Proteolytic processing of the coronavirus infectious bronchitis virus 1a polyprotein: identification of a 10-kilodalton polypeptide and determination of its cleavage sites.

Proteolytic processing of the polyprotein encoded by mRNA 1 is an essential step in coronavirus RNA replication and gene expression. We have previously reported that an open reading frame (ORF) 1a-specific proteinase of the picornavirus 3C proteinase group is involved in processing of the coronavirus infectious bronchitis virus (IBV) 1a/1b polyprotein, leading to the formation of a mature viral protein of 100 kDa. We report here the identification of a novel 10-kDa polypeptide and the involvement of the 3C-like proteinase in processing of the ORF 1a polyprotein to produce the 10-kDa protein species. By using a region-specific antiserum, V47, raised against a bacterial-viral fusion protein containing IBV sequence encoded between nucleotides 11488 and 12600, the 10-kDa polypeptide was detected in lysates from both IBV-infected and plasmid DNA-transfected Vero cells. Coexpression, deletion, and mutagenesis studies showed that this novel polypeptide was encoded by ORF 1a from nucleotide 11545 to 11878 and was cleaved from the 1a polyprotein by the 3C-like proteinase domain. Evidence presented suggested that a previously predicted Q-S (Q3783 S3784) dipeptide bond encoded by ORF 1a between nucleotides 11875 and 11880 was responsible for the release of the C terminus of the 10-kDa polypeptide and that a novel Q-N (Q3672 N3673) dipeptide bond encoded between nucleotides 11542 and 11547 was responsible for the release of the N terminus of the 10-kDa polypeptide.

[Protective effect of polysaccharide sulfate on human umbilical vein endothelial cell (HUVEC) injury in vitro].

The influence of four kinds of polysaccharide sulfate [heparin (Hep), low molecular weight heparin (LMWH), proglylene glucol mannate sulfate (MS871), chondroitin A sulfate (CAS)] on HUVEC injured by polycations and oxygen free radicals was investigated. The degree of injury was determined by checking the release of LDH to medium and 3H-TdR incorporation. The four kinds of polysaccharide sulfate were shown to protect cells from injury caused by polylysine concentration-dependently. The study also showed that the effects of Hep and CAS were stronger than those of LMWH and MS871 at the same dose level. In view of the level of LDH, the four kinds of polysaccharide sulfate can protect cells from oxygen free radical injury. The effects of Hep and CAS were more active than those of LMWH.

[Serum level of soluble interleukin-2 receptor and T-lymphocyte subpopulations in normal pregnancy].

Serum soluble interleukin-2 receptor (sIL-2R) levels and T lymphocyte subpopulations were measured in 87 cases of normal pregnancy and 109 normal non-pregnant women as control group. It was found that sIL-2R level and CD8 T subpopulation increased as compared with control group (214,600 +/- 70,400 U/L vs 162,100 +/- 84,100 U/L, P < 0.01; 37.6% +/- 5.3% vs 31.3% +/- 7.0%, P < 0.01), but the ratio of CD4/CD8 was lower than that of control group (1.2 +/- 0.2 vs 1.5 +/- 0.5, P < 0.01). No significant difference was found between CD3, CD4 subpopulations of pregnancy and that of control group (64.1% +/- 7.3% vs 66.0% +/- 9.9%, P > 0.05; 44.1% +/- 5.8% vs 43.8% +/- 9.0%, P > 0.05). And no significant relation was found between sIL-2R level and CD3, CD4, CD8 subpopulations, CD4/CD8 (r = 0.203 2, 0.207 7, 0.103 7, 0.121 4 respectly, P > 0.05). These results suggested that the changes of T lymphocytes and sIL-2R may play an important role in maintaining normal pregnancy and sIL-2R may be one of factors improving fetal survival.

Evidence for multiple adrenoceptor sites in rainbow trout vasculature.

The importance of neuronal and lumenal vascular adrenoceptors in the regulation of vascular reactivity was examined in rainbow trout (Oncorhynchus mykiss), in vivo and in vitro. In vivo, ganglionic blockade with hexamethonium or alpha-adrenoceptor blockade, with either phentolamine or prazosin, produced similar (7mmHg) decreases in dorsal aortic blood pressure. The drop in dorsal aortic pressure produced by phentolamine or prazosin was due to reduced systemic vascular resistance. Neither the alpha-adrenoceptor antagonist, phenoxybenzamine nor chemical sympathectomy with 6-hydroxy-dopamine affected dorsal aortic pressure. However, after chemical sympathectomy, phenoxybenzamine lowered dorsal aortic pressure to levels similar to that produced by either phentolamine or prazosin. Plasma epinephrine and norepinephrine concentrations increased four- and twofold, respectively, in sympathectomized fish. Sympathectomy also produced a leftward shift in the epinephrine dose/response curve of the in vitro perfused splanchnic vasculature, placing the effective catecholamine concentration well within the in vivo plasma levels. These results indicate that following chemical sympathectomy arterial blood pressure is stabilized by circulating catecholamines through the combined effect of increased plasma catecholamine concentrations and increased sensitivity of vascular adrenoceptors. Phenoxybenzamine is incapable of blocking neuronal vascular adrenoceptors but is a potent antagonist of the up-regulated adrenoceptors, suggesting that the latter are localized on the lumenal side of the vessel.

[Specific curative effects of nitrosourea drugs on tumor cells with Mer--phenotype in Chinese].

O6-Methylguanine DNA methyltransferase (O6-MT) activity and cellular sensitivity to nitrosourea drugs of 10 kinds of tumor cell strains derived from Chinese patients were measured by 3H radioactivity and colony-forming ability, respectively. The results in vitro showed that nimustine (Nim) 25 micrograms.ml-1 and carmustine (Car) 20 micrograms.ml-1 exhibited specific killing effects on Mer-phenotype tumor cells characterized by low O6-MT activity. In vivo both Nim and Car (25 mg.kg-1.wk-1 x 4 wk, ip) had specific curative ability to Mer- tumor cells implanted in nude mice. These findings suggested that assay of O6- MT activity in tumor biopsy could be used as a predictable guide to human tumor chemotherapy with nitrosourea compounds.

[Dynamic changes of fibronectin, DNA and morphology of the nuclei of experimental hepatoma in rats].

The dynamic changes of fibronectin (FN) content and nuclear features (DNA content, morphological parameters) during the development of hepatoma induced by 3'-Me-DAB were studied via computer-assisted image analysis. The results showed that the amount of FN decreased progressively and even disappeared completely during hepatocarcinogenesis. The difference between distinctive foci or nodules was highly significant (P less than 0.0001) and the DNA content was also increased coincidently during the same process.

O6-methylguanine-DNA methyltransferase and human cancer chemotherapy.

Two kinds of human tumor cell strains having different activity of O6-methylguanine-DNA methyltransferase (O6-MT) were transplanted into nude mice. Then the mice were injected intraperitoneally with bifunctional alkylating agent 1-(4-amino-2-methyl-5-pyrimidinyl) methyl-3-(2-chloroethyl)-3-nitrosourea hydrochloride (ACNU). The tumors with low O6-MT activity were quickly suppressed or cured. The result suggests that some tumors, if provisionally determined with low O6-MT activity, might be efficiently cured by treatment with ACNU. This probably opens a new way for human cancer chemotherapy.

Muscarinic action of acetylcholine in the pigeon optic tectum.

Eighty visual units were extracellularly recorded from the pigeon optic tectum, and the effects of iontophoretically applied acetylcholine and its antagonists, atropine and tubocurarine, on these units were examined. The results showed that acetylcholine in the avian tectum functions as an excitatory transmitter or modulator and acts predominantly through a muscarinic mode of action.
