Vector competences of Aedes (Ae.) vexans nipponii (nip.) and Culex (Cx.) tritaeniorhynchus to Getah virus were assessed by using a membrane feeding technique. The Getah virus was present at high titer in both species of mosquitoes after 21 days of extrinsic incubation at 28 degrees C. Infection rates on 21 post-feeding were 100 per cent (4/4) for Ae. vexans nip. at a virus dosage of 10(5.3) PFU/ml and 60 per cent (3/5) for Cx. tritaeniorhynchus at similar virus dosage. More than 10(3.5) PFU of virus was detected in salivary glands of both species of mosquitoes on day 21 of extrinsic incubation. Forty percent (2/5) of Ae. vexans nip. transmitted the virus into serum-agar after ingesting 10(4.3) PFU/ml of virus blood mixture. In experiments with Cx. tritaeniorhynchus ingesting 10(7.5) PFU/ml of virus blood mixture, 57 per cent (4/7) were able to transmit the virus to suckling mice and 59 per cent (10/17) transmitted the virus into serum-agar.

Cytochemical reactions with a panel of special stains were explored in peripheral blood and lymph node cells of animals with the adult, calf, and thymic types of bovine lymphosarcoma as well as lymphosarcoma in sheep induced with the bovine leukosis virus. These results were compared with normal lymphoid cells. Cytochemical methods included Sudan black B, periodic acid-Schiff, acid phosphatase and three esterases. Cells from ovine lymphosarcomas and a thymic type of bovine lymphosarcoma were distinctively positive to the alpha naphthyl acetate esterase reaction. These cytochemical reactions which were obtained with standard cytochemical methods should aid in the characterization and classification of lymphoproliferative disorders in animals, especially when combined with immunologic surface markers.

A 29-year-old man and a 29-year-old woman developed over fifty asymptomatic, slate-colored macules disseminated mainly on the forehead. Histologic study revealed keratinous cysts in the dermis, lined by a few layers of epithelial cells with rudimentary hair bulb-like structures. The cysts' contents were vellus hair fragments and laminated horny material. Ultrastructural studies of the second case disclosed that the keratinization of the cyst wall was similar to that of the infundibular portion of the outer root sheath. It is suggested that our two cases are a type of follicular hamartoma. Similar follicular hamartomas, eruptive vellus hair cysts have been reported previously. The cysts were located in the chest and/or extremities and had a tendency to be eliminated transepidermally. Although these clinical features were different from ours, the characteristic histologic features were the same; therefore, our cases might be a variant of eruptive vellus hair cysts.
The cell cycle of fetal lamb kidney (FLK) cultures chronically infected with bovine leukosis virus was synchronized by double thymidine block. The synchronized FLK cells were examined by production of BLV antigen and virion release by cytoplasmic immunofluorescence and syncytia forming assay, respectively. The production of BLV antigens was increased during S and G2 phases and was decreased during M and G1 phases. BLV release was associated with the M phase of FLK cells. Short term lymphocyte cultures from BLV infected cattle were treated with hydroxyurea and mitomycin C. The expression of BLV antigen and DNA synthesis of PHA stimulated lymphocytes was inhibited by both drugs.

The detection of tumor-associated antigen and B cell surface marker was attempted in lymphoid cells of peripheral blood (PBL) and lymph nodes obtained from cattle with lymphosarcoma, cattle infected with bovine leukosis virus (BLV), and clinically normal cattle. As a result, specific tumor-associated antigen was found in PBL and lymphoid tumor cells from cattle with the adult form of lymphosarcoma (ALS) using anti-ALS serum. The antigen was also detected in PBL of BLV-infected cattle. A weak reaction for the antigen was found in 2 of 5 cases of calf form. No significant reaction for the tumor-associated antigen of neoplastic lymphoid cells from ALS was found in the thymic and skin forms. The percentage of B cells in PBL and lymph nodes from ALS was higher than that in normal cattle. However, a loss of B cell surface marker was suggested in some ALS cases. A good relation was obtained between an increased B cell percentage and an appearance of the tumor-associated antigen in PBL of BLV-infected cattle. In lymphoid cells of PBL and lymph nodes from sporadic cases, the percentage of B cells was lower than that observed in normal cattle.

Lymphoid cells of peripheral blood, lymph nodes, and thymus from clinically normal cattle, cattle infected with bovine leukemia virus (BLV), and cattle with lymphosarcoma were characterized for T- and B-cell surface markers. B-cells were detected by the erythrocyte-antibody-complement (EAC) rosette test and the surface immunoglobulin (sig) immunofluorescence assay. Peripheral blood from BLV-infected cattle had a higher than normal percentage of B-cells by both EAC rosette and sig immunofluorescence assays. Lymphoid cells from tumorous lymph nodes of cattle with the adult type of lymphosarcoma had a higher than normal percentage of sig-bearing cells, but in the same cell preparation the EAC rosette-positive cells were fewer than sig-positive cells. T-cells were detected by the erythrocyte rosette test. The percentage of T-cells by this test in lymph nodes of adult type lymphosarcoma was lower than that in normal cattle. A distinctly lower than normal percentage of lymphocytes could be characterized as either B- or T-cells in lymph nodes thymus, and peripheral blood from the calf type and thymic type of lymphosarcoma.

The protein portion of endotoxin (OEP), or common antigen of Pseudomonas aeruginosa prepared from strain N10 of P aeruginosa, was examined as a possible vaccine against this organism in mink. Mink were given OEP vaccine and challenge exposed by instillation with the inoculum into the nostril. They survived after this exposure with 4 median lethal (LD50) doses of strain NC-5 (protease negative, elastase negative) or with 160 LD50 doses of strain 5 (protease positive, elastase positive). Strain NC-5 belonged to TYPE 5 AND strain 5 belonged to type 8 (according to the serotyping method of Homma). Strain N10, the source of OEP, belonged to serotype 5. Thus, the mink vaccinated with OEP vaccine were able to stand challenge exposure to both homologous and heterologous serotype strains. More mink given OEP vaccine survived the challenge exposure by intranasal spraying with virulent strain 5 than did those in the nonvaccinated control group, with a significant difference. When the surviving mink of the vaccinated group were rechallenge exposed with 300 or 3,000 LD50 doses of the same strain a month later, most of them survived.
Transformability of Leptospira was investigated with the two genetic markers, streptomycin (SM) resistance and the ability to grow in SHENBERG's synthetic medium. DNA was extracted from the two mutants of L. icterohaemorrhagiae strain Shibaura, which were SM-resistant and able to grow in SHENBERG's medium. Recipients were 5 strains of L. icterohaemorrhagiae and 1 strain of L. copenhageni. Studies taking careful consideration of the growth phase of the recipient cells, transformation medium, and incubation temperature did not produce the appearance of transformant. Transformation of L. kremastos with SM resistance also showed a negative result.
The protective attributes of the structural components of Leptospira icterohaemorrhagiae strain Shibaura were examined. Guinea pigs were immunized with the outer envelope, the cell wall, the type-specific main antigen, and the protein constituent of the outer envelope respectively. Of the 4 preparations, the outer envelope, the cell wall and the type-specific main antigen showed a considerable protective effect in this order. The protein constituent of the outer envelope was less effective. Immunization with 200 mug of the outer envelope protected guinea pigs from lethal infection but no completely from the renal carrier state. The rate of protection from renal leptospirosis by the preparations of leptospiral components and lyophilized whole cells was always inferior to the survivor rate.
