Altered expression of genes involved in hepatic morphogenesis and fibrogenesis are identified by cDNA microarray analysis in biliary atresia.

Biliary atresia (BA) is characterized by a progressive, sclerosing, inflammatory process that leads to cirrhosis in infancy. Although it is the most common indication for liver transplantation in early childhood, little is known about its etiopathogenesis. To elucidate factors involved in this process, we performed comprehensive genome-wide gene expression analysis using complementary DNA (cDNA) microarrays. We compared messenger RNA expression levels of approximately 18,000 human genes from normal, diseased control, and end-stage BA livers. Reverse-transcription polymerase chain reaction (RT-PCR) and Northern blot analysis were performed to confirm changes in gene expression. Cluster and principal component analysis showed that all BA samples clustered together, forming a distinct group well separated from normal and diseased controls. We further identified 35 genes and ESTs whose expression differentiated BA from normal and diseased controls. Most of these genes are known to be associated with cell signaling, transcription regulation, hepatic development, morphogenesis, and fibrogenesis. In conclusion, this study serves to delineate processes that are involved in the pathogenesis of BA.

Noninvasive differentiation of pseudonormal/restrictive from normal mitral flow by Tei index: a simultaneous echocardiography-catheterization study in patients with acute anteroseptal myocardial infarction.

BACKGROUND: Differentiation of pseudonormal/restrictive from normal mitral flow is still clinically problematic. Pseudonormal/restrictive flow is usually associated with left ventricular dysfunction, which can be detected by Doppler Tei index, combining systolic and diastolic function. Therefore, the purpose of this study was to test the feasibility of the Tei index to differentiate pseudonormal/restrictive from normal mitral flow. METHODS: In 26 patients with anteroseptal acute myocardial infarction and early diastolic mitral flow velocity (E) to late diastolic mitral flow velocity (A) ratio (E/A) > or = 1, left ventricular volumes; E and A; deceleration time of E; and the Tei index, defined as the sum of the isovolumic contraction and relaxation time divided by ejection time, were evaluated by Doppler echocardiography, and pulmonary capillary wedge pressure was measured by catheterization. Pseudonormal/restrictive mitral flow was defined as E/A > or = 1 associated with pulmonary capillary wedge pressure > 12 mm Hg. RESULTS: There were 19 and 7 patients with pseudonormal/restrictive and normal mitral flow, respectively. Among the indices of left ventricular function, the Tei index achieved the best correlation with pulmonary capillary wedge pressure (r(2) = 0.66, P <.0001). By setting the Tei index > or = 0.55 as the criteria for pseudonormal/restrictive mitral flow, this diagnosis had the sensitivity, specificity, and accuracy of 84%, 100%, and 88%, respectively. CONCLUSION: The Tei index allows noninvasive differentiation of pseudonormal/restrictive from normal mitral flow.

Gemmatimonas aurantiaca gen. nov., sp. nov., a gram-negative, aerobic, polyphosphate-accumulating micro-organism, the first cultured representative of the new bacterial phylum Gemmatimonadetes phyl. nov.

A phylogenetically novel aerobic bacterium was isolated from an anaerobic-aerobic sequential batch reactor operated under enhanced biological phosphorus removal conditions for wastewater treatment. The isolation strategy used targeted slowly growing polyphosphate-accumulating bacteria by combining low-speed centrifugations and prolonged incubation on a low-nutrient medium. The isolate, designated strain T-27T, was a gram-negative, rod-shaped aerobe. Cells often appeared to divide by budding replication. Strain T-27T grew at 25-35 degrees C with an optimum growth temperature of 30 degrees C, whilst no growth was observed below 20 degrees C or above 37 degrees C within 20 days incubation. The pH range for growth was 6.5-9.5, with an optimum at pH 7.0. Strain T-27T was able to utilize a limited range of substrates, such as yeast extract, polypepton, succinate, acetate, gelatin and benzoate. Neisser staining was positive and 4,6-diamidino-2-phenylindole-stained cells displayed a yellow fluorescence, indicative of polyphosphate inclusions. Menaquinone 9 was the major respiratory quinone. The cellular fatty acids of the strain were mainly composed of iso-C15:0, C16:1 and C14:0. The G + C content of the genomic DNA was 66 mol%. Comparative analyses of 16S rRNA gene sequences indicated that strain T-27T belongs to candidate division BD (also called KS-B), a phylum-level lineage in the bacterial domain, to date comprised exclusively of environmental 16S rDNA clone sequences. Here, a new genus and species are proposed, Gemmatimonas aurantiaca (type strain T-27T=JCM 11422T=DSM 14586T) gen. nov., sp. nov., the first cultivated representative of the Gemmatimonadetes phyl. nov. Environmental sequence data indicate that this phylum is widespread in nature and has a phylogenetic breadth (19% 16S rDNA sequence divergence) that is greater than well-known phyla such as the Actinobacteria (18% divergence).

Constitutively active protein kinase A disrupts motility and chemotaxis in Dictyostelium discoideum.

The deletion of the gene for the regulatory subunit of protein kinase A (PKA) results in constitutively active PKA in the pkaR mutant. To investigate the role of PKA in the basic motile behavior and chemotaxis of Dictyostelium discoideum, pkaR mutant cells were subjected to computer-assisted two- and three-dimensional motion analysis. pkaR mutant cells crawled at only half the speed of wild-type cells in buffer, chemotaxed in spatial gradients of cyclic AMP (cAMP) but with reduced efficiency, were incapable of suppressing lateral pseudopods in the front of temporal waves of cAMP, a requirement for natural chemotaxis, did not exhibit the normal velocity surge in response to the front of a wave, and were incapable of chemotaxing toward an aggregation center in natural waves generated by wild-type cells that made up the majority of cells in mixed cultures. Many of the behavioral defects appeared to be the result of the constitutively ovoid shape of the pkaR mutant cells, which forced the dominant pseudopod off the substratum and to the top of the cell body. The behavioral abnormalities that pkaR mutant cells shared with regA mutant cells are discussed by considering the pathway ERK2 perpendicular RegA perpendicular [cAMP] --> PKA, which emanates from the front of a wave. The results demonstrate that cells must suppress PKA activity in order to elongate along a substratum, suppress lateral-pseudopod formation, and crawl and chemotax efficiently. The results also implicate PKA activation in dismantling cell polarity at the peak and in the back of a natural cAMP wave.

In a subset of subjects on highly active antiretroviral therapy, human immunodeficiency virus type 1 RNA in plasma decays from 50 to <5 copies per milliliter, with a half-life of 6 months.

Three of five virally suppressed human immunodeficiency virus type I (HIV-1)-infected patients treated with highly active antiretroviral therapy and followed intensively with a supersensitive reverse transcriptase PCR assay with a lower limit of quantitation of 5 copies/ml showed statistically significant viral load decays below 50 copies/ml, with half-lives of 5 to 8 months and a mean of 6 months. This range of half-lives is consistent with the estimated half-life of the latent HIV-1 reservoir in the peripheral blood. Those patients without decay of viral load in plasma may have significant cryptic HIV-1 residual replication.

Transcription-dependent degradation of topoisomerase I-DNA covalent complexes.

Topoisomerase I (Top I)-DNA covalent complexes represent a unique type of DNA lesion whose repair and processing remain unclear. In this study, we show that Top I-DNA covalent complexes transiently arrest RNA transcription in normal nontransformed cells. Arrest of RNA transcription is coupled to activation of proteasomal degradation of Top I and the large subunit of RNA polymerase II. Recovery of transcription occurs gradually and depends on both proteasomal degradation of Top I and functional transcription-coupled repair (TCR). These results suggest that arrest of the RNA polymerase elongation complex by the Top I-DNA covalent complex triggers a 26S proteasome-mediated signaling pathway(s) leading to degradation of both Top I and the large subunit of RNA polymerase II. We propose that proteasomal degradation of Top I and RNA polymerase II precedes repair of the exposed single-strand breaks by TCR.

Enhanced osteoblast functions on RGD immobilized surface.

Many methods are currently under investigation to improve the integration of dental implants to surrounding bones. Among these methods, peptide-modified surfaces have been highlighted as one of the most promising. Our study, aimed at the cellular response to RGD-immobilized surface in vitro, investigated the basis for designing a bone-active surface coating with RGD-containing peptide. Gold-coated titanium surfaces were used as indicative control surfaces for peptide immobilization. Using self-assembly monolayer techniques, 2 types of peptides, RGDC (Arg-Gly-Asp-Cys) and RDGC (Arg-Asp-Gly-Cys), were immobilized onto the gold surfaces. Surface justification was realized through X-ray photoelectron spectroscopy and Fourier transform infrared spectra. Primary calvarial osteoblasts were cultured on RGDC, RDGC, and non-peptide-coated surfaces. Cell attachment, morphology, proliferation, and expression of osteocalcin (OC) messenger RNA (mRNA) were assessed using cell counting, immunolabeling fluorescence microscopy, and Northern blot assay. Four and 8 hours after culture, cell attachment was enhanced on RGDC surfaces. Correspondingly, increased cell spreading and significantly greater cell proliferation were also observed in cells grown on the RGDC-coated surfaces. More importantly, osteoblasts on RGDC surfaces showed earlier and significant OC mRNA expression at day 15 compared with controls having the similar expression at day 21. These results provided evidence of the enhanced functions of osteoblasts cultured on the RGDC-modified surfaces, which might be effective in improving osseointegration for dental implants.

Radioimmunotherapy with 153Sm-CEA monoclonal antibody in nude mice bearing human colon carcinoma: an experimental study.

OBJECTIVE: To observe the therapeutic effect of 153Sm-labeled CEA monodonal antibody(mAb) in nude mice bearing human colon carcinoma. METHODS: Fifteen nude mice were subjected to subcutaneous inoculation of human colon carcinoma cells, and 3 days later, they were divided into 3 groups with equal numbers to receive single high-dose injection of 11.1 MBq 153Sm-CEA mAb (therapy group), 11.1MBq 153SmCl3 (therapy control group), or 100 microl normal saline (non-treatment control group). The tumor-inhibiting effect of 153Sm-labeled CEA mAb was evaluated in terms of body weight changes and tumor volume variation 4 weeks after the treatment. Histological analysis of tumors were performed in all the groups after the all other observations were completed. RESULTS: 153Sm-CEA mAb had a significant anti-tumor effect, with a tumor inhibition rate of 74.29% at 4 weeks after treatment, while for 153SmCl3, the inhibition rate was only 15.90%. Rapid tumor growth was observed in non-treatment control group. No significant difference in the body weight changes was noted between the 3 groups. Histopathological examination revealed tumor necrosis as the evidence for radioactive damage in therapy group, which was not observed in non-treatment control group. CONCLUSION: 153Sm-CEA mAb has a strong selective inhibitory effect against colon carcinoma and may be potentially used as an agent in radioimmunotherapy.

[Tolerance of human body to simultaneous infusion of 5-fluorouracil and calcium folinate].

BACKGROUND & OBJECTIVE: It has been confirmed that calcium folinate (CF), as a chemotherapy-biochemical modulator of 5-fluorouracil (5-FU), can improve the effect of antitumor of 5-FU. But the usage and dosage of this therapy should be further studied. The purpose of this article was to study the maximum tolerated dose (MTD) and the dose limiting toxicity (DLT) of human body to simultaneously intravenous infusion of 5-FU and CF. METHODS: The cases being up to the selecting standard were treated with DDP+CF/5-FU regimen. The dose of CF and DDP for every course was fixed. The first dose of 5-FU was 500 mg x (m(2) x d)(-1),then the doses were increased by degree of 50 mg x (m(2) x d)(-1), if no severe side effect was observed, up to maximum toxicity tolerated dose. There were 3-6 cases in each degree of dose. RESULTS: Thirty-two cases were treated and given total 64 courses of the administration in all. The overall remission rate was 84.4%. The MTD of 5-FU was 800 mg x (m(2) x d)(-1). Mucositis and diarrhea were found to be the DLT. CONCLUSION: Simultaneous continuous infusion of 5-FU and CF can enhance the dosage of 5-FU, and has a better efficacy to advanced digestive tract, head and neck cancers. So the authors recommend that the dosage of 5-FU in phase II clinical trial is 700 mg x (m(2) x d)(-1). This dose can be used for 5 consecutive days and be repeated after 3 weeks.

[Role of expression inducible nitric oxide synthase-mRNA of leukocyte and nasal mucosa in allergic rhinitis].

OBJECTIVE: To investigate the relationship between expression of leukocyte inducible nitric oxide synthase (iNOS)-mRNA and allergic rhinitis (AR). METHOD: Thirty-five patients with AR and 30 healthy controls were included in this study. Expression of iNOS-mRNA in peripheral blood leukocyte was detected by in situ hybridization. NO in plasm was measured by nitrate reductase. Expression of iNOS-mRNA in nasal mucosal was detected in 8 patients with AR and 6 healthy controls. RESULT: No expression of leukocyte iNOS-mRNA in healthy controls was found. In AR patients, the positive cells were significantly increased, the positive rate reached 40.82%. Expression of iNOS-mRNA was localized at the epithelium, gland and macrophage in healthy controls. Hyperplasia and expression of iNOS-mRNA increased at epithelium, gland and macrophage in the AR patients(t = 23.17, P < 0.001). The level of plasm NO in AR group was higher than that in healthy control group (t = 27.89, P < 0.001). CONCLUSION: There is a relationship between expression of leukocyte iNOS-mRNA and the level of plasm NO in AR patients. The study provides an easy method of in situ hybridization for detecting some signal in body.

[Multifactor analysis of the reasons causing undercorrection after laser in situ keratomileusis].

OBJECTIVE: To investigate the relationship between various factors and the undercorrection after laser in situ keratomileusis (LASIK). METHODS: Of 1 391 eyes (696 cases), the undercorrection eyes grouped by left or right eye were analyzed by Chi-square test and logistic regression analysis based on age, sex, occupation, address, family history of myopia and all ophthalmologic examination results before and after LASIK. RESULTS: The results of logistic regression analysis showed that the undercorrection after LASIK of both groups was related to four factors: the duration of myopia (left eye OR = 1.076, 95% CI: 1.030 - 1.124; right eye OR = 1.093, 95% CI: 1.046 - 1.142), preoperative refractive power (left eye OR = 7.799, 95% CI: 1.755 - 34.654; right eye OR = 28.823, 95% CI: 5.750 - 144.467), preoperative best corrected vision (left eye OR = 0.000, 95% CI: 0.000 - 0.262; right eye OR = 0.000, 95% CI: 0.000 - 0.144) and the corneal thickness (left eye OR = 0.976, 95% CI: 0.965 - 0.987; right eye OR = 0.975, 95% CI: 0.964 - 0.986). CONCLUSION: Longer duration of myopia and the more refractive power seemed to be the risk factors, while the better corrected vision and the thicker thickness of cornea the protect factors of undercorrection after LASIK.

[Application of power spectral entropy to the noninvasive detection of focal ischemic cerebral injury].

A model of SD rat focal ischemic cerebral injury is presented for use in noninvasively detecting both the extent and the location of focal ischemic cerebral injury. EEG signals of ischemic region and normal region are recorded from the moment before ischemia to 30 minutes after ischemia. Then the Power Spectral Entropy(PSE) analysis of EEG is performed. Results show that the PSE of EEG signals changes greatly as a result of focal ischemic cerebral injury. The PSE of EEG signal of ischemic region is less than that before ischemia as the ischemia lasts 15 minutes. The PSE of EEG signals of ischemic region is much less than that of normal region. These indicate that the power spectral entropy of EEG signal is a good index of focal ischemic cerebral injury and the method of power spectral entropy analysis is simple and effective.

Effect of aminoguanidine on the recovery of rat hindlimb motor function after spinal cord injury.

OBJECTIVE: To study the effects of aminoguanidine (AG), the inhibitor of inducible nitric oxide synthase (NOS), on the recovery of rat hindlimb motor function after spinal cord injury and explore the possible mechanism. METHODS: Thirty rat models of spinal cord compression injury were established according to Nystrom's method. AG therapy was administered for 4 times at 1 h before and 8, 24, 36 h after the injury, and 24 h after the completion of the therapy, spectrophotography was performed to measure the content of NO and activity of NOS in the injured spinal cord, followed by flow cytometry for determining the apoptotic rate 48 h later. The evaluation of the hindlimb motor function recovery was conducted by electrophysiological method and by measuring the behavior scores. RESULTS: AG significantly decreased the NO content (from 2.2714+/-0.4239 micromol/g.pro to 0.8466+/-0.0477 micromol/g.pro, P <0.05) and NOS activity (from 0.3408+/-0.0228 U/mg pro to 0.2702+/-0.0148 U/mg pro, P <0.05) in the injured spinal cord. The apoptotic rats were also reduced (from 7.88% +/-0.79% to 3.10% +/-0.66%, P <0.05). Four weeks after the therapy, the behavior score of the rats improved from 7.1+/-4.5 to 17.3+/-4.7 (P <0.01), and the latency and amplitude of the motor evoked potentials improved from 0 ms to 8.89+/-0.91 ms and from 0 mv to 1.99+/-0.48 mv respectively, showing significant therapeutic effect of AG (P <0.05). CONCLUSION: AG can improve motor functions of injured spinal cord in rats, possibly resulting from decreased apoptotic cells of the neurons in the spinal cord in the early stages of the injury.

[Effects of La on the growth of kidney bean seedling under Cd stress].

In this study, kidney bean (Phaseolus vulgaris) was used as experiment-material, and effects of lanthanum on growth and metabolism of kidney bean seedling under cadmium stress was studied by water culture. The results showed that kidney bean seedling was chronically harmed by 30 mumol.L-1 Cd2+, however, its height and main root were reduced about 31.1% and 39.2%, meanwhile, leaf area, fresh and dry weight of leaf, stem and root were decreased 48.0%, 42.7%, 29.6%, 61.3% and 49.4% respectively. With acute damage of 50 mumol.L-1 Cd2+, physiological and biochemical characteristics of seedling changed greatly, e.g., chlorophyll content and root activity were decreased about 23.5% and 28.7%, cell membrane permeability, the malonydialdehyde (MDA) content, activities of catalase (CAT) and peroxidase (POD) were increased 5.58%, 28.6%, 0.6% and 7.0% respectively. And with longer treatment time, the damage became more serious. When 10 mg.L-1 La was used to spray on kidney bean seedling under cadmium stress one time, growth and metabolism of seedling was obviously reconditioned and the damage of Cd pollution was reduced. It is proved, by the experiment, that La can improve chlorophyll content(30.0%) and reduce cell membrane permeability (0.87%) and content of MDA(9.5%), and keep the activities of CAT (0.1%) and POD(1.6%) of kidney bean seedling.

[Evaluation of Fenton process for the treatment of sanitary landfill leachate with Response Surface Methodology].

Response Surface Methodology (RSM) was employed to investigate the effect of TOC ratio of high molecular weight organic matter (HMWOM) to low molecular weight organic matter (LMWOM), the LMWOM concentration and hydrogen peroxide to ferrous ion molar ratio on COD removal during the treatment of sanitary landfill leachate with Fenton reagents. The high and low levels of HMWOM/LMWOM, the LMWOM concentration and H2O2/Fe2+ were from 14 to 38, 975 to 3000 mg/L, and 1.75 to 4.00 respectively. Using the software of Design--Expert 5, a response surface quadratic model in terms of actual factors was obtained based on the experimental data, and COD removal contour plots were derived from the model accordingly. It showed that COD removal increased with the increase of HMWOM to LMWOM ratio, but decreased with increasing LMWOM concentration. There existed an optimal hydrogen peroxide to ferrous ion molar ratio so that the highest COD removal efficiency could be achieved.
