[Adamantane derivatives enhancing body's resistance to emergencies].

A total of 329 novel adamantane derivatives have been synthesized and pharmacologically studied. Among them, the compounds containing halogen-containing aromatic radicals in the second position show the most pronounced effect on animal resistance to the emergencies induced by its habitat and performance. N-(2-adamantyl)-N-(para-bromphenyl)amine (bromantane) possesses a low toxicity, a high ability to enhance the physical and operant working maintenance of animals, to accelerate its recovery in developed fatigue, in hyperthermia and hypoxia in particular. 2-(para-chlorobenzoylamine)adamantane (chlodantane) has the processes of a rapid-action adaptogenic agent by enhancing the resistance of animals to various physical and toxically chemical noxious agents. The two compounds have immunostimulating effects in secondary stress-induced immunodeficiencies whose mechanism of action is a membranous protective activity.

Adamantane

[Decrease and asymmetry in taste perception in patients with nerve trunk damage].

The purpose of this article was the gustatory perception state study (NaCl solution perception) in patients with one-sided damages of branchial plexus as well as of radial, facial and ulnar nervous trunks. The decrease of gustatory perception was observed in 78 (70.3%) cases, while the lack of such alterations in 33 (29.7%) patients. The gustatory perception disturbances (increase of the absolute threshold on the ipsilateral tongue's half) took place in 82.2% and 69.6% at right and left limbs damages respectively. The results presented testified the strengthening of normally existing asymmetry in gustatory perception especially in patients with left-sided denervation. The degree of gustatory perception restoration may serve as a prognostic test at surgery of such patients.

[A quantitative pharmaco-electroencephalographic analysis of the action of bromantane].

The action of the new stimulant bromantane on spectra power EEG on Fourier of sensorimotor cortex, dorsal hippocamp and lateral hypothalamus of left and right hemispheres of brain of rat in free behavior was investigated. Bromantane leads to decreases in the total and absolute powers of all frequency bands of EEG spectra, changes structural spectra in the cortex and in hippocamp--decreases the relative power of theta-band and increases the relative power of beta 1, 2-activity. The basic feature of bromantane's action is a two-phase effect (its maximum occurs 2-3 and 6-7 hours after administration), which remains up to 8 h of EEG recording. These data suggest that that bromantane has more marked and prolonged stimulant properties than other adamantane psychostimulants.

Amantadine

[The biochemical mechanisms of the decrease in work capacity in sailors exposed to high temperatures and the possible means for their pharmacological correction].

The studies conducted on experimental animals and volunteers have proved that the increase of adrenalin, histamine, and serotonin ration in blood was caused by hyperthermic conditions. Activation of hormonal systems leads to acceleration of biochemical processes and to reduction of substrates in organs and tissues (such as regenerated glutathione, histidine) which are necessary for normal health status. A conclusion is made that the metabolite depletion of biochemical reactions provoke the reduction of job performance of man in the conditions of hyperthermic influence. Methods of medicamentous therapy are proposed to maintain the normal job performance of seamen in the conditions of unfavourable influence of high temperatures.

The mink proopiomelanocortin gene: characterization of cDNA and chromosomal localization.

A cDNA library from the mink pituitary was screened using as probe a synthetic oligodeoxyribonucleotide, 5'-TTCATGACCTCCGA-3', corresponding to the endorphin region of bovine proopiomelanocortin (POMC) cDNA. As a result, several clones containing inserts complementary to POMC mRNA were identified. The sequence of one of the fragments (585 bp, 65% of the total length of mRNA) was determined. A high degree of homology (over 80%) among the primary structures of sequences from mink, man, and bovine cDNA POMC was established. With the cloned mink cDNA fragment as probe, the DNAs from mink-Chinese hamster hybrid clones were studied. The results of segregation analysis of mink POMC sequences and mink chromosomes in the mink-Chinese hamster panel allowed us to assign the POMC gene to mink chromosome 11.

[Pharmacological characteristics of a new phenyl analog of piracetam--4-phenylpiracetam].

The central neurotropic effects of 4-phenylpyracetam, a new phenyl analog of pyracetam, were studied and compared with the effects of pyracetam, morpholene and 4-phenylpyrrolidone. 4-Phenylpyracetam was found to activate the operant behavior more powerfully, to remove psychodepressant effects of diazepam, to inhibit post-rotational nystagmus, and to prevent the development of retrograde amnesia. Unlike pyracetam, 4-phenylpyracetam exhibits a specific anticonvulsant action. When given in high doses, the compound under study produces psychodepressant effects.

[Role of peripheral adrenoreactive structures in the etiology of disorders of the motor components of rat operant behavior during emotional stress].

It has been found in experiments on rats trained to avoid electric stimulation of the foots by pressing the pedal that previous immobilization and administration of adrenaline and isoprenaline (but not noradrenaline) disturbs the accuracy of the realization of motor components of operant behavior. The total beta-adrenoblockers (but not phentolamine and cardioselective beta 1-adrenoblockers) given in doses which do not cause any central effects reduced the negative action of emotional stress. It is inferred that activation of beta 2-adrenoreceptors of skeletal muscles is likely to play a role in the genesis of disturbed motor skill under emotional stress.

[Psychotropic effect of sodium hydroxybutyrate].

A conclusion was drawn on the qualitative features of the spectrum of sodium hydroxybutyrate psychotropic activity from its effect on the conditioned motor-defensive reflex, behaviour in the conflict situation, active avoidance without discriminative control in rats as well as from the lateral position test The results obtained allow the effect of sodium hydroxybutyrate in non-narcotic doses to be characterized as a hypnotic one showing elements inherent in neuroleptics and tranquilizers.

[Active avoidance without discriminating control as a model for the study of protective effect of drugs in emotional stress].

The method of plotting of hystograms of distribution according to the frequency intervals between pressing the redal (the method of active avoidance with no discriminating control) permits one to estimate slight shifts in the character of operative activity of rats occurring under the effect of emotional stress. The design of the simple multichannel apparatus for developing and studying of habits of active avoidance in several animals is presented.

[Participation of GABA and glycine in the formation of somato-sympathetic reflexes].

Experiments were conducted on cats with intact spinal cord, and after spinalization a study was made of the action of strychnine and picrotoxin (synaptic GABA and glycine antagonists) on the components of somatosynaptic reflexes caused by stimulation of the skin and muscular intercostal afferents. Induced potentials were recorded in the white ramus of the third thoracic segment. The action of strychnine and picrotoxin on the values of different reflex components was not identical; this pointed to a different localization of the inhibitory action of GABA and glycine in the central link of the somatosympathetic reflex arch.

[Effect of gamma-aminobutyric acid and phenibut on the central links of the vascular reflexes with chemo- and mechanoreceptors of the aorto-carotid zone].

Experiments were conducted on decerebrated cats. A depressive effect of gamma-aminobutyruc acid (GABA)--100--200 mg/kg and its phenyl derivative phenibut--20 mg/kg--on depressive reactions of the systemic arterial pressure and on the inhibition of spontaneous bioelectrical activity in the renal nerve occurring in stimulation of the mechanoreceptors of the carotid sinus and of the sinus and depressor nerve afferents (having a mechanoreceptor modality) was demonstrated. Pressor reactions of the systemic arterial pressure and evoked bioelectrical activity were enhanced in the renal nerve in stimulation of chemoreceptors of the carotid sinus following administration of the same GABA and phenibut doses. The data obtained are interpreted from the aspect of a deprimating action of GABA and phenibut in the area of the paramedian reticular nuclei of the medulla oblongata.
