[Placental-spermatic beta 2-globulin: its immunochemical identification, physicochemical characteristics and localization in the reproductive system].

A new antigen with beta 2-globulin mobility and M. W. of 20 kD, named placenta-sperm beta-globulin (PSBG), was identified in human early placental tissue. Using immunodiffusion method, PSBG was found in the seminal plasma with the concentration about 120 mg/l, in the amniotic fluid 1-5 mg/l, in the liquor 1-10 mg/l, and also in fetal kidney and stomach extracts. The examination of the male reproductive system revealed PSBG levels in the extracts of adults and fetus.

[Affinity chromatography of mouse and rat alpha-fetoproteins on immobilized diethylstilbestrol].

Alpha-fetoproteins (AFP) from amniotic fluid of mouse and rat demonstrate high affinity and specificity during their binding with immobilized diethylstilbestrol, which allows to isolate these two proteins by one step using the method of affinity chromatography on Sepharose with immobilized diethylstilbestrol. Meanwhile the yield of mouse AFP was 42%, and rat AFP--75%. The preliminary incubation of the amniotic fluid of rat and mouse with free estradiol results in abrupt fall of AFP outcome, which may testify to the binding of estradiol and diethylstilbestrol by the same receptor sites on AFP molecule.

[Cellular localization of human secretory beta globulin in normal and tumor tissues].

Distribution of secretory beta-globulin (S beta G) which possesses affinity for steroids was investigated immunohistochemically. Tissue specificity of S beta G, produced in adult secretory epithelial cells of the seminal vesicles, salivary glands, prostate, bronchi and mammary gland was discovered. The protein was not detected in fetal and embryonal tissues. S beta G synthesis is abnormal in neoplasms: its expression partly preserves in breast cancer cells and increases in epithelium of mammary ducts near the focus of malignancy. In lung cancer and bronchial glands cells near the focus of neoplastic transformation S beta G positive reaction was not observed.

[The content of fibronectin in the blood of patients with psoriasis].

Fibronectin, a high-molecular glycoprotein, one of acute-phase proteins, contributes to cellular proliferation and macrophagal activity regulation, and its blood plasma levels are an indicator of the reticuloendothelial system functional activity. Blood plasma fibronectin levels were measured in psoriasis patients before and after hemoperfusion. These levels were found significantly increased in the course of hemoperfusion treatment, this being associated with disappearance of psoriatic elements. A correlation between blood plasma fibronectin level and NBT test parameters, on the one hand, and severity of psoriasis course and efficacy of therapy, on the other, has been revealed.

[Immunochemical identification of onco-ovarian acid-soluble alpha-2-globulin].

Antisera were obtained by rabbits immunization with pooled extract from ovarian carcinoma and its metastases into the ome tum. Using standard test-system antigen, precipitating in agar, was identified in the tissue of 12 out of 24 primary ovarian carcinomas and in 8 out of 20 metastases. It was not revealed in adult healthy internal organs tissues and in fetal tissues, except embryonal large intestine, where it was determined in trace amounts in isolated samples. Using immunodiffusion method it wasn't determined in blood serum of healthy people and patients, pregnant women and neonates. Immunochemical identification using standard test-systems showed that this antigen is not identical with already known carcino-embryonic antigen, placental, reactive and onco-ovarian proteins. It presents acid-soluble alpha-2-globulin (ASAG-2) with MW 55 kD determined by gel-filtration and 55 kD determined by electrophoresis in 10% polyacrylamide gel with dodecylsulfate under reducing conditions. Physico-chemical and antigenic properties of ASAG-2, as we think, give the opportunity to present it as a new onco-ovarian antigen, which differs from the known proteins.

[Competitive affinity chromatography of human alpha fetoprotein on immobilized diethylstilbestrol].

Human alpha-fetoprotein (AFP) was isolated by affinity chromatography method on immobilized diethylstilbestrol from butanol extract of abortive material. Elution from the column was performed with 10% aqueous buffered butanol solution, pH 8.6. During one procedure human AFP-preparation containing about 10% of AFP and about 90% of albumin was obtained, with the yield about 60%. The preliminary incubation of extract of the abortive material with estrone raised AFP yield up to 85% with the increase of AFP content in the preparation up to 35%, and preincubation with estriol and estradiol caused the increase of the yield up to 88-92%, and AFP content in the preparation was 50% and 65%, respectively. The preincubation of human AFP with diethylstilbestrol lowers the yield of this protein, which testified to the possible binding of human AFP with free diethylstilbestrol; testosterone, hydrocortisone and desoxycorticosterone caused the increase of the yield of AFP. So the competitive variant of the affinity chromatography on immobilized diethylstilbestrol makes it possible to raise human AFP preparation purity and yield by decreasing the competition between AFP, and not binding free steroid hormones, ad albumin for immobilized diethylstilbestrol.

[Immunochemical identification and physico-chemical characteristics of specific alpha 2 globulin of human granulocytes].

The antigen which has alpha 2-globulin mobility was identified immunochemically in leukolysate and pus extract. This antigen was localized in polymorphonuclear leukocytes by the immunofluorescence technique in the blood of healthy donors. The alpha 2-globulin of granulocytes (alpha 2GG) is not identical to lactoferrin, lysozyme, granulocyte elastase, fibronectin, fetal hemoglobin, amyloid P-component of the serum. The molecular mass of alpha 2GG is equal to 50 +/- 8 Kd. in the pus extract. The biological role of alpha 2GG is unknown.

[The immunochemical identification of beta-globulin in human seminal plasma].

Using affinity chromatography there was isolated a protein from human seminal plasma, which was named beta-globulin of seminal plasma (beta-GSP). It was demonstrated, that beta-GSP possesses heterogeneity, consists of two subunits with the M. W. about 19 and 15 kD, it has electrophoretic mobility of b1-globulin and possesses affinity to immobilized 17-beta-estradiol. Relatively high concentration of beta-GSP was found in seminal plasma. Besides, this protein was determined in small amounts in saliva. One can suppose, that beta-GSP is a secretory protein.

[Serum ferritin and trophoblastic globulin in genital tumors in women].

Blood serum levels of ferritin and trophoblastic globulin were measured by immunoassay in 389 and 114 cases, respectively, suffering malignant or benign tumors of the uterus and ovary as well as in controls. Hyperferritinemia identified at serum ferritin levels in excess of 200 micrograms/l was established in 94% of cases of ovarian cancer, 57%--benign ovarian tumors, 60%--endometrial carcinoma and in 16% of patients with uterine myoma. Patients with ovarian and uterine malignancies were shown to have the highest serum ferritin levels. The study failed to establish an increase in trophoblastic globulin concentration in cases of nontrophoblastic tumors of the genitals. It is suggested that serum ferritin level be measured in patients presenting with ovarian and uterine tumors and in subjects at high risk for ovarian cancer to assure early diagnosis of disease.

[The cellular localization of the fertility alpha 2-microglobulin in tumors of the reproductive system].

Seventy-five samples of human tumors were examined immunohistochemically for fertility alpha-2-microglobulin also known as progesterone-associated protein of the endometrium. The protein was detected in 35.7% (5 of 14) endometrial cancer samples and in 20% (2 of 10) of ovarian malignancies. Tumors of the stomach, colon, breast, lung and some other sites failed to reveal the antigen with the exception of a single case of pulmonary adenocarcinoma which showed the ectopic expression of the protein. Fertility alpha-2-microglobulin is stage-specific tissue marker of the endometrium. It is expressed at the final stage of cell differentiation and is partially lost in cancer.

[Comparative physico-chemical characteristics of trophoblastic beta-1 glycoprotein preparations isolated from retroplacental blood].

Trophoblastic beta 1-glycoprotein preparations (TBG-1, TBG-2) isolated by different methods preserve their physicochemical and immunochemical properties. According to the data of disk-electrophoresis and densitometry TBG-1 preparation obtained using two-stage method is highly purified and protein yield is about 20%, while TBG-2 isolated by multi-stage method has a small number of admixture proteins.

[Detection of trophoblastic beta 1-glycoprotein in tumor tissue and blood in ovarian neoplasms].

Using immunochemical analysis methods (the reaction of precipitation in agar, immunoenzymatic method, immunofluorescence), trophoblastic beta 1-glycoprotein (TBG) concentration in tumour tissue and in the blood serum of patients with ovarian cancer was studied. By rabbits immunization with glycoprotein fraction of ovarian adenocarcinoma, dissolved in 0.6 M sulfosalicylic acid, the authors obtained antibodies to TBG. Immunoenzymatic method showed, that TBG level is raised during ovarian cancer (more than 3 micrograms/l): in 18% of tumour extracts, in 12.5% of blood sera samples and in 41.6% of cases in ascites fluid. Utilizing indirect immunofluorescence method morphological structures of trophoblastic type were identified in paraffin sections of ovarian adenocarcinoma. The authors suppose, that such structures may be responsible for TBG biosynthesis in ovarian tumours.

[Affinity chromatography of human alpha-fetoprotein on immobilized estrogens].

Human alpha-fetoprotein was isolated from abortive material with the help of affinity chromatography on immobilized estrogens. After butanol extraction from the abortive material human AFP obtained the ability for affinity binding with immobilized estrogens. The addition of estrogens to the extract of isolated AFP preparation and incubation with them did not lower AFP binding with immobilized estrogens during the experiments, using affinity chromatography. A 10% buffered aqueous butanol solution was most optimal for elution. The data obtained can suggest that AFP in biological fluids is bound to estrogens, and butanol extraction deestrogenizes human AFP. The mechanism of AFP binding to estrogens in vivo is, evidently, carried out with the help of specific unknown carrier, as AFP does not bind free estrogens.
