Construction and expression of eukaryotic expressing vector pCH510 of polypeptide CH50 and its chemotaxis and antitumor function by in vivo transfection.

To construct an eukaryotic expressing vector that expresses CH50, a recombinant Cell I-Hep II bifunctional-domain polypeptide of human fibronectin, and to investigate the chemotaxis to immune cells and the inhibitory effect on the growth of tumor by the expression of the plasmid in vivo, the plasmid was constructed by DNA recombination. Gene transfection was performed in vitro and in vivo. The expressed product was identified by Western blot. The chemotaxis after gene transfection in vivo was observed by histotomy and staining of muscle tissues. The inhibition of gene transfection on solid tumor was observed in mice. The results showed that plasmid pCH510 was constructed by the recombination of the 5'-terminal noncoding region and signal peptide coding region of human fibronectin cDNA and cDNA fragment coding CH50 polypeptide with a 3'-terminal noncoding region of human FN cDNA, and the insertion of the recombinated fragment into plasmid pcDNA3. 1. After transfection with plasmid pCH510, NIH3T3 cells could produce CH50 polypeptide. The transfection of plasmid pCH510 by the injection in muscle of mouse could produce the effects of chemotaxis on immune cells and the inhibition on the growth of solid tumor. It is concluded that plasmid pCH510 can express in cells and in vivo in mouse. The expression of the plasmid in vivo has a chemotactic effect on immune cells and can inhibit the growth of solid tumor.

Aluminum chloride pretreatment of elastin inhibits elastolysis by matrix metalloproteinases and leads to inhibition of elastin-oriented calcification.

Calcification of elastin occurs in many pathological cardiovascular diseases including atherosclerosis. We have previously shown that purified elastin when subdermally implanted in rats undergoes severe calcification and aluminum chloride (AlCl(3)) pretreatment of elastin inhibits calcification. In the present study we investigated whether matrix metalloproteinase (MMP) binding to elastin and elastin degradation is prevented by AlCl(3) pretreatment. Subdermal implantation of AlCl(3)-pretreated elastin showed significantly lower MMP-9 and MMP-2 activity surrounding the implant as compared to the control implants. AlCl(3) pretreatment also significantly inhibited elastin implant calcification at the seven-day implant period (AlCl(3)-pretreated 4.07 +/- 1.27, control 23.82 +/- 2.24 microg/mg; p<0.0001). Moreover, elastin gel zymography studies showed that gel pretreatment with AlCl(3) inhibited elastolysis by MMP-9. We also demonstrate significant suppression of MMP-2 activity in aortic wall segments of AlCl(3)-pretreated porcine bioprosthetic heart valve implants as compared to control valve implants in sheep mitral valve replacement studies. AlCl(3) pretreatment also significantly inhibited calcification of elastin in this model. Thus, we conclude that aluminum ion binding to elastin prevents MMP-mediated elastolysis and thus prevents elastin calcification.

The inhibitory effects of BmK IT2, a scorpion neurotoxin on rat nociceptive flexion reflex and a possible mechanism for modulating voltage-gated Na(+) channels.

Buthus martensi Karsch IT2 (BmK IT2), a scorpion neurotoxin, was found to display a biphasic inhibitory effect on the C component of the rat nociceptive flexion reflex by subcutaneous injection in vivo, and also on the total Na(+) currents of rat dorsal root ganglion neurons using whole-cell patch clamping. BmK IT2 blocked the tetrodotoxin-resistant (TTX-R) component of the Na(+) currents with a degree of selectivity. The partial block of the TTX-R Na(+) currents, brought about by 0.01 microg/microl BmK IT2, reversed less rapidly and completely than the partial block of the tetrodotoxin-sensitive (TTX-S) current brought about by the same concentration of BmK IT2. These results suggest that the inhibition of the rat nociceptive flexion reflex by BmK IT2 may be attributed to modulation of the different voltage-gated Na(+) channels.

Dual functions of largest NURF subunit NURF301 in nucleosome sliding and transcription factor interactions.

NURF is an ISWI complex of four proteins that uses the energy of ATP hydrolysis to catalyze nucleosome sliding. Three NURF components have been identified previously. We have cloned cDNA encoding the largest NURF subunit, revealing a 301 kDa polypeptide (NURF301) that shares structural motifs with ACF1. We have reconstituted full and partial NURF complexes from recombinant proteins and show that NURF301 and the ISWI ATPase are necessary and sufficient for accurate and efficient nucleosome sliding. An HMGA/HMGI(Y)-like domain of NURF301 that facilitates nucleosome sliding indicates the importance of DNA conformational changes in the sliding mechanism. NURF301 also shows interactions with sequence-specific transcription factors, providing a basis for targeted recruitment of the NURF complex to specific genes.

IL-4 production by human basophils found in the lung following segmental allergen challenge.

BACKGROUND: Human blood basophils secrete high levels of IL-4 following activation with specific allergen, yet their role as cytokine-producing cells in allergic lesions has not been described. OBJECTIVE: Our objective was to investigate whether and under what conditions basophils infiltrating allergic lesions in the lung secrete IL-4 in vitro. METHODS: Bronchoalveolar lavage (BAL) cells were recovered 20 hours after segmental allergen challenge. Basophils were enriched with Percoll using a protocol commonly used for blood basophils. IL-4 and histamine were measured in culture supernatants following activation with a variety of stimuli. Two-color flow cytometry was performed to detect intracellular IL-4. RESULTS: IL-4 protein was detected in all basophil culture supernatants following a 4- to 5-hour incubation in medium alone; the levels obtained did not significantly increase with the addition of anti-IgE. BAL basophils failed to release histamine in response to specific allergen but showed nearly 60% histamine release with N-formyl-methionyl-leucyl-phenylalanine, suggesting that they were desensitized to IgE-mediated stimuli as a result of their activation in vivo. Using these same conditions, IL-4 was not detected in BAL cell fractions enriched for lymphocytes and eosinophils. Ionomycin induced IL-4 secretion by BAL basophils, and this response was reduced with the addition of phorbol myristate acetate. In contrast, phorbol myristate acetate promoted the secretion of IL-4 by BAL cells enriched for lymphocytes; both findings are identical to those reported for basophils and lymphocytes purified from blood. Flow cytometry confirmed the secretion of IL-4 by BAL basophils. CONCLUSIONS: These data suggest that basophils migrating to the lung following allergen challenge represent a major source of IL-4.

Effects of prednisone on the cellular responses and release of cytokines and mediators after segmental allergen challenge of asthmatic subjects.

BACKGROUND: Systemic glucocorticoids are a major therapy for the management of allergic inflammation and asthma; however, information about their effects in vivo are limited. OBJECTIVE: This study was performed to examine the effects of prednisone on inflammatory mediators, cytokines, and cellular responses in the model of segmental allergen challenge (SAC) of allergic asthmatic subjects. METHODS: The effects of a 3-day pretreatment with oral prednisone (30 mg twice daily) on the physiologic and inflammatory responses to SAC were studied in 10 allergic asthmatic subjects in a double-blind, placebo-controlled, crossover protocol. RESULTS: Prednisone improved baseline FEV(1) by 10% and modestly inhibited the SAC-induced fall in FEV(1) at 30 minutes and at 6 to 8 hours. Five minutes after challenge, levels of histamine, PGD(2), 9alpha,11beta-PGF(2), and thromboxane B(2) increased in bronchoalveolar lavage fluid (median increase, 5- to 14-fold); prednisone did not inhibit these responses. Prednisone inhibited (median decrease, 66%-97%) the total influx of inflammatory cells, specifically eosinophils, basophils, and some subsets of T lymphocytes (CD4, CD45RA, and CD45RO cells) assessed 19 hours after SAC, but it did not inhibit the influx of neutrophils. Increases in soluble E-selectin, kinins, and albumin were also inhibited by the glucocorticoid (median decrease, 36%-74%). Prednisone treatment inhibited the appearance of mRNA, protein, or both for T(H)2 cytokines (IL-4 and IL-5), as well as for IL-2 and transforming growth factor alpha, but did not inhibit increases of immunoreactive GM-CSF in bronchoalveolar lavage fluid. CONCLUSION: These studies indicate that prednisone suppresses multiple components of allergic airway inflammation, including cell recruitment, adhesion molecule expression or release, airway permeability, and production of cytokines potentially involved in airway immunity or remodeling.

Phospholipids released from activated platelets improve platelet aggregation and endothelial cell migration.

This study was undertaken to isolate phospholipids released from activated platelets and to investigate their biological activities. Freshly washed platelets were activated with freezing/thawing, thrombin, ionophore 23187, and arachidonic acid. Thrombin was incubated with platelet-rich plasma to promote synthesis and release of phospholipids from platelets. Phospholipids in supernatants of activated platelets were extracted with butanol and separated by thin-layer chromatography. Release of phosphatidylserine (PS) and phosphatidic acid (PA) increased when platelets were treated with freezing/thawing, ionophore, and thrombin. The lysophosphatidyl ethanolamine (LPE) appeared not to be induced with freezing/thawing, but increased significantly by thrombin, ionophore, and arachidonic acid. The effects of platelet phospholipids on hemostasis and angiogenesis were studied with platelet aggregation and endothelium chemotaxis. Phospholipids isolated from thrombin-stimulated platelet-rich/platelet-poor plasmas were used as synergistic agonists in platelet aggregation and as chemotactic agents in endothelial cell migration. Several phospholipids increased chemotaxis and platelet aggregation; these were PS, PA, LPE, and sphingosine-1-phosphate. Also, chemotaxis of those phospholipids increased when combined with charcoal-stripped fetal bovine serum, suggesting that cofactors in serum enhanced phospholipid-induced cell migration. These observations suggest that activated platelets release biologically active phospholipids into the blood stream, where they may play an important role in thrombosis and angiogenesis.

Contrasting effects of thiol-modulating agents on endothelial NO bioactivity.

The bioactivity of endothelium-derived nitric oxide (NO) is an important component of vascular homeostasis that is sensitive to intracellular redox status. Because glutathione (GSH) is a major determinant of intracellular redox state, we sought to define its role in modulating endothelial NO bioactivity. In porcine aortic endothelial cells (PAECs), we depleted intracellular GSH (>70%) using 1) buthionine-(S,R)-sulfoximine (BSO), which inhibits GSH synthesis; 2) diamide, which oxidizes thiols; or 3) 1-chloro-2,4-dinitrobenzene (CDNB), which putatively depletes GSH through glutathione S-transferase activity. Cellular GSH depletion with BSO had no effect on endothelial NO bioactivity measured as A-23187-induced cGMP accumulation. In contrast, oxidation of intracellular thiols with diamide inhibited both A-23187-induced cGMP accumulation and the cGMP response to exogenous NO. Diamide treatment of either PAECs, PAEC membrane fractions, or purified endothelial nitric oxide synthase (eNOS) resulted in significant inhibition (approximately 75%) of eNOS catalytic activity measured as L-[(3)H]arginine-to-L-[(3)H]citrulline conversion. This effect appeared related to oxidation of eNOS thiols as it was completely reversed by dithiothreitol. Glutathione depletion with CDNB inhibited A-23187-stimulated cGMP accumulation but not the cGMP response to exogenous NO. Rather, CDNB treatment impaired eNOS catalytic activity in intact PAECs, and this effect was reversed by excess NADPH in isolated purified eNOS assays. Consistent with these results, we found spectral evidence that CDNB reacts with NADPH and renders it inactive as a cofactor for either eNOS or glutathione reductase. Thus thiol-modulating agents exert pleiotropic effects on endothelial NO bioactivity, and these data may help to resolve a number of conflicting previous studies linking GSH status with endothelial cell NO bioactivity.

A unique mRNA initiated within a middle intron of WHSC1/MMSET encodes a DNA binding protein that suppresses human IL-5 transcription.

Human interleukin (IL)-5 gene transcription is regulated by several transcription factor binding sites, including CLE 0, GATA, and a region from position -123 to -92 known as response element (RE)-II. By expression cloning, a partial protein was identified that bound to concatamers of RE-II. Recombinant protein derived from this initial complementary DNA (cDNA) encoding the partial protein specifically bound to RE-II-containing oligonucleotides in electromobility shift assays. The complete sequence (3,649 bp) was determined by 5' rapid amplification of cDNA ends and comparisons to existing ESTs, and found to be identical to the 3' half of Wolf-Hirschhorn syndrome candidate 1, (WHSC1; also known as Multiple Myeloma SET domain [MMSET]). The full-length protein contains an SET domain and two plant homeodomain-type zinc fingers. Transcription initiation of RE-II binding protein (RE-IIBP) messenger RNA (mRNA) uniquely occurred within the middle of WHSC1 near a region that exhibits complex mRNA splicing. RE-IIBP reactive polyclonal antisera identified proteins in human T-cell nuclear protein extracts of 62 and 66 kD that were consistent with the length of the longest open reading frame in RE-IIBP. In contrast, WHSC1 is predicted to encode a protein of 136 kD. In activated human Jurkat and murine D10.G4.1 T cells, expression of full-length and truncated forms of RE-IIBP repressed RE-II promoter activity of a 5X-RE-II luciferase reporter construct by as much as 75%. In addition, RE-IIBP expressed in activated D10.G4.1 T cells inhibited endogenous murine IL-5 production. The repressor activity of RE-IIBP is consistent with the presence of an SET domain that is found in other proteins that act as gene silencers.

[Inhibition of voltage-activated outward delayed rectifier potassium channel currents in dorsal root ganglion neurons of rats by lead].

OBJECTIVE: To study the effect of lead on voltage-activated outward delayed rectifier potassium channel currents in acute isolated dorsal root ganglion (DRG) neurons of adult rats. METHODS: Patch clamp technique was employed to record the whole cell of DRG in adult rats. RESULTS: In the experiments, delayed rectifier (IK) potassium currents channel was recorded and activation was voltage-dependent. The threshold concentration of lead (Pb2+) for IK was 1 mumol/L, which reduced potassium current by (8.6 +/- 0.8)%, 4 mumol/L of Pb2+ reduced the current by (38.6 +/- 6.2)%, and 8 mumol/L of Pb2+ reduced the current by (63.0 +/- 5.1)%. The action of lead was rapid and its effect would be reversed rapidly to various extent by washing. CONCLUSION: Higher outward delayed rectifier potassium channel currents, which could be reversed.

[Construction of transgenic rice populations by inserting the maize transponson Ac/Ds and genetic analysis for several mutants].

An efficient and rapid gene transformation system of rice mediated by Agrobacterium tumefaciens was used. Calli induced from immature and mature embryos of Zhonghua No. 11, a japonic rice variety, were cultured with the A. tumefaciens strain EHA105 harboring the superbinary plasmid pDsBar1300 or pUBITs separately, and more than 400 independent transgenic lines inserted Ds element or Ac fragment were obtained. Some visible mutants in T0 or T1 generation were found, consisting of disease resistance, albino, dwarf, male sterile, chlorosis, early heading, late heading, stripe, etc. From the phenotype analysis, a few mutants such as dwarf and male sterile seemed to be linked to the Basta resistance and the transposon.

Modulation of BmKAS-1 and BmK1-3-2 to sodium channel in rat dorsal root ganglion neurons.

OBJECTIVE: To investigate what effects BmKAS-1 (a polypeptide purified from the Chinese scorpion Buthus martensi Karsch [BmK] and named as BmK activator of skeletal-muscle ryanodine receptor) and its upstream mixture BmK1-3-2 have on Na+ channels in dorsal root ganglion (DRG) small diameter neurons. METHODS: The whole-cell patch-clamp technique was used to investigate the effects of BmKAS-1 and BmK1-3-2 on Na+ current in rat small diameter DRG neurons. RESULTS: About 50% peak Na+ current was suppressed by 10 micrograms/ml of BmK1-3-2. 1.62 micrograms/ml of BmKAS-1 also blocked 50% peak Na+ current, and there was an obvious dose-dependent relationship. CONCLUSION: Both BmK1-3-2 and BmKAS-1 have a blocking effect on Na+ channels, and this may one of the mechanisms for the analgetic effect of BmK1-3-2 and BmKAS-1.

[Estimation of insulin secretion rate in multiple sampled intravenous glucose tolerance test].

OBJECTIVE: To measure and calculate insulin secretion rate(ISR) in multiple sampled intravenous glucose tolerance test (FSIVGTT) according to the two compartment model and standard parameters of the kinetics of connecting peptide (C-P) in vivo. METHODS: 15 subjects with normal glucose tolerance and 11 type 2 diabetic patients took part in the study. Oral glucose tolerance test (OGTT) and FSIVGTT were performed. Glucose concentrations were measured by hexokinase method. Insulin (INS) and C-P concentrations were measured by radioimmunoassay. A computer procedure devised by our laboratory was used to calculate the value of ISR at each time point. RESULTS: In the control group, serum INS and C-P concentrations reached the peak at 4 min, the second peak was not obvious. In type 2 diabetes, only 5 subjects had a small peak within 4 min, two curves reached its peak between 60 and 80 min. In the control group, ISR reached its peak 2 min after injection of glucose, that was earlier than INS and C-P curves in time sequence (P = 0.000), the second obvious peak was shown at 22 min. In type 2 diabetes, an obvious but small peak of ISR was shown at 2 min and all the patients had this peak. CONCLUSIONS: It is necessary to take the process of metabolic clearance of INS and C-P into consideration, when blood concentration of INS and C-P were used as indexes to evaluate the secretion function of beta-cells. The ISR profile is more sensitive for investigation of the second phase of insulin secretion in the control group and the first phase in type 2 diabetes group.

[Study on drug resistance of Mycobacterium tuberculosis in different age group].

OBJECTIVE: To investigate drug resistance of Mycobacterium tuberculosis in different age group in recent five years, and to analyze the trends of drug resistance at present. METHODS: 1 948 cases of pulmonary tuberculosis with culture-positive were divided into youth group (18 approximately 39 years), middle age group (40 approximately 59 years) and old age group (>/=60 years). All of the strains of Mycobacterium tuberculosis were tested for resistance to INH, RFP, SM and EMB by the absolute concentration method. RESULTS: The rate of primary drug resistance in youth group was 36.1%, in middle age group 46.8%, in old age group 40.2%, there were no significant differences among them (P > 0.05). The rate of acquired drug resistance was higher in youth group than that in old age group (P = 0.019 6). The primary resistance rate to RFP in middle age group was 10.6%, much higher than that in youth group and in old age group. The acquired resistance rate to RFP in youth group was 63.6%, higher than that in middle age group and in old age group. The rate of primary multi-drug resistance in middle age group was 5.3%, higher than that in youth group (P = 0.011 2) and in old age group (P = 0.008 5). The rate of acquired multi-drug resistance in youth group was 55.2%, higher than that in middle age group (P = 0.031 9) and in old age group (P < 0.001). The drug resistance rate in using drugs 1 approximately 3 months group was 58.7%, higher than that in never using drug group (P < 0.001) and in using drugs < 1 month group (P = 0.047 2). The drug resistance rate had no significant difference between never using drug group and using drugs < 1 month group. The emergency of drug resistance in youth group was fastest in all age groups. CONCLUSIONS: Primary drug resistance or acquired drug resistance varies in different age group. It is suggested that the surveillance of drug resistance in different age group should be taken seriously, so as to provide an important basis for revising National Tuberculosis Programme (NTP).

[Apoptosis of retinal capillary cells in diabetic rats and expression of apoptosis-related genes].

OBJECTIVE: To determine the apoptosis of retinal capillary cells in early diabetic rats and observe the expression of apoptosis-related genes (bcl-2, Bax) in retinas of early diabetic rats. METHODS: 40 healthy adult Wistar rats were chosen and randomly divided into four groups: normal control (CON), diabetes mellitus one month (DM1), three months (DM3) and six months (DM6). Diabetic mellitus in rats was induced by streptozotocin (STZ) intraperitoneally. Retinal preparations of blood vessels and paraffin sections were stained by TdT-mediated dUTP nick end labeling (TUNEL) and immunohistochemical (ABC) method respectively and analyzed by computer-picture analytic system on the results of ABC method. RESULTS: Positive pericyte nuclei labeled by TUNEL were emerged in DM3 and DM6, but positive endothelial cell nuclei only in DM6. No positive reaction was seen in CON and DM1. Chromatin of TUNEL positive nuclei was distributed unequally, showing apoptotic characteristics of ring-like and crescent nuclei. In ABC staining method, two genes' (bcl-2, Bax) proteins were expressed in retinal vessels. Along with the progression of diabetes, the intensity of positive reaction was increased in degrees. Moreover, pigment epithelial cells in DM3 showed Bcl-2 positive reaction that again extended further to inner rod segments and ganglion cells in DM6, while in DM6, Bax was expressed in ganglion cells. CONCLUSIONS: The nature of pericyte dropout in early diabetic rats is apoptosis that also exists in endothelial cells; the expression of Bax and bcl-2 in retinas of early diabetic rats is enhancing.

[The effect of ketamine on the expression of HSP70 induced by endotoxin in liver of rat].

OBJECTIVE: To observe the effect of ketamine on the expression of heat shock proteins (HSP70) in liver of rat with septic shock. METHODS: Sixty-four rats were randomly allocated to control group, endotoxin group and ketamine group. The model of septic shock was used to examine the expression of HSP70 in rat liver and the TNF-alpha level of blood at 2 hours after endotoxin treatment and dying time for endotoxin group rats. The survival rates for the three groups were compared. RESULTS: There was a significant increase in the expression of HSP70 and the level of TNF-alpha in the endotoxin group as compared with those of control group (P < 0.05). A significant increase in the expression of HSP70, a decrease in the level of TNF-alpha and a significantly higher survival rate were seen in ketamine group when compared with those of endotoxin group, respectively. CONCLUSION: Ketamine could increase the expression of HSP70 in rat liver, restrain the release of TNF-alpha caused by endotoxin, and raise the survival rates of rats with septic shock.
