Genomic instability of murine hepatocellular carcinomas with low and high metastatic capacities.

AIM: To investigate the frequency of genomic instability in murine hepatocellular carcinoma (HCC) cell lines Hca/A2-P(P) and Hca/163-F(F) with low and high metastatic capacity, and to explore its association with the occurrence and metastasis of hepatocellular carcinomas. METHODS: Forty microsatellite markers were randomly selected to examine P and F cells for genomic instability using PCR-simple sequence length polymorphism (PCR-SSLP) analysis. RESULTS: Allelic genes on the chromosomes of P cell line with thirty informative microsatellite loci were paralleled to those of inbred strain C(3)H mouse, while those of F cell line with 28 loci were paralleled to those of inbred strain C(3)H mice. The frequency of microsatellite alterations was 37.5% and 42.5% in P cell line and F cell line, respectively. There were different alterations of allelic band 9 at loci between P and F cells, among which, the frequency of microsatellite alterations was most commonly seen on chromosomes 3, 7, 11 and 16. CONCLUSION: Genomic instability in mouse chromosomes 3, 7, 11 and 16 may play a more important role in the development and progression of HCC in mice. It is suggested that these two sub-clones derived from a same hepatic tumor in homozygous mouse present different genetic features.

Methylmalonic acid in amniotic fluid and maternal urine as a marker for neural tube defects.

To evaluate the implication of methymalonic acid (MMA) in the early diagnosis of neural tube defects (NTD), a quantitative assay for MMA was established by using gas chromatography-mass spectrometry with stable isotope of MMA as an internal standard. Amniotic fluid and maternal urine MMA concentration, maternal serum folate, red blood cell folate and vitamin B12 levels were measured in the middle term of NTD-affected and normal pregnancies. Amniotic fluid and maternal urine MMA concentrations in the middle term of NTD affected pregnancies (1.4 +/- 0.9 micromol/L, and 22.1 +/- 12.6 nmol/micromol creatinine) were significantly higher than that of normal pregnancies (1.0 +/- 0. 4 micromol/L, and 2.5 +/- 1.1 nmol/micromol creatinine). There was no significant difference between normal and NTD pregnancies for serum folate, red blood cell folate and vitamin B12 levels. The results suggested that MMAs in amniotic fluid and maternal urine are sensitive markers for early diagnosis of NTD. Vitamin B12 is an active cofactor involved in the remethylation of homocycteine and its deficiency is an independent risk factor for NTD. MMA is a specific and sensitive marker for intracellular vitamin B12 deficiency. This study suggests that it is necessary to monitor the vitamin B12 deficiency and advocates vitamin B12 supplementation with folate prevention program.

Prolactin-releasing peptide affects pain, allodynia and autonomic reflexes through medullary mechanisms.

Prolactin-releasing peptide (PrRP) and neuropeptide FF (NPFF) are RF-amide peptides expressed in brain areas involved in pain modulation. NPFF displays multiple effects on acute, inflammatory and neuropathic pain. The potential role of PrRP in pain was addressed by intrathecal and intracerebral injections of PrRP on pain-related responses in both neuropathic and normal rats. Particularly in the dorsal medulla, PrRP produced significant antinociception in normal rats and an antiallodynic effect in neuropathic rats. To understand the basis of PrRP-induced pain modulation, distributions of PrRP, PrRP receptor, and NPFF were compared in the rat central nervous system. PrRP and NPFF mRNA were expressed in different parts of the nucleus of the solitary tract. In the medulla, PrRP receptor mRNA expression was abundant only in area postrema. Of the peptides studied, only NPFF mRNA was found in the dorsal horn of the spinal cord and spinal nucleus of the trigeminal nerve. PrRP-immunoreactivity corresponded to the mRNA distribution. Even if the neuronal groups producing NPFF and PrRP were distinct, the fiber networks immunoreactive for PrRP and NPFF overlapped. The results show that PrRP modulates nociception due to supraspinal rather than spinal action, and that its antinociceptive mechanism differs from that previously characterized for NPFF.

Clinical analysis of pericardial devascularization by preserving vagus trunks in 42 patients with portal hypertension.

Pericardial devascularization (PD) is less of a burden and provides better bleeding control to patients compared to shunt procedures, and so has been widely used in portal hypertension in China. However, because the vagus trunks are interrupted during surgery, patients easily suffer from postoperative stomach adynamia. Based on our understanding from autopsy of the path of vagus trunks along the distal oesophagus, we designed the operative procedure of PD by preserving vagus trunks (PDPVT) to treat portal hypertension. Between May 1991 and January 2003, patients with portal hypertension were treated surgically using PDPVT (n = 42), single PD (n = 32), or PD with pyloroplasty (PD+PP; n = 16). Operative mortality was 2.4% in the PDPVT group and 6.3% in both the PD and PD+PP groups (p > 0.05). The postoperative rebleeding rate was 9.5% in the PDPVT group and 12.5% in both the PD and PD+PP groups (p > 0.05). There were no differences in operative time and estimated blood loss between the three groups (p > 0.05). The recovery time for gastroenteric function was shorter with PDPVT (mean, 3.5 days) than with PD (mean, 5.7 days) and PD+PP (mean, 4.2 days; p < 0.02). Incidences of early satiety and enterogastric reflux were significantly lower in the PDPVT group (both 4.8%) than in the PD group (46.9% and 18.8%) and PD+PP group (12.5% and 100%; p < 0.005). Incidences of retention of gastric juice, diarrhoea and late gallstones were 12.5%, 15.6% and 7.1%, respectively, in the PD group, and 12.5%, 18.8% and 6.3%, respectively, in the PD+PP group, but none of these were observed in the PDPVT group. Since it preserves vagus trunks, PDPVT can maintain normal stomach dynamics and physiological function of hepatobiliary and gut systems better than PD and PD+PP, thus reducing incidences of postoperative complications. Thus, PDPVT is superior to PD and PD+PP in the treatment of portal hypertension.

Influence of IL-1 beta and TNF-alpha on Fas expression of human retinal pigment epithelial cells in vitro.

PURPOSE: To observe Fas expression change of cultured human retinal pigment epithelial (RPE) cells by IL-1 beta and TNF-alpha. METHODS: With flow cytometry, immunohischemistry, and color imaging system, Fas expressions by exposure to IL-1 beta and/or TNF-alpha were measured. RESULTS: The gray degree values of Fas expression were 67.5 +/- 6.1 in IL-1 beta + TNF-alpha-treated group, 80.1 +/- 9.2 in IL-1 beta-treated group, and 70.4 +/- 6.4 in TNF-alpha-treated group, respectively. There were significant differences (P < 0.005) compared with control group (107.0 +/- 10.2). Flow cytometry showed that 15.0% cultured human RPE cells expressed Fas. Fas-positive in IL-1 beta, TNF-alpha, and IL-1 beta + TNF-alpha-treated groups expressed was 28.1%, 34.5%, and 65.2%, respectively. CONCLUSION: IL-1 beta, TNF-alpha, and combining both of them can up-regulate Fas protein expression, which may contribute to more Fas (+) cells in proliferative vitreoretinopathy (PVR). Minimizing this process by means of inducing apoptosis of Fas (+) proliferative cells of Fas/FasL pathway is a future preventive and therapeutic possibility for PVR.

[The detection of nanobacteria infection in serum of healthy Chinese people].

OBJECTIVE: Nanobacteria, a new kind of bacteria found by a Finnish scholar, is considered to relate to many human diseases like nephrolithiasis. However, there are no data available on nanobacteria infection in Chinese people. METHODS: Nanobacteria was detected in serum of 336 cases of healthy adults in Southern China by enzyme-linked immunosorbent assay (ELISA) and immunohistochemistry stain. The monoclonal antibody of nanobacterum was supplied by Kuipio University of Finland. RESULTS: Nanobacteria infection rates were 27 (8.0%), 19 (5.7%) in the healthy adults by ELISA and immunohistochemistry stain respectively. No difference was shown between the 2 methods and between male and female, statistically. Age and sex did not seem to be related to the infectious risk of nanobacteria. However, the infectious rate was lower in those below 30-year-old than that of people over 60-year-old (P < 0.05). CONCLUSION: Nanobacteria was existed in the serum of Chinese healthy people with an infectious rate of 8.0%.

[Diagnosis by ultrasonography and CT scan in orbital vascular tumors and malformations].

OBJECTIVE: To analyze the value of ultrasonography (standardized A/B scan) and CT scan in the diagnosis of orbital vascular tumors and vascular malformations. METHODS: One hundred and eight cases of orbital vascular tumors and malformations were studied, including 98 cases examined by ultrasonography, 108 cases examined by CT scan and 94 cases studied by orbital operation and pathological examination. There were 54 cases of cavernous hemangioma, 26 cases of varix, 13 cases of venous hemangioma, 6 cases of lymphangioma, 5 cases of infantile hemangioma and 4 cases of others disorders. RESULTS: The tumor could be detected in ultrasonography in 96/98 cases (98%), the false negative rate of ultrasonography was 2% (2/98). The nature of the tumor could be determined by ultrasonography in 93% (91/98) cases pre-operatively. The tumor could be found by CT in all cases (108/108). The nature of the tumor could be determined by CT in 92% (99/108) cases pre-operatively. CONCLUSIONS: Combined ultrasonography with CT scan can estimate the nature, extent and location of orbital vascular tumors and malformations and can be useful for selection of the route of orbital operation.

[Reductive dechlorination of atrazine by Ni/Fe bimetallic particles].

Dechlorination of atrazine under acidic conditions was studied by using Fe and Ni/Fe bimetallic particles respectively for the confirmation of the catalytic role of Ni/Fe. Effect of pH value, Ni/Fe proportion and dosage of metal powder were discussed as well. The results indicated that Ni/Fe bimetallic particles was very effective for dechlorination of atrazine, with the dechlorination efficiencies above 90% by using 1.22% (W/W) Ni/Fe at pH value of 2.0 in 30 minutes, very superior to Fe particle with the dechlorination efficiencies only 22.21% in 90 minutes under the same conditions. By the comparison of the surface morphology of both Fe and Ni/Fe and experimental results, catalytic mechanism of Ni/Fe bimetallic particles was also proposed.

[Studies on the infectivity by porcine endogenous retrovirus with porcine skin fibroblast in vitro and in vivo].

To understand the infectivity by porcine endogenous retrovirus with porcine skin fibroblast cell in vitro and in vivo, porcine skin fibroblast cell established by our laboratory were co-cultured with neo/HEK293 cell for the infection of PERV in vitro, and were subcutaneously transplantated to SCID (severe combined immuno-deficiency) mice for the infection of PERV in vivo, laying the foundation for valuation of biologic safety of xenotransplantation. The event of neo/HEK293 cells infected by PERV occurred during co-culture of porcine skin fibroblast cells with neo/HEK293 cells, expanding the rang of the infection of porcine endogenous retrovirus. After pig cells transplantated subcutaneously in SCID mice, the microchimerism (78.57%) of pig cells occurred widely, and there was phenomena of integration of PERV provirus (85.71%) in several organs or tissues remote from the injected sites, indicating infection of PERV in SCID mice in vivo, yet, there is no evidence of active viral replication in analysis of PERV env RNA of these tissues or organs. The infectivity of porcine endogenous retrovirus with porcine skin fibroblast cells in vitro and in vivo were demonstrated in this study, and there is no evidence of active viral replication of PERV in vivo. It is an important event of biologic safety which attention must be paid to potential hazard of infectious interspecies transmission of PERV during xenotransplantation.

[Assessment of total body water of patients on hemodialysis with urea kinetic model].

OBJECTIVE: To propose a method to evaluate the total body water (TBW) of patients on hemodialysis with urea kinetic model (UKM), and compare it with body surface bio-impedance spectrum (BIS) analysis. METHODS: We enrolled 24 adult patients with end stage renal disease (ESRD) without hyper-catabolism in our dialysis center. All of them had been on hemodialysis for more than 3 months. TBW was measured with BIS analysis immediately before and after dialysis session, and one hour after hemodialysis session. Spent dialysate was collected; blood samples were taken before and one hour after hemodialysis session, TBW before hemodialysis session were calculated by UKM. RESULTS: Patients were 6 men and 18 women, the average age was (51.2+/-13.5) years and the average time on dialysis was (33.2+/-36.7) months. Causes of ESRD included chronic glomerulonephritis (8 patients), diabetic nephropathy (1 patients), hypertensive renal damage (1 patients), interstitial nephritis(two patients), chronic pyelonephritis (two patients). The average ultrafiltration volume was (2.7+/-1.0) L (0.5-4.4 L) . Plasma urea concentrations were (23.06+/-5.76) mmol/L and (8.15+/-2.06) mmol/L before and one hour after hemodialysis session, respectively. There was no significant difference between TBW measured immediately after and one hour after hemodialysis session with BIS analysis [(29.9+/-8.8) L and (29.8+/-8.6) L, respectively; average difference was (0.1+/-0.9)L, P=0.70]. These two measurements correlated very well (Pearson r=0.99, P<0.05). There was no significant difference between UKM and BIS analysis [(31.4+/-7.3) L and (31.3+/-8.6) L, respectively; average difference was (0.2+/-4.5) L, P=0.87]. Results of these two methods correlated very well (Pearson r=0.86, r(2)=0.73,P< 0.05). CONCLUSION: TBW measured with UKM in patients on hemodialysis provides theoretic basis for fluid status evaluation and dry body weight assessment.

[mtDNA mutations in mouse tumors].

OBJECTIVE: To investigate variations of mtDNA in mouse tumors and to explore the relationship between mtDNA mutations and murine carcinogenesis. METHODS: Variations of D-loop, ND3 and tRNAIle + Glu + Met gene fragments of mtDNA from six mouse tumor cell lines were analyzed by PCR-RFLP and PCR-SSCP techniques. RESULTS: ND3 and tRNAIle + Glu + Met gene fragments of mtDNA from the tumors showed no variations at 27 endonuclease sites. The D-loop of mtDNA from Hca-F demonstrated an additional endonuclease site of Hinf I in contrast to the inbred mouse. Upon PCR-SSCP analysis, the D-loop of mtDNA was found to possess mutations in 4 of 6 tumors. CONCLUSION: D-loop appears to be the hot spot for tumor mtDNA mutations, which may contribute to the carcinogenesis of murine tumors.

[Clinical study of subcutaneous administration of gonadotropin-releasing hormone agonist every six weeks in girls with idiopathic central precocious puberty].

OBJECTIVE: Gonadotropin-releasing hormone agonist (GnRHa) is widely used for the treatment of precocious puberty, but there was no identical regime for the dosage and the interval of administration. The aim of this study was to assess the feasibility of subcutaneous administration of Triptorelin with a prolonged interval (six weeks) for the suppression of pituitary-gonadal axis and clinical signs in girls with idiopathic central precocious puberty (ICPP). METHODS: Forty-six girls with ICPP were enrolled in this trial and were divided into two groups at random, with 26 patients in group A and 20 in group B. There were no significantly differences in chronological age, height, weight, age at clinical onset of puberty and bone age between the two groups before treatment. Triptorelin (Decapeptyl, 3.75 mg) was administered subcutaneously (SC) at 6 weeks intervals in group A or intramuscularly (IM) at 4 weeks intervals in group B. Both forms were given for more than 12 months consecutively. During the period of treatment, the clinical parameters including height, weight, pubertal stage, bone age, uterine volume and ovarian size were documented. The serum levels of luteinizing hormone (LH), follicle stimulating hormone (FSH) and estradiol (E(2)) were monitored using immunoenzymometric assay. RESULTS: Both treatment regimes can completely suppressed the pituitary-gonadal axis. The clinical signs of pubertal development regressed or stabilized in all patients. Breast developments, as reflected by Tanner staging, were regressed. Uterine volume decreased after treatment, however, this decrease did not reach statistical significance. Mean ovarian volume did not change significantly during treatment. The height velocity was significantly decreased from a pretreatment value of 6.3 +/- 1.4 cm/year to 5.8 +/- 1.2 cm/year in group A and 6.7 +/- 1.3 cm/year to 5.4 +/- 1.0 cm/year in group B, respectively. The rate of skeletal maturation was reduced significantly during treatment. The ratio of bone age/chronological age was 1.2 +/- 0.2 or 1.3 +/- 0.3 at the onset of therapy and decreased significantly after the treatment to 0.7 +/- 0.2 or 0.9 +/- 0.1, respectively. The predicted adult height increased significantly and progressively during therapy. The levels of serum LH, FSH and E(2) were returned to the prepubertal condition after 6 - 8 weeks treatment and remained suppressed for the duration of treatment. No significantly side effects of therapy were noted. The most common adverse event during SC treatment was that a non-irritating, 1 cm in diameter mass can be palpated at the site of subcutaneous injection in the abdominal wall of group A patients. It gradually decreased in size, and disappeared after 8 - 12 weeks. Two girls had minimal withdrawal virginal bleeding episodes after the first injection. CONCLUSION: Both IM and SC triptorelin administration were clinically effective. They can induce profound suppression of hypothalamic-pituitary-gonadal axis while stabilizing height velocity, slowing bone maturation and increasing predicted adult height. These results suggest that subcutaneous injection of triptorelin in 6 weeks intervals at a dosage of 3.75 mg was a safe and acceptable regime for ICPP with good efficacy.

[Analysis of presence and difference of sequence of porcine endogenous retrovirus in DNA genomes from peripheral blood white cells of pig and mRNA in tissues from mini-pigs].

The aim is to investigate biological features of porcine endogenous retrovirus (PERV) of pigs in China and to provide basic parameters for evaluation of biological safety of xenotransplantation from pig to human. In this study, basic biologic features of PERV of DNA of peripheral blood white cells of 12 species of domestic pigs in China were examined by polymerase chain reaction; Analysis of difference of PERV gene sequence was investigated by sliver-stained single stranded conformational polymorphism and restriction fragment length polymorphism; Difference of PERV mRNA from a variety of tissue of two species of mini-pig was analysized by reverse transcriptase-PCR and semi-quantitative analysis. These results showed that Sequences of PERV-A, -B genes are ubiquitous in DNA genome of peripheral blood white cells in the examined pigs; There was no single stranded conformational polymorphism in PERV-A, -B gene sequences. There is the phenomenon of restriction fragment length polymorphism of sequence of PERV env gene in DNA genome of peripheral blood white cells in some pigs of the examined pigs. The PERV-A, -B and -C are expressive in a variety of tissues from two species of mini-pig, higher in kidneys, lymph nodes and livers, lower in pancreas and brains. It is deserved that the phenomenon of restriction fragment length polymorphism of PERV and the absence and mismatch of base of sequence of PERV-A may induce potential hazard of infection of PERV during xenotransplantation.

[Construction of the plant expression vector with hepatitis a capsid protein fusion gene and genetic transformation of Citrus. Sinensis Osbeck].

The use of edible plants for the production and delivery of vaccine proteins could provide an economical alternative to fermentation systems. The construction of the plant expression vector pBI121-A was reported, which contained a fusion gene encoding hepatitis A capsid proteins. The gene was located between the left and right Ti border sequences under the control of CaMV35S promoter. The vector was identified via PCR and restriction enzyme analysis and was introduced into Agrobacterium tumerifacience LBA4404. The transgenic Citrus plants were produced by Agrobacterium-mediated transformation of epicotyl segments.13 putatively transformed plants through the kanamycin selection were micrografted onto the seedlings. The presence and integration of the transgene had been verified by PCR analysis. The result showed that five transformants were integrated and the transformation efficiency was 4.1%.

Rapid treatment of atrazine-contaminated water by nickel/iron bimetallic system.

The utility of nickel/iron in the remediation of atrazine-contaminated water was investigated. The experimental results showed that nickel/iron had effective catalytic activity in dechlorinating atrazine under acidic conditions. The dechlorination reaction approximately followed the first-order kinetics under the experimental conditions (nickel/iron: 1.0 g/250 ml; C(atrazine) = 20.0 mg/L), the reaction rate increased with decreasing pH value of the reaction solution and increasing the proportion of Ni : Fe within 2.95%. For condition with 2.95% nickel/iron, the reaction rate constants were 0.07518 (R = 0.9927), 0.06212 (R = 0.9846) and 0.00131 min(-1) (R = 0.9565) at pH = 2.0, 3.0 and 4.0, respectively. HPLC analysis was used to monitor the decline of atrazine concentration.

Pyridoacridine alkaloids inducing neuronal differentiation in a neuroblastoma cell line, from marine sponge Biemna fortis.

A new and three known pyridoacridine alkaloids were isolated from the Indonesian marine sponge Biemna fortis as neuronal differentiation inducers against a murine neuroblastoma cell line, Neuro 2A. The chemical structure of the new compound, labuanine A (1), was determined by spectroscopic study and chemical conversion. These pyridoacridine alkaloids induced multipolar neuritogenesis in more than 50% of cells at 0.03-3 micro M concentration. Compound 3, which showed the strongest neuritogenic activity among them, also induced increase of acetylcholinesterase, a neuronal marker in Neuro 2A and arrested cell cycle at the G2/M phase.
