Enrichment of gene-coding sequences in maize by genome filtration.

Approximately 80% of the maize genome comprises highly repetitive sequences interspersed with single-copy, gene-rich sequences, and standard genome sequencing strategies are not readily adaptable to this type of genome. Methodologies that enrich for genic sequences might more rapidly generate useful results from complex genomes. Equivalent numbers of clones from maize selected by techniques called methylation filtering and High C0t selection were sequenced to generate approximately 200,000 reads (approximately 132 megabases), which were assembled into contigs. Combination of the two techniques resulted in a sixfold reduction in the effective genome size and a fourfold increase in the gene identification rate in comparison to a nonenriched library.

Prevalence of vaginal colonization by drug-resistant Candida species in college-age women with previous exposure to over-the-counter azole antifungals.

We enrolled 382 college-age women in a cross-sectional survey to investigate the relationship between use of over-the-counter (OTC) azole-based antifungal drugs and vaginal colonization by drug-resistant Candida. This study showed no correlation (P=.506) between previous OTC exposure and colonization of drug-resistant Candida in vaginal flora. However, a small number of resistant Candida species isolates were obtained from women with a history of multiple exposures to OTC antifungals; given the widespread use of these products, this may be an emerging concern.

Evaluation of problem solving and communication skills of persons with schizophrenia.

Although much research has shown positive outcomes of teaching problem solving and communication skills to persons with schizophrenia, the amount of research on the assessment of these two sets of skills has been modest. The current study demonstrates an effective model for their assessment and includes a previously neglected procedure to determine if subjects attended to the skill prompts. It replicates and expands prior findings on problem solving and communication skills. Results indicated that persons with schizophrenia were generally deficit in problem solving skills. Persons without schizophrenia seemed to use more behavioral information when deciding if a problem existed and they were better able to differentiate between appropriate and inappropriate solutions to problems. Persons with schizophrenia were found to be deficit in communication skills that facilitate interaction and showed a style that subordinated their wants to those of others.

Linkage of protonation and anion binding to the folding of Sac7d.

The temperature, pH, and salt dependence of the folding of recombinant Sac7d from the hyperthermophile Sulfolobus acidocaldarius is mapped using multi-dimensional differential scanning calorimetry (DSC) and folding progress surfaces followed by circular dichroism. Linkage relations are derived to explain the observed dependencies, and it is shown that the data can be explained by the linkage of at least two protonation reactions and two anion binding sites to a two-state unfolding process. Circular dichroism spectra indicate that a native-like fold is stabilized at acid pH by anion binding. An apparent binding isotherm surface (folding progress versus pH and salt) is used to obtain intrinsic chloride binding constants as a function of pH for both sites. A saddle is predicted in the folding progress surface (progress versus temperature and pH) at low salt with a minimum near pH 2 and 20 degrees C with approximately 25% of the protein folded. The position of the saddle is sensitive to the intrinsic delta C degrees of unfolding and provides a third measure of delta C degrees independent of that obtained by a Kirchoff plot of DSC data and chemical denaturation. The observed enthalpy of unfolding approaches zero near the saddle making the unfolding largely invisible to DSC under these conditions. The linkage analysis demonstrates that the delta C degrees for unfolding obtained from a Kirchoff plot of DSC data should be distinguished from the intrinsic delta C degrees of unfolding. It is shown that the discrepancy between the free energy of unfolding for Sac7d obtained by DSC and that obtained by chemical denaturation may be explained by the linkage of protonation and anion binding to protein folding. The linkage analysis demonstrates the limitations of using the delta Hcal/ delta Hvh ratio an indication of two-state unfolding.

A single-bedtime-dose self-medication system.

A medication system in use at the Florida Mental Health Institute is based on the unit-dose procedure and weekly reviews of patients' medications. The program is aimed at reducing the use of polypharmacy and of concentrates and stabilizing patients on a single daily bedtime dose of medication. Once patients are stabilized on a single dose, they are trained by the nurses and through a drug group to manage their medications independently after discharge. The system has produced savings of up to 90 per cent of medication costs per patient and is gaining acceptance in other mental health facilities in the state. The authors believe it has the potential for even greater therapeutic and financial benefits by reducing the recidivism that occurs when discharged patients discontinue their drugs.

Grocery purchases as a function of obesity and assumed food deprivation.

Six hundred and four grocery shoppers were observed unobtrusively at three supermarkets to investigate the relationship between grocery purchasing and the factors of obesity, assumed food deprivation, social class, and meal hour. It has been suggested that shoppers will purchase more if they shop prior to eating rather than after (Stuart & Davis, 1972), although such a notion conflicts with Schachter's (1971) internal-external hypothesis and with the conclusions of one field study (Nisbett & Kanouse, 1969). We found that female shoppers who shopped prior to standard meal times consistently purchased more than those who shopped after meal times (P less than 0.001), regardless of social class or meal hour. Males displayed the same tendency though differences were not significant. No obese-normal differences were found for either sex. A negative relationship between obesity and social class was found amoung females (P less than 0.001).
