Characterization of MVP and VPARP assembly into vault ribonucleoprotein complexes.

Vaults are barrel-shaped cytoplasmic ribonucleoprotein particles composed of three proteins: the major vault protein (MVP), the vault poly(ADP-ribose)polymerase (VPARP), and the telomerase-associated protein 1, together with one or more small untranslated RNAs. To date, little is known about the process of vault assembly or about the stability of vault components. In this study, we analyzed the biosynthesis of MVP and VPARP, and their half-lives within the vault particle in human ACHN renal carcinoma cells. Using an immunoprecipitation assay, we found that it took more than 4h for newly synthesized MVPs to be incorporated into vault particles but that biosynthesized VPARPs were completely incorporated into vaults within 1.5h. Once incorporated into the vault complex, both MVP and VPARP were very stable. Expression of human MVP alone in Escherichia coli resulted in the formation of particles that had a distinct vault morphology. The C-terminal region of VPARP that lacks poly(ADP-ribose)polymerase activity co-sedimented with MVP particles. This suggests that the activity of VPARP is not essential for interaction with MVP-self-assembled vault-like particles. In conclusion, our findings provide an insight into potential mechanisms of physiological vault assembly.

The effects of magnesium sulfate on fetal rats of FGR and the expression of caspase-3 in the placenta of maternal rat.

To investigate the effect of magnesium sulfate on the fetal rats of FGR and the expression of caspase-3 in the placenta of maternal rat; To explore the mechanism of using magnesium sulfate to cure the FGR. Establish model of FGR by a way of passive smoking: giving the maternal rats different agent of magnesium sulfate by subcutaneous injection: low agent group (300 mg/kg), high agent group (600 mg/kg). Concentration of magnesium sulfate was monitored. The expression of caspase-3 was measured by RT-PCR and immunohistochemistry technology. Both of the concentrations of magnesium sulfate in high and low agents group are higher than the FGR group (P < 0.01); the weight of the placenta and fetal rat in high agent group are higher than the FGR group (P < 0.05 and P < 0.01); the expression of mRNA and protein of caspase-3 in the two agent group is higher than the FGR group (P < 0.05 respectively); concentration of magnesium sulfate in the maternal rat blood correlate to the weight of fetal rat (r = 0.899, P = 0.038) and the expression of caspase-3 in the placenta of maternal rat (r = -0.747, P = 0.033; r = - 0.915, P = 0.001). The research suggests that the weight of fetal rat could be increased by treatment of magnesium sulfate. Because it would imfrmove the placental function by depressing the expression of caspase-3.

Disruption of murine Mus81 increases genomic instability and DNA damage sensitivity but does not promote tumorigenesis.

The Mus81-Eme1 endonuclease is implicated in the efficient rescue of broken replication forks in Saccharomyces cerevisiae and Schizosaccharomyces pombe. We have used gene targeting to study the function of the Mus81-Eme1 endonuclease in mammalian cells. Mus81-deficient mice develop normally and are fertile. Surprisingly, embryonic fibroblasts from Mus81(-/-) animals fail to proliferate in vitro. This proliferation defect can be rescued by expression of the papillomavirus E6 protein that promotes degradation of p53. When grown in culture, Mus81(-/-) cells have elevated levels of DNA damage, acquire chromosomal aberrations, and are hypersensitive to agents that generate DNA cross-links. In contrast to the situation in yeast, murine Mus81 is not required for replication restart following camptothecin treatment. Mus81(-/-) mice and cells are hypersensitive to DNA cross-linking agents. Cross-link-induced double-strand break formation is normal in Mus81(-/-) cells, but the resolution of repair intermediates is not. The persistence of Rad51 foci in Mus81(-/-) cells suggests that Mus81 acts at a late step in the repair of cross-link-induced lesions. Despite these defects, Mus81(-/-) mice do not show increased predisposition to lymphoma or any other malignancy in the first year of life.

[Establishing a parabiosis animal model of two-way paradigm].

OBJECTIVE: To establish a parabiosis model between allogenic conspecific adult mice to study two-way paradigm. METHODS: Fifty-four female Balb/c mice and 54 male C57BL/6 mice were paired and equally divided into 3 groups, namely group 1 with normal saline (NS) injection, group 2 with injections of spleen cells and cyclophosphamide (CP), and group 3 injected with spleen cells, CP, and cyclosporin A (CsA). The treatments were performed by injecting the spleen cells from one of the mice in a pair into the other via tail vein and vise versa, and two days after the operation, CP (150 mg/kg) was injected intraperitoneally. Intraperitoneal CsA (30 mg/kg daily) injection was given starting from 2 days before till 7 days after the operation. Twelve of the 18 pairs of parabiosis mice in each group were separated after 1 week, and part of the skin were transplanted to each other. The maintenance of parabiosis was observed in the other 6 pairs of parabiosis mice were observed. Mixed lymphocyte reaction (MLR) and delayed-type hypersensitivity (DTH) were observed and studied in the separated mice. RESULT: The duration of parabiosis maintenance and skin survival of the group 3 was significantly longer than those in the other two groups, and group 3 showed suppressed MLR and DTH. CONCLUSION: With the application of immunosuppressants, we have successfully established the two-way paradigm model in mice.

[Application of laminar air flow techniques in burn treatment].

OBJECTIVE: To evaluate the value of laminar flow in the treatment of burns. METHODS: The air in the laminar flow chamber and the wound tissues of the patients were sampled for bacterial detection. The number and stains of bacterial colony from different classes of laminar air flow chambers at different time points were inspected and compared. RESULTS: The bacterial number was 0 in the laminar flow chamber of 1000 grade, which was obviously different from that in the public area. The mortality was obviously decreased in the laminar air flow chamber with shorter treatment time and hospitalization. No wound infection occurred and the wounds healed smoothly in all these patients. CONCLUSION: The application of laminar air flow can be helpful for the treatment of severe burns.

[Studies on micromorphological characters of fruit coats in cultivated Fructus Trichosanthis in Shandong Province].

OBJECTIVE: To explore the affinity relation among land race of Trichosanthes kirilowii and to provide evidence tor the classification and authentication. METHOD: Using scanning electron microscope, characteristics of the pericarp surface were studied comparatively. RESULT: The pericarp surface of the cultivated Fructus Trichosanthis in Shandong showed the characteristics of generality and diversity. CONCLUSION: The results of the study could be used for the identification of the cultivated Fructus Trichosanthis in Shandong.

A critical role for p27kip1 gene dosage in a mouse model of prostate carcinogenesis.

In human prostate cancer, the frequent down-regulation of p27(kip1) protein expression is correlated with poor clinical outcome, yet p27(kip1) rarely undergoes mutational inactivation. Here, we investigate the consequences of reducing or eliminating p27(kip1) function for prostate carcinogenesis in the context of a mouse modeling lacking the Nkx3.1 homeobox gene and the Pten tumor suppressor. Unexpectedly, we find that triple mutant mice heterozygous for a p27(kip1) null allele (Nkx3.1(+/- or -/-); Pten(+/-); p27(+/-)) display enhanced prostate carcinogenesis, whereas mice that are homozygous null for p27(kip1) (Nkx3.1(+/- or -/-); Pten(+/-); p27(-/-)) show inhibition of cancer progression. Expression profiling reveals that Cyclin D1 is highly up-regulated in compound p27(kip1) heterozygotes, but is down-regulated in the compound p27(kip1) homozygous mutants. Using RNA interference in prostate cancer cell lines with distinct p27(kip1) gene doses, we show that prostate tumorigenicity depends on levels of p27(kip1) and that the consequences of p27(kip1) gene dosage can be attributed, in part, to altered levels of Cyclin D1. Our findings suggest that p27(kip1) possesses dosage-sensitive positive as well as negative modulatory roles in prostate cancer progression.

Difference in skeletal muscle function in males vs. females: role of estrogen receptor-beta.

Male skeletal muscles are generally faster and have higher maximum power output than female muscles. Conversely, during repeated contractions, female muscles are generally more fatigue resistant and recover faster. We studied the role of estrogen receptor-beta (ERbeta) in this gender difference by comparing contractile function of soleus (mainly slow-twitch) and extensor digitorum longus (fast-twitch) muscles isolated from ERbeta-deficient (ERbeta(-/-)) and wild-type mice of both sexes. Results showed generally shorter contraction and relaxation times in male compared with female muscles, and ERbeta deficiency had no effect on this. Fatigue (induced by repeated tetanic contractions) and recovery of female muscles were not affected by ERbeta deficiency. However, male ERbeta(-/-) muscles were slightly more fatigue resistant and produced higher forces during the recovery period than wild-type male muscles. In fact, female muscles and male ERbeta(-/-) muscles displayed markedly better recovery than male wild-type muscles. Gene screening of male soleus muscles showed 25 genes that were differently expressed in ERbeta(-/-) and wild-type mice. Five of these genes were selected for further analysis: muscle ankyrin repeat protein-2, muscle LIM protein, calsequestrin, parvalbumin, and aquaporin-1. Expression of these genes showed a similar general pattern: increased expression in male and decreased expression in female ERbeta(-/-) muscles. In conclusion, ERbeta deficiency results in increased performance during fatigue and recovery of male muscles, whereas female muscles are not affected. Improved contractile performance of male ERbeta(-/-) mouse muscles was associated with increased expression of mRNAs encoding important muscle proteins.

An expressive three-mode principal components model for gender recognition.

We present a three-mode expressive-feature model for recognizing gender (female, male) from point-light displays of walking people. Prototype female and male walkers are initially decomposed into a subspace of their three-mode components (posture, time, and gender). We then apply a weight factor to each point-light trajectory in the basis representation to enable adaptive, context-based gender estimations. The weight values are automatically learned from labeled training data. We present experiments using physical (actual) and perceived (from perceptual experiments) gender labels to train and test the system. Results with 40 walkers demonstrate greater than 90% recognition for both physically and perceptually labeled training examples. The approach has a greater flexibility over standard squared-error gender estimation to successfully adapt to different matching contexts.

[Expression of transient receptor potential channel 4 in striatum and hippocampus of rats is increased after focal cerebral ischemia].

This paper was designed in middle cerebral artery occlusion (MCAO) model of rats, to explore the role of transient receptor potential channel 4 (TRPC4) as Ca(2+) selective channel by detecting the changes of the expression of TRPC4 in different parts of cerebral tissues under the condition of focal cerebral ischemia. The rats were sacrificed after MCAO surviving time 6 h, 12 h, 1 d, 3 d. As determined by Western blot, the expressions of TRPC4 in striatum and hippocampus of 12 h, 1 d, 3 d groups were significant higher than that in the control group (P<0.05). Immunohistochemical staining showed that the TRPC4 immunoreactive substances were present in the membrane of neurons. Compared with the control group, immunostaining positive cells increased in hippocampus and striatum of cerebral ischemia groups. The TRPC4 immunostaining positive cells increased significantly in 1d-group and 3d-group (P<0.05). It suggests that as a Ca(2+) selective channel, the variance of the expression of TRPC4 may play a role in acute and delayed neuronal injury in focal cerebral ischemia.

[Detection of microsatellite instability in primary gastric cancer using fluorescence-labelled primers multiplex PCR].

To assess the role of microsatellite instability(MSI) on chromosome 19 in the carcinogenesis of gastric cancer. 26 microsatellite markers on chromosome 19 were amplified by Fluorescence-Labelled Primers Multiplex PCR in 44 paired gastric cancer tissues for MSI. The amplified fragments ran in PAGE gel by ABI 377 sequencer and the gel images were analyzed by GeneScan and Genotyper. Meanwhile, MSI of four of all microsatellite sites were also analyzed using PCR-SSLP-silver stain method. MSI was observed in 22 of 26(84.62%) microsatellite sites. D19S402 showed the highest frequency(22.73%). The total frequency of MSI was 8.3%. In 44 gastric cancer samples, 34(77.27%) cases showed MSI at one or more than one site. MSI, as a possible mechanism of carcinogenesis, might play an important role in human primary gastric cancer. MSI detections will become an integral part of the routine classification of all gastrointestinal tumors in the very near future.

The effect of low dose gamma irradiation on the differentiation and maturation of monocyte derived dendritic cells.

The effects of gamma-irradiation on the differentiation of peripheral blood monocytes (PBM) into monocyte derived dendritic cells (MDC), their maturation, and subsequent ability to present antigen to T cells was studied. Undifferentiated MDC were more sensitive to gamma-irradiation induced apoptosis than mature MDC. Irradiation of immature MDC with 5 Gy of gamma-rays down regulated the expression of the costimulatory receptors CD80/CD86 and may compromise their ability to capture and present antigen. By contrast, gamma-Irradiation of mature MDC did not affect the expression of CD86/CD80, and HLA-DR. Gamma-irradiation increased the apoptosis of MDC; but did not affect the ability of mMDC to stimulate autologous MLR. T cell proliferative response in the MLR and in response to tetanus antigen was reduced when gamma-irradiated primary DC1 were used to either stimulate or present antigen to T cells.

RNA interference inhibition of Mus81 reduces mitotic recombination in human cells.

Mus81 is a highly conserved endonuclease with homology to the XPF subunit of the XPF-ERCC1 complex. In yeast Mus81 associates with a second subunit, Eme1 or Mms4, which is essential for endonuclease activity in vitro and for in vivo function. Human Mus81 binds to a homolog of fission yeast Eme1 in vitro and in vivo. We show that recombinant Mus81-Eme1 cleaves replication forks, 3' flap substrates, and Holliday junctions in vitro. By use of differentially tagged versions of Mus81 and Eme1, we find that Mus81 associates with Mus81 and that Eme1 associates with Eme1. Thus, complexes containing two or more Mus81-Eme1 units could function to coordinate substrate cleavage in vivo. Down-regulation of Mus81 by RNA interference reduces mitotic recombination in human somatic cells. The recombination defect is rescued by expression of a bacterial Holliday junction resolvase. These data provide direct evidence for a role of Mus81-Eme1 in mitotic recombination in higher eukaryotes and support the hypothesis that Mus81-Eme1 resolves Holliday junctions in vivo.

A surface acoustic wave biosensor concept with low flow cell volumes for label-free detection.

Special surface acoustic wave (SAW) devices using horizontally polarized surface shear waves can be operated in water. They allow an easy detection of molecules with biological relevance (e.g., proteins) via direct detection of the adsorbed mass. The transducer structures of conventional SAW devices are usually connected to the electronics by bond wires. In consequence, flow cell volumes can hardly be designed smaller than 50 microL. A new type of SAW device that is coupled capacitively with the electronics enables the reduction of flow cell volumes down to 60 nL, which decreases sample consumption and reduces the length of the measurement cycles down to a few minutes. To create an immunosensor, the SAW devices first are coated with a thin parylene layer for creating a sensor surface that is chemically homogeneous. Then OptoDex, a dextran containing both photoactive and functional groups is immobilized photochemically. Finally, antibodies are coupled via conventional EDC/NHS chemistry. The technique has been used to monitor urease binding at anti-urease-coated SAW devices in real time and with good resolution. Because of the simple sensor handling and the economical sample use, the new SAW device is particularly suitable for the design of an array.

Specific regulation of lipocalin-type prostaglandin D synthase in mouse heart by estrogen receptor beta.

Estrogens have important physiological roles in the cardiovascular system. We use DNA microarray technology to study the molecular mechanism of estrogen action in the heart and to identify novel estrogen-regulated genes. In this investigation we identify genes that are regulated by chronic estrogen treatment of mouse heart. We present our detailed characterization of one of these genes, lipocalin-type prostaglandin D synthase (L-PGDS). Northern and Western blot analysis revealed that L-PGDS was induced both by acute and chronic estrogen treatment. Northern blot analysis, using estrogen receptor (ER)-disrupted mice, suggests that L-PGDS is specifically induced by ERbeta in vivo. In further support of ERbeta-selective regulation, we identify a functional estrogen-responsive element in the L-PGDS promoter, the activity of which is up-regulated by ERbeta, but not by ERalpha. We demonstrate that a one-nucleotide change (A to C) in the L-PGDS estrogen-responsive element affects receptor selectivity.

[Effects of Fränkel II appliance on sagittal dimensions of upper airway in children].

OBJECTIVE: The purpose of this study was to investigate the effects of Fränkel II appliance on the upper airway of children. METHODS: The subjects consisted of 20 patients with Angle Class II, division I malocclusion (10 males and 10 females). The ages of all cases were ranged in 9-10 years. Each experimental subject was also served as self-control and cephalometric analysis was conducted. RESULTS: Compared with the results of pretreatment, the distance of PNS-Ba, SPP-SPPW, P-T, V-LPW and the Mcnamara line increased remarkably. CONCLUSION: The Fränkel II appliance can improve the sagittal dimensions of the upper airway in children.
