Genetic and linkage analysis of cleistogamy in soybean.

Early maturing cultivars of soybean [Glycine max (L.) Merr.] native to the shores of the Sea of Okhotsk (Sakhalin and Kuril Islands) and eastern Hokkaido (northern Japan) have been used in breeding for chilling tolerance. These cultivars have a strong tendency to produce cleistogamous flowers throughout their blooming period. This study was conducted to determine the genetic basis of cleistogamy in an early maturing cultivar, Karafuto-1, introduced from Sakhalin. Genetic analysis was performed using F1 plants, the F2 population, and 50 F3 families produced by crossing between Karafuto-1 and a chasmogamous cultivar, Toyosuzu. F2 plants had chasmogamous flowers, indicating that chasmogamy was dominant to cleistogamy. Analysis of F2 populations and F3 families generated segregation data that was close to a two-gene model with epistatic interactions, although a portion of the pooled F3 data on the frequency of chasmogamous segregants from cleistogamous families significantly deviated from the model. The results suggested that a minimum of two genes with epistatic effects were involved in the genetic control of cleistogamy. Furthermore, cleistogamy was associated with early flowering in the F2 and F3 populations. A gene for cleistogamy was linked to one of the recessive genes responsible for insensitivity to incandescent long daylength.

Preventive effect of dapsone on renal scarring following mannose-sensitive piliated bacterial infection.

Renal scarring has been thought to occur in the later stages of chronic pyelonephritis. We previously reported that mannose-sensitive (MS) piliated bacteria promoted renal scarring, which was prevented by antioxidants. The preventive effect of diaphenylsulfone (dapsone), which has a scavenging activity on active oxygen species, on renal scarring was examined. Female Sprague-Dawley rats were inoculated with clinical isolates of Serratia marcescens which had both MS and mannose-resistant pili or with recombinant strains which had MS pili on their surface; they were then administered 20 mg/kg of dapsone or not. Dapsone significantly suppressed scarring following infection of the kidney. The bacterial counts in the kidneys were not different in dapsone-treated and nontreated rats. We conclude that dapsone is effective in preventing renal scarring, and it is suggested that the clinical use of this drug may prevent renal scar formation following pyelonephritis.

Malignant fibrous histiocytoma with epithelial differentiation?

A case of recurrent soft part sarcoma with focal areas showing epithelial differentiation in the right thigh in a 78-year-old woman is reported. The primary tumor consisted of myxoid areas and solid areas, in which relatively uniform epithelioid tumor cells were arranged in sheets, whereas pleomorphism and a storiform pattern appeared in the recurrent tumors. Thus this tumor was suspected to be a malignant fibrous histiocytoma. However, further studies showed unexpected ultrastructural and immunohistochemical features. Cytokeratin immunoreactivity and tonofilamentlike structures probably indicated epithelial differentiation of some tumor cells. From the clinical and histological findings, this tumor should be identified as a malignant fibrous histiocytoma with phenotypic expressions of epithelial cell.
