[Effects of lotensin and nitrendipine on plasma fibrinogen and platelet aggregation in hypertensive patients].

Plasma fibrinogen and platelet aggregation were measured by turbidimetric immunoassay, turbidimetry in 47 hypertensive patients and 20 normotensive control subjects. Among the 47 hypertensives, 24 cases were received lotensin and 23 nitrendipine. The plasma fibrinogen was increased and platelet aggregation enhanced in hypertensive patients before treatment. Platelet aggregation decreased after 8 weeks of treatment with lotensin or nitrendipine respectively. Lotensin decreased plasma fibrinogen whereas nitrendipine did not. It was concluded that both lotensin and nitrendipine decreased platelet aggregation; lotensin decreased plasma fibrinogen but nitrendipine did not.

[Relationship between sex hormone levels and blood lipids/immunity in perimenopausal women].

To investigate the relationship between sex hormone levels and blood lipids/immunity and to evaluate the therapeutical effects of nylestriol, 96 women without coronary heart disease(CHD) were studied during their perimenopausal period. The estimation of serum biochemical components included serum 17 beta-estradiol(E2), testosterone(T), total cholesterol(TC), triglyceride (TC), high density lipoprotein cholesterol(HDL-C), low density protein cholesterol(LDL-C), apolipoprotein A-I(ApoAI), apolipoprotein B(ApoB), lipoprotein (a)[Lp(a)], immunoglobulin G(IgG), and IgG antibody against cardiolipin(ACAIgG). Thirty-six postmenopausal volunteers were divided into two groups and randomized to treat with either 2 mg nylestriol or placebo. In postmenopausal women, serum levels of E2, E2/T, HDL, and ApoAI decreased, while those of T, TC, TG, LDL, ApoB, Lp(a), IgG, and ACAIgG increased. Serum level of E2 was positively correlated to HDL-C and negatively correlated to TC, ApoB, LDL, IgG, and ACAIgG. Serum level of T was positively correlated to LDL, IgG, and ACAIgG and negatively correlated to HDL. In the nylestriol group, as compared with the results before treatment, serum levels of TG, TC, LDL, IgG, and ACAIgG decreased and that of HDL-C increased after treatment. We conclude that estradiol is a protective factor of CHD, whereas testerone is a dangerous factor. After menopause, the imbalance of the estradiol/testosterone ratio increases the incidence of CHD. Nylestriol is an effective substitute for estradiol to prevent CHD in post menopausal women.

[Changes of plasma endothelin-1 in patients with congestive heart failure and the influence of metoprolol].

To investigate the alterations of plasma endothelin-1(ET-1) in patients with congestive heart failure(CHF) and the effects of metoprolol on it, plasma ET-1 and norepinephrine(NE) were measured in 43 patients using radioimmunoassay and high-performance liquid chromatography methods. Twenty-four patients were treated with metoprolol plus the routine therapy while the others were received the routine therapy only. The findings were that levels of plasma ET-1 and NE increased before the treatment, and decreased after the treatment with metoprolol for 1 month. There was no alteration of plasma ET-1 or NE in the control group.

[Study on telomeric association in nasopharyngeal carcinoma].

The chromosomal telomeric association in peripheral blood lymphocytes from 25 untreated patients with nasopharyngeal carcinoma (NPC) and in NPC cell lines (CNE) was investigated by using chromosome G banding technique. The cell telomeric association rate of patients (35.07%) was found to be higher than that of controls(20.27%) (P < 0.01), while in the two groups the distributions of chromosomes involved were approximately the same (P > 0.05). The telomeric association rates of patients were significantly higher than those of control in chromosomes 1, 2, 3, 4, 7, 11, 15, and 17(P < 0.05). In NPC lines (CNE), 94% of the cell division phases showed telomeric association. Chromosomes 2, 3, 5, 11 and 16 had higher frequencies of association than other chromosomes. The most frequent telomeric association was single chromatid association.

[Expression of inducible nitric oxide synthase in atherosclerotic rabbits].

Atherosclerotic lesions contain vascular smooth cells and thus may have an increased capacity for generation of nitric oxide (NO) by inducible NO synthase (iNOS). Rabbits (n = 8) were raised for eight weeks by the atherogenic diet, then the aortic rings were captured. Contractive responsiveness to norepinephrine, changes of CGMP, expression of iNOS of aortic rings were examined in vitro. There was a substantial decrease in contractile responsiveness to norepinephrine (NE) (P < 0.05), a decrease of CGMP (P < 0.01) and an increase of expression of iNOS (P < 0.05), after L-arginine or sodium nitroprusside treatment. The contractile responsiveness to NE was attenuated (P < 0.01). The CGMP was increased ((P < 0.01). After NG-nitric arginine (L-NNA) treatment, those reaction were reversed. It implied that L-arginine/NO pathway obstructure might likely be another mechanism of atherosclerosis and atherosclerotic arteries and had an increased capacity for generation of NO by iNOS.

Opening blood-brain-barrier by intracarotid infusion of papaverine in treatment of malignant cerebral glioma.

OBJECTIVE: To find an effective method for the treatment of malignant cerebral glioma by opening the blood-brain-barrier (BBB). METHODS: The concentration of methotrexate (MTX) in tumor was measured in 18 patients with cerebral glioma, and 155 patients with malignant glioma were treated by intracarotid infusion of papaverine and Bis-chloronitrosourea (BCNU). RESULTS: After intracarotid infusion of papaverine for reversible opening of BBB, the MTX concentration in tumor tissue was 1810 +/- 380 ng/g, higher than that in patients without papaverine infusion (997 +/- 126 ng/g). Eighty-nine patients with glioblastoma and 66 patients with anaplastic astrocytoma received intracarotid infusion of papaverine and BCNU (250 mg) for an average of 2.14 times. The mean survival time of these patients was 114.5 weeks, median survival time was 140 weeks, and five-year survival rate was 22.97%. CONCLUSION: The combined chemotherapy with papaverine and BCNU through intra-arterial infusion is simple, safe and effective in treating malignant cerebral glioma so long as the tumor is sensitive to the drugs.

[Determination of clozapine and its N-demethylated metabolite in serum by gas chromatography-mass spectrometry with selected ion monitoring].

This article reports a highly sensitive method specific for determination of clozapine(CLP) and its N-demethylated metabolite (DCLP) in serum by gas chromatography-mass spectrometry with selected ion monitoring. A 0.5 mL serum was alkalized with 0.5 mL 1.0 mol/L sodium hydroxide and was extracted with 5 mL ether, then the organic phase was transferred to another tube containing 1 mL 0.1 mol/L hydrochloric acid. After being shaken and centrifuged, the organic phase was discarded, and 10 microL 100 micrograms/L diazepam (DAP) was added to the water phase as internal standard, then 0.5 mL 1.0 mol/L sodium hydroxide and 5 mL ether were added. The extract was evaporated to dryness and then was acylated with 50 microL trifluoroacetic anhydride under 50 degrees C for 45 min. The excess of the reagent was evaporated and the residue was disolved in 20 microL acetone. Take 2 microL for gas chromatography-mass spectrometry analysis. Chromatographic conditions were: column DB-1 (30 m x 0.25 mm x 0.25 microns), injector 260 degrees C, interface 250 degrees C, column oven with temperature program: [equation: see text] Helium carrier gas flow rate was 1.8 mL/min. The mass spectrometer was adjustd to record the ion m/z352 for CLP and DCLP, and m/z 256 for DAP in quantitation. The minimum detection limits were 0.1 microgram/L for CLP and 0.2 microgram/L for DCLP. The standard curves of CLP and DCLP were linear between 1-128 micrograms/L (rCLP = 0.998, rDCLP = 0.999). Recoveries of two substances were better than 83%, and the RSD values were less than 10%. This method has been successfully used in clinical pharmacokinetic study of CLP before and after CYP1A2 inhibitor used. The results indicate that the metabolism of clozapine is strongly affected by CYP1A2 activity.

[Electrophoretic analysis of protein composition in gingival crevicular fluid].

OBJECTIVE: To seek the protein composition related to periodontal diseases. METHODS: 86 gingival crevicular fluid (GCF) samples were collected by rinsing the sulgingival sulci from subjects with healthy gingiva (20), gingivitis (24) and periodontitis (42). The protein composition were examined by sodium dodecyl sulphate polyacrylamide gel electrophoresis (SDS-PAGE) on a Phast System. The gels were scanned and analyzed using Phast Image with computer. RESULTS: In inflammatory condition, the serum derived proteins with MW 82,000, 77,000, 17,000 occurred more frequently than in healthy gingiva, and the occurrence and abundance of non-serum derived proteins with MW 41,000, 21,000, 12,000 were also increased in inflammatory condition compared with healthy gingiva. CONCLUSION: The high levels of proteins with MW 82,000, 41,000, 21,000, 12,000 are close related to the periodontal inflammatory status.

[The role of nitric oxide in hyperdynamic circulation in portal hypertensive rats].

OBJECTIVE: To evaluate the role of nitric oxide (NO) in hyper-hemodynamics in portal hypertension, we investigated the effects of nitric oxide synthase inhibitor L-NMMA with or without L-arginine, the substrate for enzyme NO synthase on the systemic and splanchnic hemodynamics in portal hypertensive rats. METHOD: Ninety-seven male Sprague-Dawley rats were divided into four groups: intrahepatic portal hypertension (IHPH, n = 23) by injection of CCl4, prehepatic portal hypertension (PHPH, n = 26) by stenosis of the portal vein, end-to-side portacaval shunt (PCS, n = 23), and sham-operated controls (SO, n = 25). Animals of each group were divided into three subgroups: systemic administration of L-NMMA (50 mg/kg BW, 1 ml), L-NMMA (50 mg/kg BW, 0.5 ml) plus L-arginine (30 mg/kg BW, 0.5 ml), and normal saline solution (1 ml). The radioactive microsphere method was used for hemodynamic study. RESULT: There was the characteristic of hyperdynamic circulatory state including increased cardiac output and splanchnic blood flow, decreased mean arterial blood pressure, total peripheral vascular resistance and splanchnic vascular resistance in IHPH, PHPH and PCS rats. The hyperdynamic circulatory state in IHPH, PHPH and PCS rats could be effectively reversed by L-NMMA to the resting values of hemodynamics in SO rats, but not the levels after administration of L-NMMA in SO rats, indicating that there is excessive production of NO in portal hypertensive and PCS rats. Administration of L-arginine counterbalanced the effects of L-NMMA on hyperdynamic circulatory state in portal hypertensive and PCS rats. CONCLUSION: NO is an important mediator of hyperdynamic circulation in portal hypertension.

[Nitric oxide in pathogenesis of cirrhotic portal hypertension in rats and its hemodynamic effects].

OBJECTIVE: To measure serum nitrite and nitrate levels as an index for in vivo nitric oxide generation in rats at different stages during the development of cirrhosis with portal hypertension. METHOD: Plasma endotoxin and hemodynamic parameters were measured. The effects of NO-monomethyl-L-arginine (L-NMMA), an inhibitor of NO biosynthesis were investigated in rats with cirrhosis. RESULT: Significant increase was noted in serum nitrite/nitrate and plasma endotoxin compared with controls at each stage of cirrhosis. At each stage, the mean arterial pressure (MAP) and splanchnic vascular resistance (SVR) were significantly lower than those of controls, whereas portal venous pressure (PVP) and portal vascular resistance (PVR) significantly increased and portal venous flow (PVF) was not markedly different from that of the controls. In addition, the serum nitrite/nitrate levels significantly were correlated with endotoxemia and PVP. Intravenous bolus administration of L-NMMA (25 mg/kg) significantly increased MAP, PVR and SVR in cirrhotic rats. L-NMMA also significantly decreased PVF but did not alter PVP and IVCP. CONCLUSION: The excessive synthesis and release of NO plays a key role in the induction of portal hypertension.

[Immunohistochemical studies on wholemounts of the cornea and iris-ciliary body after corneal transplantation].

OBJECTIVE: To study the phenotype of the cells involved in the pathogenesis of allograft rejection after corneal transplantation. METHODS: Wholemounts of the cornea and iris-ciliary body were isolated and prepared from normal rats and those after penetrating corneal transplantation. Immunohistochemical stainings were carried out on these wholemounts using monoclonal antibodies to CD3 (T cell), CD4 [T-helper (Th) cell], CD8 [T-suppressor (Ts) cell], macrophages, dendritic cells, B lymphocytes, major histocompatibility complex (MHC) class II antigen and transforming growth factor-beta (TGF-beta). RESULTS: All the cells mentioned above except B lymphocytes were positive and present in a small number in the peripheral cornea and limbus of normal rats. A massive influx of CD3, CD4, CD8 positive cells, macrophages, dendritic cells, MHC class II and TGF-beta positive cells not only into cornea but also into the iris was observed 7 days after allograft corneal transplantation. Whereas no B cell was found in the allograft. Such a cellular response was not found in the rats in which corneal transplantation with autologous cornea was performed. CONCLUSION: T cells, Th cells, Ts cells, macrophages, dendritic cells, MHC class II positive cells and TGF-beta positive cells are involved in the pathogenesis of corneal allograft transplantation. The involvement of the iris and ciliary body may promote or aggravate the allograft rejection.

[Determination of oligopeptide by IR matrix-assisted laser desorption ionization mass spectrometry].

Luteinizing releasing hormone analog (LRH-A), renin substrate tetradecapeptide and angiotensin I were analyzed with infrared-matrix assisted laser desorption ionization/Fourier transform ion cyclotron resonance mass spectrometry (IR-MALDI/FTICR MS). 2,5-Dihydroxybenzoic (DHB) and succinic acid (SA) were used as matrices. The m/z of oligopeptides were determined with an error less than 4 x 10(-6). A series of characteristic fragment ions of peptides were obtained by increasing the laser power density appropriately. Their elemental compositions and the first order structures were confirmed. A sensitive, rapid and accurate method was established for the determination of oligopeptide.

[The mechanisms of antioxidant against inflammatory lesion in animal oral mucosa. II. A study of glutathione peroxidase].

In order to understand the mechanisms of superoxide dismutase (SOD) against inflammatory lesion in hamster cheek pouch, glutathione peroxidase (GPx) activity in the cheek pouch mucosa was assayed in different time. The efficacy of GPx scavenging lipid peroxide was also measured. In the SOD group GPx activity decreased significantly in the early stage (8 h). But the level of lipid peroxide in the SOD group not increase at the same time. The results of the model group were contrary. It was implied that exigenous SOD scavenged lipid peroxide in the oral mucosa sufficiently in stead of GPx.

[Detection of HFRSV in Eulaelaps shanghaiensis and Ornithonyssus bacoti by using in situ hybridization].

AIM: To provide molecular biological evidence of transmission of hemorrhogic fever with renal syndrome virus (HFRSV) by gamasid mites, Eulaelaps shanghaiensis and Ornithonyssus bacoti. METHODS: Frozen sections of the gamasid mites 10 days and more than 100 days after biting suckling mice inoculated with HFRSV were in situ hybridized with dig-labelled HFRSV cDNA probes. RESULTS: RNA was detected in frozen sections of Eulaelaps shanghaiensis, after biting suckling mice inoculated with Hantaan (76-118) and Seoul (UR) virus, respectively. Most of the fine granules of the virus RNA were located in the nuclei, cytoplasm and nuclear membrane of cells of brain cortex, caeca and genitalia of the mites. In situ hybridization results showed that 17 of 31 mites in Hantaan group and 10 of 23 mites in Seoul group were positive. The virus RNA was still detected in tissues of the mites on d132 for Hantaan group and on d102 for Seoul group after infection, respectively. Among 20 Ornithonyssus bacoti detected 12 were positive on d10 after biting suckling mice inoculated with Hantaan virus, and the virus RNA was mainly found in the cells of genitalia (Figs. 1-13). CONCLUSION: Both Eulaelaps shanghaiensis and Ornithonyssus bacoti could be infected with HFRSV by biting HFRSV-positive mice. E. shanghaiensis could be infected with both Hantaan and Seoul virus, and the two types of HFRSV were found to be maintained in the mites for 132 days and 102 days, respectively. These confirmed that E. shanghaiensis and O. bacoti are suitable vectors and reservoirs of both Hantaan and Seoul virus and might play an important role in the cross transmission of the two types of HFRSV.
