Effect of co-administration of interferon inducer, polyriboinosinic acid-polyribocytidylic acid, with SKF 525-A on hepatic drug metabolizing enzymes of rats.

The protective effect of SKF 525-A on the suppression of cytochrome P-450 content and monooxygenase activities by treatment with CoCl2 and polyriboinosinic acid-polyribocytidylic acid [poly(I.C] was compared as a part of studies of suppression of drug metabolizing enzymes by interferon inducers. Induction of heme oxygenase activity by CoCl2 and poly (I.C) was not altered by simultaneous treatment with SKF 525-A. Depression of cytochrome P-450 content and benzphetamine N-demethylase activity by treatment with CoCl2 was prevented by co-treatment with SKF 525-A. This effect was explained by the prevention of release of heme from cytochrome P-450 by forming metabolic intermediate complexes with metabolites of SKF 525-A. On the other hand, poly(I.C) significantly suppressed P-450 content and benzphetamine N-demethylase and benzo [a] pyrene hydroxylase activities, even under simultaneous treatment with SKF 525-A. This inhibition by poly (I.C) was accompanied by weak staining of proteins corresponding to cytochrome P-450 in SDS gel electrophoresis. In addition, the activity of non-heme enzyme, 4-hydroxybiphenyl glucuronyltransferase, was suppressed by treatment with poly (I.C) but not by CoCl2-treatment. These findings strongly suggested that, unlike CoCl2, poly (I.C) suppressed cytochrome P-450 content and monooxygenase activities due to decreased synthesis or increased degradation of the apoprotein of cytochrome P-450 with slight contribution of the induced heme oxygenase.

[Effect of anxiolytic drugs on the development and healing process of stress ulcer: application of endoscopic observation].

The present study was conducted to investigate the effect of anxiolytic drugs on the development and healing process of gastric mucosal lesions (GML). In order to produce the GML in mice and rats, we employed a communication box paradigm which can differentially induce a physical and psychological stress. The following drugs (2.5, 50.0, 7.5 mg/kg for mice and 5.0, 10.0 mg/kg for rats) were administered orally: diazepam, clotiazepam, ethyl loflazepate and vehicle (0.5% CMC solution). To define the drug effects on the healing process of GML in rats, we applied an endoscopic technique using the ultra-thin fiberscope. The endoscopic observation was performed under ether anesthesia at 1, 24, 36 and 60 hr after the termination of stress. Both diazepam and ethyl loflazepate significantly suppressed the development of GML in mice, while the preventive effect of clotiazepam was less potent than those of the other anxiolytics. In rats, only ethyl loflazepate showed a significant preventive effect. The curative effect of clotiazepam on the healing process of GML in rats was clearly demonstrated, whereas diazepam was ineffective on the healing process. Ethyl loflazepate also showed a tendency to facilitate the healing process of GML. The present study suggests that the application of the endoscopic technique to rats may provide a potential tool for evaluating the curative effect of drugs on the GML.

[Effect of mazindol on glucose absorption in everted rat small intestine].

Effect of an anorexiant, mazindol, on glucose absorption was investigated. Ten weeks-old female Sprague-Dawley rats were divided into mazindol treated (fed on powder diet containing 100 mg/kg of mazindol) and control groups. Four weeks later, experiments of continuous observation of glucose absorption and glucose transport were performed in each group using the everted sac method. During 180 min of continuous observation of glucose absorption, significantly lowered glucose concentrations of the serosal medium were observed in the mazindol treated group at oral and caudal ends of the upper, caudal end of the middle, and caudal ends of the lower small intestine, whereas no significant differences in glucose concentrations of the mucosal medium were observed between the two groups. After 60 min incubation for monitoring the glucose transport, significantly decreased glucose concentrations of serosal medium were observed in mazindol treated group at oral and caudal ends of the upper, caudal ends of the middle and caudal ends of the lower small intestine, whereas no significant differences in glucose concentrations of mucosal medium were observed. The results suggested that there is little effect on glucose absorption, but the metabolism of glucose or the remaining glucose in the small intestinal wall is increased by mazindol treatment.

[Effects of chromium compounds on the respiratory system. Part 4. Long-term inhalation of chromic acid mist in electroplating to ICR female mice].

In order to determine the effect on the respiratory system of long-term inhalation of chromic acid in electroplating, histopathological examination was performed on mice (ICR, female, n = 50) exposed to chromic acid mist for 12 months (30 min/d, 2 d/wk), by using the authors' miniaturized chromium electroplating system. After 12 months' exposure, the mice were kept for 6 months without exposure. In the upper respiratory tract, round perforations were found in the nasal septum in 6 mice that were sacrificed or died after 10 months' exposure. The perforations were located near the point of the septum, the thinnest part of the nasal septum, and were of pin-hole sige. On the epithelium of the trachea and bronchus, loss of cilia, proliferation of goblet cells or basal cells, and squamous metaplasia were observed in almost all of the exposed mice. These changes appeared to be more advanced in mice exposed for a longer period. In the lung, development of benign adenoma was observed in one mouse in the 6- to 9-months' exposure group, in three in the 10- to 14-months' exposure group and in one in the group examined after 15 to 18 months. Furthermore, adenocarcinomas were found in two mice, one of which died at 17 months and the other was sacrificed 18 months after initiation of exposure. In conclusion, the finding that adenomas and adenocarcinomas were observed in mice exposed to chromic acid mist suggests the need to give careful attention to the possibility of respiratory cancers in chromium electroplating workers.

[Carcinogenicity in mice by inhalation of benzotrichloride and benzoyl chloride].

Benzotrichloride and benzoyl chloride are suspected to be causative agents of lung cancer and maxillary malignant lymphoma of workers employed in factories producing benzoyl chloride. The present study was undertaken to evaluate the carcinogenicity in mice of inhaling benzotrichloride and benzoyl chloride. Mice inhaled benzotrichloride and benzoyl chloride vapor for 30 min/d for 2 d/wk for 5 months, and each animal was followed for several month without subsequent exposure. Tumor developed in the lung, skin and lymphatic tissues at high incidences in mice inhaling benzotrichloride. By benzotrichloride vaporized at 50 degrees C, the incidence of pulmonary tumors was 53.1% (17/32, p less than 0.001), that of skin tumors was 25% (8/32, p less than 0.02), and that of malignant lymphoma was 25% (8/32, p less than 0.02) observed at the 10th month after exposure. These are significantly higher than that observed in control mice. In mice exposed to benzotrichloride vaporized at room temperature, the incidence of pulmonary tumors was 81.1% (30/37), that of skin tumors was 27.0% (10/37), and that of malignant lymphoma was 10.8% (4/37) observed at the 15th month after exposure. On the other hand, by benzoyl chloride vaporized at 50 degrees C, the incidence of pulmonary tumors was 10.7% (3/28) and that of skin tumors was 7.1% (2/28), but these incidences did not show any significant difference from the controls. These results suggest that the carcinogenicity of benzotrichloride is much higher than benzoyl chloride and that benzotrichloride is the primary cause of malignancies developing among workers engaged in manufacturing benzoyl chloride.

Effects of several factors on urinary excretion of methamphetamine and its metabolites in rats.

Effects of dose, route of administration, and sex and strain differences on the metabolism of methamphetamine in vivo were investigated in rats. Excretion of p-hydroxymethamphetamine was decreased with increasing dose of methamphetamine, while that of unchanged methamphetamine and amphetamine increased with dose. Subcutaneous injection instead of oral administration of methamphetamine increased the urinary excretion of unchanged drug and amphetamine, and decreased the excretion of p-hydroxymethamphetamine. No marked sex difference was observed in the urinary excretion of methamphetamine and metabolites. The Donryu strain of rats excreted unchanged methamphetamine and p-hydroxylated metabolites to a lesser extent than did Sprague-Dawley and Wistar strains.

Effects of chronic inhalation exposure to diesel exhaust on the development of lung tumors in di-isopropanol-nitrosamine-treated F344 rats and newborn C57BL and ICR mice.

Long-term exposure to diesel exhaust from a small diesel engine was carried out in order to elucidate the effects of exhaust on the development of lung tumors in F344 rats and newborn ICR and C57BL mice. The deposition of inhaled particles progressed in accordance with duration of exposure in both sets of the animals. No evidence of lung tumor formation was detected following exposure to diesel exhaust alone in rats. However, the combined treatment with DIPN and diesel exhaust led to an overadditive effect in the development of lung tumors. There was a tendency for the diesel-exposed mice to have higher incidences of lung tumors compared with non-exposed mice. The results obtained in the present study suggest that diesel exhaust may act as a factor relating to human respiratory carcinogenesis.

Idiopathic right ventricular dilation. Special reference to "arrhythmogenic right ventricular dysplasia" and analogous lesions.

Two autopsy cases which showed marked depletion of the right ventricular musculature of the heart accompanied with marked infiltration of the adipose tissue were reported. The first cases was an 18-year-old female who died of right sided congestive heart failure after about 4-years clinical course. The autopsy disclosed marked dilation of the right atrium and ventricle. The entire free wall of the right ventricle was markedly thin. Microscopically, most of the myocardial fibers of the right ventricle were replaced by fat and fibrous tissue. The second case, a 15-year-old boy, whose identical twin was previously diagnosed as arrhythmogenic right ventricular dysplasia designated by Fontaine et al., died suddenly during exercise. He showed no cardiac symptoms but electrocardiogram was abnormal. Autopsy revealed majority of the myocardial fibers of the right ventricular free wall were replaced by fatty tissue. In both cases, fatty infiltration was mainly noticed at the epicardial side and some myocardial fibers remained in the fatty tissue showed hypertrophic and/or degenerative changes. Review of the literature on similar cases showing depletion of the right ventricular musculature including so-called adult's Uhl anomaly, ARVD and dilated right ventricular myocardiopathy was conducted and the relationship of the present cases with these lesions was discussed.

Evaluation of "M-point" in hepatic artery to identify left medial segment of liver. Angiographic study.

The hepatic arteries of 122 patients were analysed on angiography to identify the left medial segment of the liver. Left medial arterial branches were classified into three types: type I arising from the left hepatic artery on the umbilical portion of the portal vein; type II arising from proximal portion of left hepatic artery before reaching the umbilical portion of the portal vein; type III arising from right hepatic artery. Incidence of each type is 37.2%, 35.8% and 27.0%, respectively. The artery frequently kinks at the right side of the umbilical portion of the portal vein and a total of incidence is 68% and that of each type is 23.5%, 89.8% and 100%, respectively. We call this characteristic kinking point of the left medial arterial branches, the "M-point".

Iodine-131 metaiodobenzylguanidine intra- and extravesicular accumulation in the rat heart.

In order to establish the appropriate time for [123I]MIBG human myocardial imaging to assess the adrenergic nerve activity, the time courses of metaiodobenzylguanidine (MIBG) intra- and extravesicular accumulation in the rat heart were estimated by using [131I]MIBG and reserpine. In the heart, the intravesicular accumulation was relatively constant, while the extravesicular accumulation decreased rapidly from 5 min to 6 hr. The intravesicular percentage of the total cardiac tissue concentration reached a plateau value of 50% at 4 hr after i.v. injection of [131I]MIBG. In the spleen, similar time courses were observed as those in the heart, both of these organs being richly innervated by adrenergic nerves. Along with the time activity difference previously observed in the human hearts, these results suggest that at 4 hr post i.v. injection, [123I]MIBG myocardial imaging will best express the neuronal accumulation of the tracer and may be useful for the assessment of adrenergic function in various pathological conditions of the human heart.
