Tissue tropism and species specificity of poliovirus infection.

Paralytic poliomyelitis is caused by the destruction of neurons in the central nervous system during the lytic replication of poliovirus. The human gene for the poliovirus receptor has been cloned recently, and transgenic mice generated that are susceptible to this virus. This has allowed the critical amino acids that determine the specificity of poliovirus to be identified.

Assessment of bone density in the distal radius with computer assisted X-ray densitometry (CXD).

A modified and improved radiographic absorptiometry of the distal radius which enables on-site analysis, called computer assisted X-ray densitometry (CXD), was evaluated from the viewpoint of quality assessment. Its precision and the correlation with dual energy X-ray absorptiometry (DXA) was evaluated in 12 volunteers (mean age 44.7 years). The profile of CXD-measured radial bone mineral density (RBMD) from 142 subjects (75 premenopausal and 67 postmenopausal women, mean ages 44.9 and 50.6 years, respectively) were compared with previous data by other methodologies of bone mineral analysis. The intra-assay coefficient of variation (CV) was 0.617%, the inter-assay CV was 2.064%, and the inter-observer CV was 0.673%. The correlation between CXD-measured RBMD and DXA-measured RBMD was of statistical significance (r2 = 0.733, P < 0.01). The correlation of CXD-measured RBMD with age, height or weight corresponded well with previous reports. CXD-measured RBMD and DXA-measured vertebral bone mineral density (VBMD) also had a significant positive correlation, but their correlation was not so close (r2 = 0.149, P < 0.01). The discriminative ability of osteoporosis by CXD was of acceptable level (odd's ratio = 5.72, P < 0.05), when assessed by comparison with bone dystrophy score (BDS) on the plain vertebral radiogram. Although some problems remain in technical standardization, CXD could be an easy, inexpensive, and widely applicable alternative of non-weight bearing cancellous bone densitometry.

A novel peptide probe for studying the transbilayer movement of phosphatidylethanolamine.

Ro09-0198 is a cyclic peptide isolated from Streptoverticillium griseoverticillatum which recognizes strictly the structure of phosphatidylethanolamine (PE) and forms an equimolar complex with the phospholipid on biological membranes. To use the peptide as a probe for analyzing the transbilayer movement of PE, we labeled the amino-terminal amino acid of the peptide with biotin without changing either the reactivity or specificity of the peptide. The amount of the peptide bound to the membrane was measured by enzyme linked immunosorbent assay (ELISA) after extraction of the peptide from the membrane. The peptide showed a strict temperature-dependent binding to human erythrocytes and the binding increased with increasing temperature. Since the peptide bound to PE in a temperature-independent manner and the binding to membrane PE is not affected by membrane proteins, the present temperature-dependent binding of the peptide to the cell membranes was likely to reflect temperature-dependent translocation of PE. The binding of the peptide to erythrocytes differed greatly among animal species. The peptide also showed temperature-dependent binding to a human histocytic lymphoma cell line, U937, suggesting that the peptide will provide a novel and convenient probe for analyzing the transbilayer movement of PE in eukaryotic cells.

Accentuated temporal line on the frontal skull radiograph: a sign of hyperparathyroidism.

PURPOSE: To determine the importance of a temporal line (the marginal line for the attachment of the temporal muscle to the skull) that is accentuated on frontal skull radiographs of hyperparathyroid patients owing to subligamentous bone resorption under the temporal muscle. MATERIALS AND METHODS: Radiographs from skeletal surveys of 134 surgically treated patients with primary (n = 102) or secondary (n = 32) hyperparathyroidism (HPT) and frontal skull radiographs of 63 age-matched control patients were reviewed. RESULTS: An accentuated temporal line was the most frequent finding (29.4% of cases) in primary HPT, followed by subperiosteal bone resorption in the hand (8.6%), salt-and-pepper appearance of the skull (3.5%), and rugger-jersey spine (1.1%). In secondary HPT, an accentuated temporal line became less obvious as subperiosteal bone resorption advanced. This finding was not seen in the control group. CONCLUSION: An accentuated temporal line is another radiographic indication of bone resorption in HPT.

Datura stramonium agglutinin released histamine from rat peritoneal mast cells that was inhibited by pertussis toxin, haptenic sugar and N-acetylglucosamine-specific lectins: involvement of glycoproteins with N-acetylglucosamine residues.

The N-acetyl glucosamine (GlcNAc)-specific lectin Datura stramonium agglutinin (DSA) rapidly and sugar-specifically released histamine from rat peritoneal mast cells, and pertussis toxin (IAP) inhibited it, suggesting that DSA activated mast cells via an IAP-sensitive G protein pathway. The additive effects of DSA and basic secretagogues such as compound 48/80 that activate IAP-sensitive G protein directly suggest that they shared the same mechanism of action including involvement of the IAP-sensitive G protein. Using lectin-blotting, blots of the corresponding glycoproteins detected by DSA diminished by haptenic sugar or pretreatment of the cells with N-glycosidase F, suggesting that the binding of DSA was responsible for the mast cell activation. The other GlcNAc-specific lectins such as Phytolacca americana mitogen, Solanum tuberosum agglutinin and wheat germ agglutinin (WGA) inhibited the histamine release induced by DSA, suggesting that these lectins were antagonists, but DSA was an agonist. Sialic acid-specific Macckia amurensis mitogen (MAM) inhibited the histamine release, and neuraminidase-treatment decreased mast cell activation induced by DSA. At least four mast cell glycoproteins that have affinity to DSA, WGA and MAM and are sensitive to neuraminidase-treatment were detected by lectin-blotting. Some of them may be binding sites coupled to histamine release including the IAP-sensitive G protein pathway. DSA is a useful tool for studying signal transduction of mast cells including the involvement of the IAP-sensitive G protein.

MR of pulmonary hamartoma: pathologic correlation.

We describe the magnetic resonance (MR) appearance with pathologic correlation of six pulmonary hamartomas. All six tumors showed intermediate signal intensity on T1-weighted images and high signal intensity on T2-weighted images. Four of the six tumors showed a lobulated appearance separated by septa on precontrast T1-weighted or T2-weighted images. Gadolinium-enhanced T1-weighted images showed marked enhancement of the septa that separated the tumors into less well enhanced lobules; this was seen in all six tumors. Comparison between MR images and pathologic specimens showed that the regions with less enhancement corresponded to core cartilaginous tissue and septa; areas of marked contrast enhancement corresponded to cleft-like branching mesenchymal connective tissue that dipped into the cartilaginous core. MR images correlated well with the surgically resected tumors.

[MR imaging of fatigue fractures: characteristics and value for differential diagnosis].

The magnetic resonance (MR) appearance of five fatigue fractures was analyzed in an attempt to isolate characteristic MR findings which would facilitate the differentiation of this entity from bone neoplasms and osteomyelitis. The bone lesions reviewed for comparison included three osteoid osteomas, eight osteosarcomas, two malignant lymphomas and four osteomyelitis. Four characteristics useful for differentiation were found: 1) All fatigue fractures showed a smooth and continuous periosteal reaction with a superficial layer of high signal intensity on T 2-weighted images, while all malignant bone tumors and osteomyelitis showed irregular, interrupted or undetectable periosteal reaction, and osteoid osteomas showed no superficial high signal on periosteal reactions. 2) In all fatigue fractures and osteoid osteomas, abnormal signals in the bone marrow were ill defined, whereas they were well defined in all bone neoplasms and one of four osteomyelitis. 3) None of the fatigue fractures or osteoid osteomas was associated with extraosseous components. 4) Fatigue fractures showed little signal change in the surrounding soft tissues, while most of the bone tumors and osteomyelitis were accompanied by high signal in the adjacent soft tissues on T 2-weighted images. These MR characteristics are helpful in differentiating fatigue fracture from bone neoplasms and osteomyelitis, enabling unnecessary surgical procedures to be avoided.

Identification of novel members of G-protein coupled receptor superfamily expressed in bovine taste tissue.

Using reverse transcription-polymerase chain reaction (RT-PCR) and degenerate oligonucleotide primers, we amplified novel members of two different subfamily of G-protein coupled receptor (GCR) superfamily from bovine taste tissue. Type A receptor clones composed of multiple cDNA clones had significant similarity with putative olfactory receptor subfamily, while a single type B clone had significant similarity with peptide receptor subfamily. Physiological functions of these receptors in taste cells are discussed.

Improvement of blood compatibility on cellulose dialysis membrane. I. Grafting of 2-methacryloyloxyethyl phosphorylcholine on to a cellulose membrane surface.

A methacrylate with a phospholipid polar group, 2-methacryloyloxyethyl phosphorylcholine (MPC), was grafted on cellulose membrane for haemodialysis in an aqueous medium using cerium ion (Ce4+) as an initiator. The effects of the concentrations of MPC and Ce4+, and degassing of feed solution on the grafting of MPC on the surface and the membrane properties such as permeability and mechanical strength were examined. The grafted MPC composition depended on the concentrations of both the monomer and initiator in the feed solution. When the grafted MPC distribution was controlled by the monomer concentration, the permeability of the membrane decreased with an increase in grafted MPC distribution. On the other hand, the permeability was not changed from the original membrane's value when the MPC distribution was regulated by Ce4+ concentration. The tensile strength of the membrane did not change during the grafting of MPC and this indicated that the grafting had taken place in the amorphous region of the cellulose. These results suggested that this method is a promising way to improve the blood compatibility of a cellulose membrane without having an adverse effect on the haemodialysis membrane.

Improvement of blood compatibility on cellulose dialysis membrane. 2. Blood compatibility of phospholipid polymer grafted cellulose membrane.

The blood compatibility of a cellulose haemodialysis membrane whose surface was grafted with a methacrylate having a phospholipid polar group, 2-methacryloyloxyethyl phosphorylcholine, was evaluated with attention to platelet adhesion to the membrane surface and complement activation induced by the membrane. When the original cellulose membrane came in contact with platelet-rich plasma for 30 min, numerous platelets adhered to the surface and aggregated. On the other hand, the membrane grafted with 2-methacryloyloxyethyl phosphorylcholine effectively suppressed platelet adhesion and activation. This effect became more pronounced with increasing surface distribution. Especially, the 2-methacryloyloxyethyl phosphorylcholine grafted membranes, whose distribution exceeded 0.27, completely inhibited platelet adhesion, even when the contact time was 180 min. Moreover, the complement activation was also reduced with increased 2-methacryloyloxyethyl phosphorylcholine distribution on the surface of the membrane.

Thin-rim enhancement in Gd-DTPA-enhanced magnetic resonance images of tuberculoma: a new finding of potential differential diagnostic importance.

The article describes rim enhancement seen on Gd-DTPA-enhanced magnetic resonance (MR) images of two patients with tuberculoma. Pathologic examination of the surgical specimens disclosed that the peripheral portion of the tuberculoma, which showed contrast enhancement, was composed mainly of a fibrous capsule and epithelioid granulomas and that the central zone, which showed no contrast enhancement, was composed of caseous necrotic material. In a retrospective study of MR images from 20 consecutive cases of surgically proven bronchogenic carcinoma, none showed a thin enhancing rim on postcontrast MR images. Although confirmation of these findings must await further experience, the postcontrast MR findings described here may prove to be of value in distinguishing tuberculoma and possibly other granulomatous lesions from bronchogenic carcinoma.

MR imaging of thymoma: radiologic-pathologic correlation.

MR images in 17 patients with surgically proved thymomas (12 malignant and five benign) were reviewed and compared with pathologic specimens. In nine cases, MR images of excised specimens also were made and evaluated. On T2-weighted images, 11 of 12 malignant thymomas had an inhomogeneous signal intensity, half with and half without a lobulated internal architecture. None of the five benign thymomas had a lobulated internal architecture, and they all had a moderately or slightly inhomogeneous signal. Cystic regions and/or hemorrhage were noted pathologically, and corresponded to areas of inhomogeneous high signal intensity seen on T2-weighted images. Examination of the excised specimens in malignant thymomas showed that the lobulated configuration seen in the tumors was caused by thick fibrous septa. Our experience suggests that, although calcification cannot be identified, MR is helpful in making a differential diagnosis of mediastinal tumors and in determining malignancy of thymoma.

Intrathoracic neurogenic tumors: MR-pathologic correlation.

An MR-pathologic correlation of nine intrathoracic neurogenic tumors (three neurofibromas, four schwannomas, and two ganglioneuromas) was performed to characterize their MR appearances. The three types of intrathoracic neurogenic tumor were found to have characteristic appearances on MR images that should be helpful when establishing the differential diagnosis of posterior mediastinal tumors. The MR and histologic features of these tumors are illustrated and described in this essay.
