Protection of mice against the lethal toxicity of a lipopolysaccharide (LPS) by immunization with anti-idiotype antibody to a monoclonal antibody to lipid A from Eikenella corrodens LPS.

We produced anti-idiotype antibodies to antibody to lipid A from Eikenella corrodens. The ALA-1 monoclonal antibody (immunoglobulin M [IgM] isotype), which had already been produced in our laboratory (T. Kato, I. Takazoe, and K. Okuda, Infect. Immun. 57:656-659, 1989), had reacted strongly with lipid A from E. corrodens, Escherichia coli, and Salmonella minnesota. Four anti-idiotype monoclonal antibodies to ALA-1 (Ab1), designated A2LA-1 (IgG1 isotype), A2LA-2 (IgG2a isotype), A2LA-3 (IgG2a isotype), and A2LA-4 (IgG3 isotype), which recognized the idiotype Ab1, were produced. A2LA-1, A2LA-2, and A2LA-3 were capable of over 61% inhibition of ALA-1 reactivity to E. coli J5 lipid A in an enzyme-linked immunosorbent assay system. The sera of mice and rabbits immunized with the anti-idiotype antibodies revealed that the internal image anti-idiotype antibody induced the production of IgG antibodies that cross-reacted with or bound to lipid A. These studies indicate that A2LA-1 and A2LA-2 contained an antigenic epitope that mimicked lipid A. Immunization of mice with A2LA-1 resulted in prevention of lethal toxicity from E. coli J5 lipopolysaccharide.

[A rapid diagnosis (SK-013) for periodontitis based on the enzymatic activity of periodontopathic bacteria. Study on the characterization of the SK-013 and the enzyme specificity].

We present studies for development of an enzymatic diagnostic method for periodontitis. A rapid and sensitive diagnostic method named SK-013 was provided by Sunstar Inc. to evaluate the peptidase activities specifically derived from periodontopathic bacteria such as Bacteroides gingivalis, Bacteroides forsythus and Treponema denticola and some strains of Capnocytophaga species. The results obtained indicated the specificity of the enzymatic reaction in this system.

[A rapid diagnosis (SK-013) for periodontitis based on the enzymatic activity of periodontopathic bacteria. The relationship between the enzymatic activity (SK-013) and B. gingivalis, B. forsythus, T. denticola in subgingival microflora].

SK-013 was developed for rapid detection of the peptidase activity in subgingival plaque samples. The purpose of this study was to determine whether SK-013 could be a marker for the presence of periodontopathic bacteria including Treponema denticola, Bacteroides gingivalis and Bacteroides forsythus which produce trypsin-like enzyme. Subgingival plaque samples were taken with 3 paper points from 10 clinically healthy sites and 30 periodontal diseased sites. The SK-013 activity of plaque sample was assayed and the cells of T. denticola, B. gingivalis and B. forsythus in the sample were counted by immunofluorescence technique. In diseased sites, both the SK-013 activity and the cell counts of these organisms were significantly higher than those in healthy sites. The proportions and cell counts of these organisms and the SK-013 activity were closely correlated with clinical parameters including Gingival Index, Plaque Index, and Probing depth. The correlation between the presence of these organisms and the SK-013 activity was significant. Correlation coefficients between the presence of T. denticola and the SK-013 activity were higher than those of others.

Inhibitory effects of aqueous extract of cacao bean husk on collagenase of Bacteroides gingivalis.

Natural materials containing tannin were examined to determine whether they possessed any inhibitory activity against collagenase obtained from the culture supernatant of Bacteroides gingivalis. The collagenolytic activity was determined with Collagenokit CLN-100 (Collagen Technological Co., Tokyo) using FITC-conjugated collagen type 1. Among the test samples, only an aqueous extract of cacao (Theobroma cacao) bean husk strongly inhibited the bacterial collagenase. This inhibitory potency was at the same level as that of tetracycline-HCl.

Clinical, microbiological, and immunological studies following initial preparation in adult periodontitis.

This study investigated the clinical, microbiological and immunological changes induced in adult periodontitis by initial preparation. Microflora in periodontal lesions was mainly examined by cultural methods, and serum IgG antibody levels were measured by an enzyme linked immunosorbent assay. Black-pigmented gram negative anaerobes, especially Porphyromonas (Bacteroides) gingivalis, was predominant in periodontal pockets with adult destructive periodontitis. Significantly elevated serum IgG antibody levels to P. gingivalis was found in high frequency, and these serum IgG antibody levels correlated with the presence of P. gingivalis in periodontal pockets, not with other bacteria tested. Total-CFU was significantly reduced and clinical conditions were improved after supragingival plaque control, but the microflora themselves showed few changes. Although reduction of total-CFU and changes in microflora were observed following scaling and root planing, it was found to be difficult to completely eliminate periodontopathic bacteria from periodontal pockets. Although serum IgG antibody levels to P. gingivalis was significantly reduced following initial preparation, the levels were still elevated from those of the controls. It was found that the presence of P. gingivalis was correlated with clinical conditions, including bleeding on probing and residual pocket depth following initial preparation.

Studies of the antibacterial activity of plant extracts and their constituents against periodontopathic bacteria.

Plant extracts and their constituents were tested for antibacterial activity against periodontopathic bacteria, including Actinobacillus, Capnocytophaga, Fusobacterium, Eikenella and Bacteroides species. The essential oils of two labiatae plants, Mosla chinensis Maxim. and Pogostemon cablin Benth., and five terpenoids, hinokitiol, thymol, carvacrol, patchoulialcohol and pogostone, showed antibacterial activity. The terpenoids were especially effective against Bacteroides species.

Clinical, microbiological and immunological studies on recurrent periodontal disease.

The present study was performed to evaluate the clinical, microbiological and immunological aspects in the early stages of recurrent periodontal disease. After clinical monitoring of pockets with recent evidence of disease recurrence, microbiological samples for cultural analysis, serum and gingival crevicular fluid (GCF) samples were taken for IgG antibody analysis from 14 sites, 7 in 6 recurrent periodontitis patients and 7 in 7 periodontally healthy control subjects. IgG responses of serum antibody to 8 gram-negative bacterial strains were compared with those of GCF sampled from the recurrent site. The results clearly demonstrated the predominance of Bacteroides gingivalis in most subgingival plaque samples during the early stages of disease recurrence; the mean proportions of B. gingivalis were significantly different from those of the healthy sites (p less than 0.05). 4 out of 6 serum samples showed the elevated antibody responses to B. gingivalis 381; and this was closely correlated to homologous infection by this micro-organism in recurrent sites. Elevated serum antibody responses were also noted to Eikenella corrodens 1073 and Actinobacillus actinomycetemcomitans Y4. However, no relationship with homologous infection was found for A. actinomycetemcomitans. 3 out of 6 GCF samples had greater antibody titers than the serum, suggesting local antibody synthesis by the gingival cells in the recurrent pockets. The present study showed that B. gingivalis might play an important role in the pathogenesis of disease recurrence in its early stages.

Structural analysis of lipopolysaccharides from Eikenella corrodens by use of murine monoclonal antibodies.

The structure of lipopolysaccharides (LPSs) of Eikenella corrodens was analyzed with prepared murine monoclonal antibodies. A common core epitope was found in three of seven LPS preparations from E. corrodens strains and Fusobacterium nucleatum ATCC 25586. Four E. corrodens LPSs were found to possess an O-side-chain epitope which cross-reacted with LPSs from Fusobacterium necrophorum ATCC 25286 and Capnocytophaga ochracea M-12. Lipid A of E. corrodens LPS shared an epitope common among LPSs from various gram-negative rods.

Antimicrobial action of natural substances on oral bacteria.

The antimicrobial action of natural substances was investigated in vitro against oral bacteria including Streptococcus sp., Actinomyces sp., Actinobacillus sp., Bacteroides sp., Capnocytophaga sp., Eikenella sp., Fusobacterium sp. and Propionibacterium sp. Among the natural substances tested, hinokitiol was the most inhibitory to oral bacteria. Cinnamon bark oil, papua-mace extracts, and clove bud oil in spice extracts were also inhibitory against many oral bacteria. Egg white lysozyme exhibited antimicrobial action against the periodontitis associated bacteria.

Evaluation of a model for short-term clinical testing of cariogenicity.

Conventional mirror and probe examination and laser fluorescence were used to score caries-like alterations on the buccal surfaces of posterior teeth in 60 panelists, allotted to one of three groups. All subjects refrained from oral hygiene for five days. In one group each subject consumed 12 sucrose candies daily between meals and in the second group each subject similarly consumed 12 palatinose (isomaltulose) candies. The third group served as a control. At the end of the experimental period the tooth surfaces reexamined. Conventional and laser fluorescence scoring techniques showed an increase in the number of caries-like alterations for all groups, but there were no significant differences between the groups. For all groups, the laser method gave significantly greater scores throughout. The findings imply that certain aspects of the early development of enamel caries in subjects regularly using fluorides must be considered in the design of a short-term cariogenicity test. It is concluded that sensitive quantitative methods for the registration of caries are necessary in such tests.

Protective efficacy of active and passive immunizations against experimental infection with Bacteroides gingivalis in ligated hamsters.

The protective efficacy of immunization against Bacteroides gingivalis infection was examined in hamsters. Whole cells or extracted hemagglutinin of B. gingivalis 381 was injected with incomplete Freund adjuvant into the inguinal regions of hamsters. Two weeks after the rats received a booster injection, cotton threads were tied coronally to the gingival margins of the mandibular first molars, and then a streptomycin-resistant B. gingivalis 381R' strain was inoculated into the rats' oral cavities. The subcutaneous immunizations resulted in slight reductions in the numbers of B. gingivalis on the ligature threads compared with the sham-immunized group. Peroral administration of whole cells of B. gingivalis to hamsters elicited salivary immunoglobulin responses, but no reduction of B. gingivalis 381R' colonization was found in this group. Repeated passive immunizations with rabbit antiserum to B. gingivalis into the oral cavities of the hamsters resulted in a reduction in the number of organisms in the periodontal region.

Recent changes in the dental curriculum in Japan.

Information on curriculum changes was obtained from 25 of Japan's 29 dental schools. Twelve of the schools reported changes in the last 5 years. There has been some extension of inter-disciplinary teaching, more integration of previously separate courses and improved orientation at the beginning of new courses. Japanese dental education has not been influenced by significant changes in dental disease for the changes noted in other industrialized countries have not yet made an impact in Japan. Among aims for the future are (1) to shift curriculum emphasis from dental caries to periodontal disease, (2) to develop a better understanding of the dental needs of the elderly (3) and to improve the comprehension of relevant general health problems. An extensive internship programme has recently been introduced in Japan.
