Somatostatin receptor subtype 2 knockout mice are refractory to growth hormone-negative feedback on arcuate neurons.

The pulsatile nature of GH release is apparently regulated by alternating sequential changes in two hypothalamic hormones, GH releasing hormone (GHRH) and somatostatin. Entrainment of this pulsatility appears to involve GH-mediated negative feedback. Recently a new receptor involved in GH release was cloned. Activation of this receptor by GH-releasing peptides and MK-0677 initiates and amplifies GH pulsatility and is associated with increased Fos immunoreactivity and electrical activity in GHRH containing arcuate neurons. We show that pretreating mice with GH blocks activation of these neurons by MK-0677. Similarly, octreotide inhibited the action of MK-0677. To determine whether this GH-mediated negative feedback on GHRH neurons was direct, or by GH stimulation of somatostatin release from periventricular neurons, we selectively inactivated the gene for one of the five specific somatostatin receptor subtypes (subtype 2). In the knockout mice, both GH and octreotide failed to inhibit MK-0677 activation of arcuate neurons. GH did, however, increase Fos immunoreactivity in the periventricular nucleus, consistent with GH stimulation of somatostatin release from periventricular neurons. Thus, GH-mediated negative feedback involves signaling between periventricular and arcuate neurons with the signal being transduced specifically through somatostatin subtype 2 receptors.

Effects of diet and exercise in preventing NIDDM in people with impaired glucose tolerance. The Da Qing IGT and Diabetes Study.

OBJECTIVE: Individuals with impaired glucose tolerance (IGT) have a high risk of developing NIDDM. The purpose of this study was to determine whether diet and exercise interventions in those with IGT may delay the development of NIDDM, i.e., reduce the incidence of NIDDM, and thereby reduce the overall incidence of diabetic complications, such as cardiovascular, renal, and retinal disease, and the excess mortality attributable to these complications. RESEARCH DESIGN AND METHODS: In 1986, 110,660 men and women from 33 health care clinics in the city of Da Qing, China, were screened for IGT and NIDDM. Of these individuals, 577 were classified (using World Health Organization criteria) as having IGT. Subjects were randomized by clinic into a clinical trial, either to a control group or to one of three active treatment groups: diet only, exercise only, or diet plus exercise. Follow-up evaluation examinations were conducted at 2-year intervals over a 6-year period to identify subjects who developed NIDDM. Cox's proportional hazard analysis was used to determine if the incidence of NIDDM varied by treatment assignment. RESULTS: The cumulative incidence of diabetes at 6 years was 67.7% (95% CI, 59.8-75.2) in the control group compared with 43.8% (95% CI, 35.5-52.3) in the diet group, 41.1% (95% CI, 33.4-49.4) in the exercise group, and 46.0% (95% CI, 37.3-54.7) in the diet-plus-exercise group (P < 0.05). When analyzed by clinic, each of the active intervention groups differed significantly from the control clinics (P < 0.05). The relative decrease in rate of development of diabetes in the active treatment groups was similar when subjects were stratified as lean or overweight (BMI < or > or = 25 kg/m2). In a proportional hazards analysis adjusted for differences in baseline BMI and fasting glucose, the diet, exercise, and diet-plus-exercise interventions were associated with 31% (P < 0.03), 46% (P < 0.0005), and 42% (P < 0.005) reductions in risk of developing diabetes, respectively. CONCLUSIONS: Diet and/or exercise interventions led to a significant decrease in the incidence of diabetes over a 6-year period among those with IGT.

[Insulin autoantibodies (IAA) in sera of individuals with negative values in IAA radioimmunoassay].

Insulin autoantibidy(IAA) is one of the most important markers for detection of population at risk to develop into Type I diabetes. In screening the subjects and following up their conditions, the authors re-examined some sera with IAA radioimmnoassasy by using different concentrations of unlabeled insulin. The results of these subjects were compared with those of normal controls and IAA positive diabetics. It was confirmed that there was actually a large amount of IAA in the sera of individuals with negative values in standard IAA radioimmunoassay. Only in this group, the levels of IAA varied with increased amount of unlabeled insulin (P < 0.01). Should similar events be confronted, it is advisable to increase the amount of unlabeled insulin in IAA radioimmunoassay on purpose to avoid missed diagnosis.

[Volume loading experiment in dogs with large area infarction of right and left ventricles].

In order to study volume loading effect on large area myocardial infarction (MI) in right and left ventricles, large area MI in both ventricles and cardiogenic shock in 12 dogs were induced by occluding coronary arteries. Left ventricular systolic pressure (LVSP) and +/- dp/dt max. dropped markedly by 54%, 51% and 47% respectively, whereas right ventricular systolic pressure (RVSP) and +/- dp/dt max. fell by 9%, 25% and 27% respectively. The condition was obviously worse by rapid volume loading (dextran, 30 ml/kg, i.v. in 20 min.) in group I (n = 6), leading to increased retrograde beat of right ventricle, further decrease of +/- dp/dt max. in both ventricles, significant increase of right atrial pressure (RAP) and left ventricular and diastolic pressure (LEVDP) (2.9 +/- 0.2 kPa, P < 0.01 and 5.0 +/- 0.3 kPa, P < 0.001, respectively), and even ventricular fibrillation. Shock was reversed by combined treatment of dopamine (10 micrograms/kg.min) and glyceryl trinitrate (1 microgram/kg.min) in group II (n = 6) in 30 min, showing evident increase of arterial pressure, cardiac output, LVSP and +/- dp/dt max. without rise in RAP and LVEDP.

[Application of atracurium to critical care patients in ICU].

Fifty-two patients entered ICU after heart operation with cardiopulmonary bypass. These patients with ventilation atracurium (ATC) were given 3 micrograms-6 micrograms/kg/min for 5-48 hours. No complications happened. After ATC was used, the patients could open eyes, and cooperate with the doctor when they regain consciousness. The circulation index was normal. The limbs of these patients relaxed. In the 52 cases, 30 did't use sedatives, and 22 used senall dose of sedatives for sleep at night. We conclude that ATC does not require hepatic or renal function for termination of effect and action is terminated by the "Hoffman elimination" the ester hydrolysis. The time of effect is short and there is no cumulation effect. There is no vagal or ganglionic blocking activity, so ATC does not influence circulation and myocardial function. Maintenance relaxation keeps ventilation steady and oxygen supply normal.

[Optimazation of conditions for stereoselective reaction by enzyme-catalyzed acetylation].

Optically active endo-tricyclo-[5,2,1, 0(2,6)]deca-8-en-3,5-diol-5-acetate is a very useful chiral synthon for the synthesis of optically active natural products. In this paper, the optimal conditions for the synthesis of this compound was established by orthogonality test. Under these conditions, the chemical percentage yield of acetylation of (+) endo-tricyclo-[5,2,1,0(2,6)] deca-8-en-3,5-diol-5-acetate and its % e.e. can reach up to 81% and 98.3% respectively. The optimazation of the conditions is efficient for the reaction of enzyme-catalyzed esterification.

[Bone histomorphometric changes in ovariectomized goat at different time courses].

Using histopatholoical method, we investigated the bone histomorphometric changes in ovariectomied goats at different time courses. Eight goats, female, aged one and a half years, were randomly divided into two groups, the control and the ovariectomy (OVX). Iliac crest biopsies were separately processed before ovariectomy and at 60 days, 120 days and 180 days after ovariectomy. The histomorphometric changes were observed. It was found that the bone structure in trabecular bone remained relatively constant in the control group throughout the whole study. In contrast, the trabecular quantity was slightly decreased and the trabeculae were disconnected in OVX at 60 days after ovariectomy. The bone structure was characterized by the thinning of the trabeculae, the further discontinuance and quantity loss of trabeculae, and the anlargement of marrow cavity at 120 days to 180 days after ovariectomy. Apparently, these bone structural changes manifested with typical osteopenia pathological changes. Our results may serve as a basis for the design of further studies using OVX goat as an animal model for postmenopausal osteoporosis.

[Culture of human corneal epithelial cells].

PURPOSE: To select culture technique of human epithelial cells for grafts. METHODS: Epithelial cells of limbus were collected by improved enzymatic disaggregation or explant technique growth curves and to calculate doubling time. DNA synthesis of cells were measured with 3H-thymidine incorporation and liquid scintillation techniques. RESULTS: Doubling time of cells which were collected by enzymatic disaggregation and by explant technique were 54.15 +/- 4.28 h and 67.88 +/- 1.96 h (P < 0.01). Cellular DNA synthesis of the former was more active (P < 0.01). CONCLUSIONS: Epithelial cells which were collected by improved enzymatic disaggregation from limbus include stem cells of corneal epithelium. These cells show more active cellulde proliferation and are fitter for use as grafts.

Analysis of the antigen-antibody specificity in the semen of patients with Neisseria gonorrhoeae.

This study was done to define the human genital immune response to infection with Neisseria gonorrhoeae. The semen specimens were obtained from 15 patients with uncomplicated gonococcal infection in the acute and convalescent phases and 15 men with uninfected control. After precipitated with amoniasulfate, the semen was tested against the outer membrane protein of gonococcal isolates from the same patients to examine antigen-antibody interactions by use of the western blot technique. The antibodies in the semen reacted with more gonococcal antigens in the acute phase than in the convalescent phase. IgA in the semen reacted with more antigens than did IgG in the same specimens. The predominant reacted antigens were protein I, protein II, 46-48, kD, 14-16 kD and 88-90 kD protein.

[Primary study on the use of ovariectomized rabbits for establishment of osteoporosis model].

Purebred New Zealand female rabbits were used in this study for demonstrating the feasibility of establishing rabbit's primary osteoporosis model. Bilateral ovariectomies were performed. Five to eight months after operation, we observed the BMD, weight, hair colour and living condition of the model. After killing the rabbits, we made biomechanical detection and pathologic histomorphometry using the bones. The weight of bone ash, the content of calcium and phosphorus, and the weight of metra were determined. The results showed that BMD, bone bending stress, weight of bone ash, content of calcium and phosphorus in ash, and weight of metra all decreased (P < 0.05); the number of trabecula decreased and the bone cortex became thinner. All these parameters evidenced the primary founding of rabbit's osteoporosis model. This research provides a basis for studying the pathogenesis of primary osteoporosis and evaluating the efficacy of pharmacotherapy.

[Changes in the expression of alpha- and beta-myosin heavy chain gene during the development of renal hypertension in rats].

The changes in the expression of cardiac alpha- and beta-myosin heavy chain (MHC) gene of the left ventricle were investigated in two-kidney, one-clip (2K1C) renal hypertensive rats. The results were as follows: (1) When blood pressure was increased, the left ventricle became hypertrophic, alpha-MHC gene expression was reduced and beta-MHC gene expression was increased in 2K1C renal hypertensive rats. (2) When the animal was treated with captopril, angiotensin converting enzyme inhibitor 4 W after operation and then 8 W with removal of the ischemic kidney, the blood pressure was decreased with attendant regression of left ventricular hypertrophy, while the increase in beta-MHC mRNA level was attenuated and the inhibition of alpha-MHC mRNA level was reduced. The above results suggest that the rise in arteral pressure is an important factor in the left ventricular hypertrophy and the MHC gene switch. Renin angiotension system may be involved in the cardiac hypertrophic and MHC gene switch during the development and maintenance of 2K1C renal hypertension.

Resistance to cytotoxic drugs in DNA mismatch repair-deficient cells.

Loss of DNA mismatch repair is a common finding in many types of sporadic human cancers as well as in tumors arising in patients with hereditary nonpolyposis colon cancer. The effect of the loss of DNA mismatch repair activity on sensitivity to a panel of commonly used chemotherapeutic agents was tested using one pair of cell lines proficient or deficient in mismatch repair due to loss of hMSH2 function and another due to loss of hMLH1 function. 6-Thioguanine and N-methyl-N'-nitro-N-nitrosoguanidine, to which these cells are known to be resistant, were included in the panel as controls. The results were concordant in both pairs of cells. Loss of either hMSH2 or hMLH1 function was associated with low level resistance to cisplatin, carboplatin, and etoposide, but there was no resistance to melphalan, perfosfamide, 5-fluorouracil, doxorubicin, or paclitaxel. The results are consistent with the concept that the DNA mismatch repair proteins function as a detector for adducts produced by 6-thioguanine, N-methyl-N'-nitro-N-nitrosoguanidine, cisplatin, and carboplatin but not for melphalan and perfosfamide. They also suggest that these proteins play a role in detecting the DNA damage produced by the binding of etoposide to topoisomerase II and propagating signals that contribute to activation of apoptosis.
