A simple and rapid radioimmunoassay for human interleukin-6.

Studies were undertaken to develop a sensitive radioimmunoassay for human IL-6 using commercially available reagents. The assay utilized a polyclonal rabbit anti-IL-6 binding to iodinated IL-6; the reaction was carried out in solution and the immune complexes were precipitated using anti-rabbit IgG coupled to magnetic particles. Using a format where sample IL-6 was added in advance of the radiolabelled IL-6, the working range of the assay was found to fall between 0.1 and 10 ng/ml (IC50 around 1 ng/ml) for a 50 microliters sample volume, and was run overnight. However, the assay could be completed in 2 h, using a direct competitive format when less sensitivity is required (working range 1.5-25 ng/ml, IC50 12 ng/ml). The RIA correlated directly with a standard functional assay for IL-6 (proliferation of the mouse hybridoma B9.9) for both recombinant IL-6 and natural IL-6 from human monocytes. The total assay variance was less than 12% and no reactivity with interleukins-1-5 was found. Using the RIA, IL-6 produced in culture by human monocytes in response to various stimuli (LPS, IL-1, dibutyryl cAMP) was measured.

Bucladesine

A Pascal computer program for digitizing lateral cephalometric radiographs.

The authors describe a new program for cephalometric analysis which uses a commonly available microprocessor (computer) and digitizing pad to register fifteen commonly identified cephalometric landmarks to produce a meaningful analysis which is printed out for permanent or hard-copy record. Conventional and digitizing errors of cephalometric measurement are reviewed, with a discussion of the advantages of computer-assisted programs. The authors describe a program that uses vectors and vector algebra and the capabilities of the Pascal computer language to determine angular measurements and distances. It is suggested that computer-assisted cephalometric programs will likely be widely used in the near future, providing the orthodontist with a superior method of cephalometric analysis with respect to accuracy and speed of completion.

Comparison of media for direct isolation and transport of Shigellae from Fecal specimens.

Xylose-lysine-deoxycholate (XLD) agar, SS agar, and MacConkey agar for isolating shigellae from fecal specimens were compared. XLD agar was superior to both SS agar and MacConkey agar for isolating Shigella sonnei, and both XLD and SS agar were superior to MacConkey agar for isolating S. flexneri. Direct plating of the fecal specimens in the field resulted in a greater yield of shigellae as compared to transporting specimens to the laboratory either in holding media or enrichment broth. Buffered glycerol saline was superior to other transport media evaluated, yielding 83% of shigella isolates when plated within 48 hr as compared to direct plating. The combination of XLD agar and SS agar is recommended for direct isolation of shigellae, and, whenever possible, these solid media should be taken to the bedside and inoculated directly.
