Molecular cloning and functional expression of a gene encoding an antiarrhythmia peptide derived from the scorpion toxin.

From a cDNA library of Chinese scorpion Buthus martensii Karsch, full-length cDNAs of 351 nucleotides encoding precursors (named BmKIM) that contain signal peptides of 21 amino acid residues, a mature toxin of 61 residues with four disulfide bridges, and an extra Gly-Lys-Lys tail, were isolated. The genomic sequence of BmKIM was cloned and sequenced; it consisted of two exons disrupted by an intron of 1622 bp, the largest known in scorpion toxin genomes, inserted in the region encoding the signal peptide. The cDNA was expressed in Escherichia coli. The recombinant BmKIM was toxic to both mammal and insects. This is the first report that a toxin with such high sequence homology with an insect-specific depressant toxin group exhibits toxicity to mammals. Using whole cell patch-clamp recording, it was discovered that the recombinant BmKIM inhibited the sodium current in rat dorsal root ganglion neurons and ventricular myocytes and protected against aconitine- induced cardiac arrhythmia.

Quadruplex structures of RNA 14-mer, r(GGAGGUUUUGGAGG) and DNA 14-mer, d(GGAGGTTTTGGAGG).

We have determined solution structure of r (GGAGGUUUUGGAGG) (R14) by NMR; the RNA 14-mer forms an intra-strand parallel quadruplex with a G-tetrad and a hexad, in which a G-tetrad core is augmented by association of two A residues. The quadruplex further forms a dimer through stacking interaction between the hexads. We also synthesized the corresponding DNA 14-mer, d (GGAGGTTTTGGAGG) (D14), and examined its properties and structure by CD, gel electrophoresis, and NMR. The CD and gel electrophoretic studies suggest that D14 forms a quadruplex entirely different from that of R14 in the presence of K+ ions; two molecules of D14 form a quadruplex containing antiparalle strands and diagonal loops. The NMR study also gave the results that are consistent with such structure: alternate glycosidic conformation, 5'G(syn)-G(anti)3', and characteristic chemical shift data.

Basolateral membrane expression of the Kir 2.3 channel is coordinated by PDZ interaction with Lin-7/CASK complex.

The basolateral membrane sorting determinant of an inwardly rectifying potassium channel, Kir 2.3, is comprised of a unique arrangement of trafficking motifs containing tandem, conceivably overlapping, biosynthetic targeting and PDZ-based signals. In the present study, we elucidate a mechanism by which a PDZ interaction coordinates one step in a basolateral membrane sorting program. In contrast to apical missorting of channels lacking the entire sorting domain, deletion of the PDZ binding motif caused channels to accumulate into an endosomal compartment. Here, we identify a new human ortholog of a Caenorhabditis elegans PDZ protein, hLin-7b, that interacts with the COOH-terminal tail of Kir 2.3 in renal epithelia. hLin-7b associates with the channel as a part of a multimeric complex on the basolateral membrane similar to a basolateral membrane complex in C. elegans vulva progenitor cells. Coexpression of hLin-7b with Kir 2.3 dramatically increases channel activity by stabilizing plasma membrane expression. The discovery identifies one component of the sorting machinery and provides evidence for a retention mechanism in a hierarchical basolateral trafficking program.

A new indicator cell line to monitor human foamy virus infection and stability in vitro.

In order to determine the human foamy virus (HFV) infection in vitro conveniently, we developed a baby hamster kidney cell (BHK)-21-derived indicator cell line (BHK-HFVLTR-EGFP) containing a plasmid that encodes the enhanced green fluorescent protein (EGFP) driven by the HFV long terminal repeat promoter. The viral trans-activator Bel-1 protein can induce EGFP expression, so the HFV titer could be determined by counting the corresponding EGFP-positive cells. This foamy-virus-activated EGFP expression (FAE) assay was about 50 times more sensitive than the traditional focus plaque assay by end-point dilution. Additionally, the results of the FAE assay can be obtained rapidly within 2 days, whereas the determination of focus developing needs 2 weeks. Moreover, a linear relationship was found between the fluorescence intensity and the titer of inoculated HFV. In brief, the FAE assay is a rapid, easy, sensitive and quantitative method for monitoring and investigating HFV infection. Using the indicator cell line BHK-HFVLTR-EGFP, we examined the effects of repeated freeze-and-thaw cycles and temperature on HFV stability by the FAE assay. Such information about HFV infection and stability would be valuable for HFV applications.

Resveratrol induces growth inhibition, S-phase arrest, apoptosis, and changes in biomarker expression in several human cancer cell lines.

PURPOSE: We examined the effects of the phytochemical resveratrol in six human cancer cell lines (MCF7, SW480, HCE7, Seg-1, Bic-1, and HL60). EXPERIMENTAL DESIGN AND RESULTS: Resveratrol induced marked growth inhibition in five of these cell lines, with IC(50) values of approximately 70-150 microM. However, only partial growth inhibition was seen in Bic-1 cells. After treatment with 300 microM resveratrol for 24 h, most of the cell lines were arrested in the S phase of the cell cycle. In addition, induction of apoptosis was demonstrated by the appearance of a sub-G(1) peak and confirmed using an annexin V-based assay. Cyclin B1 expression levels were decreased in all cell lines after 48 h of treatment. In SW480 cells, cyclin A, cyclin B1, and beta-catenin expression levels were decreased within 24 h. There was a decrease in cyclin D1 expression after only 2 h of treatment, and this persisted for up to 48 h. This decrease was partially blocked by concurrent treatment with the proteasome inhibitor calpain inhibitor I. Using a luciferase-based reporter assay, resveratrol did not inhibit cyclin D1 promoter activity in SW480 cells. Furthermore, using a reverse transcription-PCR-based assay, only a higher dose of resveratrol (300 microM) appeared to decrease cyclin D1 mRNA. Seg-1 cells expressed basal levels of cyclooxygenase-2 (cox-2), which was further induced by resveratrol. Neither basal levels nor induction of cox-2 was detectable in the remaining cell lines. Thus, cox-2 does not appear to be a critical target of this compound. CONCLUSIONS: These studies provide support for the use of resveratrol in chemoprevention and cancer therapy trials. Cyclin D1, cyclin B1, beta-catenin, and apoptotic index could be useful biomarkers to evaluate treatment efficacy.

[One year observation on effects of estrogen, tamoxifen combined with or without fluoride on preventing bone loss in ovariectomized rats].

OBJECTIVE: To compare the effects of one year treatment of estrogen and tamoxifen, either alone or combined with fluoride on bone metabolism in ovariectomized rats. METHODS: One hundred and forty two virgin female Sprague-Dawley rats were ovariectomized (OVX) or sham operated at 6 months of age, and randomly divided into 7 groups (19 - 21 rats each): (1) sham-operated controls; (2) OVX vehicle controls; (3) estrogen group; (4) fluoride group; (5) tamoxifen group; (6) fluoride plus estrogen; (7) fluoride plus tamoxifen. The treatments lasted for one year. Bone mineral density (BMD) measurement, bone histomorphometry analysis (lumbar vertebrae) and biomechanical test (3-point-bending test on right femur) were performed before and after treatment. The histological change of uterus were also determined at the same time. RESULTS: (1) Twelve months of ovariectomy, the OVX group had significantly lower total body (279 mg/cm(2)), lumbar vertebra (232 mg/cm(2)) than all other groups (total body 286 - 298 mg/cm(2), lumber 251 - 266 mg/cm(2), P < 0.05). Estrogen group (216 mg/cm(2)) had higher BMD than tamoxifene group (195 mg/cm(2)) in midshaft of femur. (2) After 4 months of ovariectomy, only the two combination groups kept the maximum load (145 N) of right femur over the level of OVX group (118 N); after 12 months of treatment, the maximum load of OVX group [(108 +/- 13) N] was significantly different from all other groups (132 - 155 N); both maximum load and elastic load of estrogen group were significantly higher than that of tamoxifen group. (3) No evidence of dysmineralization was found by bone histomorphometry. CONCLUSIONS: Estrogen has more potent effects on keeping bone mass and bone strength as compared with tamoxifene. The combined therapy (estrogen + fluoride or tamoxifen + fluoride) resulted in better bone strength than the single ones.

[Experimental investigation on sulfur emission properties of micro-pulverized coal].

Micro-pulverized coal(0-20 microns) combustion is a new kind of pulverized coal combustion technology. Four Heshan sulfur coal samples with different mean particles diameter were combusted, using a TGA-FTIR made in Germany. Micro-pulverized coal self-desulfurization and adding CaO desulfurization characteristics were investigated. Experimental result proved that micro-pulverized coal SO2 emission was less than coarse particles, and with the same Ca/S molar ratio 3 micro-pulverized coal can obtain higher sulfur capture efficiency. Its unique desulfurization property provide a new idea path for reduce sulfur releasing from pulverized coal combustion.

[Analysis of prognostic factors for 63 patients with brain metastasis from lung cancer after radiochemotherapy].

BACKGROUND & OBJECTIVE: Brain metastasis is often found in the patients with lung cancer. Radiotherapy is regular and effective method, and it aims at palliating symptoms and prolonging survival time. However, now there are different viewpoints on protocols of radiotherapy and prognostic factors. A retrospective analysis was used to evaluate the results of treatment for 63 cases with brain metastasis from lung cancer and explore the prognostic factors. MATERIALS AND METHODS: Sixty-three patients of brain metastasis from lung cancer from Jan. 1994 to Dec. 2000 were studied retrospectively. All of them have received radiotherapy (RT) and 42 cases received combined chemotherapy. The median dose of radiotherapy was 40Gy(24-62.4 Gy). Mean survival times from two factors were compared by t-test and the cumulative survival rate was analyzed by Kaplan-Meier estimates, and the influencing factors were screened by Cox proportional hazard model. RESULTS: The mean survival times were 9.5 months in the radiochemotherapy group and 6.3 months in radiotherapy alone group, 9.3 months in good response group and 5.0 months in the poor response group (P = 0.043), 10.8 months in the > or = 50 Gy group and 6.7 months in the < 50 Gy group(P = 0.028), and 10 months in > or = 1.8 Gy fraction group and 4.8 months in the < 1.8 Gy fraction group (P = 0.015). Survival analysis results suggested that the patients with combined chemotherapy and good response have better prognosis. CONCLUSIONS: Combined chemotherapy and efficacy at the end of treatment are independent prognosis factors of brain metastasis from lung cancer.

[Effects of anti-ABL tyrosine kinase intrabody on the growth of K562 cells in nude mice].

OBJECTIVE: To study the effects of anti-ABL tyrosine kinase intrabody on the growth of human chronic myelogenous leukemia (CML) cells in nude mice. METHODS: A recombinant retroviral vector MSCV-ibE-IRES-eGFP was constructed to express intracellular single-chain antibody (intrabody) against ABL tyrosine kinase domain in CML cells. K562 cells were transduced with the retrovirus, eGFP+ cells were then selected by fluorescence-activated cell sorting (FACS). The intrabody mRNA expression was determined by reverse transcription (RT)-polymerase chain reaction (PCR). BCR/ABL and c-ABL protein tyrosine kinase (PTK) activity in the cells was examined. Transduced cells and control group K562 cells were transplanted into nude mice respectively and the tumor sizes were dynamically observed. RESULTS: K562-ibE cell was obtained. Expression of the BCR/ABL and c-ABL protein tyrosine kinase activity of harvested K562-ibE cells were markedly inhibited. At 14, 21 and 28 days after cell injection, the tumor volumes of experimental mice were obviously smaller than that of control mice, about one half of the control groups (P < 0.05). CONCLUSION: The growth of K562-ibE cells was significantly inhibited in vivo. It is possible that inhibition of the BCR/ABL protein tyrosine kinase activity by the intrabody blocked BCR/ABL signal transduction pathway, promoted apoptosis and reduced tumorigenicity of K562 cells in vivo.

[Effects of liposomes formulation and preparation method on the stability of acyclovir palmitate liposomes].

AIM: To study the effects of various liposomes formulations and preparation methods on the stability of acyclovir palmitate (ACV-C16) liposomes on storage at 4 degrees C and 25 degrees C over a 6 months period. METHODS: The mean particle size, Zeta potential, pH and leaking ratio of ACV-C16 liposomes were the parameters chosen to indicate the stability of liposomes. All of the parameters were compared among various lipid compositions [egg lecithin/cholesterol/hosphatidylserine (PC/CH/PS), egg lecithin/cholesterol/stearylamine (PC/CH/SA), egg lecithin/cholesterol/cholesteryl sulphate (PC/CH/CS), bovine brain ceramides/cholesterol/palmitic acid/cholesteryl sulphate (CM/CH/PA/CS)], different preparation methods (film dispersing, reverse phase evaporation, dehydration/rehydration), charges (positive, negative), as well as among multilamellar vesicles liposomes (MLV), large unilamellar vesicles liposomes (LUV) and dehydration/rehydration vesicles liposomes (DRV). RESULTS: An analysis of various parameters led to the conclusion that the stability of liposomes followed the order of PC/CH/CS > CM/CH/PA/CS > PC/CH/PS > PC/CH/SA at the same storage conditions; the positively charged system showed the most unstable delivery system of liposomes as compared to the other three systems. As far as stability was concerned, LUV liposomes proved to be superior to MLV liposomes and DRV liposomes, and the modified reverse phase evaporation method of Szoka provided the best preparation method. The stability in systems was enhanced when systems were stored at 4 degrees C as compared to storage at 25 degrees C. CONCLUSION: The stability of liposomes was significantly interrelated with lipid composition of various liposomes, preparation method and different storage conditions.

[Methods of gene therapy for inner ear diseases].

OBJECTIVE: To study an effective, safety and convenient method for gene transfer and introduction to inner ear. METHOD: Ad GFP were transferred into inner ears of 30 guinea pigs through round window, basal turn of cochlea and cerebrospinal fluid respectively. Auditory brainstem response (ABR)and otoacoustic emission of distortion products (DPOAE) were assayed to these animals preoperatively and postoperatively, then transfection and expression of Ad-GFP in inner ears and structure of cochlear hair cells were observed. RESULT: Transfection and expression of Ad-GFP were found in spiral ganglion of cochlea, stria vascularis and Cortisorgan through all of the three methods. The expression products were in the highest level at the third day after operation. But the operation through round window and basal turn of cochlea could lead to bleeding and coagulum deposited in parietalgyrus, which could result in hair cells injury. Changes of ABR threshold of these animals were not significant. But amplitude of DPOAE decreased significantly in animals through round window and basal turn of cochlea pathways. There was no hair cells injury and changes of DPOAE through cerebrospinal fluid pathway. CONCLUSION: The study suggested the operations through round window and basal turn of cochlea could lead to bleeding in cochleas, which resulted in change of DPOAE amplitude. It is a safe and effective method to transfer through cerebrospinal fluid.

A triboluminescent europium(III) complex.

The crystal structure of (4,4'-dimethyl-2,2'-bipyridyl)tris[3,3,3-trifluoro-1-(2-thenoyl)propan-2-onato]europium(III), or more commonly (4,4'-dimethyl-2,2'-bipyridyl)tris(2-thenoyltrifluoroacetonato)europium(III), [Eu(C8H4F3O2S)3(C12H12N2)], has been determined. Crystals of the complex emit vivid red light when scratched or fractured. This triboluminescent activity seems to correlate with the non-centrosymmetric crystal structure and disorder of the thienyl rings and CF3 groups which is present here and in similar compounds. While modeling the thienyl-ring disorder, it was noted that the bond angle at the C atom replaced by S is a sensitive sign of even small rotational ring disorder. The coordination geometry of the Eu(III) ion can be described as square antiprismatic, with coordination by the six O atoms of the three chelating beta-diketonate ligands and the two N atoms of the neutral bipyridyl ligand.

How did the introduction of managed care for the uninsured in Iowa affect the use of substance abuse services?

Concerns about access under managed care have been raised for vulnerable populations such as publicly funded patients with substance abuse problems. To estimate the effects of the Iowa Managed Substance Abuse Care Plan (IMSACP) on substance abuse service use by publicly funded patients, service use before and after IMSACP was compared; adjustments were made for changes in population sociodemographic and clinical characteristics. Between fiscal years 1994 and 1997, patient case mix was marked by a higher burden of illness and the use of inpatient, residential nondetox, outpatient counseling, and assessment services declined, while use of intensive outpatient and residential detox services increased. Findings were similar among women, children, and homeless persons. Thus, care moved away from high-cost inpatient settings to less costly venues. Without knowing the impact on treatment outcomes, these changes cannot be interpreted as improved provider efficiency versus simply cost containment and profit maximization.
