Expression of cecropin CMIV fusion protein in E. coli under T7 promoter.

A synthetic gene coding for cecropin CMIV from Bombyx mori was cloned into plasmid pEZZ318 and fused to a DNA segment encoding the signal peptide of Staphylococcal protein A and IgG binding domain. The fusion gene was then subcloned into expression vector pET11c under the control of T7 promoter and expressed in E. coli. The fusion protein did not exhibit any antibacterial activity either in cell lysate or in medium. After cleavage from the fusion protein with CNBr the biological activity of recombinant cecropin CMIV was obtained.

Cytokine receptor signal transduction and the control of hematopoietic cell development.

The cytokine receptor superfamily is characterized by structural motifs in the exoplasmic domain and by the absence of catalytic activity in the cytosolic segment. Activated by ligand-triggered multimerization, these receptors in turn activate a number of cytosolic signal transduction proteins, including protein tyrosine kinases and phosphatases, and affect an array of cellular functions that include proliferation and differentiation. Molecular study of these receptors is revealing the roles they play in the control of normal hematopoiesis and in the development of disease.

Expression and localization of prolactin messenger ribonucleic acid in the human immune system.

Pituitary PRL is involved in immunoregulation. Also, a PRL-like molecule is secreted by peripheral blood mononuclear cells. In this study, we examined tissues of the human immune system to evaluate if the PRL gene is expressed and to determine the location and type of cells involved in its synthesis. To evaluate the expression of PRL messenger RNA (mRNA) in normal and abnormal human lymphoid tissues, we used RT-PCR to generate a specific 276-bp product from normal human thymus, spleen, tonsil, lymph node, and lymphoid tumors. Restriction enzyme digestion confirmed that this PCR product was expressed PRL. Furthermore, we developed a specific and sensitive nonisotopic in situ hybridization technique for PRL mRNA, and cells containing PRL mRNA were found in each tissue of the human immune system. Also, PRL mRNA was widely distributed throughout neoplastic tissue from a thymoma and lymphomas where mitogenic and anti-apoptotic properties of PRL could be involved in tumor progression. PRL mRNA was localized in lymphocytes, epithelial cells, and vascular endothelial cells. The presence of PRL mRNA in vascular endothelium cells suggests other roles for PRL in these tissues in addition to immunomodulation. In conclusion, the presence of PRL mRNA in human lymphoid tissue implies that locally synthesized PRL may play a critical role in immunocompetence by providing an important regulatory signal to the microenvironment of human lymphoid organs.

Localization of growth hormone messenger ribonucleic acid in the human immune system--a Clinical Research Center study.

GH is an immunomodulatory factor that can be synthesized and secreted by mononuclear leukocytes. To determine if GH can be produced by the human immune system, we assessed the presence of GH messenger RNA (mRNA) in both normal and abnormal human lymphoid tissues by RT-PCR and nonisotopic in situ mRNA hybridization. The predicted PCR product of 16lbp from human thymus, spleen, tonsil, lymph node, thymoma, and T and B cell lymphomas was similar to the product amplified from the human pituitary. Restriction enzyme digestion confirmed that complementary DNA generated using GH primers originated from GH mRNA. Furthermore, we performed in situ hybridization to localize the GH mRNA-positive cells. A hybridization signal for GH mRNA was found in all tissues examined. The distribution patterns of GH in normal lymphoid tissues suggested that GH can be produced by lymphocytes, as well as endothelial cells and other cell types. Also, GH mRNA-positive cells were distributed diffusely throughout T and B cell lymphomas and a thymoma. Our results demonstrate GH gene expression in normal and neoplastic human lymphoid tissues including nonlymphoid cells. This finding supports the hypothesis that the human immune system is an extrapituitary site of GH gene expression that may serve as an autocrine/paracrine factor in immunomodulation.

Self-reported automobile accidents involving patients with obstructive sleep apnea.

We developed a routine survey instrument, which included data on self-reported motor vehicle accidents (MVA), among 253 patients who attended the UCLA Sleep Disorders Center to evaluate whether patients with diagnosed sleep apnea syndrome (SAS) have a higher ratio of MVAs. We used unconditional multiple logistic regression to determine the odds ratio between MVA and SAS, adjusted for the following covariates: age, sex, work shift, daytime nap, alcohol and coffee intake, and history of neurologic diseases. Thirty-one percent of patients with SAS compared with 15% of patients without SAS reported at least one MVA (p < 0.01). The adjusted odds ratio found through logistic multiple regression analysis was 2.99 (p < 0.01). The results demonstrate that patients with SAS had a higher ratio of self-reported MVA than did those without SAS and that the characteristics of falling asleep at inappropriate times and driving past destinations were two good indicators of probability of having an MVA. Nevertheless, sleep apnea is a very treatable condition; once treated the risk factors diminish considerably.

[Effect of zhuyu huatan decoction for cellular and humoral immunity in cerebral hemorrhage].

Fifty-one patients of cerebral hemorrhage were observed on controlled study which were treated with the Zhu-Yu Hua-Tan Decoction (ZYHTD) and Western medicine. The results revealed that ZYHTD could reduce the serum IgG (P < 0.05), and lymphocyte transformation rate (P < 0.05). Meanwhile, the patient's health condition of treated group was better than the control group in the fields of eliminating brain hematoma, evaluating patients condition score in TCM and compromised neurologic function (P < 0.05). It is assumed that there is positive significance in anti-inflammatory effect and promoting the absorption of brain hematoma in the acute phase of cerebral hemorrhage.

Rejoining and misrejoining of radiation-induced chromatin breaks. I. experiments with human lymphocytes.

Fluorescence in situ hybridization with a composite probe for human chromosome 4 and a probe that stained all centromeres was used to study gamma-ray induced breakage, rejoining and misrejoining in prematurely condensed chromosomes in human lymphocytes. Dose-response curves for the induction of all types of aberrations in prematurely condensed human chromosomes 4 were determined immediately after irradiation and after 8 h postirradiation incubation. In addition, aberrations were measured after various incubation times from 0 to 18 h after a dose of 7 Gy. Unrejoined chromosome breaks were the most frequent type of aberration observed immediately after irradiation. Approximately 15% of total aberrations observed were chromosome exchanges. After 8 h postirradiation incubation, the frequency of breaks in prematurely condensed chromosomes declined to about 20% of the initial value, and chromosomal exchanges became the most frequent aberration. Results of metaphase analysis were similar to those for prematurely condensed chromosomes after 8 h incubation with the exception that a significantly lower frequency of fragments was observed. Symmetrical and asymmetrical interchanges were found at similar frequencies at all doses. No complex exchanges were observed in lymphocyte chromosomes immediately after exposure. They accounted for about 1% of total exchanges in metaphase chromosomes at doses <3 Gy and about 14% at 7 Gy. Incomplete exchanges amounted to approximately 15% of total exchanges at all doses. The kinetics of break rejoining was exponential, and the frequency of exchanges increased with kinetics similar to that observed for the rejoining of the breaks. This increase in the total exchanges as a function of the time between irradiation and fusion was due to a rapid increase in reciprocal interchanges, and a slower increase in complex exchanges; the frequency of incomplete exchanges increased initially, then decreased with prolonged incubation to the level observed in metaphase. It is concluded that the formation of each kind of chromosome aberrations follows different kinetics.

Rejoining and misrejoining of radiation-induced chromatin breaks. II. Biophysical Model.

A biophysical model for the kinetics of the formation of radiation-induced chromosome aberrations is developed to account for the recent experimental results obtained with a combination of the premature chromosome condensation (PCC) and fluorescence in situ hybridization (FISH) techniques. In this model, we consider the broken ends of DNA double-strand breaks (DSBs) to be reactant and make use of the interaction distance hypothesis. The repair/misrepair process between broken ends is suggested to consist of two steps; the first step represents the two break ends approaching each other, and the second step represents the enzymatic processes leading to DNA end-to-end rejoining. Only the second step is reflected in the kinetics observed in experiments using PCC. The model appears to be able to fit existing data for human cells. It is shown that the kinetics of the formation of chromosome aberrations can be explained by a single rate that characterizes both rejoining and misrejoining of DSBs, suggesting that repair and misrepair share the same mechanism. Fast repair (completed in minutes) in a subset of DSBs is suggested as an explanation of the complete exchanges observed with PCC in human lymphocytes immediately after irradiation. The fast repair component seems to be absent in human fibroblasts.

Effect of 764-3 on aggregation and calcium movements in aequorin-loaded human platelets.

Washed human platelets were loaded with the Ca(2+)-sensitive photoprotein, aequorin, using hypoosmotic shock treatment-technique. Then aggregation and cytoplasmic ionized calcium concentration ([Ca2+]i) changes in response to collagen or thrombin were measured simultaneously in the aequorin-loaded human platelets with a Platelet Ionized Calcium Aggregometer. 764-3, an active component isolated from the Chinese medicinal herb Salvia Miltiorrhiza Bge, inhibited platelet [Ca2+]i rise as well as aggregation evoked by collagen or thrombin in the presence of extracellular Ca2+. After the extracellular Ca2+ was removed by addition of EGTA, collagen or thrombin, causing no aggregation, still elicited platelet [Ca2+]i rise which reflected Ca2+ mobilization from intraplatelet stores. Under this condition, 764-3 could also suppress platelet [Ca2+]i rise. Analysis shows that 764-3 inhibits platelet Ca2+ influx and Ca2+ mobilization with similar potency, which accounts for its suppression of platelet [Ca2+]i rise, and must contribute to its inhibition of platelet aggregation.

[Effect of ginger-processing on l-ephedrine contents in rhizoma Pinelliae].

The contents of l-ephedrine in Rhizoma Pinelliae have been determined to be approximately 3.44 x 10(-3). The contents vary with the five different processing methods in the following order, Pinellia boiled with ginger juice and alum > raw Pinellia > Pinellia dipped in ginger juice > Pinellia boiled only with ginger juice > Pinellia dipped in alum solution. The alum solution and the Pinellia boiled only with ginger juice bear most strongly on the l-ephedrine contents of Pinellia.

[Post-thyroidectomy osteoporosis].

Thyroxine and calcitonin secreted by the thyroid gland are reduced in 26 cases patients with thyroid cancer after total or subtotal thyroidectomy. Since calcitonin participates in bone mineral metabolisim, bone mineral density (BMD) was examined in these patients treated in the past 10 years. It was found that serum calcitonin level was decreased, and the degree of reduction positively correlated with BMD (r = 0.78, P < 0.005). Osteoporosis developed about 10 years of age earlier. Most patients did not constantly receive thyroid tablets after excision of the thyroid. The resulting hypothyroidism led to ovarian dysfunction in female patients as shown by lowered level of estradiol. It is therefore indicated to administer thyroid tablet and calcitonin to prevent osteoporosis.

[A study on lens epithelial cell DNA damage induced by hydrogen peroxide in rat and its repair].

OBJECTIVE: The induction and repair of DNA single-strand breaks (SSBs) were measured in primary cultured infant rat lens epithelial cells (RLECs) following hydrogen peroxide (HP) exposure, and the influential factors were detected. METHODS: Utilizing nick translation methodology which utilizes incorporation of labeled nucleotides at the sites of DNA SSBs to sensitively quantitate the damage, we studied the effects of temperature, doses of HP and trace amount of selenium on cell survival. RESULTS: The number of DNA SSBs at 37 degrees was higher than that at 4C (P < 0.01). Significant numbers of SSBs were detected after being insulted by as low as 36.4 mumol/L hydrogen peroxide for 12 minutes 4 degrees (P < 0.05). Repair rapidly initiated and almost completed in 60 minutes after mild damage, and it was unable to repair when the DNA damage was induced by HP at toxic concentration. Trace selenium was added to the culture and incubated for 24 hours, then the number of SSBs was significantly decreased (P < 0.01). CONCLUSIONS: At 37 degrees HP may induce DNA SSBs in RLECs. HP induces the SSBs in a dose dependent manner. The DNA SSBs can not be repaired after severe damage. Trace selenium can decrease the degree of susceptibility to oxidative damage in infant RLECs.

Conformation of an analog of human follicular gonadotropin releasing peptide TF14 in solution by 2D-NMR.

Since the discovery of hF-GRP, several analogs have been synthesized in order to see their effects on the gonadotropin releasing activity, either as agonists or antagonists to this peptide. TF14 is one of these analogs, whose 14th position in the primary sequence is Phe instead of Asn in hF-GRP, while its activity is doubled. 2D-NMR (TOCSY, ROESY) was used to determine the conformation of TF14 in solution. Compared with hF-GRP, the whole peptide is in a non-typical more extended conformation, which may give some clue to the relation between structure and function of these two peptides.

[Orthogonal experiment design in the optimization of processing technology for Rhizoma Pinelliae by ginger and alum].

Based on the L9(3)1 orthogonal design with the amount of ginger, amount of alum and decocting time as working factors, and using the method of recording comprehensive scores, an experimental study has been made on the optimization of processing technology for Rhizoma Pinelliae by ginger and alum as stipulated in the Pharmacopoeia. The result shows that the best process is to use 15 kg of ginger juice and 8 kg of alum per 100 kg of Pinellia ternata and decoct them together for 2-3 hours till the juice is fully absorbed by Rhizoma Pinelliae.

Analysis of truncated forms of Bombyx mori glycyl-tRNA synthetase: function of an N-terminal structure in RNA binding.

Bombyx mori glycyl-tRNA synthetase (GRS) was expressed as the full length protein and as N-terminally and C-terminally truncated forms. The intact enzyme and forms with deletions of 12, 27, 46, and 55 N-terminal residues were expressed, purified, and characterized. All were active, having 15-25% of both pyrophosphate exchange activity and aminoacyl-tRNA synthetase activity compared to wild type enzyme. Active site titration indicated that this difference in activity was not the result of production of inactive enzyme. Sedimentation and gel filtration experiments indicate that the N-terminally deleted forms and the wild type enzyme were dimers. Deletion of 55 N-terminal residues did not result in significant effects on the Michaelis constants for ATP, glycine, or tRNA, while deletion of 108 N-terminal residues and two internal 64- and 200-residue deletions generated inactive forms. Five forms with C-terminal deletions of 24, 37, 59, 162, and 327 amino acid residues were soluble and intact but lacked detectable pyrophosphate exchange activity or aminoacyl-tRNA synthetase activity. The C-terminal sequence may be required for catalysis or to maintain a stable structure. Zone electrophoresis demonstrated the wild type enzyme bound both tRNA(Gly) and noncognate tRNA(Ala). Deletion of 55 N-terminal residues resulted in altered binding of tRNA(Gly) and eliminated binding of tRNA(Ala). The first 55 N-terminal residues are not essential for catalysis, dimerization, or substrate binding in aminoacylation but are required for RNA binding not associated with aminoacylation.

Structural predictions for the ligand-binding region of glycoprotein hormone receptors and the nature of hormone-receptor interactions.

BACKGROUND: Glycoprotein hormones influence the development and function of the ovary, testis and thyroid by binding to specific high-affinity receptors. The extracellular domains of these receptors are members of the leucine-rich repeat (LRR) protein superfamily and are responsible for the high-affinity binding. The crystal structure of a glycoprotein hormone, namely human choriogonadotropin (hCG), is known, but neither the receptor structure, mode of hormone binding, nor mechanism for activation, have been established. RESULTS: Despite very low sequence similarity between exon-demarcated LRRs in the receptors and the LRRs of porcine ribonuclease inhibitor (RI), the secondary structures for the two repeat sets are found to be alike Constraints on curvature and beta-barrel geometry from the sequence pattern for repeated beta alpha units suggest that the receptors contain three-dimensional structures similar to that of RI. With the RI crystal structure as a template, models were constructed for exons 2-8 of the receptors. The model for this portion of the choriogonadotropin receptor is complementary in shape and electrostatic characteristics to the surface of hCG at an identified focus of hormone-receptor interaction. CONCLUSIONS: The predicted models for the structures and mode of hormone binding of the glycoprotein hormone receptors are to a large extent consistent with currently available biochemical and mutational data. Repeated sequences in beta-barrel proteins are shown to have general implications for constraints on structure. Averaging techniques used here to recognize the structural motif in these receptors should also apply to other proteins with repeated sequences.
