxycycline was administered. Scanning electron microscopy demonstrated a decreased granulocyte adherence to glass as well as a decrease in number of filopods per cell in the presence of doxycycline.
A new doxycycline preparation, doxycycline polymethaphosphate sodium complex (DMSC), was compared to doxycycline in a cross-over study on six volunteers. The serum levels attained were of the same magnitude, but DMSC was shown to influence the serum bactericidal effect to a lesser degree. In vitro investigations of the influence on chemotaxis showed that DMSC even in high concentrations did not interfere with the spontaneous or induced migration to the same extent as doxycycline.
