[The relationship between plasma tumor necrosis factor alpha levels and complications after allogeneic bone marrow transplantation].

OBJECTIVE: To ascertain the implication of tumor necrosis factor alpha (TNF alpha) in the pathogenesis of acute graft-versus-host disease (aGVHD). METHODS: Plasma TNF alpha levels were assessed by ELISA in 243 sera samples from 40 patients who had undergone allogeneic bone marrow transplantation (BMT). Plasma TNF alpha levels were measured before BMT and at different time points after BMT. The results were correlated with aGVHD, infection and conditioning regimen. RESULTS: Plasma TNF alpha levels were significantly higher in patients with grade II and grades III-IV aGVHD (0-4.90) micrograms/L and (0.25-4.21) micrograms/L, respectively than in those with grade 0 or I aGVHD, but there was no clear correlation between plasma TNF alpha levels in patients with grade II and grade III-IV aGVHD. Plasma TNF alpha levels in patients with infection were not statistically different from those in patients without infection. The conditioning regimen did not cause a significant rise in TNF alpha levels. CONCLUSION: It is indicated that TNF alpha may be useful for the diagnosis of aGVHD and for differentiation between aGVHD and other BMT related complications such as infection.

[The transferability of acetochlor and butachlor in soil].

The transferability of acetochlor and butachlor in soil was studied by soil thin layer chromatography. Acetochlor and butachlor were dropped on the glass plate and spreaded soil on the glass plate was collected per 2 cm, then acetochlor and butachlor were analyzed quantitatively by HPlC. When river water was as the spread solution, Rf(relative flow) of acetochlor and butachlor in the Haidian loam were 0.116 and 0.031 respectively, Rf of acetochlor and butachlor in the Baiyangdian sandy loam were 0.147 and 0.032 respectively. When 30 mg.L-1 dodecylbenzene sulfonic acid sodium salt solution was as the spread solution, Rf of acetochlor and butachlor in the Haidian loam were 0.159 and 0.034 respectively. Acetochlor's transferability was weak and the gradation of its transferability was II grade, while butachlor's was more weak and the gradation was I grade. Anionic surfactant solution can promote pesticides to transfer. Cationic surfactant solution can impede pesticides to transfer.

[The change in the plasma contents of adrenomedullin and endothelin in burn patients during early postburn stage].

OBJECTIVE: To observe the change in the plasma contents of adrenomedullin (ADM) and endothelin (ET) in burn patients, so as for us to explore their roles in the pathogenesis in the physiological disturbance in burn patients. METHODS: The plasma concentrations of ADM and ET were determined at 6, 12, 24 and 48 postburn hours (PBHs) in 40 burn patients with radioimmunoassay (RIA) and compared with those of 25 healthy people. RESULTS: The postburn plasma levels of ADM and ET in burn patients increased immediately after injury, and they were positively correlated to burn area. The plasma ADM reached peak level at 12 PBHs and decreased thereafter, but remained at higher level than that in healthy people at 48 PBHs (P < 0.01). Plasma ET increased to top level at 6 PBH and decreased thereafter. But it remained at high level at 48 PBHs in severe burn patients (burned area larger than 45% TBSA) (P < 0.05 approximately 0.01). The plasma levels of ET and ADM increased proportionally and their ratio was close to that in normal control, but it decreased obviously after 6 PBH in all groups (P < 0.01). CONCLUSION: There were evident postburn changes in plasma ADM and ET contents, and the results implied that ADM and ET played some roles in the development of postburn physiological disturbance.

Immunity induced by DNA vaccine of plasmid encoding the rhoptry protein 1 gene combined with the genetic adjuvant of pcIFN-gamma against Toxoplasma gondii in mice.

OBJECTIVE: To construct the eukáryotic expression recombinant plasmid, pcIFN-gamma, as a genetic adjuvant and observe the immune responses elicited by pcDNA3-rhoptry protein 1 (pc-ROP1) combined with pcIFN-gamma against Toxoplasma gondii (T. gondii) infection in mice. METHODS: A fragment of the IFN-gamma gene was directly inserted into the pcDNA3 plasmid and identified by two restriction endonucleases digestion. pcIFN and pcROP1 DNA was injected into the left leg muscle of mice at a dosage of 100 micrograms, and a booster vaccination was given at the same dosage after two weeks. Control groups were injected with pcDNA3 blank plasmid or normal saline. At 30, 50 and 70 days after booster injection, kinetic tests were carried out: MTT assay for the proliferation response of T lymphocyte cells and the activity of NK cells, sandwich ABC-ELISA for the determination of IFN-gamma, IL-2 and IL-10; a serum enzymetic aassay for nitric oxide (NO) in sera and ELISA for the titer of IgG antibody in sera. RESULTS: The recombinant plasmid, pcIFN-gamma was constructed. The proliferation response of spleen T lymph cells, NK cell killing activity, and serum levels of IFN-gamma, IL-2 and NO in mice injected with pcROP1 and pcIFN-gamma were higher than in those injected with pcROP1 alone. There was no difference in IgG antibody levels between the two groups. CONCLUSION: The genetic adjuvant, pcIFN-gamma, could enhance the cellular immune response induced by DNA vaccine of pcROP1 in mice against Toxoplasma gondii infection.

Construction of a recombinant plasmid harbouring the rhoptry protein 1 gene of Toxoplasma gondii and preliminary observations on DNA immunity.

OBJECTIVE: To observe the immune responses elicited in BALB/c mice by a DNA vaccine. A gene encoding rhoptry protein 1 (ROP1) from Toxoplasma gondii (T. gondii) was cloned into vector pcDNA3. METHODS: Amplifyied gene fragments coding for ROP1 from the genomic DNA of T. gondii ZS2 were inserted into cloning vector, pUC18, and sub-cloned into pcDNA3. Mice were injected at a dosage of 100 micrograms recombinant plasmid DNA by intramuscular injection and boosted after 2 weeks. pcDNA3 and normal saline were used as control. 30, 50 and 70 days after the second immunization, NK cell activity, T lymphocyte proliferation and sub-clusters and serum IgG antibody were assayed. RESULTS: The specific gene fragment coding for ROP1 was amplified and a pcROP1 recombinant was constructed. At 30 days after immunization, the spleens of the mice were obviously enlarged evidently. NKC activity and the proliferation of spleen T lymphocytes seen on MTT assay were higher in pcROP1 group than in the controls. The number of CD4+ T cells exhibited no obvious increase compared with that of the control, but CD8+ T cells were obviously increased (P < 0.05). At 90 days after vaccination, the titer of IgG antibody in the serum of vaccinated mice was positive (1:100). CONCLUSION: pcROP1 was constructed and it could elicit both cellular and humoral immune responses in immunized mice.

The effect of volume replacement on serum protein concentration during cardiopulmonary bypass.

Although controversy exists concerning the optimal total protein and colloid osmotic pressure that should be maintained during cardiopulmonary bypass (CPB), the primary volume expanders remain albumin and 6% hetastarch. The purpose of this study was to quantify the effect of adding boluses of volume replacement agents under various conditions to total serum protein values during CPB. A standard CPB circuit was utilized in eight 45-kg swine that had a priming volume (physiologic saline solution) of 2309 +/- 245 mL. Volumetric alterations occurred throughout the CPB period by the addition of combinations of physiologic saline solution, 6% hetastarch or 5% swine albumin. Pre- and postadministration samples were assayed for total serum protein, total protein, and albumin throughout the CPB period and at pre- and postvolume administration times. There was a significant decline in total serum protein with the initiation of CPB (6.14 +/- 0.49 g/dL vs. 3.40 +/- 0.43 g/dL, p < .0001). Addition of 12.5 g of swine albumin (N = 5) to two different swine increased total serum protein significantly when compared to adding 500 mL of 6% hetastarch (N = 6) (swine albumin 12.4 +/- 6.3% vs. hetastarch 3.3 +/- 2.1%, p < .005). A reduction in total serum protein occurred after hemodilution with varying amount of physiologic saline solution: 250-450 mL (7.4 +/- 4.5%), 451-650 mL (9.6 +/- 5.6%), and 651-1050 mL (19.4 +/- 4.0%). In summary, knowledge of total serum protein concentration and estimated circulating blood volume can be used to guide albumin and hetastarch administration following hemodilution.

[A survey of the current status and distribution of cataract in the elderly].

OBJECTIVE: To study the current status and distribution of cataract in the ageing population. METHODS: A total number of 8,252 elderly aged 60 years and above in the urban and rural areas of Beijing, Shanghai, Guangzhou, Chengdu, Xi'an and Shenyang were studied, using cluster random sampling methods. RESULTS: The crude and adjusted rates of cataract prevalence in the elderly were found to be 46.8% and 42.8% respectively. However, the self-reported crude prevalence was 19.7% with only 42.1% of that shown by medical examination. The prevalence rates increased with ageing (P < 0.01), to have shown 27.9%, 41.3%, 53.2%, 67.5% and 68.0% in the age groups of 60-, 65-, 70-, 75-, 80-, 85- years old respectively (P < 0.01). There was difference in the prevalence among areas, with highest (77.9%) in the urban areas in Guangzhou and in the rural areas in Beijing (67.3%) (P < 0.01). The prevalence differed in gender as well: higher in women (49.1%, adjusted) than in men (35.6%, adjusted). Logistic regression analysis showed that the crude prevalence was correlated with ageing, profession and area (P respective < 0.01). CONCLUSION: The prevalence of cataract was high in the elderly and increased with ageing with differences in areas, and professions. It was low when self-reporting shown by medical examination, suggesting prevention and treatment of cataract in the elderly be strengthened.

[Culture and identification of fetal human retinal photoreceptors].

OBJECTIVE: To investigate the methods of isolation and culture of fetal human retinal photoreceptors. METHODS: Fetal human retinal photoreceptors were obtained by enzyme gradation digestion, cultured in media conditioned by retinal pigment epithelial cells (RPE-CM), identified by morphologic observation and immunohistochemically stained with rod specific opsin 4D2 (Rho 4D2) antibody. The digested retinal tissues were tested by hematoxylin-eosin (HE) staining. RESULTS: Enzyme gradation digestion could obtain relatively pure retinal photoreceptors. These cells were stained positively for Rho 4D2 antibody, and survived in RPE-CM for a long time. CONCLUSION: Fetal human retinal photoreceptors obtained by enzyme gradation digestion can survive in RPE-CM for a long time and express rod specific opsin. This culture system will provide a source for human retinal photoreceptor transplantation and further studies of photoreceptors in vitro.

[Effect of hepatitis C virus nonstructural protein NS3 on telomerase activity].

OBJECTIVE: To study the effect of hepatitis C virus nonstructural protein NS3 (HCV NS3) on telomerase activity and its carcinogenesis, and to observe the practical use of in situ telomerase activity labeling. METHODS: NIH3T3 cells were transfected with plasmid pRcHCNS3-5prime prime or minute (expressing HCV NS3 C-terminal deleted protein), pRcHCNS3-3prime prime or minute (expressing HCV NS3 N-terminal deleted protein) and blank plasmid pRcCMV. Positive clones, 11 with plasmid pRcHCNS3-5prime prime or minute, 11 with pRcHCNS3-3prime prime or minute and 8 with plasmid pRcCMV were harvested respectively. Streptavidin-peroxidase conjugated method (SP) was used to detect the expression of HCV NS3 protein in NIH3T3 cells transfected with plasmid pRcHCNS3-5prime prime or minute and pRcHCNS3-3prime prime or minute. Telomerase activity was detected by in situ telomerase activity labeling method and telomerase PCR ELISA technique in the transfected and non-transfected NIH3T3 cells. RESULTS: HCV NS3 protein was expressed in the cells transfected with plasmid pRcHCNS3-5prime prime or minute or plasmid pRcHCNS3-3prime prime or minute. The positive signals of HCV NS3 protein were localized in the cytoplasm of transfected NIH3T3 cells, and the signal intensity of the former was stronger (chi(2) = 6.667 P < 0.05). There was significant difference on telomerase activity between each group (F = 134.083 P < 0.01). Telomerase activity in all 11 clones with plasmid pRcHCNS3-5prime prime or minute was stronger than cells with plasmid pRcHCNS3-3prime prime or minute (P < 0.01), whereas telomerase activity in NIH3T3 cells transfected with the plasmid pRcCMV or non-transfected NIH3T3 cells were weaker than the cells with the plasmid pRcHCNS3-3prime prime or minute. The expression level of HCV NS3 protein were correlated significantly with the strength of telomerase activity (rs = 0.8084 P < 0.01). The results obtained with in situ telomerase activity labeling corresponded to those with telomerase PCR ELISA technique (rs = 0.50196 P < 0.01). CONCLUSIONS: (1) HCV NS3 protein may activate telomerase through endogenous mechanism to induce host cells transformation. (2) The effect of HCV NS3 C-terminal deleted protein on telomerase activity in host cells may be more intense than that of HCV NS3 N-terminal deleted protein. (3) In situ telomerase activity labeling is a reliable technique for studying pathological morphology and telomerase activity in tissues and cells.

[A study of bone development and metabolism in childhood].

OBJECTIVE: To explore the regularity of bone development and metabolism in childhood. METHODS: A survey was made to collect and analyze the data on bone biochemistry, the density of lumbar vertebrae and the X-ray examination of calcaneus in 224 healthy children aged 3-15 years in the urban area of Chengdu. RESULTS: Comparative analysis of serum and urine calcium and phosphate concentrations showed no significant difference between the age groups. The serum alkaline phosphatase (ALP) and bone alkaline phosphatase (BALP) were found to increase with age and reach their peak values in children aged 7-12 years, and their values were significantly higher than that of adults (P < 0.05). The density of lumbar vertebrae and the trabecular bone of calcanesus also developed with age. CONCLUSION: The measurements of the bone biochemistry, bone density and the trabecular bone of calcaneus which reflect the bone development and metabolism have their special regularity in childhood. The criteria in use for adulthood is not fit for the evaluation in childhood. The data obtained can be used in clinical diagnosis and treatment of the bone metabolic diseases of children.

[Effects of XW630 on mechanical properties and trabecular structure parameters of bone tissue in ovariectomized rats].

OBJECTIVE: The study aimed at investigating the effects of a new anti-osteoporotic drug, XW630 on promoting the osteogenic activity of bone tissue in ovariectomized rats. METHODS: Thirty-six female SD rats, three months old, were randomly divided into four groups: sham-operated group (sham), the ovariectomized group (OVX), the esterone-therapy group (OVX + CFT) and the XW630-therapy group (OVX + XW630). Three rats in each group were killed on the 30th, 60th, and 90th day after the operation. The femur of one side was taken for the three-point bending resistance test, and the bilateral tibias were taken for the HE stain and scanning electronic microscopy (SEM) examination. RESULTS: The HE and SEM results indicated that the bone loss appeared in the OVX group 30 days after the operation, which lasted for the whole observing period. While after application of ester-one and XW630, there were different degrees of bone repairing around the trabeculae, and the osteogenic activity was obviously active in the XW630 group than that in the esterone group. During the experimental period, the three-point bending resistance of the OVX group gradually decreased and there was a significant difference compared with that of the sham group (P < 0.01), while the three-point bending resistance of the two therapy group gradually increased and there was significant difference between the OVX group and the XW630 group (P < 0.05 and P < 0.01). CONCLUSION: XW630 can effectively promote osteogenic action and prevent osteoporostic fracture in ovariectomized rats.

[The effects of perfluorodecalin on the anterior chamber angle and intraocular pressure in rabbits].

OBJECTIVE: To observe the effects of purfluorodecalin on the anterior chamber (AC) angle and intraocular pressure (IOP) in rabbits. METHODS: 0.05 ml perfluorodecalin was injected into right AC of 17 white rabbits and the same does of balanced salt solusion(BSS) was given to the controll group (6 rabbits). The IOP was measured before and 1, 2, 3, 4, 8, 12, 16 weeks after the injection. The treatment rabbits were sacrificed on the week of 1, 2, 4, 8 and 16 postoperatively. The eyes were enucleated and the histological examination was performed. RESULTS: There's no statistic IOP difference in the controll group before and after the injection of BSS. In treatment group, the IOP is similar to the preoperation level (11.62 +/- 6.35 mmHg) on the 7th postoperative day (10.82 +/- 3.17 mmHg, P < 0.05). A increment of IOP was measured at the week of 2 to 4, 8, 12, and 16 postoperatively (P < 0.01). Perfluorodecalin, having a 'fish eggs' or bubbles appearance, was noted in the inferior AC. Histopathologically, transparent single or fused vesicles and macrophages were accumulated at the AC angler in treatment rabbits. Some lymphocytes and neutrophils were aslo noted here. Trabeculae degenerated and intertrabecular space increased. CONCLUSIONS: The anterior chamber injection of perfluorodeclin could lead to a longstanding increase of IOP. Inflammatory reaction and trabeculae degeneration were seen in the anterior chamber angler.
