Establishment of a new human lymphoma line that secretes plasminogen activator.

A new cell line, designated RC-K8, was established from the peritoneal effusion of a patient with histiocytic lymphoma. The RC-K8 had human male karyotype with 14q+ and was found to have B-cell (B1) and monocyte/macrophage (intracytoplasmic alpha subunit of S-100 protein) markers. A fibrin plate method demonstrated that RC-K8 cells release plasminogen activator in culture.

Burkitt's lymphoma in Japan.

The clinicopathological features of 18 Japanese patients with Burkitt's lymphoma are reviewed. Six patients were 12 years of age or younger. Seven patients presented with abdominal masses and seven with jaw tumours. Thirteen patients manifested blood and/or marrow involvement at the time of initial diagnosis (five patients) or later in the course of the disease (eight patients). Most of the patients had a rapid clinical course in spite of combination chemotherapy. The malignant cells from all 17 patients examined exhibited a B-cell phenotype; the surface immunoglobulin was IgM in 15 and IgG in two. Two patients had tumours positive for Epstein-Barr virus-determined nuclear antigen while the other 13 patients tested lacked this antigen in their tumours. Chromosome analysis revealed a t(8;14) translocation in 11 patients, a 14q+ without an identifiable donor chromosome in two, a t(2;8) translocation in two, and a t(8;22) translocation in one.

Detection and characterization of simian retroviruses homologous to human T-cell leukemia virus type I.

Lymphoid cell lines were established from five different species of monkeys which were positive in antibodies cross-reactive with human T-cell leukemia virus type I (HTLV-I) and were shown to contain provirus sequences homologous to HTLV-I. Gene-specific probes of HTLV-I, gag, pol, env, pX, and LTR, hybridized efficiently with monkey DNAs from these cell lines under stringent conditions, indicating that the proviruses are very similar to HTLV-I along with whole viral genomes. However, the preliminary restriction mapping turned out the difference between simian retroviruses and HTLV-I and also among simian retroviruses. These findings suggest a common ancestor of simian and human retroviruses, but exclude the recent interspecies transmission between monkeys and humans.

Altered transcription of the c-abl oncogene in K-562 and other chronic myelogenous leukemia cells.

Expression of the cellular abl (c- abl ) oncogene was studied in K-562 and other chronic myelogenous leukemia (CML) cells and cell lines by means of Northern blot hybridization. In contrast to non-CML cells, which contained 7.4- and 6.8-kilobase abl -related transcripts, the CML cells contained a predominant and novel 8.2-kilobase abl -related RNA. In addition, the levels of abl -related message were up to eight times higher in CML cell lines from patients at the blast crisis stage of the disease compared with CML cells obtained during the chronic phase and with non-CML cells.

Heterotransplantation and maturation of a chronic myelogenous leukemia cell line (KCL-22) in vivo.

Heterotransplantation was performed to study the maturational capacity of a new Ph1 chromosome-positive chronic myelogenous leukemia cell line (KCL-22). Five million KCL-22 cells of undifferentiated blasts were implanted intraperitoneally into 15 immunosuppressed newborn hamsters. Of the 15 hamsters, 10 that survived developed granulocytic tumors 16-61 days after implantation. The tumor cells were composed of a mixed population of undifferentiated blasts, neutrophils, and eosinophils and had a human female karyotype with double Ph1 chromosomes identical to that found in the KCL-22 cell line used for transplantation. These findings indicate that Ph1 chromosome-positive cells can be induced to mature along the neutrophil-eosinophil lineage after growth in the heterologous hosts.

Granulocytic sarcoma presenting as a mediastinal tumor. Report of a case and cytological and cytochemical studies of tumor cells in vivo and in vitro.

An unusual case of granulocytic sarcoma in a 23-year-old man is reported. The patient initially presented with mediastinal tumor and was diagnosed clinically as having thymoma. The patient was treated by radiotherapy and surgical removal of the tumor. Histology of the excised tumor had been nondiagnostic because of extensive fibrous changes. Eight months later, the patient developed pleural effusion on the right, which soon was followed by blood and bone marrow pictures consistent with acute promyelocytic leukemia. In vitro culture of pleural effusion cells unexpectedly gave rise to a continuously growing peroxidase-positive myeloid cell line. Autopsy revealed the recurrent mediastinal tumor to be positive for intracytoplasmic naphthol AS-D chloroacetate esterase and lysozyme activity. From these findings, the patient retrospectively was diagnosed as having mediastinal granulocytic sarcoma, which terminated in pleural effusion and acute promyelocytic leukemia.

Ultrastructural study of type C virus particles in phytohemagglutinin-stimulated lymphocytes from healthy adults seropositive to adult T-cell leukemia-associated antigens.

PHA-stimulated peripheral lymphocytes from six healthy adults consisting of four family members of an adult T-cell leukemia (ATL) patient and two blood bank donors, seropositive to ATL-associated antigens (ATLA), were all positive for the expression of ATLA and ATL-associated virus (ATLV). These results revealed that anti-ATLA-positive persons were healthy carriers of ATLV and that the numbers of ATLV observed in each culture were more proportional to the percentage of ATLA-positive lymphocytes than anti-ATLA titers of each person. Virus particles detected were C-type in morphology, and essentially similar to those observed in MT-2 and other ATL-related cells, but rather uniform in size, mostly 120-130 nm in diameter. Furthermore, it is of interest that tubuloreticular structures and striated fusiform structures, which were found in fresh or short term cultured ATL cells, were observed in some cultures of lymphocytes from anti-ATLA-positive healthy persons.

An atypical von Willebrand's disease with hyperreactivity of platelet aggregation.

Two family members (daughter and mother) with a bleeding disorder showed prolonged bleeding time and activated partial thromboplastin time associated with decreased plasma levels of factor VIII procoagulant activity, factor VIII-related antigen, and factor VIII-ristocetin cofactor activity. The ristocetin-induced platelet agglutination (RIPA) was enhanced, ADP-, collagen- and Ca ionophore-induced platelet aggregation were also increased at low concentrations of these compounds. In the mother, spontaneous platelet aggregation (SPA) was also observed. Contrary to type II B von Willebrand's disease (vWd), pseudo-vWd and platelet-type vWd, the patients did not show any increased binding of factor VIII/vWf to platelets in the presence of ristocetin. The RIPA in normal controls were inhibited by addition of antifactor VIII antiserum to the platelet-rich plasma, but not in cases 1 and 2. In this atypical vWd, therefore, the hyperreactivity of platelet aggregation may be due to an intrinsic abnormality of the platelets, but not to any enhanced interaction between plasma factor VIII and the platelets.

Detection of HLA-D-region-associated antigens on the surface of adult T-cell leukemia virus particles by immunoelectron microscopy.

To search for lymphocyte marker antigens on the surface of human T-cell leukemia virus (HTLV), an immunoelectron microscopic study was performed on a HTLV-producing human T-cell line, MT-2, using monoclonal antibodies, such as anti-Leu-1, -Leu-2b, -Leu-3a, -Leu-5, -Leu-10 and -HLA-DR and OKIal. The reactivity of each antibody with MT-2 cells was tested by the immunoperoxidase method at the light microscopic level. OKIal, anti-HLA-DR and -Leu-10 gave positive results. At the ultrastructural level, the surface of HTLV as well as the plasma membranes of MT-2 cells were labeled with ferritin by the monoclonal antibodies OKIal, anti-HLA-DR and -Leu-10, but not by anti-Leu-1 and -Leu-3a. These findings suggest that HLA-D region -associated antigens are common antigenic determinants shared by the surface of HTLV and the plasma membranes of MT-2 cells. These antigens on the virus surface are probably picked up selectively from the plasma membranes and may play an important role in the interaction of HTLV and target T-cells.
