Lens epithelial cell proliferation and cell density in human age-related cataract.

PURPOSE: To discuss the potential effect of the lens epithelial cell proliferation in age-related cataract. METHODS: In vitro cell proliferation was assayed by MTT method to evaluate the lens epithelial cell density, index, and proliferation capacity in normal lens and all kinds of age-related cataract. Capsulotomy specimens from all kinds of patients who underwent cataract phacoemulsification extraction surgery were compared with the lens epithelial specimens from non-cataract lenses of Eye Bank eyes. RESULTS: Lens epithelial cell density of central anterior capsule (LECD) in female normal lens was higher than that in male, LECD in nuclear cataract (> NIII) was higher than that in normal lens, but in the mature cortical cataract, LECD was lower. Mitotic index of three kinds of age-related cataracts in vivo had no statistical difference, neither did cell proliferation capacity of cultivated cells in vitro. CONCLUSION: The individual difference of lens epithelial cell density and proliferation capacity in vivo may be an important underlying cause for senile cataract in the cellular level, especially for nuclear cataract.

[A histochemical and immunohistochemical study of cornea and adjacent bulbar conjunctiva in Mooren's ulcer].

OBJECTIVE: To investigate immunopathologic changes of cornea and adjacent bulbar conjunctiva in Mooren's ulcer, and roles of macrophage, T lymphocyte, and VCAM-1 in the pathogeny of Mooren's ulcer. METHODS: The specimens of the cornea lesions and their adjacent bulbar conjunctiva taken from 18 active Mooren's ulcer cases who received resection of conjunctiva and lamellar keratoplasty surgery were studied with histochemical and labelled streptavidin biotin (LSAB) immunohistochemical techniques to investigate the expressions of CD68, CD3, CD4, CD8, CD1a, VCAM-1, HSP70, and TNF in the affected cornea and adjacent conjunctiva. RESULTS: Under the histochemical examination, infiltration of neutrophils, plasma cells, lymphocytes, and eosinophils were found in the affected cornea and conjunctiva. Degeneration and necrosis of corneal epithelium, Bowman's layer, collageneous fiber in stromal layer, and edema of conjunctiva were shown. Infiltration of a large number of macrophages and T lymphocytes in the affected cornea and adjacent conjunctiva were shown in LSAB immunohistochemical staining. The main infiltrating T lymphocytes were the CD4 positive cells. The CD4/CD8 ratio was significantly higher than that of normal contrals (P < 0.01). VCAM-1 was strongly expressed in the affected corneal and conjunctival epithelium. There were no significant differences for expressions of CD1a, HSP70, and TNF between the pathological specimens and normal contrals. CONCLUSION: The large number of infiltrating macrophages are responsible for the immunopathological damage caused by T lymphocyte infiltration, aberrant production of cytokikens and aberrant expression of HLA-DR antigen of Mooren's ulcer. There is an aberrant expression of VCAM-1 in the affected corneal and conjunctival epithelium. The role of VCAM-1 in the immunopathogenesis of Mooren's ulcer needs to be studied.

[The electron microscopic study of AchR and ultrastructure of neuromuscular junction in recurrent laryngeal nerve reinnervation].

OBJECTIVE: The purpose of this study was to observe the distribution of Acetylcholine receptor (AchR) and ultrastructural changes of the neuromuscular junction after laryngeal reinnervation. METHODS: alpha-bungarotoxin-horseradish peroxidase (alpha-BT-HRP) conjugate electron microscopic immunocytochemical methods and computer analysis of intensity of immunocytochemical reaction were applied to observe the distribution and relative quantity of junctional N-AchR. RESULTS: The results showed that the synaptic nerve terminals and presynaptic membrane were segmentally disrupted and almost disappeared completely and were replaced by Schwann's cells at 3 weeks after neurorrhaphy. At 6 weeks, some of the regenerating axons grew to the neuromuscular junctions and all the muscle fibers were reinnervated at 12 weeks. But the original postsynaptic membranes were only partially in contact with the regenerated nerve terminals. These conditions persisted until 18 weeks after neurorrhaphy, when remyelination was found in the vicinity of the neuromuscular junctions. The N-AchR reaction product on the postsynaptic membrane were increased greatly about 1.28 times more than the normal at 3 weeks after neurorrhaphy. The reactive products decreased greatly which was only 0.47% of that in the normal at 6 weeks. It remained low until 12 weeks after neurorrhaphy, which was 0.72% of the normal side. It increased obviously to 0.83 times of normal sides at 18 weeks and didn't changed there after. CONCLUSION: The state of reinnervation determines the quantity of neuromuscular junctional N-AchR.

Arginine vasopressin gene expression in supraoptic nucleus and paraventricular nucleus of hypothalamous following cerebral ischemia and reperfusion.

BACKGROUND: Our previous studies indicated that the increased arginine vasopressin (AVP) in ischemic brain regions of gerbils could exacerbate the ischemic brain edema. This experiments is further clarify the relation between AVP and cerebral ischemia at the molecular level. METHODS: The contents of AVP, AVP mRNA, AVP immunoreactive (ir) neurons in supraoptic nucleus(SON) and paraventricular nucleus (PVN) after cerebral ischemia and reperfusion were respectively determined by radioimmunoassay (RIA), immunocytochemistry (II C), situ hybridization and computed image pattern analysis. RESULTS: The contents of AVP in SON, PVN were increased, and the AVP ir positive neurons in SON and PVN were also significantly increased as compared with the controls after ischemia and reperfusion. And there were very light staining of AVP ir positive neurons in the other brain areas such as suprachiasmatic nucleus (SC) and periventricular hypothalamic nucleus (PE), but these have no significant changes as compared with the controls. During different periods of cerebral ischemia (30 approximately 120 min) and reperfusion (30 min), AVP mRNA expression in SON and PVN were more markedly increased than the controls. CONCLUSIONS: The transcription of AVP gene elevated, then promoting synthesis and release of AVP in SON, PVN. Under the specific condition of cerebral ischemia and reperfusion, the activity and contents of central AVP increased abnormally is one of the important factors which causes ischemia brain damage.

[Spectrophotometric determination of trace cadmium in polyferric sulphate flocculent].

A new sensitive method for determining trace cadmium in polyferric sulphate flocculent is proposed in the paper. The new sensitive method is spectrophotometric method. The color reagents are the butylrhodamin B and potassium iodide. The surfactant is the polyvingl alcohol(PVA). The maximum absorbency wavelength is 607 nm. The molar absorptivity is 4.99 x 10(5) L.mol-1.cm-1. The sandell sensitivity is 2.25 x 10(-4) micrograms Cd2+.cm-2. The Beer's law is obeyed for Cd(II) in the range of 0-1.5 micrograms.25 mL-1. The recovery is 96%-105% and the sample processing procedure is simple. This method has been applied to determination of trace Cd(II) in polyferric sulphate flocculent. The determining results are satisfactory.

[Study on the photoinduced-color changing about novel azobenzocrown ether derivatives 16ABC-OH and 16ABC-NH2 in water solution and Langmuir-Blodgett film].

Two kinds of azobenzocrown ether derivation 16ABC-OH and 16ABC-NH2 have been synthesized, and their photoinduced color changing phenomena has been investigated in water solution, the reaction of 16ABC-OH is the photochemical Cis-trans-isomerization, and it is a first-order reaction, but the reaction of 16ABC-NH2 photochemical Cis-trans-isomeriation is very complex; and it is not a first-order reaction. In Langmuir-Blodgett film of 16ABC-OH, there is not molecular self-assembly phenomenon, the part of Cis-16ABC-OH become to iso-16ABC-NH2, but in the film, 16ABC-NH2 form H-Polymer by self-assembly, and the photochromic Cis-trans-isomerization is forbidded.

The synthetic somatostatin analogue, octreotide, ameliorates acute and delayed intestinal radiation injury.

PURPOSE: Reducing intraluminal proteolytic activity attenuates intestinal radiation toxicity. This study assessed whether pharmacological inhibition of exocrine pancreatic secretion protects against early and delayed radiation enteropathy in a preclinical rat model. METHODS AND MATERIALS: Rat ileum was sham-irradiated or exposed to 16 once-daily 4.2 Gy fractions of X-radiation. Vehicle or somatostatin analogue (octreotide, 2 microg/kg/hr) were administered from 2 days prior to 10 days after the end of irradiation. Mucosal injury was monitored noninvasively by assessment of granulocyte transmigration. Radiation injury was assessed at 2 weeks (early phase) and 26 weeks (chronic phase) using quantitative histopathology, immunohistochemistry, and morphometry. RESULTS: Octreotide decreased granulocyte transmigration (p<0.0006), reduced accumulation of myeloperoxidase-positive cells at 2 weeks (p = 0.0002), attenuated structural injury at 2 weeks (p = 0.04) and 26 weeks (p = 0.02), preserved mucosal surface area at 2 weeks (p = 0.0008) and 26 weeks p = 0.0008), and reduced intestinal wall thickening at 26 weeks (p = 0.002). Octreotide did not affect granulocyte transmigration, histology, or mucosal surface area in sham-irradiated controls. CONCLUSION: These results demonstrate the importance of consequential mechanisms in the pathogenesis of chronic radiation enteropathy. Short-term octreotide administration ameliorates acute radiation-induced mucosal injury, as well as chronic structural changes, and should be subject to further preclinical and clinical testing.

Characterization of the electrophysiological and morphological properties of rat central amygdala neurons in vitro.

Forty central amygdala neurons labelled with 2% Neurobiotin were categorized according to their distinctive bioelectrical membrane properties and classified physiologically by their hyperpolarized resting membrane potential (-74 mV), short duration medium afterhyperpolarization (239.2 ms), and non-accommodating response as Type A neurons (63%; N = 25/40), or as Type B neurons (37%; N = 15/40) by their depolarized resting membrane potential (-66 mV), long slow-afterhyperpolarization (1.8 s), and accommodation response. Visualized within subnuclei of the central amygdala, Neurobiotin-labelled Type A neurons were medium-size cells [16.5 +/- 3 x 10.7 +/- 2 mu m; length x width] with smooth, spine-free ovoid, pyramiform, and fusiform perikarya. Aspinous primary dendrites gave rise to distal dendrites covered with numerous small pedunculated spines; density of spines ranged from sparse to abundant. Type B central amygdala neurons were larger cells [23.9 +/- 5 x 14.9 +/- 4 mu m] with smooth, aspinous ovoid, polygonal, and pyramiform somata. Dendrites were aspinous and covered with variably sized varicosities. Two distinct populations of neurons exist within the central amygdaloid complex: the medium-size, spine-laden Type A defined neuron with its non-accommodating electrophysiological response and the larger aspinous, varicosity-laden Type B defined neuron with its accommodating response. In contrast to their neighboring "cortical-like" amygdala neurons, central amygdala neurons possess a "striatal-like" cytoarchitecture and electrophysiology.

Mice lacking both presenilin genes exhibit early embryonic patterning defects.

Genetic studies in worms, flies, and humans have implicated the presenilins in the regulation of the Notch signaling pathway and in the pathogenesis of Alzheimer's Disease. There are two highly homologous presenilin genes in mammals, presenilin 1 (PS1) and presenilin 2 (PS2). In mice, inactivation of PS1 leads to developmental defects that culminate in a perinatal lethality. To test the possibility that the late lethality of PS1-null mice reflects genetic redundancy of the presenilins, we have generated PS2-null mice by gene targeting, and subsequently, PS1/PS2 double-null mice. Mice homozygous for a targeted null mutation in PS2 exhibit no obvious defects; however, loss of PS2 on a PS1-null background leads to embryonic lethality at embryonic day 9.5. Embryos lacking both presenilins, and surprisingly, those carrying only a single copy of PS2 on a PS1-null background, exhibit multiple early patterning defects, including lack of somite segmentation, disorganization of the trunk ventral neural tube, midbrain mesenchyme cell loss, anterior neuropore closure delays, and abnormal heart and second branchial arch development. In addition, Delta like-1 (Dll1) and Hes-5, two genes that lie downstream in the Notch pathway, were misexpressed in presenilin double-null embryos: Hes-5 expression was undetectable in these mice, whereas Dll1 was expressed ectopically in the neural tube and brain of double-null embryos. We conclude that the presenilins play a widespread role in embryogenesis, that there is a functional redundancy between PS1 and PS2, and that both vertebrate presenilins, like their invertebrate homologs, are essential for Notch signaling.

The Alzheimer's disease amyloid precursor protein modulates copper-induced toxicity and oxidative stress in primary neuronal cultures.

The amyloid precursor protein (APP) of Alzheimer's disease can reduce copper (II) to copper (I) in a cell-free system potentially leading to increased oxidative stress in neurons. We used neuronal cultures derived from APP knock-out (APP(-/-)) and wild-type (WT) mice to examine the role of APP in copper neurotoxicity. WT cortical, cerebellar, and hippocampal neurons were significantly more susceptible than their respective APP(-/-) neurons to toxicity induced by physiological concentrations of copper but not by zinc or iron. There was no difference in copper toxicity between APLP2(-/-) and WT neurons, demonstrating specificity for APP-associated copper toxicity. Copper uptake was the same in WT and APP(-/-) neurons, suggesting APP may interact with copper to induce a localized increase in oxidative stress through copper (I) production. This was supported by significantly higher levels of copper-induced lipid peroxidation in WT neurons. Treatment of neuronal cultures with a peptide corresponding to the human APP copper-binding domain (APP142-166) potentiated copper but not iron or zinc toxicity. Incubation of APP142-166 with low-density lipoprotein (LDL) and copper resulted in significantly increased lipid peroxidation compared to copper and LDL alone. Substitution of the copper coordinating histidine residues with asparagines (APP142-166(H147N, H149N, H151N)) abrogated the toxic effects. A peptide corresponding to the zinc-binding domain (APP181-208) failed to induce copper or zinc toxicity in neuronal cultures. These data support a role for the APP copper-binding domain in APP-mediated copper (I) generation and toxicity in primary neurons, a process that has important implications for Alzheimer's disease and other neurodegenerative disorders.

Linkage of the BH4 domain of Bcl-2 and the nuclear factor kappaB signaling pathway for suppression of apoptosis.

Nuclear factor (NF) kappaB is a ubiquitously expressed transcription factor whose function is regulated by the cytoplasmic inhibitor protein, IkappaBalpha. We have previously shown that IkappaBalpha activity is diminished in ventricular myocytes expressing Bcl-2. (de Moissac, D., Mustapha, S., Greenberg, A. H., and Kirshenbaum, L. A. (1998) J. Biol. Chem. 273, 23946-23951). In view of the growing evidence that the conserved N-terminal BH4 domain of Bcl-2 plays a critical role in suppressing apoptosis, we ascertained whether this region accounts for the underlying effects of Bcl-2 on IkappaBalpha activity. Transfection of human embryonic 293 cells with full length Bcl-2 resulted in a significant 1.9-fold reduction in IkappaBalpha activity (p < 0.006) with a concomitant increase in DNA binding and 3.4-fold increase in NFkappaB-dependent gene transcription (p < 0. 022) compared with vector transfected control cells. In contrast, no significant change in IkappaBalpha activity was detected with either a BH4 domain deletion mutant (residues 10-30) or BH4 domain point substitution mutants, I14G, V15G, Y18G, K22G, and L23G (p = 2.77). However, a small 0.60-fold decrease (p < 0.04) in IkappaBalpha activity was noted with the BH4 mutant I19G, suggesting that this residue may not be critical for IkappaBalpha regulation. Furthermore, adenovirus-mediated delivery of an IkappaBalpha mutant to prevent NFkappaB activation impaired the ability of Bcl-2 to suppress apoptosis provoked by TNFalpha plus cycloheximide in ventricular myocytes. The data provide the first evidence for the regulation of IkappaBalpha by Bcl-2 through a mechanism that requires the conserved BH4 domain that links Bcl-2 to the NFkappaB signaling pathway for suppression of apoptosis.

Copper levels are increased in the cerebral cortex and liver of APP and APLP2 knockout mice.

The pathological process in Alzheimer's disease (AD) involves amyloid beta (Abeta) deposition and neuronal cell degeneration. The neurotoxic Abeta peptide is derived from the amyloid precursor protein (APP), a member of a larger gene family including the amyloid precursor-like proteins, APLP1 and APLP2. The APP and APLP2 molecules contain metal binding sites for copper and zinc. The zinc binding domain (ZnBD) is believed to have a structural rather than a catalytic role. The activity of the copper binding domain (CuBD) is unknown, however, APP reduces copper (II) to copper (I) and this activity could promote copper-mediated neurotoxicity. The expression of APP and APLP2 in the brain suggests they could have an important direct or indirect role in neuronal metal homeostasis. To examine this, we measured copper, zinc and iron levels in the cerebral cortex, cerebellum and selected non-neuronal tissues from APP (APP(-/-)) and APLP2 (APLP2(-/-)) knockout mice using atomic absorption spectrophotometry. Compared with matched wild-type (WT) mice, copper levels were significantly elevated in both APP(-/-) and APLP2(-/-) cerebral cortex (40% and 16%, respectively) and liver (80% and 36%, respectively). Copper levels were not significantly different between knockout and WT cerebellum, spleen or serum samples. There were no significant differences observed between APP(-/-), APLP2(-/-) and WT mice zinc or iron levels in any tissue examined. These findings indicate APP and APLP2 expression specifically modulates copper homeostasis in the liver and cerebral cortex, the latter being a region of the brain particularly involved in AD. Perturbations to APP metabolism and in particular, its secretion or release from neurons may alter copper homeostasis resulting in increased Abeta accumulation and free radical generation. These data support a novel mechanism in the APP/Abeta pathway which leads to AD.

Involvement of caspases in proteolytic cleavage of Alzheimer's amyloid-beta precursor protein and amyloidogenic A beta peptide formation.

The amyloid-beta precursor protein (APP) is directly and efficiently cleaved by caspases during apoptosis, resulting in elevated amyloid-beta (A beta) peptide formation. The predominant site of caspase-mediated proteolysis is within the cytoplasmic tail of APP, and cleavage at this site occurs in hippocampal neurons in vivo following acute excitotoxic or ischemic brain injury. Caspase-3 is the predominant caspase involved in APP cleavage, consistent with its marked elevation in dying neurons of Alzheimer's disease brains and colocalization of its APP cleavage product with A beta in senile plaques. Caspases thus appear to play a dual role in proteolytic processing of APP and the resulting propensity for A beta peptide formation, as well as in the ultimate apoptotic death of neurons in Alzheimer's disease.
