Mutation in and lack of expression of tyrosinase-related protein-1 (TRP-1) in melanocytes from an individual with brown oculocutaneous albinism: a new subtype of albinism classified as "OCA3".

Most types of human oculocutaneous albinism (OCA) result from mutations in the gene for tyrosinase (OCA1) or the P protein (OCA2), although other types of OCA have been described but have not been mapped to specific loci. Melanocytes were cultured from an African-American with OCA, who exhibited the phenotype of Brown OCA, and his normal fraternal twin. Melanocytes cultured from the patient with OCA and the normal twin appeared brown versus black, respectively. Melanocytes from both the patient with OCA and the normal twin demonstrated equal amounts of NP-40-soluble melanin; however, melanocytes from the patient with OCA contained only 7% of the amount of insoluble melanin found from the normal twin. Tyrosinase- related protein-1 (TRP-1) was not detected in the OCA melanocytes by use of various anti-TRP-1 probes. Furthermore, transcripts for TRP-1 were absent in cultured OCA melanocytes. The affected twin was homozygous for a single-bp deletion in exon 6, removing an A in codon 368 and leading to a premature stop at codon 384. Tyrosine hydroxylase activity of the OCA melanocytes was comparable to controls when assayed in cell lysates but was only 30% of controls when assayed in intact cells. We conclude that this mutation of the human TRP-1 gene affects its interaction with tyrosinase, resulting in dysregulation of tyrosinase activity, promotes the synthesis of brown versus black melanin, and is responsible for a third genetic type of OCA in humans, which we classify as "OCA3."

Interrelationship between magnesium and potassium in preventing myocardial ischemia-reperfusion arrhythmia.

OBJECTIVE: To assess the interrelationship between Mg2+ and K+ in preventing reperfusion arrhythmia (RA). MATERIAL AND METHODS: 72 rat hearts were randomly divided into 6 equal groups, and perfused with various combinations of Mg2+ and K+ in the perfusate by Langendorff method. Each heart was subjected to coronary occlusion for 15 minutes followed by 12-minute reperfusion. The incidence of RA was recorded. The concentration of K+ and malondialdehyde (MDA) in the coronary effluent before and after reperfusion were measured with increasing concentrations of Mg2+ and fixed concentration of K+. RESULTS: All forms of ventricular arrhythmias occurred after reperfusion but with decreased incidence by increasing Mg2+ and K+. When both Mg2+ and K+ were in high level, no VF appeared on reperfusion. However, when they were both in low level, 100% of hearts developed severe arrhythmia. The concentration of K+ and MDA in the coronary effluent was markedly increased compared with that before reperfusion, and the loss of K+ and malondialdehyde (MDA) decreased with increasing Mg2+. CONCLUSIONS: Mg2+ may prevent or reduce the incidence of RA by maintaining the integrity of intracellular K+ and by reducing the lipid peroxide reaction.

Relationship between chondroitin sulfate proteoglycan and coronary atherosclerosis in the youth.

OBJECTIVE: To study the relationship between chondroitin sulfate proteoglycan (CSPG) and coronary atherosclerosis in youth. MATERIAL AND METHODS: Immunohistochemistry of chondroitin sulfate proteoglycan and SM-alpha-actin was performed on 39 human coronary arteries, including normal vessels and vessels with fatty streaks, intermediate lesions and plaques. Some of the lesional vessels were taken for beta-lipoprotein immunohistochemical study. CSPG contents were quantitated by image analyzer and the coronary arteries from one heart were processed for immunoelectron microscopy. RESULTS: The range of CSPG distribution was extensive and CSPG area density was increased significantly in the intima with atherosclerotic lesions. The increase of CSPG was parallel to the numbers of smooth muscle cell (SMC) with positive SM-alpha-actin reaction and was likely related to the phenotype transformation of SMC. Similar distribution of beta-lipoprotein and CSPG were found within the lesional regions. CONCLUSIONS: The increase of CSPG in atherosclerotic lesion may be an important factor resulting in the deposition of lipids in the vascular wall so as to promote the formation and development of atherosclerosis.

Pharmacokinetics and tissue disposition of a chimeric oligodeoxynucleoside phosphorothioate in rats after intravenous administration.

Antisense oligonucleotides represent a novel therapeutic principle for designing drugs against various diseases. Oligonucleotides can be chemically modified to improve their pharmacokinetics and in vivo stability, and it is important to understand the effect of these modifications. In the present study, the pharmacokinetics of a 25-mer phosphorothioate oligonucleotide containing four contiguous, internucleotide, methylphosphonate linkages at the 3'- and 5'-ends (chimeric oligonucleotide) were determined in rats after i.v. administration of the 35S-labeled oligonucleotide at a dose of 30 mg/kg. Plasma disappearance of the oligonucleotide could be described by a two-compartment model, with half-lives of 0.38 and 52.9 hr. The intact chimeric oligonucleotide was detected in plasma up to 6 hr after dosing. Urinary excretion represented the major elimination pathway, with approximately 21% of the administered dose being excreted within 24 hr and 35% being excreted over a 240-hr period after dosing. The majority of the radioactivity in urine was associated with the intact oligonucleotide within 6 hr after dosing and with increasing degradation products thereafter. Fecal excretion was a minor elimination pathway. The oligonucleotide was widely distributed in tissues, with the majority of the radioactivity in most tissues being intact up to 48 hr after dosing. Compared with oligodeoxynucleotide phosphorothioates, the chimeric oligonucleotide was significantly more stable in vivo. The presence of intact oligonucleotide in plasma and tissues even 12 hr after dosing is a significant advantage over an "all"-phosphorothioate analog. Thus, the chimeric oligonucleotide could provide a longer duration of action as an antisense agent after its administration.

Transformation of Pleurotus sapidus protoplasts by electroporation.

The total DNA of Pleurotus ostreatus was successfully transferred into the protoplasts of monokaryotic mycelia of Pleurotus sapidus by electroporation. The transformants were selected by their clamp connection of dikaryocyte, and the transformation frequency and transformation rate were 8.2 x 10(-5) and 3.6%, respectively. Analyzing the esterase isozyme, it was found that the transformants showed some isozyme bands of P. ostreatus and P. sapidus, and new non-parental bands. This proved that the recombination occurred between the DNAs of P. sapidus and P. ostreatus. The fruit bodies of the transformants showed morphological changes.

[The PCR amplification, cloning, sequencing, expression in E. coli of gene encoding endoflagella subunit protein (fla B) from Leptospira interrogans serovar lai].

A pair of oligonucleotide primers were designed by ourselves to amplify the endoflagella gene of L. interrogans serovar lai. A fragment about 840 bp was generated with PCR and inserted into plasmid pUC8 after the fragment and pUC8 were digested respectively with Bam HI and Pst I. A recombinant plasmid (designated as pLF1) was obtained. SDS-PAGE analysis indicated that a 33 kd was expressed in E. coli JM103 harboring pLF1 and the expression level of the protein was 11% of total bacterial soluble proteins. Western blot analysis showed that the protein band could be recognized by the antiserum against the endoflagella (Axiall filament) of Leptospira interrogans serovar lai. Nucleotide seguence data showed an open reading frame encoding 282 aminoacids residues, corresponding to a protein of molecular weight 33.6 kd. The G + C content of endoflagella subunit protein gene was 48 mol%. Therefore, the G + C content of the leptospiral fla B Gene is significantly higher than the reported 39 mol% G + C content of leptospiral genome of L.interrogans serovar lai but similar to the G + C of the Treponema pallidum genome. Comparison of the deduced endoflagellar subunit protein (fla B) amino acid sequence with flagellins from other bacteria revealed a high level of identity with the Treponema pallidum fla B proteins. Immunization/protection experiment was performed on the model of BALB/c mice and showed that the survival rate in the group JM103-pLF1 was higher than that in the group JM103-pUC8, but statistically the difference between them was significant (P < 0.05) and pLF1 did not induce significant levels of agglutinating antibodies against L.interrogans serovar lai.

[Gene expression of bFGF and intimal hyperplasia of autologous vein grafts in rats].

OBJECTIVE: To establish a model of autologous vein graft through translating jugular vein to aortic artery in 80 Wistar rats, and detect gene expression of basic fibroblast growth factor (bFGF). METHOD: In situ hybridization and reverse transcription polymerase chain reaction (RT-PCR) were used. RESULTS: The expression of bFGF mRNA was more decreased than that of normal artery and vein after transplantation, up to 1 week, the level of bFGF gene expression decreased to the lowest (P < 0.05). Two weeks after transplantation, bFGF transcripts recovered. CONCLUSION: bFGF released from dead and damaged cells of autologous vein graft not only is an important factor that promotes SMC proliferation, but also is a potent stimulator of endothelial regrowth.

[Anatomic observation of the dorsal metacarpal arteries].

Anastomosis between the first to the fourth dorsal metacarpal arteries was observed in 43 hands of adult corpses. It was found that 86% of the first dorsal metacarpal artery arises from the radial artery, 14% is replaced by the main artery of thumb or superficial branches of the radial arteries; eighty-six percent of the second dorsal metacarpal artery originates from the dorsal carpal artery. Sixty-seven percent of the third and the fourth dorsal metacarpal arteries is formed by anastomosis of the dorsal carpal artery and its deep palmar arch. The first to the fourth dorsal metacarpal arteries have 4 to 8 cutaneous branches and proximal and distal anastomoses. It is concluded that the dorsal metacarpal arteries are characterized with multiple sources and abundant anastomoses around the metacarpophalangeal joints, which can be used with advantage in the repair of soft tissue defects of the area.

[Asphyxia of the newborn delivered by elective cesarean section and gastroesophageal reflex and aspiration].

OBJECTIVE: To analyse the causes, clinical manifestation, management and prevention of gastroesophageal reflex and aspiration (GER-A) in the infants delivered by cesarean section. METHOD: The clinical data of 6 infants with asphyxia resulted from GER-A, delivered by elective cesarean section were retrospectively analysed. RESULTS: GER-A occurred during the period from 5 to 12 minute after birth, and its main clinical manifestation was airway obstruction. Among the patients, four cases recovered completely after treatment, one was diagnosed as having aspiration pneumonia and the other died of severe bronchospasm. CONCLUSIONS: The occurrence of this complication was probably related to the mode of delivery by cesarean section, the underdevelopment of effective barrier for preventing GER-A at very early stage and the inadequate management of the newborn. It is suggested that the prevention and proper treatment of GER-A was important in lowering neonatal morbidity and mortality.

[Research on the recombinant plasmid pDJH2 of L. interrogans serovar lai: sequencing and alignment with other known bacterial Omp sequence].

The Leptospira whole cell vaccine (LWCV) currently used in China is safe and effective, out the immunity following vaccination with two doses of the fluid medium vaccine is of low order. The duration of immunity conferred by this vaccine is rather short, six months or at most one year. Therefore, it is necessary to develop new generation vaccines against Leptospirosis for the developing world. In this paper we report the sequencing of the insert fragment of pDJH2 from genomic DNA of L. interrogans sevovar lai strain 017 and its alignment with other bacterial omp sequences. A genomic library of Leptospira interrogaans serovar lai strain 017 was constructed with the plasmid vector pUC18. A recombinant plasmid designated pJDH2 was screened from the genomic library. Inserted fragment of pDH2 is 1.9 kb by gel electrophoresis. Immunization/protection was studied in BALB/c mice model. The results showed highly significant difference between pDJH2 and pUC18 (control). Inserted fragment of pDJH2 DNA sequencing was performed by Dr Yan Zhengxin (Max-Planck-Institut for Biology. Tubingen, Germany). Insert fragment was cloned into pBluescript II KS-(stratagene) and sequenced by using AB1 (Applied Bio Systems, Model 373A). Two open reading frames of 565 and 662 nucleotides were identified. There were identifiable initiation codons, terminators, Shine-Dalgano ribosome combining site, Pribnow boxes and Sextama boxes within the 2 sequenced regions. Nucleotide sequences were analysed using Gene Work, a suit of computer program developed by Department of Biochemistry St. Jude Children's Research Hospital Memphis. U.S.A. The results of formatted alignment showed the predicted nucleotide sequence of ORF1 of the serovar lai had significant similarity with ORF2 (49.36%). L. kirschneri ompL1 (49.26%), Borrelia burgdoferi omp (48.97%), Treponema phagedenis omp (47.3%); Salmonella typhimurium ompC(46.87%), Yersinia enterocolitica ompH (46.7%), Leptospira borgpeterseni pfap (46.3%), and Serratia marcescens omp (43.3%). The close relationship of the pDJH2 ORF1 and ORF2 nucleotide sequences from Leptospira kirschneri ompL 1 is apparent. Whether the recombinant pDJH2 will prove useful for vaccine development remains to be tested.

[Subclones of fragment DNA recombinant rpDJH2 of L. interrogans serovar lai strain 017 and it's expression of high level in E. coli].

Fragment of 1.9 kb recombinant DNA of pDJH2 was linked with vectors pT7-7 and pRSETs. Then they were transformed into E. coli JM109 (DE3) respectively. Expression of subclones was achieved in E. coli JM109 (DE3) with IPTG inducement. SDS-PAGE showed that the molecular weights of products were 68kd and 23 kd respectively. The amount of production seemed to be higher than that of the outer membrane proteins of L. interrogans serovar strain 017 in nature. Immunoblotting of pDJt and pDJrB2 (both are subclones) with the specific antiserum of anti-OMP of L. interrogans serovar lai strain 017 and the experiment of initiative immuno-protection in guinea pigs showed both protein-68 kd and 23 kd might be the antigens of immuno-protection on the outer membrane of L. interrogans serovar lai strain 017.

[Mannital induced acute renal failure].

14 cases of mannitol-induced acute renal failure were reported. The dosage of mannitol used varied widely. In all cases serum Na+, HCO3- were decreased, K+ and BUN increased significantly. Serum osmolality was measured in 5 cases. The osmolal gap was increased greatly, 77.4mOsm/kg. H2O in average. The increase of osmolal gap may play an important role in acute renal failure by causing intensive renal vasocontraction. Monitoring of serum osmolality or osmolal gap can help to prevent mannitol intoxication. The decrease of serum Na+ may be a warning sign of increased osmolal gap. Hemodialysis is the best way for the treatment of mannitol-induced acute renal failure.

Cardiovascular effects of total soyasaponin in central nervous system and its relationship with monoamine transmitters.

AIM: To study the cardiovascular effect of total soyabeans saponins (TS) in brain and its relationship with monoamines. METHODS: After injection of TS (75 micrograms) into ventriculus lateralis cerebri (VLC) the changes of blood pressure (BP) and heart rate (HR) were observed and the contents of monoamines both in peripheral blood and brain (telencephalon, diencephalon, brainstem) were measured respectively by HPLC-ECD and fluorophotometry. RESULTS: After injection of TS into VLC, BP rise from 11.59 +/- 0.84 to 14.59 +/- 0.69 kPa; HR increased from 411 +/- 21 to 465 +/- 14 bpm; the contents of NE and E in peripheral blood increased from 6 +/- 3 to 64 +/- 44, from 6 +/- 2 to 38 +/- 34 nmol/L plasma, respectively, NE in brainstem increased from 14 +/- 0 to 18 +/- 3 nmol/g wet tissue respectively, but the contents of 5-HT in the 3 areas measured in the experiment decreased: in telecephalon from 9 +/- 1 to 5 +/- 1, in diencephalon from 14 +/- 2 to 7 +/- 2, in brainstem from 14 +/- 3 to 6 +/- 1 nmol/g wet tissue. CONCLUSIONS: The cardiovascular effects of TS in CNS were involved in the monoamine transmitters.

Self-association of CPV3, an avian thymic parvalbumin.

The avian parvalbumin called CPV3 readily forms disulfide-linked oligomers. Sedimentation data presented herein reveal that CPV3 also undergoes noncovalent self-association. Interestingly, the noncovalent interaction is promoted by either Ca2+ or Mg2+, whereas covalent complex formation displays an absolute requirement for the Ca(2+)-bound protein. Apo-CPV3 exhibits an apparent sedimentation coefficient of 2.08 S at 20 degrees C, in 0.15 M NaCl, 0.025 M HEPES-NaOH, pH 7.4. This value increases to 2.85 S or 3.16 S with addition of 1.0 mM Ca2+ or 5.0 mM Mg2+, respectively. Least-squares analysis of sedimentation equilibrium data suggests that 100 microM apo-CPV3 is primarily a mixture of monomeric and dimeric forms. With the addition of Ca2+, the equilibrium becomes exclusively monomer-trimer, with negligible amounts of dimer. A comparable distribution is observed in the presence of Mg2+.

CD14 enhances cellular responses to endotoxin without imparting ligand-specific recognition.

Binding of the lipid A portion of bacterial lipopolysaccharide (LPS) to leukocyte CD14 activates phagocytes and initiates the septic shock syndrome. Two lipid A analogs, lipid IVA and Rhodobacter sphaeroides lipid A (RSLA), have been described as LPS-receptor antagonists when tested with human phagocytes. In contrast, lipid IVA activated murine phagocytes, whereas RSLA was an LPS antagonist. Thus, these compounds displayed a species-specific pharmacology. To determine whether the species specificity of these LPS antagonists occurred as a result of interactions with CD14, the effects of lipid IVA and RSLA were examined by using human, mouse, and hamster cell lines transfected with murine or human CD14 cDNA expression vectors. These transfectants displayed sensitivities to lipid IVA and RSLA that reflected the sensitivities of macrophages of similar genotype (species) and were independent of the source of CD14 cDNA. For example, hamster macrophages and hamster fibroblasts transfected with either mouse or human-derived CD14 cDNA responded to lipid IVA and RSLA as LPS mimetics. Similarly, lipid IVA and RSLA acted as LPS antagonists in human phagocytes and human fibrosarcoma cells transfected with either mouse or human-derived CD14 cDNA. Therefore, the target of these LPS antagonists, which is encoded in the genomes of these cells, is distinct from CD14. Although the expression of CD14 is required for macrophage-like sensitivity to LPS, CD14 cannot discriminate between the lipid A moieties of these agents. We hypothesize that the target of the LPS antagonists is a lipid A recognition protein which functions as a signaling receptor that is triggered after interaction with CD14-bound LPS.
