[Monoclonal antibodies to human spleen ferritin. I. Preparation and study of the interaction with isoferritin and apoferritin].

Three monoclonal antibodies to human spleen ferritin were produced and their interaction with soluble and immobilized ferritins studied. An immunoassay was developed to monitor the interaction of soluble biotinylated ferritin and the antibody with subsequent separation of the soluble complexes on streptavidin-cellulose. Analysis of immunoreactivities of a series of isoferritins (human liver, spleen, heart and equine spleen ferritins) revealed that all the three monoclonal antibodies bound to only human L-type ferritins (spleen and liver ferritins), suggesting a high species- and tissue specificity of these antibodies. The monoclonal antibodies were specifically directed against conformation-dependent antigenic determinants as could be evidenced from the lack of their binding to the subunits of the dissociated ferritin. The affinity of the monoclonal antibody for the ferritin deprived of iron (apoferritin) was higher than that for native ferritin due to the greater conformational flexibility of the apoferritin molecule. The latter property may underly a complete loss of immunoreactivity by the apoprotein adsorbed on the polystyrene surface as a result of conformational changes induced in the apoferritin molecule by adsorption. These findings provide additional support for recognition by all of the three antibodies of conformational antigenic epitopes in the ferritin molecule.

[Cyclic GMP phosphodiesterase from bovine retina. Amino acid sequence of beta-subunit and nucleotide sequence of corresponding cDNA].

Primary structure of beta-subunit of the cyclic GMP phosphodiesterase has been determined by the parallel analysis of the protein amino acid sequence and the corresponding cDNA nucleotide sequence. The beta-subunit contains 852 amino acid residues, its molecular mass is 98291 Da. A significant homology is found between beta- and alpha-subunit of the cGMP phosphodiesterase.

[The choice of the optimal aeration conditions for the stab cultivation of Bacillus thuringiensis H14 266/2-1].

Optimal conditions of aeration with depth regime of cultivation of Bacillus thuringiensis H14 266/2-1, their effect on the output of biomass, endotoxin and activity of bacteria have been studied. The aeration conditions are optimized using the mathematical method of experiment planning. It is established that the air supply favours an increase of the bacillary biomass and insecticide activity for the whole cultivation period. In the first day of fermentation it is inexpedient to increase to rate of the medium agitation. To obtain a high titre of cells of the studied bacilli and to retain their activity a specific air consumption is recommended amounting to 0.5 1/1/min for the first 12h with an increase to 1.01/1/min in the following hours of fermentation, the constant rate of the medium agitation being 500 rev/min.

[Intensification of the growth and development of Bacillus thuringiensis H14 266/2-1 by optimizing the nutrient medium].

The mathematical method of experimental design was used to develop a new enzymic medium for cultivation of Bacillus thuringiensis H14 266/2-1, a bactoculicide producer. The optimized medium based on corn flour enzyme lysate as a carbon source and fodder yeast enzyme lysate as a source of nitrogen amine made it possible to increase twice the titre biomass yield for 24 h cultivation as compared to the initial medium. The above medium does not yield to the initial production medium in the insecticide activity.

[L-forms of aerobic spore-forming bacteria isolated from urologic patients].

New data are presented on the ability of different aerobic spore-forming bacteria isolated from the organism of urological patients to produce L-forms of these microorganisms in the presence of penicillin and ampicillin. Bacillus cereus is shown to be the most resistant to these antibiotics.
