cTrans: generating polypeptide databases from cDNA sequences.

cTrans is a comprehensive utility used to generate polypeptide databases from cDNA sequences. The goal is achieved through integrating four main functions, including retrieving sequences of species of interest from the downloaded packages from dbEST of GenBank, format conversion, checking and deleting vector and adaptor contamination, and translating the cDNA sequences in all six frames and selecting specific translations for database construction in a user-defined length threshold. In addition, this utility is also applicable to cDNA sequences produced by users themselves.

Differential proteomic analysis of proteins in wheat spikes induced by Fusarium graminearum.

Scab, caused by Fusarium graminearum, is a serious spike disease in wheat. To identify proteins in resistant wheat cultivar Wangshuibai induced by F. graminearum infection, proteins extracted from spikes 6, 12 and 24 h after inoculation were separated by 2-DE. Thirty protein spots showing 3-fold change in abundance when compared with treatment without inoculation were characterized by MALDI-TOF MS and matched to proteins by querying the mass spectra in protein databases or the Triticeae EST translation database. Based on their volume profiles, these proteins were classified into four categories. The first one fell off rapidly at the initial inoculation and then rose at 12 or 24 hai, the second one decreased considerably after inoculation and remained at low level, the third one rose at the initial inoculation and then declined at 12 or 24 hai, the forth one showed steady increase after inoculation and maintained at a high level. Many of the proteins identified in the first two categories are related to carbon metabolism and photosynthesis. While most of proteins identified in the last two categories are related to stress defense of plants, indicating that proteins associated with the defense reactions were activated or translated shortly after inoculation.

[Investigation of microcystin pollution on Dong Lake, Wuhan City].

OBJECTIVE: To investigate the level of pollution of microcystin and its changing in freshwater lake and fish. METHODS: Samples of freshwater and fish were collected from Dong Lake in Wuhan City. Samples were tested using ELISA. RESULTS: All freshwater samples collected in July and October contained blue-green algae and it had become the dominant species. For the same waterblooming lake, MC content was significantly lower in July than that in October (P < 0.05). MC content in fish muscle was significantly lower than that in liver (P < 0.05).

[Microcystin contamination of fish on Poyang Lake in Jiangxi province].

In order to investigate the level of microcystin contamination of fish and its dynamic change in fish body, samples of freshwater as well as fish were collected from Poyang lake in Jiangxi province in July and October in 2000 respectively. Microcystin were tested for all samples via ELISA method. The results showed that: all freshwater samples collected in July and October contained blue-green alga and it has become a dominant alga. Microsystin content was higher in freshwater collected in October than that of in July. The average content level of microcystin in freshwater of river bank and Yongxiu river was 888.8 ng/ml, being ten times higher than that of water of Bang lake 1, Bang lake 2 and Big lake pool (83.6 pg/ml). The highest level of microcystin was 1036.9 pg/ml in freshwater of Yongxiu river. Microcystin could be detected in all samples of fish muscle and fish liver. The level of microcystin in muscle of fish collected in October was 1.2 to 2.0 times higher of that collected in July, and microcystin in liver of fish collected in October was 2.0 to 20 times higher of that collected in July. The average level of microcystin in fish liver in river bank and Yongxiu river was 27.2 ng/g, being significantly higher than that of in Bang lake1, Bang lake 2 and Big lake pool (2.8 ng/g).

[Investigation of concentration of microcystin in raw materials and finished products of spires health food].

In order to investigate the concentration of micromystin in raw materials and finished products of spires health food, we collected 33 water samples from water resources, 160 water samples from culturing pool, 86 samples of Spires slurry and 70 samples of powders of Spires raw materials from 7 Spires manufacturers in Jiangsu, Yunnan, Fujian and Guandong provinces which are the main spires producing areas from July to August 2002. At the same time, we also collected 71 samples of 19 finished products of Spires health foods in markets of above-mentioned 4 provinces. Microcystin were tested for all samples by ELISA. The results show: microcystin were not detected in 12 tap water samples, were detected in 6 of 9 under water samples, 8 of 12 surface water samples; microcystin concentration of water samples from culturing pool was 207.9 pg/ml and Spires slurry was 31.9 ng/g, the former is 153 times as the latter; powder of Spires raw material was 206.4 ng/g. The average concentration of microcystin of all Spires health food samples from market was 317.2 ng/g, there into, microcystin was 142.7 ng/g in a tables and 222.6 ng/g in capsule, respectively. The results suggested: we shouldn't ignore the risk of exposuring microcystin when consuming Spires health food, it is necessary to further study and set a standard of limited microcystin in Spires health food.

[Toxicity of microcystin and its bioaccumulation effect].

Microcystins are cyclic heptapeptide hepatotoxins from certain cyanobacteria. It can inhibit hepatic protein phosphatase 1 and 2A activities, break the balance between protein phosphorylation and phosphorylysis, causing liver damage even necrocytosis. Furthermore, it is a strong tumor promoter. Yet, its hazard received little study. This paper presents a latest review of toxicity and bioaccumulation of Microcystins.
