Clinical therapeutic effect of drug-separated moxibustion on chronic diarrhea and its immunologic mechanisms.

In the present paper it was reported that 24 cases of ulcerative colitis and 23 cases of chronic colitis were treated by drug-separated moxibustion and the total effective rates were 95.83% and 95.65% respectively, which were markedly superior to that of western medicine group. Observation in 15 cases of ulcerative colitis showed that drug-separated moxibustion could inhibit humoral immunity and elevate cellular immune function. The therapy possibly regulated the abnormal ratio of T-lymphocyte subgroups and corrected the abnormal state of autoimmunity of patients, so that pathological changes of intestinal mucosa were corrected effectively.

[Expression of transforming growth factor beta 1 in keratinocytes of oral submucous fibrosis tissue].

To explore significance of transforming growth factor beta 1 (TGF beta 1) in the pathogenesis of oral submucous fibrosis (OSF), TGF beta 1 mRNAs in keratinocytes of the paraffin embeded tissues of 25 OSF cases, 5 normals (NOR) and 10 oral lichen planus (OLP) were determined by the in situ hybridization technique. The result showed that there was an expression of TGF beta 1 mRNA in keratinocytes of 15 OSFs (60%). The positive expression of TGF beta 1 mRNA was mainly in kerationcytes of early and middle stage OSF. There was no expression in that of 5 NORs and 10 OLPs. The study suggests that keratinocytes of OSF tissue may synthesize and release TGF beta 1 which may play an important role in the pathogenesis of OSF and participate as a mediator in the pathogenetic process of OSF.

[Successful 48-72 h cold storage of dog kidneys with HX-1 solution].

HX-1 solution was used in this study to determine its effects on long term kidney preservation. 20 female and male mongrel dogs were randomly divided into control (HC-WCU solution) and experimental (HX-1 solution) groups. Kidneys were flushed with HC-WCU or HX-1 solution and stored at 0.4 degree C for 48 or 72 h. After being preserved, the kidney was implanted in the right groin of the animal and anastomosed to the iliac vessles. This procedure was immediately followed by a contralateral nephrectomy. The experimental findings were: 1. The maximum serum creatinine levels were 642.60 +/- 158.60 mumol/L and 686.20 +/- 154.04 mumol/L respectively in HC-WCU solution subgroups for 48 or 72 h cold storage. But the maximum serum creatinine levels were 448.32 +/- 36.90 mumol/L and 524.60 +/- 109.38 mumol/L respectively in HX-1 solution subgroups. 2. 80% of the kidneys were viable after 48 h storage in HC-WCU kidney solution, but 100% viable when stored in HX-1 solution: 40% of the kidneys were viable after 72 h storage in HC-WCU solution, but 60% viable when stored in HX-1 solution. 3. Histologically the kidney of dogs that died of renal failure was damaged worse in HC-WCU solution group than that in HX-1 solution group. These results suggest that HX-1 solution is superior to HC-WCU solution for preserving kidneys and HX-1 solution may store up the kidney of dogs to 48-72 h.

Potassium channels on cardiomyocytes of selenium iodine deficiency rats and iodine deficiency rats.

Using diet control method to feed the weaning male Wistar rats with selenium and iodine deficiency Keshan endemic area food for 8 weeks to set up animal model. Singal channel recording in cell attached model was used to measure cardiac cell membrane potassium channel conductances, which coincide with the cardiac cell membrane potassium channel conductances in normal Wistar rats. The potassium channel conductance on selenium and iodine deficiency rat cardiac cell membrane is showing current-voltage increasing lineally in the range of clamping voltage 0 +/- 30 mV with channel conductance of 43.4 pS. The channel current does not increase depending on the clamping voltage that is showing the rectifying characteristic and the channel current amplitude can be augmented by added KIOa 5 mmol/L in bath solution. A kind of inward rectifying potassium channel activity was recorded, but this channel activity disappeared after lasting 6-10 minutes only. Then an inward rectifying potassium channel with the conductance of 14.2 pS was activated by KIOa 5 mmol/L introduced to bath solution. Both conductances are less than that of normal Wistar rats.

[Experimental study on aqueous areca nut extracts inducing oral submucous fibrosis in rats. I. Observation of histomorphology].

The purpose of this study was to establish the animal model of oral submucous fibrosis (OSF) in buccal mucosa of SD rats by submucosa injecting and surface painting aqueous areca nut extracts (AANE). The animals of all groups were killed randomly in the 12th, 16th, 22nd and 28th week. The buccal mucosae were collected, and were detected by light microscope and transmission electron microscope. The results were obtained: at different stages, the OSF-like lesions appeared in different degrees among some animals of the model groups, including atrophic epithelium with loss or partial loss of rate pegs, inflammation, accumulation and disorder of collagen fibre in juxta-epithelial region, along with concomitant muscle degeneration. The lesions didn't reverse obviously themselves after the authors stopped using AANE. Histomorphologic changes didn't appear in any animal of the control groups. The results indicated that the typical and stable OSF animal model were induced rapidly by submucosa injecting and surface painting AANE.

[Experimental study on aqueous areca nut extracts inducing oral submucous fibrosis in rats. II. Effection of mast cells on collagen metabolism].

The effection of mast cells (MC) on collagen metabolism of oral submucous fibrosis (OSF) induced by aqueous areca nut extracts (AANE) in rats was studied by transmission electron microscope, histochemical and biochemical analyses. The results revealed that there was a close relationship between MC and fibroblats (FB) in the pathogensis of OSF; MC which appeared in the buccal mucosa in the model groups increased significantly and became more active in the function. A great deal of collagen compacted in the buccal mucosa in the model groups. The contents of tissue hydroxyproline (Hyp) in the model groups were obviously higher than those in the control groups. Moreover, the longer the time of irritation continued, the higher the contents were. The number of MC had a higher positive correlation with the contents of tissue Hyp at the same time. Total serum Hyp had no significant difference among all the groups and any stages. The results indicated that some aqueous components of areca nuts might disturb collagen metabolism by accumulation and activation of MC.

[Isolation and determination of the seeds of Pachyrrhizus errosus protein by high performance gel filtration chromatography (GFC)].

From the seeds of Pachyrrhizus errosus, three protein constituents, namel PE1, PE2 and PE3, have been isolated and purified by extraction with 5mmol/L phosphate saline (0.9% NaCl) buffer (PB) at pH 7.2, and S-Sepharose Fast Flow Column (2.6cm x 15cm) chromatography which eluted with 5mmol/L phosphate buffer (pH 7.0) containing 1mmol/L NaCl. Three proteins were burther separated on two connected Protein-Pak 60+Protein-Pak 125 [7.5mm x 39cm, 10microm] columns with mobile phase of 0.2mol/L phosphate buffer (pH 6.5). The flow rate was kept constant at 0.8mL/min by YSB-2 type high press pump. The effluent was monitored at a wavelength of 280nm on photodiode array detector. These three proteins are proved to be homogeneous by SDS-PAGE, IEF and HPGFC experiments, and all present the typical absorption spectra in ultraviolet region. The moleculer weights of the three proteins are approxiamtely 33000D, 14500D and 14000D respectively by SDS-PAGE. But as using HPGFC analysis, the MW value of PE2 is 28000D. This indicates PE2 may be composed of two chains joined by disulfide bond, which is further proved from the latter amino acid composition analysis. The isoelectric points of three proteins are 4.5, 6.5 and 7.5 respectively by using IEF. The amion acids compositions of the three proteins were determined with OPA post-column derivatization/fluorescence detection.

[The determination of vitamin D in fortified milk and other foods by high performance liquid chromatography (HPLC)].

An HPLC procedure for determination of vitamin D(VD) in fortified milk and food has been developed. The samples were saponified by adding KOH, ethanol and vitamin C at 75 degrees C for 30 min. The digest was extracted with hexane in multistep manner. The extract was washed with 5% KOH, distilled water and 55% ethanol, and evaporated under reduced pressure at 50 degrees C. The residue was dissolved in hexane, transferred to a coentrifuge tube, evaporated under N2 and then dissolved in 200 microL mobile phase. The latter was injected into a silica column (10 microm, 30 cm x 4 mm) for cleanup, eluted with mixture of cyclohexane-hexane containing 0.8% isopropanol and detected at UV 254nm. VD fraction was collected according to the retention time of standard, evaporized under N2, redissolved in 100 microL MeOH, and separated on a C18 column (10 microm, 30 cm x 4 mm) with MeOH/H2O (98:2) as eluent. VD2 and VD3 were eluted as a mixture under the conditions. Recoveries of the method were 94.8% to 99.7%. The average CV (n = 5) for within and between day measurements were 1.6% and 2.1% respectively. This method was already used to determine the content of vitamin D in milk powder, pulvis elementi compound, mixed proteins, dried meat floss, humanized milk power, children health drink, milk wheat powder and fortified milk. The method is simple, rapid and accurate.

Testing for treatment related trend with partially exchangeable clustered data.

When testing for treatment related effect, it is common to focus on data that describe some primary response, even though an experiment might yield data on secondary, less conspicuous effects. An example of such an experiment can be found in developmental toxicity studies, where a primary response might be the presence of skeletal malformation in a fetus, and a secondary effect might be reduced weight. In such studies, weight reduction may occur not only among malformed fetuses, but also among normal fetuses. In this paper, we use a likelihood ratio procedure to construct treatment related trend tests for developmental experiments with such outcomes. We assume that within each cluster, data are partially exchangeable and we use the presence or absence of malformations as categorical covariates. Two models are considered for the covariance structures: In the general model, partially exchangeable covariance matrices are used for each cluster within dose groups. In the homogeneous model, a common exchangeable covariance matrix is used for all clusters. Maximum likelihood estimates (MLEs) of unknown parameters in the general model are obtained by directly maximizing the log-likelihood function through careful numerical consideration, while MLEs of parameters of teh homogeneous model are obtained through an iterative procedure. Two applications to developmental toxicity data are described.

Induction of chromosome aberrations in human cells by charged particles.

Chromosome aberrations induced by high-energy charged particles in normal human lymphocytes and human fibroblasts have been investigated. The charged particles included 250 MeV/nucleon protons, 290 MeV/nucleon carbon ions and 1 GeV/nucleon iron ions. The energies of the charged particles were higher than in most of the studies reported in the literature. Lymphocytes were stimulated to grow immediately after irradiation, while fibroblasts were incubated at 37 degrees C for 24 h for repair. Chromosomes were collected at the first mitosis after irradiation and chromosome aberrations were scored using the fluorescence in situ hybridization (FISH) technique with a whole-chromosome 4 probe. Chromosome aberrations were classified as reciprocal exchanges, incomplete exchanges, deletions and complex exchanges. The relative biological effectiveness (RBE) for each type of aberration was calculated by dividing a dose of 4 Gy by the dose of the charged particles producing the same effect as 4 Gy of gamma rays. Results of this study showed that complex aberrations have the highest RBE for radiation of high linear energy transfer (LET) for human lymphocytes, but for fibroblasts, the greatest effect was for incomplete exchanges. For both lymphocytes and fibroblasts, iron ions induced a similar fraction of aberrant cells.
