The effect of dihydrotestosterone and culture conditions on proliferation of the human prostatic cancer cell line LNCaP.

Cell density, nutritional state, and serum factors modify the growth response of LNCaP human prostatic cancer cells to dihydrotestosterone. Evaluation of growth response to dihydrotestosterone requires logarithmic transformation of cell count or thymidine incorporation data. Under conditions of dose response, growth increases with cell density but no significant interaction of dihydrotestosterone with cell density was found under optimal culture conditions. The frequency of media change was a significant factor in modulating dose response. When cells from cultures maintained at different feeding periods were plated at different cell densities of (trypan blue) viable cells, significant effects of plating density on dihydrotestosterone response were found. Dihydrotestosterone protects cells under the adverse effects of media deprivation. Under the extreme adverse effects of serum deprivation, cells respond to dihydrotestosterone even under conditions of increasing cell loss. The effects of dihydrotestosterone on final cell density were significant. In the absence of serum, the elongated cells of LNCaP assume a round shape, but many remain adherent to the culture dish and can be restored to normal morphology by serum. A number of growth factors fail to restore normal morphology that was completely restored by a combination of fibronectin and dihydrotestosterone. We have not developed a practicable serum-free system for LNCaP.

Pituitary extract causes aggregation and differentiation of rat mammary tumor MTW9/Pl cells.

Alkaline pituitary extracts (PE) of the mammosomatotropic tumor MtTW10 differentiated cultured PRL-independent rat mammary tumor cells MTW9/Pl. Within 24 h, MTW9 cells growing in suspension began to aggregate and adhere to the plastic dish. Cultured dispersed MTW9 cells were undifferentiated, but PE treatment resulted in organoid aggregates that exhibited glandular luminal structures and periodic acid-Schiff-positive material consistent with basement membrane formation. Morphometric examination of organoids demonstrated a reduction in nuclear and cell perimeters compared to those of untreated cells. Electron microscopy showed that the treatment resulted in polarization, nuclear changes, junctional complexes, secretory spaces, microvilli, and basement membrane formation. A disaggregated undifferentiated tumor now appeared as a differentiated adenocarcinoma. PE induced expression of laminin and milk protein, but failed to increase fibronectin expression. Extracts of MTOM (a variant of MtTW10 which secretes little PRL) and bovine pituitary also produced aggregation and adhesion of MTW9. A number of tissue extracts, growth factors, and hormones failed to produce such aggregation. Laminin, but not fibronectin, produced aggregation and adhesion similar to those produced by PE. Cycloheximide inhibited the aggregation effect of PE, but not that of laminin. PE was mitogenic for MTW9, but inhibition of proliferation by vinblastine did not inhibit the aggregation induced by PE. These observations suggest that the pituitary contains a novel factor that stimulates matrix synthesis, resulting in differentiation, possibly laminin induced.

Effect of verapamil on acute coxsackievirus B3 murine myocarditis.

The effect of verapamil (Ver) on CVB3 murine myocarditis was investigated. It was found that Ver could aggravate the myocardial inflammation, increase the viral replication in myocardium, and raise mortality in mice with viral myocarditis when the drug was injected within the first 6 days after the CVB3 inoculation.

Tumors of the heart. An analysis of 79 cases.

There were 79 cases of cardiac tumors seen from 1957 to July 1988. 49 (62.0%) of them were benign and 30 (38.0%) malignant. All the 49 benign tumors except 2 were surgically excised and found to be myxoma. Of them, 18 patients were male and 31 female. 85.7% of the tumors were located in the left atrium, 12.2% in the right atrium and 2.0% in the left ventricle. Palpitation, dyspnea, chest oppression, fever, episodes of syncope and hemiplegia, cough, diastolic and systolic murmurs at the apical or tricuspid area were the common symptoms and signs. Atrial fibrillation was found only in 2 cases. Echocardiographic findings were diagnostic while ECG and X-ray findings were nonspecific. Four patients died after operation. Of the 30 cases of malignant tumors, 15 were secondary tumors metastasized mainly from the lung or mediastinal malignancies. Of 11 primary tumor cases (7 males and 4 females), 3 were malignant lymphoma, 2 mesothelioma of pericardium, 2 malignant myxoma, 1 angiosarcoma, 1 leiomyosarcoma, 1 fibrosarcoma and 1 rhabdomyosarcoma. Another 4 cases were not studied histopathologically. The clinical manifestations, ECG and X-ray findings of the 11 primary tumors were nonspecific but echocardiography was helpful to the diagnosis. Six patients were operated on and 1 died during hospitalization.

Effect of dexamethasone on Coxsackievirus B2-infected rat beating heart cells in culture.

The effects of dexamethasone on cultured rat beating heart cells experimentally infected with coxsackievirus B2 were investigated at an early stage (1-3 days) post challenge. Changes in the release of the cardiac enzyme, aspartate amino-transferase (AST), beating %, cytopathic effect (CPE), virus titre, ultrastructure and alterations of the electrical activity were evaluated. The protective effects of dexamethasone on infected cells were abolished except the release of AST at 5 days post infection. These results suggest that steroids may benefit patients with severe myocarditis if conventional therapy for protection of the myocardium and modulation of immunologic function are concomitantly given.

[Influence of dexamethasone on electrical activities in cultured rat beating myocardial cells infected with Coxsackie B-2 virus].

The effects of dexamethasone (Dex) on electrical activities in cultured rat beating myocardial cells infected with 100 TCID-50 Coxsackie B-2 virus (CB2V) was evaluated by conventional intracellular microelectrode technique. The frequency began to increase, the beating % decreased, and multiform arrhythmias were shown in the infected group 24 h post-challenge. Meanwhile, the cytopathic effect (CPE) appeared rapidly from 1+ to 3+. In the infected and Dex-treated group, the beating % was higher and the arrhythmias and CPE were less than in the infected group at the same intervals. The numbers of non-beating cells increased parallel to the incubation time in the infected group. Decreases of maximal diastolic potential (MDP), maximal upstroke rate (Vmax), overshoot (OS) and action potential amplitude (APA), and abbreviation of action potential duration (APD50 and APD100) in infected and Dex-treated group were less than those in control group during 24-96 h post-challenge. Premature beats, tachycardia, bradycardia and fibrillation occurred in the early stages after infection. It is surmised that steroids can probably save the lives of patients with severe myocarditis if Dex was supplemented.

Changes in plasma levels of tissue plasminogen activator and its inhibitor in aged myocardial infarction patients.

Plasma levels of tissue plasminogen activator (tPA) antigen, tPA activity and plasminogen activator inhibitor (PAI) activity were determined in 40 cases of acute myocardial infarction (AMI) at the onset and 2-4 weeks after the onset of the disease. 24 healthy subjects served as controls. At the onset of AMI, mean plasma level of tPA antigen was significantly higher than that of control subjects (13.5 +/- 4.8 ng/ml vs 6.4 +/- 2.0 ng/ml), with the highest elevation in the group greater than 65 years of age. Mean plasma level of tPA activity was significantly lower than in the controls (1.1 +/- 0.4 IU/ml vs 1.6 +/- 0.4 IU/ml), especially in patients over 65 years of age. Mean plasma level of PAI activity was significantly higher than in the controls (17.5 +/- 12.0 AU/ml vs 9.0 +/- 4.6 AU/ml), especially in patients over 65 years of age. At 2-4 weeks, mean plasma levels of tPA and PAI activities of AMI patients had dropped to 1.5 +/- 0.6 IU/ml and 9.5 +/- 8.9 AU/ml, respectively, not significantly different from controls. The tPA antigen had come down to 11.0 +/- 5.4 ng/ml, but still higher than in the controls. The possible mechanism of these phenomena was discussed.

Treatment of experimental Coxsackie B-3 viral myocarditis with Astragalus membranaceus in mice.

A murine model system for observing the effect of Astragalus Membranaceus (AM) on experimental myocarditis caused by Coxsackie B-3 virus (CB3V) was developed in 4-week-old male BALB/C mice. Gross, histopathologic and ultrastructural examinations of the infected-AM treated group showed that the severity and involved area of the myocardial lesions became milder and smaller than those in the infected-NS treated mice. The total lesion area, and the total lesion area/total myocardial area examined (%) and virus titer in the former group were also smaller and lower than those in the latter group. The results suggest that AM is effective in the inhibition of Coxsackie B virus propagation and protection of myocardium in mouse myocarditis.

Effect of astragalus membranaceus on electric activities of cultured rat beating heart cells infected with Coxsackie B-2 virus.

Astragalus membranaceus (AM) which has a protective effect on rat beating heart cells infected experimentally with Coxsackie B-2 virus was evaluated on the basis of changes in morphologic and electric activity of the cells. Rhythm, beating frequency, beating percentage, cardiac cellular damage and cytopathic effects (CPE) were monitored every 24 h after challenge; electric activities parameters were measured by conventional intracellular microelectrode technique. Significant protective effects were demonstrated when AM was given in the early period of infection. The results suggest that AM should be valuable in preventing and treating acute myocarditis caused by Coxsackie B virus.

[Studies on the systematic status of a new species of Drosophila (sophophora)--from morphological to molecular level].

Bock and Wheeler (1972) reviewed the Drosophila melanogaster species group in which more than 70 species were classified into a number of subgroups. One of them is the takahashii species subgroup which is a large subgroup with 12 species. We collected many specimens of drosophilid fly in Mt. Huaping, Guangxi Province and Mt. Siming, Zhejiang Province in 1983, among which taxon A and taxon B were thought to be a new species, belonging to the takahashii subgroup and named as D. liui sp. n. However, after Prof. T. Okada examined this new species, he thought it is the same as the known species D. nepalensis Okada, 1955 from Nepal and later Dr. J. Toda examined the same species and thought it was the known species D. prostipennis Lin, in Bock and Wheeler 1972 from Taiwan and/or D. trilutea Bock and Wheeler 1972 from Borneo, so that D. liui sp. n. is considered as the species incertae sedis. In the present paper, we studied 6 species of this subgroup: D. takahashii, D. lutescens, D. liui sp. n., D. nepalensis, D. prostipennis and D. trilutea. Our studies from morphological to molecular level provided the evidences which can support the description of a new species, hence we draw the conclusion that D. liui sp. n. is a new member of the takahashii subgroup.

Two nearby sites bind zen protein independently.

The region upstream from the zerknullt (zen) gene contains three sites that specifically bind the zen protein product of the gene. Evidence for these binding sites was obtained by the filter binding technique and the DNase footprinting technique. The filter binding technique was used to scan various segments of DNA for the presence of possible specific binding sites. Segments that were selectively retained by the filter binding technique invariably contained one or more specific binding sites according to the DNase footprinting technique. Two of the zen protein binding sites were spaced only 30 base pairs apart. These sites could be separated without any loss in their specific binding properties. It is concluded that these two sites function independently in the binding of zen protein.

Genetic differentiation in populations of Drosophila melanogaster from the Peoples' Republic of China: comparison with patterns on other continents.

Geographic variation at eight allozyme loci in eight natural populations of Drosophila melanogaster from the People's Republic of China (P.R.C.) was collated with data from Japanese populations and compared with previous reports for other continents. G6pdF, Est-6(1.00) and AdhS were significantly correlated with latitude. Whilst the variation in Est6(1.00) was opposite to that previously reported, AdhS showed latitudinal clines consistent with data from the northern and southern hemispheres. A thermostable variant, AdhFChD, was found at high frequency in the southern P.R.C. populations and it is suggested that the mutation occurred in this region and was then dispersed to other continents.

Rapid purification of creatine kinase MB isoenzyme on preparative polyacrylamide slabs.

We describe a procedure for purification of creatine kinase (EC 2.7.3.2) MB isoenzyme (CK-MB) from human cardiac muscle by preparative electrophoresis on polyacrylamide gel. From a 50-g portion of human myocardium we isolated 21 mg of CK-MB, which had a specific creatine kinase activity of 405 kU/g. The CK-MB exhibited an enzyme band on polyacrylamide gel electrophoresis (PAGE) with staining for enzyme activity. The preparation showed single protein bands on sodium dodecyl sulfate--PAGE, pore gradient electrophoresis, and isoelectric focusing electrophoresis, the relative molecular masses of the subunit and of the whole enzyme being 43,000 and 86,000, respectively, and the isoelectric point being 5.0. In addition, the purified CK-MB showed desirable immunological specificity and affinity (K = 2.4 x 10(11) L/mol) when measured by radioimmunoassay.

The relationship between serum lipid levels and nutrient intake in healthy inhabitants of urban and rural Shanghai.

The serum lipid levels (mean +/- SD) of 3312 healthy urban and rural inhabitants over 40 years of age in Shanghai were studied. The results were: cholesterol 166.7 +/- 30.9 mg/ml, triglyceride 81.1 +/- 42.3 mg/dl, HDL-cholesterol (HDLc) 57.4 +/- 11.6 mg/dl, HDL2c 20.6 +/- 9.8 mg/dl and HDL3c 37.1 +/- 7.8 mg/dl. Dietary surveys were carried out in 512 of these subjects and their average daily nutrient intake was estimated to be: total calories 2,577 Kcal, proteins 656 g (50.7 g of vegetable proteins), fat 69.1 g (P/S ratio 0.70), carbohydrates 423 g, cholesterol 282 mg, dietary fibres 23.5 g, sodium 4993 mg, potassium 2,618 mg and magnesium 575 mg. The serum lipid levels of urban and rural inhabitants were compared. Total cholesterol levels were significantly higher in urban inhabitants of all age groups and of both sexes, while triglyceride levels were significantly higher in urban inhabitants of certain age groups. However, HDLc levels, particularly HDL2c levels were significantly higher in rural inhabitants of most age groups and of both sexes. The above differences were probably the result of differences in dietary patterns and nutrient intake between urban and rural inhabitants.

[Effect of Astragalus membranaceus on electrical activities of coxsackie B-2 virus-infected rat myocardial cells in culture].

Beating Myocardial cell cultures of neonatal rats were prepared in vitro and infected with coxsackie B-2 virus. The cells were evaluated in the post-infected period for changes in beating percentage and cytopathic effect (CPE), alterations in the electrical activities by standard microelectrode techniques, and the protective effect of Astragalus membranaceus (AM) on coxsackie B-2 virus-infected neonatal rat myocardial cell cultures was observed. The beating percentage began to decrease in the infected group at 24 hr and only 27.9 +/- 18.6% was beating at 96 hr after virus challenge, premature beats, tachycardia and fibrillation occurred commonly during the experiment. Meanwhile the CPE appeared rapidly from 1+-3+ at the same interval. Resting potential, action potential amplitude, duration and rate of uptake were shown a significant decrease through 24-96 hr (P less than 0.01). In contrast, the beating and electrical activities were nearly normal and less CPE was shown in myocardial cells treated with AM 1 hr after virus challenge through 24-96 hr (P less than 0.05). The results suggest that AM may be valuable in prophylaxis and treatment of acute coxsackie B-2 virus caused myocarditis.
