Evaluation of the monocyte channel of the Hemalog-D: correlation with visual microscopy.

Poor correlation between Hemalog-D and standard microscopy monocyte counts has been reported. To achieve a better correlation between the monocyte figures of the two systems, the Hemalog-D includes an alarm signal: the remainder. The machine monocyte count can be adjusted to approach the visual count: if the remainder is greater than +5, add the machine remainder to the machine monocyte count; if the remainder is between -5 and -10, subtract the machine remainder from the Hemalog-D monocyte count. This simple addition or subtraction of remainders expands the range of white cell differential counts which can be accepted from the Hemalog-D without subsequent visual microscopic study of blood films.

Comparative in vitro activity of 1-oxa-beta-lactam (LY127935) and cefoperazone with other beta-lactam antibiotics against anaerobic bacteria.

The in vitro activity of 1-oxa-beta-lactam (LY127935), cefoperazone (T-1551), cefuroxime, cefsulodin, cefaclor, cefotaxime, and cefoxitin on 85 anaerobic clinical isolates (30 Bacteroides, 30 Clostridium, 25 Peptococcaceae) was simultaneously determined by the agar dilution test in two different media, Brucella Agar (Difco Laboratories) and Wilkins-Chalgren agar. In Wilkins-Chalgren agar, 90% of Bacteroides were inhibited by (micrograms per milliliter): LY127935, 0.5; T-1551, 64; cefoxitin or cefuroxime, 8; cefsulodin or cefotaxime, 32; and cefaclor, 128. All Clostridia were inhibited in Wilkins-Chalgren by (micrograms per milliliter): LY127935, 4; T-1551, 2; cefoxitin, 6; cefuroxime, 0.12; cefsulodin, 0.5; cefaclor, 1; and cefotaxime, 8. All Peptococccaceae were inhibited by T-1551, cefsulodin or cefotaxime at 4 microgram/ml and by cefoxitin or cefuroxime at 1 to 2 microgram/ml. With cefaclor at 8 microgram/ml, 92% of strains were inhibited, and LY127935 at 16 microgram/ml only inhibited 64% of strains. LY127935 was the most active of the antibiotics tested against Bacteroides, showing good activity against Clostridia and poor activity on Peptococcaceae, whereas T-1551 was more active against Peptococccaceae and had similar activity against Clostridia and poor activity on Bacteroides. There are no significant differences between minimal inhibitory concentrations obtained in Brucella Agar and those obtained in Wilkins-Chalgren.

Determination of plasma malondialdehyde-like material and its clinical application in stroke patients.

Plasma malondialdehyde-like material (MDA-LM) was evaluated in 138 normal subjects and in a group of 57 stroke patients using a modification of the method of Smith et al. (1976). The basal level of MDA-LM in the control group was 35 mumol/l with a range of 22-50 mumol/l. Values above 50 mumol/l were found in 80% of the patients suffering from subarachnoid haemorrhage, in 68% of those with cerebral thrombosis, and in 17% with transient ischaemic attacks. None of the patients with cerebral embolism, intracerebral haematoma, or lacunar infarct had values above 50 mumol/l. Significant statistical differences were found between the control group and all the patients except those with lacunar infarcts.

Dysfunctional plasminogen in full-term newborn.

Full-term newborns (FTN) plasminogen was evaluated by different techniques (affinity chromatography, immunologic technique, casein method, and chromogenic substrate). The functional activity of the FTN plasminogen was about 50% of that of the adult. This suggests the possible existence of a functional anomaly of plasminogen in FTN. FTN plasminogen aminoacids were studied, and, besides small qualitative anomalies, a decrease in amino acid residues per mole of protein and a different N-terminal amino acid were detected.

Effects of somatostatin on gastric mucosal histamine and serotonin content in restrained rats.

The effects of continuous intravenous perfusion of somatostatin on the incidence of ulcers and the histamine and serotonin gastric mucosal content in rats subjected to 12 h of restraint were studied. 51 sets of Sprague-Dawley male rats (102 animals) were divided at random into two groups. Those in group I were immobilized and treated with an intravenous infusion of isotonic saline solution. Animals in group II were also immobilized and given intravenous cyclic somatostatin dissolved in isotonic saline solution; the initial dose was 25 microgram and a continuous infusion of 2.5 microgram/kg/h was administered thereafter. A decrease in the ulcer index was observed in the rats perfused with somatostatin (p less than 0.01) as well as a decrease in the histamine (p less than 0.001) and serotonin (p less than 0.01) gastric mucosal contents. These effects of somatostatin on restraint-induced stress ulcer in rats are probably to be explained by microcirculatory changes in the gastric mucosa determined by the vasoactive amines (histamine and serotonin). They constitute an important factor in the pathogenesis of these lesions.

Platelet adhesiveness in ischemic heart disease.

The adhesiveness of platelets to collagen and glass was studies ex vivo in groups of patients with ischemic heart disease (IHD) and acute myocardial infarction (AMI). Adhesiveness was increased to glass and normal collagen in both groups of patients. No relation was found between reinfarction rate and adhesiveness in the acute stage of AMI. Electrophoretic mobility of platelets was the same in the AMI patients and a normal control group. It suggested that the increased 'retention' of platelets in IHD patients might be due more to platelet hyperaggregation or to hypersensitivity to platelet aggregating agents than to altered adhesiveness.

Alterations in the coagulation and fibrinolysis system in pregnancy, labour and puerperium, with special reference to a possible transitory state of intravascular coagulation during labour.

Various tests of hemostasis were carried out during pregancy, labour and the puerperium in a group of 259 women. Determinations were carried out in the 1st, 2nd and 3rd trimesters, in the period of dilatation, the expulsion period, the period of expulsion of the placenta and the immediate postpartum period of labour and on each of the first 5 days of the puerperium. It was confirmed that during pregnancy there is an elevation of the fibrinogen degradation products (FDP) levels with a proportional increase in the numbers of positive protamine sulfate and ethanol tests. The proportion of positive protamine sulfate and ethanol tests reaches a maximum in the expulsion of the placenta coinciding with the presence of soluble complexes heavier than fibrinogen as detected by polyacrylamide gel electrophoresis and by column chromatography. All this indicates that there is a transitory intravascular coagulation produced during labour reaching its maximum at the time of birth and tending to become normalized in the first few days of the puerperium.

Isolation of fibrin(ogen) degradation products from normal plasma by affinity chromatographic study.

An attempt was made to develop a method to isolate directly the fibrinogenfibrin (FDP and/or fdp) degradation products from plasma by means of small chromatographic columns of activated Sepharose 4-B coupled with antifibrinogen serum. The study of the material adsorbed was performed by polyacrylamide gel electrophoresis in the presence of sodium dodecyl sulphate (PAGE-SDS). Four electrophoretic bands with antigenic capacity against antifibrinogen serum were observed. The study of their molecular weights, of their polypeptide composition after reduction, and of their immunological response against antisera anti-D and anti-E allowed their identification as fibrinogen, D-dimer fragment, and fragment E, respectively. The possibility of using this technique for the differential diagnosis between a primary fibrinogenolysis and a secondary fibrinolysis in a thrombotic process is suggested, as well as its use in the control of thrombolytic therapy.
