Isolation and characterization of the dnaKJ operon from Actinobacillus actinomycetemcomitans.

The dnaKJ operon of Actinobacillus actinomycetemcomitans Y4 was cloned by the DNA-probing method using synthetic oligonucleotides designed on the basis of two conserved regions in DnaK/hsp70 proteins and sequenced by inverse PCR. The sequenced region was shown to contain two open reading frames coding for proteins analogous to DnaK and DnaJ.

[The experimental study on absorption and utilization of calcium from oyster shell powder mixed with fruit juice].

A new kind of calcium powder was prepared from the shell of oyster and the bones of other sea lives, in which fruit juice was mixed. This calcium powder and other four different forms of calcium (i.e., active calcium, gluconate, lactate, and phosphate) were fed to 4 groups of Wistar rats for 30 days. 3 days calcium metabolism test was done. The absorption and retention rates of calcium powder were 69.2% and 95.1% in the experimental group, which were higher than those in other calcium supplement groups (P < 0.05). The weight of femurs and the total calcium content of femurs had significantly increased in calcium powder group (P < 0.05). These findings indicated that calcium from calcium powder, which was prepared from sea lives and mixed fruit juice was more effective.

[Management of acute arterial occlusion of limbs: an analysis of 105 cases].

105 cases of acute arterial occlusion of limbs were admitted to our hospital from march 1982 to September 1996. We compared the treatment results of the patients with acute embolism and acute thrombosis. The limb-salvage rate of 86.8% in patients with embolic occlusion was higher than that of 57.9% in patients with thrombotic occlusion (P < 0.01), while the mortality rate of 8% patients associated with thrombosis was lower than that of 15.7% with embolism (P < 0.01). The results of limb salvage and mortality were related to different etiology of acute arterial occlusion, embolism and thrombosis.

[Therapy for 29 sarcoma of the breast].

We discuss the best therapeutic method for sarcoma of the breast. The data of all cases treated from March 1964 to May 1994 were analyzed retrospectively. Of all cases, 7 died, 9 recurred, and 15 survived more than 15 years and 5 over 20 years. The follow-up time was 4 months to 25 years. Most sarcomas were pathologically proved. The best therapeutic method was operation. The sarcomas of all cases must be extensively resected including surrounding tissues. The recurrent sarcoma should be resected again. The axillary lymphnodes should not be eliminated when lymphnodes is not discovered.

[The vascular pattern of the endolymphatic duct, endolymphatic sac and its anatomic differences in guinea pigs].

In order to study the vascular pattern of the endolymphatic duct and sac, endolymphatic duct and sac were examined with vascular Indian ink injection and image analysis. The Results were as follows: 1. In the 20 temporal bones, 17 (85%) had posterior meningeal artery (PMA) and posterior vestibular artery (PVA) supply and the rest (3 specimens, 15%) had no PVA supply; 2. The distribution frequency of PMA in the endolymphatic sac was much higher than that of PVA(P < 0.01), but the distribution of PMA and PVA in the endolympatic duct were not different (P > 0.05). The conclusions is that there are anatomic differences in vascular supply and pattern of the endolymphatic duct and sac, PMA is the main vascular structure in the endolymphatic sac.

[Correlativity among biological characteristics of Gardenia jasminoides Eills].

The relationship among certain biological characteristics, fruit yield and quality of G. jasminoides was studied, and the special features, closely correlateied with fruit production and quality, as well as with the steadiness of heredity were found out. According to these features the G. jasminoides produced in Sichuan province has been divided into 6 types.

[Influence of sulphur smoking on the content of coumarins in the root of Angelica dahurica (Fisch. ex Hoffm.) Benth. et Hook.f.var. formosana (Boiss) Shan et Yuan].

The content of coumarins in the root of Angelica dahurica (Ad) treated in different time spans with smoke of sulphur was determined by HPLC. The result showed that in nine hours before Ad was smoked thoroughly the content of coumarins reduced rapidly with the increase of time, and there after, the reduction was not obvious. After being treated for nine hours, the total coumarins were reduced by 75.3% and after forty hours the ratio reached 77.9%.

[Separation and determination of flavonols in the flowers of Carthamus tinctorius by RP-HPLC].

Carthamus tinctorius L. is a kind of traditional Chinese medicine used in the treatment of uterine congestion, cardio-vascular disease, thrombosis and so on. Eight flavonoids have been isolated from its flowers. A reversed phase HPLC method for the separation and determination of the three flavonols: rutin (I), quercetin(II) and kaempferol(III) in the flowers of Carthamus tinctorius L. is described. The column was 25 cm x 4.6 mm ID, packed with YWG-C18(10 microns). The mobile phase was a mixture of MeOH-H2O-H3PO4(48.5:51.5:0.25, pH 3.5). The flow rate was 1.0 ml.min-1. Daidzein was used as an internal standard. UV detection was performed at 360 nm. The relative standard deviation of the method was 1.1-2.6%. The calibration curves showed good linearity over the range of 0.11-0.80 microgram, gamma = 0.9995-0.9998 and detection limits were 2.0-20.0 ng. The recoveries were 97.8%-98.9% for the three flavonols. The method is simple, fast, sensitive and accurate and has been applied to the analysis of Carthamus tinctorius L. of different sources. It can also be used for the quality control of safflower preparations for clinical uses.

Pulmonary leiomyosarcoma.

Three cases of pulmonary leiomyosarcoma were presented. The characteristic clinical features were described with review of literature. In comparison with bronchogenic carcinoma, the leiomyosarcoma has some characteristics: 1) On chest X-ray, it usually appears as a sharply demarcated, even density round mass, growing rapidly within the lung, it unusually accompanies with hilar or mediastinal lymph node metastasis; 2) The preoperative cytological or pathological diagnosis is difficult either by sputum smear or by bronchoscopic biopsy or by fine needle percutaneous aspiration biopsy; 3) Pathological differential diagnosis of leiomyosarcoma of lung from anaplastic lung cancer is difficult. In conclusion, the primary pulmonary leiomyosarcoma is a rare malignant tumor, detecting the present illness seriously, paying attention to the chest X-ray films characterize, early surgical resection is the only way to get diagnosis and effective treatment method.

Diagnosis and treatment of stage-III peripheral squamous cell carcinoma of right upper lung.

During a 12-year period, 44 patients were surgically treated for stage-III peripheral squamous cell carcinoma of the right upper lobe. The mean time from the discovery of the shadow in the right upper lobe to the confirmation of the diagnosis was 2 months. All patients underwent the resection of the right upper lobe with the dissection of the right mediastinal lymph nodes, among them, 4 patients had sleeve lobectomy of the right upper lobe. P-TNM staging: 23 cases were in stage-IIIa, 21 cases in stage-IIIb. The frequency of the postoperative complications was 20.4% (9/44 cases). The 1-year survival rate of all patients was 61% (27/44 cases). None of stage-IIIb patients lived longer than 3 years. The postoperative 3- and 5-year survival rates of stage-IIIa cases were 65.2% (15/23 cases) and 21.7% (5/23 cases) respectively. In this article, the patients' clinical features, the causes of delayed-diagnosis, the operative and postoperative management were discussed. We suggest that the stage-IIIa patients should be given surgical treatment, whereas, for the patients of stage-IIIb, palliative operation was given just for the purpose of reliving the symptoms.

[Clinical and pathological study on liver fibrosis in viral hepatitis].

49 patients with viral hepatitis were studied on the correlationship between liver fibrosis and the level of serum precollagen type III (PC III), pre-collagen III pre-peptide (P III P), Laminin (LN), hyaluronic acid (HA) and collagen type IV (IV-C), as well as relationship between them and the expression of LN, IV-C, a-smooth muscle actin (a-SMA) and cytokeration (CK) in the liver tissues examined by immunohistochemistry. Results showed that the level of PC III, P III P, LN, HA and IV-C in the groups of liver cirrhosis and chronic severe hepatitis were higher than that in the groups of acute and chronic hepatitis. High correlation was found between the level of these markers (especially the level of the IV-C) and the degree of fibrosis in the group of chronic hepatitis. So were the expression of LN, IV-C, a-SMA and CK in the liver tissue. The results also showed that the liver fibrosis correlated with inflammatory reaction in the liver. So the authors suggested that more attention should be paid to the inflammatory reaction of the liver for prevention and treatment of liver fibrosis.

[Flame atomic absorption spectrometry determination of zinc and ferrum in enamel].

The amount of zinc and-ferrum in enanmel was determined by flame AAS. For twenty six healthy men, the average of zinc and ferrum were 325.6 +/- 84.8 and 34.7 +/- 9.1 microg/g, respectively. For fifty four patients with dental caries disease, the average of zinc and ferrum were 269.9 +/- 63.5 and 38.7 +/- 10.0 microg/g, respectively. The difference between healthy men and patients of zinc and ferrum rate was insignificant. As to the determination method, the rate of recovery varied from 97.0 to 101.5% for zinc and from 97.2 to 102.4% for ferrum. The RSDs fo Zn and Fe are less than 2.60 and 2.10% respectively.

Cholesterol deposition in macrophages: foam cell formation mediated by cholesterol-enriched oxidized low density lipoprotein.

Oxidized low density lipoprotein (LDL) is thought to mediate the transformation of macrophages to cholesterol-rich foam cells. Yet convincing evidence for this process is lacking in vitro. We suggest that oxidized LDL-mediated foam cell formation is not seen in vitro because the cholesteryl ester content of LDL particles (oxidized in the presence of transition metals) is dramatically reduced. Thus, if oxidized LDL could be cholesterol-enriched prior to its addition to macrophages, this lipoprotein would be made more capable of inducing the cellular deposition of cholesteryl esters. When we enriched cupric sulfate-oxidized LDL with cholesterol by incubation of this lipoprotein with unesterified cholesterol/phosphatidylcholine liposomes and added it to mouse peritoneal macrophage cultures, we found that: a) the enrichment of oxidized LDL with cholesterol did not alter the extent of oxidized LDL degradation; b) the cells accumulated massive amounts of cholesteryl ester (148 microg/mg cell protein) and unesterified cholesterol (260 microg/mg cell protein) after 24 h of incubation; and c) Sephacryl S-1000 chromatography of the cholesterol-enriched oxidized LDL verified the formation of large oxidized LDL-unesterified cholesterol/phosphatidylcholine complexes. These results demonstrate that oxidized LDL, when cholesterol-enriched, can mediate the formation of macrophage foam cells in culture

Detection of prostate cancer relapse with prostate specific antigen monitoring at levels of 0.001 to 0.1 microG./L.

PURPOSE: The development of prostate specific antigen (PSA) assays with detection limits of approximately 0.001 microgram./l. is technically feasible. We examined if serum PSA changes of 0.001 to 0.1 microgram./l. for up to 3 years after radical prostatectomy have any clinical value. MATERIALS AND METHODS: We studied 148 patients with a postoperative PSA of less than 0.1 microgram./l. by a conventional PSA assay. At least 3 serial serum samples were collected per patient along with detailed clinicopathological features. Serial serum samples were analyzed for PSA with the ultrasensitive method. Associations between increase in serum PSA and clinicopathological features were analyzed with the unconditional logistic regression model. RESULTS: After establishing a set of interpretative criteria, we divided the patients into 51 with biochemical relapse, 93 who were free of relapse and 4 with equivocal status. Between the groups with and without relapse there was no difference in year of surgery, age at operation or length of followup. Compared to patients without relapse, those with biochemical relapse were likely to have positive surgical margins (p < 0.01), larger tumor volumes (p < 0.01), greater preoperative PSA (p = 0.03) and disease extending outside the prostate (p = 0.02). The relative risks for biochemical relapse estimated by a univariate logistic regression model were 3.1 (95% confidence interval 1.39 to 6.82, p < 0.01) for positive surgical margin, 3.4 (95% confidence interval 1.46 to 8.13, p < 0.01) for tumor volume, 2.3 (95% confidence interval 1.08 to 5.02, p = 0.03) for high preoperative PSA and 2.7 (95% confidence interval 1.12 to 6.26, p = 0.03) for extraprostatic tumor extension. At multivariate analysis with the same model the associations between positive surgical margins and biochemical relapse (relative risk 2.95, p = 0.04) and tumor volume (relative risk 3.36, p = 0.03) remained significant. These associations were still observed when we analyzed a subset of patients classified as having biochemical relapse based on PSA changes of 0.001 to 0.08 microgram./l. CONCLUSIONS: Increases in postoperative serum PSA at levels of 0.001 to 0.1 microgram./l. after radical prostatectomy are associated with clinicopathological features of poor prognosis. Monitoring postoperative cases with a highly sensitive PSA assay (detection limit 0.001 microgram./l.) could offer a simple and effective means of detecting clinically important biochemical relapse early after radical prostatectomy. These patients may be suitable for early intervention when effective treatments for relapse become available.

Matrix stimulates the proliferation of human corneal endothelial cells in culture.

PURPOSE: Extracellular matrices were tested for their ability to support the adhesion and proliferation of human corneal endothelial cells. METHODS: Human corneal endothelial cells were plated onto tissue culture dishes coated with purified fibronectin or a matrix elaborated by cultured bovine corneal endothelial cells. The presence of human cells in the cultures was confirmed by karyotyping. Cell size at increasing passage number was analyzed, and cellular response to growth factors was assessed using a 96-well microtiter plate assay. RESULTS: When tissue culture dishes were coated with fibronectin, the cells attached to the dish but grew slowly. Human corneal endothelial cells plated onto the matrices elaborated by bovine corneal endothelial cells attached to the culture dish and grew to fill the flask. At confluence, the cells had a hexagonal morphology similar to that seen in vivo. Karyotype analysis showed that the cells were of human and not bovine origin. The time required for senescence in culture was dependent on the age of the donor cornea. The bovine matrices enhanced the proliferative response of human corneal endothelial cell cultures to endothelial cell growth supplement and keratinocyte growth factor. Epidermal growth factor and hepatocyte growth factor stimulated human cell proliferation in a dose-dependent fashion, regardless of the substratum on which the cells were plated. CONCLUSIONS: The use of substratum elaborated by bovine corneal endothelial cells has proved useful in the preparation of human endothelial cell cultures from juvenile and adult donors. The method has been used to establish cultures from more than 50 donors from age 1 day to 76 years.

[Study on the recognition and evolutionary genetics of the courtship song of species in Drosphila nasuta species subgroup].

The subgroup of Drosophila nasuta consists of 14 species, subspecies and taxa. It has a wide distribution in area of Indo-Pacific Ocean. The courtship song of species in nasuta subgroup is recorded for the first time in the paper. Some parameters of temporal pattern of pulse song were measured: the inter burst interval (IBI), the interpulse interval (IPI), the length of a pulse train (PTL), the number of pulses in a train (PN), the length of a pulse (PL) and the length of a cycle (CL). Computer analysis techniques were used to make spectral analysis of sine song. A three-dimensional power spectra of the sound was made. In this subgroup, the male of D. pulauna and Taxon-F didn't produce any sound signals during courtship, suggesting that the visual stimuli play an important role in mating process. By analysing the courtship sound signals of other species, subspecies and taxa, the sounds produced by courting male could be described as pulse song or as sine song. By studying the sounds of intra- or interspecific F1 hybrids we have found that the parameters of the temporal pattern of pulse song, for example, the mean value of IPI is controlled by X-linked and autosomal polygene and the sine song frequency is predisposed to the maternity. The phylogenetic tree of the subgroup is constructed based on the temporal pattern of the pulse sounds in different species, subspecies and taxa. The relationship within the members of the Drosophila nasuta subgroup is discussed.
