CS-514, a competitive inhibitor of 3-hydroxy-3-methylglutaryl coenzyme A reductase: tissue-selective inhibition of sterol synthesis and hypolipidemic effect on various animal species.

CS-514 is a tissue-selective inhibitor of 3-hydroxy-3-methylglutaryl coenzyme A reductase, a key enzyme in cholesterol synthesis. For the microsomal enzyme from rat liver, the mode of inhibition is competitive with respect to hydroxymethylglutaryl-CoA, and the Ki value is 2.3 X 10(-9) M. CS-514 also strongly inhibited the sterol synthesis from [14C]acetate in cell-free enzyme systems from rat liver and in freshly isolated rat hepatocytes; the concentrations required for 50% inhibition were 0.8 ng/ml and 2.2 ng/ml, respectively. On the other hand, the inhibition by CS-514 was much less in the cells from nonhepatic tissues such as freshly isolated rat spleen cells, and cultured mouse L cells and human skin fibroblasts. In addition, the cellular uptake of 14C-labeled CS-514 by isolated rat spleen cells or mouse L cells was less than one-tenth of that by isolated hepatocytes. These differences between hepatic and nonhepatic cells were further confirmed by the fact that CS-514 orally administered to rats inhibited sterol synthesis selectively in liver and intestine, the major sites of cholesterogenesis. CS-514 markedly reduced serum cholesterol levels in dogs, monkeys and rabbits, including Watanabe heritable hyperlipidemic (WHHL) rabbits, an animal model for familial hypercholesterolemia in man, but did not reduce those in rats and mice. In the former case, preferential lowering of atherogenic lipoproteins was observed in all of the animals tested. The biliary neutral sterols significantly decreased, whereas the amount of biliary bile acids was not affected by administration of the drug to dogs.

Effect of pulsing electromagnetic fields on DNA synthesis in mammalian cells in culture.

DNA synthesis in Chinese hamster V79 cells was significantly enhanced when they were exposed to weak, pulsing electromagnetic fields generated by specific combinations of the pulse width (25 microseconds), frequency (10, 100 Hz) and magnetic intensity (2 X 10(-5), 8 X 10(-5) T). Conversely the DNA synthesis of cells in the fields at 4 X 10(-4) T was repressed to 80% of that in controls not exposed to the fields.

Chromatid aberrations in a novobiocin-resistant subline of Chinese hamster V79 cells after exposure to novobiocin.

We examined effects of novobiocin alone or in combination with gamma-irradiation, on the frequencies of chromatid-type aberrations in a novobiocin-resistant subline of Chinese hamster V79 cells (NOVOr-1). NOVOr-1 cells were significantly resistant to novobiocin, as compared to wild-type V79 (WT) cells, with respect to cell survival and DNA synthesis. Survival responses of WT and NOVOr-1 cells to gamma-rays in the range 2-10 Gy differed only slightly and the number of chromatid aberrations produced by irradiation at 1 Gy was fairly comparable in the two cell types. Post-irradiation treatment of cells with novobiocin at concentrations exceeding 200 micrograms/ml significantly increased the number of chromatid gaps plus breaks in WT cells as compared with NOVOr-1 cells. With 200 micrograms/ml the increase was 1.2-fold (t-test, P less than 0.05) and with 400 micrograms/ml, 2.3-fold (P less than 0.01) the number produced in NOVOr-1 cells.

Microdosimetric considerations of effects of heavy ions on microorganisms.

An estimation of dose around the trajectory of an ion has been made by use of Tabata and Ito's energy deposition algorithm for electrons which takes into account transmission coefficient. The result of the calculation, as well as Butts and Katz's dose, is successfully applied to the interpretation of inactivation cross sections of vegetative cells of E. coli Bs-1, and E. coli B/r and of B. subtilis spores for He, C and N ions.

Increase in the frequency of gamma-ray induced chromosomal aberrations in mammalian cells by post-treatment with novobiocin.

The effect of novobiocin (an inhibitor of DNA topoisomerase and polymerase) on the frequency of chromosomal aberrations was examined in Chinese hamster V79 cells irradiated with gamma-rays in the plateau phase of growth and subcultured in the presence of novobiocin until the first mitosis after irradiation. Novobiocin alone affected cell survival, DNA synthesis and the mitotic frequency of unirradiated cells in a dose-dependent manner, without causing any significant increase in the frequency of chromosome- or chromatid-type aberrations. The frequency of chromosome-type aberrations induced by gamma-radiation was not influenced by novobiocin at 200 microM, but the frequency of chromosome deletions (but not rings and dicentrics) showed a two-fold increase when 300 microM novobiocin was present. Irradiation produced a low level of chromatid-type aberrations and post-treatment with novobiocin at concentrations greater than 100 microM significantly increased the frequency of chromatid gaps and breaks. The results support the idea that different radiation-induced lesions lead to chromosome- as opposed to chromatid-type aberrations.

Radiation-induced alterations in binding of concanavalin A to cells and in their susceptibility to agglutination.

Cell susceptibility to agglutination mediated by a plant lectin, concanavalin A (Con A), and the binding capacity of Con A to cells following gamma-irradiation have been examined in mouse myeloid leukaemia cells cultured in suspension. Irradiation caused an immediate decrease in the amount of Con A bound to the cell surface, whereas susceptibility of irradiated cells to agglutination by Con A was unchanged when compared to that of the unirradiated cells. Post-irradiation incubation of cells at 37 degrees C resulted in a temporary, more than 1.3-fold increase in cell susceptibility to agglutination 60 min after irradiation, whereas binding capacity of cells for Con A gradually recovered following irradiation, reaching a comparable level to that of unirradiated cells 3 h after irradiation. Cell susceptibility to agglutination by Con A does not depend strongly on its binding capacity.

Device of anoxic chamber system and repair of potentially lethal damage of anoxic cells in vitro.

In order to study radiation responses of anoxic plateau phase cells in vitro, an air-tight anoxic chamber was devised in which Chinese hamster V-79 or Swiss mouse NIH3T3 cells were deoxygenated by replacing the air with saturatedly humidified N2 gas. The cells as adhered on the culture plate, which was contained in the anoxic chamber, were x-irradiated. The deoxygenated condition in the anoxic chamber was maintained as long as necessary. Oxygen enhancement ratio obtained by the anoxic chamber was about 2.6. Repair of potentially lethal damage of the anoxic plateau phase cells was observed to the same extent of that of the oxic cells.

[A case of successfully resected abdominal aortic aneurysm complicated by arteriocolic fistula].

A 80 year-old man was admitted to our hospital with profuse rectal bleeding. Two weeks prior to admission he was diagnosed as having abdominal aortic aneurysm when he had first herald of rectal bleeding. Aortoenteric fistula was strongly suspected and patient underwent emergency operation. Bilateral axillofemoral bypass were first constructed in order to avoid contamination in hte peritoneal cavity. On laparotomy aneurysm was contaminated because of arteriocolic fistula and resection of aortoiliac aneurysm and Hartmann procedure were performed. In the postoperative course, formation of an intra-abdominal abscess, obstruction of the left axillo-femoral bypass graft and enlargement of a residual aneurysm occurred. The prolonged antibiotic therapy was successful and additional repair operations were performed. Abdominal aortic aneurysm complicated with arteriocolic fistula is a rare but lethal disease. To our knowledge, only 13 cases have been reported in the literature including our case. Eleven patients were operated but only 3 survived. Construction of an extra-anatomic bypass, avoidance of an anastomosis in unprepared bowel and aggressive use of antibiotics are recommended because fecal contamination to the aneurysm is inevitable in most cases.

Effect of novobiocin on the frequencies of chromatid-type aberrations and sister-chromatid exchanges following gamma-irradiation.

The effect of novobiocin on the frequencies of chromatid-type aberrations and SCEs was examined in Chinese hamster V79 cells which were exposed to gamma-rays and post-treated with novobiocin. While no chromatid aberrations were induced in the unirradiated cells by novobiocin, the frequency of SCEs was slightly increased by treatment with novobiocin alone. Irradiation of G2 cells produced multiple chromatid-type aberrations and post-treatment of the irradiated cells with novobiocin resulted in a significant increase of the aberrations, including chromatid gaps and breaks. In contrast, novobiocin failed to increase the frequency of SCEs induced by gamma-rays when the irradiated cells were post-treated with novobiocin.

Changes in nuclease sensitivity of mammalian cells after irradiation with 60Co gamma-rays.

Changes in sensitivity of mouse BALB/c 3T3 cells in the plateau phase to digestion with micrococcal nuclease were examined following gamma-irradiation. Immediately after irradiation, cell nuclei were more sensitive to micrococcal nuclease compared to unirradiated nuclei. However, there were no detectable changes in length of basic repeating subunits of 182 base pairs of DNA, which include the nucleosome cores consisting of approximately 140 base pairs of DNA, which When the cells were incubated at 37 degrees following irradiation, the sensitivity of cell nuclei to the nuclease first increased then decreased, reaching a similar level to unirradiated nuclei 6 h after irradiation. Both the initial increase and the subsequent decrease in sensitivity of nuclei to micrococcal nuclease were prevented when 15 microM novobiocin was present during the post-irradiation incubation, suggesting a possible involvement of type II DNA topoisomerase in repair of DNA lesions induced by gamma-rays.

Differential recovery from potentially lethal damage in normal human lung fibroblasts after irradiation with 60Co gamma-rays and accelerated N-ion beam.

PLD recovery after irradiation with accelerated N-ion was examined and compared with that after irradiation with 60Co gamma-rays in normal human lung fibroblasts (IMR-90). No shoulder region was observed in the survival curve of the cells after N-ion irradiation. Ratios of D0 values before and after a 6-hour incubation in plateau phase were 1.1 and 1.8 for N-ion irradiation and gamma-ray irradiation, respectively. The results show that recovery after irradiation with high-LET radiation is slow and much less than after low-LET irradiation.

Two active forms of valyl-tRNA synthetase from Bacillus subtilis: alteration related to the early stages of sporulation.

Two enzymatically active forms, E-I and E-II, of valyl-tRNA synthetase [EC 6.1.1.9] from cells at various stages in the life cycle of Bacillus subtilis 168 LTT, germinated cells, vegetative cells (t-0.5), sporulating cells (t0, t1, t3, and t4), forespores and mature spores, were analyzed by hydroxyapatite column chromatography. The E-II activity was detected in the main fraction of valyl-tRNA synthetase during the life cycle of B. subtilis 168 LTT. The high activity of E-II at t0 decreased rapidly in the stationary and sporulating phases. On the other hand, the E-I activity increased in the early sporulating stage and was about twofold higher at t3 than at t0. After t3, this activity also decreased rapidly and was not detected in forespores and mature spores. The relative amount of E-I at t0 was 3.4% of the total valyl-tRNA synthetase activity eluted from the hydroxyapatite column, 12.9% at t1 and 29.2% at t3, but it was less than 10% at t4 and in germinated cells. The alteration in E-I and E-II activities was also observed in cells of B. subtilis NIG 1121 (spo+), W23 and 168W, but not in any asporogenous mutant strain studied. These results show that the alteration in the valyl-tRNA synthetase activity appears only during the early stages of sporulation and is closely related to the sporulation of B. subtilis.
