[Trial of pentamidine in the treatment of trypanosomiasis of sheep inoculated with Trypanosoma brucei brucei].

An experimental study was carried out using 5 Trypanosoma brucei brucei subcutaneously infected sheep. Pentamidine treatment appeared in the CSF. Two animals were injected daily for 10 days and they died with drug intoxication. Two animals received drug every 3 days for 15 days: one of them died, the other was cured after 3 weeks of set-back. This preliminary study should be completed by experimentation made over a more important period of time.

[Antiparasitic activity of Cis-Pt (II) pentamidine in experimental trypanosomiasis of sheep caused by Trypanosoma brucei brucei].

The failure of trypanocidal therapeutics have made it a duty to use other classes of molecules which show a comparable trypanocidal efficacy with well-known medicines. In this study, the authors have evaluated the trypanocidal activity of a Cis-Platinum (II) salt, DNA-intercalating medicine used in antineoplastic therapeutics, associated with pentamidine: Cis-Pt (II) pentamidine. After primary screenings on a murine model of T. brucei brucei, Cis-Pt (II) pentamidine was evaluated on an experimental sheep trypanosomiasis model, standardized by the Institute of Neurological Epidemiology and Tropical Neurology of Limoges (France). Cis-Pt (II) pentamidine was able to cure infected mice, without relapse, after a single subcutaneous injection of 12.5 mg.kg-1 body weight. In sheep trypanosomiasis, the same treatment was used successfully during the first stage of infection. Neurological stage susceptibility and pharmacokinetics in sheep will be evaluated in a further study.

[Treatment of human African trypanosomiasis].

The melarsoprol is still the only active drug during the nervous stage of African human trypanosomiasis, its prescription still relies on empiric rules and because of its toxicity, it appears necessary to study in gaseous phase chromatography and mass spectrometry, its pharmacokinetic in the serum and the cerebrospinal fluid (CSF) of the patients at the evolutive stage of the illness. Because of the early penetration of the Trypanosoma into the central nervous system, new therapeutic investigations should only search molecules capable to pass through the blood brain barrier: this in unfortunately not always the case, the rifamycins diffusing easily into CSF, they seem to offer a reliable way of research. For these therapeutic investigations, sheep is an easy to handle experimental model and of a reasonable cost.

Animal lentivirus replication and reverse transcriptase inhibitors.

We summarize the pathogenesis of animal lentiviruses (visna-maedi virus, caprine arthritis and encephalitis virus, and equine infectious anemia virus), which have raised considerable interest since the discovery of human lentiviruses. The human lentiviruses possess structural, genetic, and clinical properties similar to those of animal lentiviruses. We describe the different mechanisms of and the principal work on reverse transcriptase inhibitors of animal lentiviruses, such as HPA-23, phosphonoformate, or 2',3'-dideoxynucleosides. Animal viruses of this family may serve as models for infection with human lentiviruses such as human immunodeficiency virus, the etiologic agent of AIDS.
