[Mechanisms of arbidole's immunomodulating action].

The immunomodulating activity of arbidole was studied in cultured cells, animals, and human beings. Arbidole was shown to have effects on nonspecific defense factors, on its capacity to induce interferon and activate phagocytes in particular. Arbidole-treated patients with lower baseline immunity showed improvement in immunological parameters (in the counts of CD4 and CD8 lymphocytes, B lymphocytes, in the levels of serum immunoglobulins). Arbidol produces a high preventive and therapeutical effects in influenza A and B and other acute respiratory viral infections, prevents postinfluenza complications, reduces the incidence of exacerbations of chronic diseases in postinfluenza patients. In influenza, the therapeutical efficiency of the drug appears as decreases in intoxication, the severity of catarrhal syndrome, shorter fever and disease in general. Arbidole is beneficial for patients with second immunodeficiency, in those with recurrent herpes infection or chronic bronchitis. After arbidole treatment regimen, postoperative immunological parameters became normal in cardiac surgical patients, which suggests that the drug should be used in cardiac surgical care. The agent showed no side effects in any case.

[Effect of bacterial toxins on the mitogen-induced increase of the Ca2+ concentration in the cytoplasm of rat thymocytes. The role of N proteins].

The effect of bacterial toxins, modifying the activity of regulatory N proteins of adenylate cyclase and probably other systems, on the mitogen-induced changes of cytosolic free Ca2+ concentration ([Ca2+]i) has been studied using Ca2+ fluorescent probe quin-2. It is shown that treatment of thymocytes with cholera toxin, E. coli heat-labile (HL) toxin or pertussis toxin abolishes the concanavalin A (con A)-induced rise of [Ca2+]i. The inhibitory effect of cholera and HL toxins can be explained by the toxin-induced rise of intracellular cAMP. The effect of pertussis toxin indicates the involvement of N proteins in the action of con A receptor and in generation of Ca2+-signal during the mitogenic activation of thymocytes.

[Interferon-inducing and immunostimulating activity of bonafton in an experiment].

It was shown for the first time that the antiviral drug bonafton administered orally to nonlinear albino mice in single doses of 5, 12.5 and 25 mg/kg induced production of interferon in the animal blood serum. The maximum interferon titer of 160-320 IU/ml was observed 18 hours after the drug administration in a dose of 12.5 mg/kg. In low doses of 5 to 12.5 mg/kg bonafton increased the nonspecific resistance of the mice to experimental viral infections when administered orally in single doses not earlier than 2 weeks prior to the contamination. The ability of the drug to stimulate the host protective forces probably plays a certain role in the mechanism of its therapeutic action in severe viral infections of man such as severe recurring ophthalmic herpes, genital herpes, Behçet's disease, Melkersson-Rosenthal syndrome and others.

[Effect of quinine, a blocker of Ca2+-activated K+-channels, on lymphocyte activation by mitogens].

Quinine inhibits mitogenesis at the same concentration (10(-4) M) as that which blocks Ca++-dependent potassium transport in lymphocytes. Lower quinine concentrations (10(-8)-10(-6) M) induce a comitogenic effect which is most pronounced when Ca++-ionophore A23187 is used as a mitogen. Thus, activation of Ca++-dependent K+-channels is not necessary to trigger mitogenesis, but is important for further stages of the process.

[The dual nature of lymphocyte interaction with allogenic stem cells].

The interaction of lymphocytes from mouse lymph nodes with allogeneic stem cells was studied using exogenous colony formation inhibition test. Dual nature of the interaction was revealed: great amounts of lymphocytes inhibited, while small amounts stimulated colony formation. This dependence holds true for macro- and microcolonies as well as for erythrocyte and granuloid microcolonies in the bone marrow during fixation on day 8 and 11 after cell mixture transplantation.

[Comparative study of the action of Vibrio cholerae enterotoxin, thermolabile Escherichia coli toxin and a Salmonella typhimurium filtrate on the small intestine of laboratory animals].

The results of the comparative study of the effects produced by V. cholerae enterotoxin E. coli thermolabile toxin (TL-toxin) and S. typhimurum 415 filtrate on the loops of the small intestine of rabbits and C57BL/6 mice are presented. The results obtained in mice were considerably more uniform than those obtained in rabbits. For the first time the dose-effect curve for V. cholerae enterotoxin and TL-toxin, obtained on both animal species, was found to be bell-shaped. The concentration relationship for dibutyryl-cAMP proved to be similar in character. The authors suggest that these effects are based on the specific relationship between the cellular processes, controlling the transport of electrolytes and water, and the level of cAMP.

[Interaction of the lymph node and splenic lymphocytes of mice in inactivation of allogeneic hematopoietic stem cells].

A study was made of interaction of mouse spleen and lymph node lymphocytes in inactivation of allogeneic stem cells. It was established that T lymphocytes of the lymph nodes and spleen lymphocytes do not interact on combined administration; their action is of additive nature. B lymphocytes of the lymph nodes have a regulating activity both in respect to T lymphocytes of the lymph nodes and lymphocytes of the spleen. The stem cells serve as target. Depending on the stem cells/B lymphocytes ratio B lymphocytes are capable of exerting either helper or suppressor action.

[Regulating role of the B-lymphocytes in lymphocyte interaction with allogeneic hematopoietic stem cells].

The preparative separation of lymph-node cells according to their charge and the study of the activity of each fraction have revealed that B-lymphocytes in the lymph nodes of mice have a regulating function in the phenomenon of the inactivation of allogeneic hematopoietic stem cells. The analysis of the dependence of effects, obtained in various experiments, on the ratio of B-lymphocytes and hematopoietic stem cells indicates that stem cells serve as targets for B-lymphocytes and the most probable mechanism of the above-mentioned effects is the direct interaction of the cells, one regulatory B-lymphocyte being capable of interacting with one target cell. Depending on the ratio of B-lymphocytes and stem cells, the inactivating effect may be either suppressed (the suppressor activity of B-lymphocytes) or enhanced (the helper activity of B-lymphocytes). The quantitative characteristics of the regulating activity of B-lymphocytes have been introduced.

[Characteristics of the B lymphocytes participating in the reaction of allogeneic stem cell inactivation].

In this work the B-cells of mouse lymph nodes are characterized. B-cells produce a helper effect on the capacity of the T-lymphocytes of the lymph nodes for inactivating nonsyngeneic stem cells. The study has revealed that the genetic heterogeneity of the B-lymphocytes does not lead to the abolition of their helper activity. B-lymphocytes of "B-mice" have also been shown to be capable of enhancing the inactivating activity of T-cells.

[Effect of substances that alter the level of intracellular cAMP on the reaction of human lymphocytes blast transformation].

Effects of cholera toxin, cholera anatoxin, theophylline and dibutyryl cyclic AMP (db-cAMP) on the PHA-induced DNA synthesis have been studied. It was shown that toxin, db-cAMP and theophylline suppressed mitogen-induced 3H-TdR incorporation and blast-transformation in a dose-related manner. When both toxin and theophylline were given together to PHA-stimulated lymphocytes, the inhibitory effects of these two agents were additive. The inhibitory effects of db-cAMP and theophylline were additive too. This inhibitory effects could be due to the elevation of intracellular cAMP. Cholera anatoxin exerted an inhibitory action on the PHA-stimulated lymphocytes. The inhibitory effects on lymphocyte DNA synthesis by anatoxin and theophylline were not additive. These results suggest that the effects of cholera toxin are mediated by cyclic AMP, but effects of cholera anatoxin may be associated with some other mechanism.
