Stimulation of spawning behavior in castrated rainbow trout (Oncorhynchus mykiss) by 17 alpha,20 beta-dihydroxy-4-pregnen-3-one, but not by 11-ketoandrostenedione.

The onset of spawning behavior in male salmonids can be closely correlated to circulating levels of the progestin 17 alpha,20 beta-dihydroxy-4-pregnen-3-one (20-P). However, evidence that 20-P plays a causal role in the development and maintenance of spawning behavior in male salmonids is largely lacking. The purpose of this study was to investigate the effect of 20-P implants on the spawning behavior in castrated rainbow trout in the presence of a sexually active female. Mature rainbow trout were placed in five experimental groups: sham-operated and castrated implanted with either Silastic capsules containing 20-P, 11-ketoandrostenedione (11-KA), 11-KA + 20-P or empty capsules. Five to 6 weeks after implantation, all males were tested individually in a stream channel in the presence of a nesting ovulated female. Following behavioral observations, blood samples were taken from all tested males. Implants of 20-P and 11-KA resulted in increased plasma levels of 20-P and 11-ketotestosterone, respectively. Sham males exhibited the strongest behavioral response, both in the frequency of quiver bouts and attention toward the female. Castrated controls and 11-KA males showed no quiver response and reduced attention toward the female, while the behavioral responses shown by the 20-P and 20-P/11-KA males were intermediate. The results indicate that 20-P, but not 11-androgens, plays a significant role in the stimulation of spawning behavior in male rainbow trout.

Zinc ions in synthetic carbonated hydroxyapatites.

Carbonated hydroxyapatites were synthesized at pH 7.0 and 9.0 with a carbonate content up to 9.2%, in the presence of Zn2+ ions. The uptake of Zn was found to be proportional to the amount of Zn added at precipitation. Chemical, lattice constant, microprobe and thermal analyses indicate incorporation of Zn into the apatite lattice.

Metabolism of androstenedione and 11-ketotestosterone in the kidney of the three-spined stickleback, Gasterosteus aculeatus.

Stickleback kidneys were incubated with tritiated androstenedione (A4) or 11-ketotestosterone (OT). After the A4 incubations the following steroids were found, testosterone (T), 5 beta-androstane-3,17-dione (5 beta Ad), etiocholanolone (Et), 5 beta-dihydrotestosterone (5 beta DHT), 5 beta-androstane-3 alpha,17 beta-diol (5 beta A3 alpha 17 beta diol), androsterone, 5 alpha-androstane-3,17-dione, 5 alpha-androstane-3 alpha,17 beta-diol (5 alpha A3 alpha 17 beta diol), as well as glucuronides of T, Et, 5 beta DHT, 5 beta A3 alpha 17 beta diol, and 5 alpha A3 alpha 17 beta diol. The metabolites found in the largest amounts were usually T, 5 beta Ad, Et, and the glucuronides of T, Et, and especially of 5 beta A3 alpha 17 beta diol. These results suggest the main pathway to be A4-5 beta Ad-Et-5 beta A3 alpha 17 beta diol-5 beta A3 alpha 17 beta diol-glucuronide or Et-Et-glucuronide-5 beta A3 alpha 17 beta diol-glucuronide. The formation of 5 beta-reduced compounds was larger in postbreeding males and females than in breeding males. The opposite was the case for 5 alpha-reduced compounds. The total formation of glucuronides was lower in the breeding males than in the other groups. After the OT incubations 11-ketoandrostenedione, 17 beta-hydroxy-5 beta-androstane-3,11-dione (tentatively identified), and OT-glucuronide were found. 17 beta-Hydroxy-5 alpha-androstane-3,11-dione was not present.

Effects of castration and androgen treatment on aromatase activity in the brain of mature male Atlantic salmon (Salmo salar L.) parr.

Brain homogenates from male Atlantic salmon parr aromatized tritiated androstenedione to estrogens. The aromatase activity in homogenates of whole brains from castrated male parr was lower than that in homogenates from sham-operated male parr in autumn. This was also found in homogenates of the telencephalon and diencephalon, but not in homogenates of the tectum opticum. Treatment of castrated males with testosterone (T) and 11-ketoandrostenedione (OA) increased the aromatase activity in whole brains. T, but not OA, also significantly increased aromatase activity in all brain parts. In spring, half a year after the peak breeding period, mature parr males had a lower aromatase activity in telencephalon, diencephalon and tectum than that found in castrated males in autumn, whereas immature males displayed an even lower level of activity.

Metabolism of E1 and E2 in Ishikawa endometrium carcinoma cells: influence of TNF alpha.

The metabolism of estrone (E1) and estradiol (E2) by Ishikawa endometrial carcinoma cells and its alteration by TNF alpha treatment was studied. Whereas this cell line practically does not respond to estrogens, it is very sensitive to TNF with respect to growth inhibition and other parameters. E2 and E1 were found to be metabolized by this cell line whereby the main metabolite for both estrogens was estriol. TNF significantly increased the rate of E1 and E2 conversion.

Carbonate content in developing human and bovine enamel.

The present study describes the detailed changes in carbonate distribution throughout the different stages of development in human and bovine enamel, from early formation to maturation. Twenty-two human maxillary and mandibular deciduous anterior teeth and 46 bovine teeth were studied. The major mineral ions, calcium and phosphorus, were also analyzed to facilitate comparison of carbonate content with progressing mineralization. The results showed that as enamel matured and mineral concentration increased, carbonate concentration decreased. The observed decrease in percent carbonate per weight enamel mineral may be due to dilution by an influx of relatively carbonate-free mineral, and could, in part, explain the observed increase in crystallinity of enamel mineral as the tissue matures.

Transforming growth factor-beta and ovarian carcinoma cells: regulation of proliferation and surface antigen expression.

Transforming growth factor-beta (TGF-beta) is a multifunctional peptide regulating several processes in ovarian cells. The growth of ovarian carcinoma cell lines (OVCAR-3, HTB-77, 2780 and CRL-1572) was reduced by TGF-beta in a dose related manner. The antiproliferative activity was not improved by combination with other biological response modifiers. Treatment with TGF-beta augmented the expression of interferon-gamma induced class I and II antigens of the major histocompatibility complex. The presentation of another antigen namely the tumor marker CA-125 on the cell surface was markedly reduced by TGF-beta.

Thermal decomposition of developing enamel.

The decomposition of forming, maturing, and mature enamel was studied between room temperature and 1,000 degrees C by powder X-ray diffraction and infrared absorption methods. In mature dental enamel, carbonate decomposition proceeds relatively fast until 500 degrees C and at a slower rate beyond it. In forming and maturing enamel, decomposition is faster and is completed around 800 degrees C. The formation of beta-Ca3(PO4)2 is observed in dental enamel at 500 degrees C. At 1,000 degrees C, the apatite phase in forming and maturing enamel transforms almost completely to beta-Ca3(PO4)2, whereas in mature enamel, even at 1,000 degrees C, only partial decomposition occurs. Infrared results show the appearance in dental enamel of (1) A-type carbonate at room temperature and in the 500-900 degrees C range, in addition to the commonly observed B-type carbonate, and (2) intermediate CO2 molecules during carbonate decomposition (200-500 degrees C).

Seasonal changes in and effect of castration/androgen replacement on the plasma levels of five androgens in the male three-spined stickleback, Gasterosteus aculeatus L.

The seasonal changes in plasma levels of the androgens 11-ketotestosterone (OT), testosterone (T), 11 beta-hydroxytestosterone (OHT), 11-ketoandrostenedione (OA), and 11 beta-hydroxyandrostenedione (OHA) were measured in the male three-spined stickleback (Gasterosteus aculeatus L). OT was the dominant plasma androgen in the breeding season in summer and was the only androgen that peaked during this period. The levels of OT correlated closely with the development of male secondary sexual characters and reproductive behavior. T and OHT were low in all seasons, whereas OHA and OA displayed the highest levels in early winter. During the postbreeding period, the time of active spermatogenesis, all measured steroids were low. Castration resulted in an almost complete loss of plasma OT and reduced T, whereas OHT, OHA, and OA were not reliably influenced. Androstenedione implants in castrated fish increased plasma T and OA implants increased plasma OT, suggesting a nontesticular site of conversion.

Conversion of 11-ketoandrostenedione to 11-ketotestosterone by blood cells of six fish species.

Blood cells from Baltic salmon, Salmo salar, three-spined stickleback, Gasterosteus aculeatus, eel pout, Zoarces viviparus, crucian carp, Carassius carassius, African catfish, Clarias gariepinus, and reedfish, Calamoichthys calabaricus, were incubated with tritiated 11 beta-hydroxyandrostenedione (OHA) or 11-ketoandrostenedione (OA). In all fish there was conversion of OA to 11-ketotestosterone (OT), indicating that 17 beta-hydroxysteroid dehydrogenase activity was present in the blood cells. On the other hand, OHA was not converted to 11 beta-hydroxytestosterone in any fish. The addition of serum to the incubates largely prevented the OA-OT conversion by salmon blood cells.

Isolation, identification, and characterization in Israel of Brucella melitensis biovar 1 atypical strains susceptible to dyes and penicillin, indicating the evolution of a new variant.

During characterization by conventional biotyping tests of numerous Brucella melitensis isolates obtained in Israel in the last 2 years, we identified several strains of the biovar 1 serotype that are atypically susceptible to dyes and penicillin. Their coisolation from two brothers in a family that rears sheep and from the milk of one of their sheep and the prevalence of such strains in two distinct geographical zones in Israel provide epidemiological support for the notion that a new variant has been identified.

Effects of castration on pituitary gonadotropic cells of the male three-spined stickleback, Gasterosteus aculeatus L., under long photoperiod in winter: indications for a positive feedback.

Male three-spined sticklebacks caught in winter were castrated or sham-operated and subsequently kept under long photoperiod at about 20 degrees for a month. With this treatment the sham-operated fish attained breeding condition. The gonadotropic cells of the sham-operated fish contained significantly more dilated endoplasmic reticulum and fewer granules than those of the castrated fish, indicating a higher secretory activity of the gonadotrops in the sham-operated fish. These findings suggest the existence of a physiological positive feedback within the gonadal-pituitary axis of the male stickleback when stimulated into its breeding condition by long photoperiod.

Modulation of secretion of human chorionic gonadotropin by biologic response modifiers on term placenta and choriocarcinoma cells.

The effects of biologic response modifiers such as interferon-gamma, tumor necrosis factor alpha (TNF), and retinoic acid on the human chorionic gonadotropin (hCG) secretion of cultured choriocarcinoma cells (JAR) and term placenta have been studied. Although the proliferation of JAR cells was not inhibited by these agents, retinoic acid and TNF markedly increased both the intracellular levels as well as the secreted amounts of hCG. In the case of the term placenta, only retinoic acid increased the hCG secretion into the culture medium, whereas interferon-gamma and TNF both markedly reduced secretion. The cytostatic agent etoposide (VP-16) was able to augment the hCG secretion on the choriocarcinoma cells but did not alter its production on term placenta. The The data presented indicate different mechanisms of regulation of hCG secretion in the normal and malignant trophoblast.

Aromatase activity in the brain of the three-spined stickleback, Gasterosteus aculeatus. III. Effects of castration under different conditions and of replacement with different androgens.

Non-breeding stickleback males were gonadectomized or sham-operated in winter. They were then exposed to a high temperature and either a short photoperiod which did not induce breeding or to a longer, stimulatory, photoperiod. In the latter experiment empty Silastic capsules or capsules filled with the aromatizable androgen, androstenedione or the non-aromatizable androgen, 11-ketoandrostenedione were implanted intraperitoneally in the operated fish. Furthermore, males that were already in breeding condition were gonadectomized or sham-operated in late spring. Brains were freeze-sectioned and punches from the nucleus preopticus-nucleus anterioris periventricularis region were incubated with [19-3H]-androstenedione. After extraction of steroids with CH2CI2, the tritium activity remaining in the water phase gives a measure of aromatization as tritiated water and formic acid are formed when [19-3H]-androstenedione is aromatized to oestrogens. The aromatase activity was higher in sham-operated than in castrated fish in all three experiments. Both androstenedione and 11-ketoandrostenedione abolished the effect of gonadectomy on aromatization.

Effects of retinoids and interferon-gamma on cultured breast cancer cells in comparison with tumor necrosis factor alpha.

The combination of retinoic acid or tumor necrosis factor alpha (TNF-alpha) with interferon gamma (IFN-gamma) resulted in a synergistic amplification of the anti-proliferative effect of IFN-gamma on cultured breast cancer cells. Retinoic acid could be replaced by other biologically active retinoids. This synergism was also observed for the induction of 2'-5'-oligoadenylate-synthetase, an enzyme which is not expressed constitutively on BT-20 human breast cancer cells and not inducible by retinoic acid or TNF-alpha alone. However, both substances augmented the IFN-gamma-mediated expression. On the other hand, only TNF-alpha and not retinoic acid was able to increase the IFN-gamma induced expression of HLA-DR on the cell surface. Both cytokines antagonized the IFN-gamma effect on detachability of cultured BT-20 cells. The combinations of retinoic acid with IFN-gamma increased the down-regulation of specific binding sites for 125I-IFN-gamma. On the other hand, IFN-gamma exerted no effect on the concentration of the cytoplasmic binding protein for retinoic acid. Data obtained in this study demonstrate a different pattern of action between retinoic acid and TNF-alpha regarding their synergism in combination with IFN-gamma.

Human interferon-gamma increases adhesion of cultured carcinoma cells to the substratum.

Effects of human recombinant-DNA derived interferon-gamma and -alpha 2 on the adhesion of cultured breast cancer cells (BT-20, ZR-75.1, MCF-7, 734-B and Hs-578-T), larynx carcinoma cells (HEP-2), epidermoid carcinoma cells (KB), lung carcinoma cells (CCL 185), and ovarian carcinoma cells (1847) to the surface of cell culture plastic dishes were studied. Layered cells were detached after a 3-day treatment with interferon either by trypsin-EDTA, trypsin, protease or cooling to 4 degrees C. Treatment with interferon-gamma (500 unit/ml) significantly increased the incubation time for trypsin-EDTA, EDTA and at 4 degrees C necessary to bring cells into suspension for the 4 cell lines BT-20, ZR-75.1, MCF-7 and HEP-2. Interferon-alpha 2 was not able to induce a similar effect. Reattachment of interferon-gamma treated ZR-75.1 cells was not increased after harvesting by trypsinization or EDTA action. Decreased adhesion of cultured cells is associated with transformation and the effects of interferon-gamma may be explained by reinforced normal phenotype. Interferon-gamma induced adhesion was not associated with other interferon effects especially the anti-proliferative activity or modulation of surface antigens.

Characterization of an interferon-resistant mutant of the human breast cancer cell line BT-20.

The human breast cancer cells BT-20 were treated for 18 months in the continuous presence of interferon-gamma (HuIFN-gamma; 500 U/ml). These cells have become completely resistant to HuIFN-gamma and interestingly also to IFN-alpha 2. However, the expression of HLA-DR and the regulation of cell adhesion to tissue culture plastic remained partially under the domain of HuIFN-gamma. A reduced number of IFN-gamma binding sites in comparison to the wild-type cells were observed on the IFN-resistant BT-20 cells.
