[Studies on the viability of pathogenic amebae of the genus Naegleria in water of different sources (author's transl)].

Water specimens taken from the public water supply (A), a swimming-pool (B), and an inland lake (C) were infected with pathogenic amebae of the genus Naegleria (strain TY/Richmond). The specimens were stored at various temperatures for different periods. The water was then filtered and examined for naegleriae by animal inoculation. The results summarized in table 1 indicate that pathogenic naegleriae are viable at least for some weeks in warm tap and natural waters. Water from indoor swimming-pools however, may be a less suitable medium for the viability of pathogenic naegleriae.

[On micromorphological changes of bradyzoites of toxoplasma cysts in cerebral tissue after storage under freezing at--20 degrees C (author's transl)].

Toxoplasma bradyzoites in cerebral tissue are no longer infectious and able to replicate after 24 hrs of storage under freezing at--20 degrees C. No intact bradyzoites could micromorphologically be demonstrated in the cysts. Clear changes in the structure and lesions of the cell walls and basic substance of the cysts, and of the bradyzoites are held responsible for the destruction of the parasites being surrounded by host tissue.

[Course of cellular immunity in urologic neoplasms].

Apart from a purely qualitative evidence concerning the presence of a specific sensibilisation it is possible to estimate also its strength or the blocking factors appearing in the serum of tumour carriers by means of the electrophoresis mobility test. By this it seems to be possible to make an evidence concerning the immunological reaction position of the tumour patient with the possibility to make conclusions as to the prognosis of the patient.

[A contribution to the early diagnosis and differential diagnosis of bladder neoplasms by means of immunological methods].

According to the present examinations it is pretty probable that already in early histological forms of the carcinoma of the urinary bladder the EM-test is able to indicate immunologically malignity, i.e. that the papillary non-infiltrating carcinoma of the transitional cell as well as the carcinoma in situ of the urinary bladder (23) also immunologically behave like tumors of the bladder. In differential-diagnostic questions concerning the organ localisation of the primary tumor the EM-test seems to give valuable references. Histologically not unequivocally malignant tumors of the bladder with positive results in the EM-test should be submitted to an exacter examination and control.

Immunization of NMRI mice against virulent Toxoplasma gondii. Differing efficacy of eleven cyst-forming Toxoplasma strains.

Mice were infected with eleven cyst-forming Toxoplasma strains of varying virulence and challenged 1 month later with highly virulent BK strain parasites. The early cellular reaction in vivo was estimated by collecting the peritoneal exudates 24 h after challenge. This consisted of 54.34--77.61% lymphocytes and 0-8.88% infected macrophages in the eleven immunized groups in contrast to 27.17% lymphocytes and 18.64% infected macrophages in the control group. The peritoneal exudate 72 h after challenge comprised of 55.88-73.86% lymphocytes and 0-6.97% infected macrophages compared to 25.88% lymphocytes and 92% infected macrophages in the control group. Following the virulent challenge, deaths occurred in seven of the eleven groups immunized with live strains and ranged 5-42% at the end of a 6-week observation period. The most virulent of the Toxoplasma strains used - Alt and Gail - gave higher mortality while the least virulent ones - 558, 1070, K8 and KSU - were solidly resistant. The significantly different mortality obtained with strains Alt, Gail and Witting in a similar experiment performed at an interval of 7 months is discussed.

[Diagnostic significance of identification of anti-entamoeba histolytica antibodies (author's transl)].

Examinations were performed with the indirect immunofluorescence test (IFAT) and the indirect haemagglutination test (IHAT). Negative titers (less than 1:5) do not exclude an infection with E. histolytica. Titers up to 1:80 in both reactions are found in patients with negative stool specimens or indicate an asymptomatic infection of the intestinal lumen, an incipient or past symptomatic amoebiasis. IFAT values of 1:160 together with a negative or positive IHAT are also suggestive of the two last-mentioned types of infection. These titers are also found in patients with gastrointestinal disorders of other orgin. IFAT values higher than 1:60 with a positive IHAT are probably due to an invasive amoebiasis. Titres are, however, only diagnostic pointers.

[In vitro susceptibility of bacteroidaceae to cefoxitin and cephalothin (author's transl)].

The activity of the new cephamycin C derivatives cefoxitin against 102 clinical isolates of Bacteroidaceae (95 Bacteroides strains and 7 Fusobacterium-Sphaerophorus cultures) was determined by tube dilution and standardized agar diffusion tests and compared to that of cephalothin. Cefoxitin was more active than cephalothin against 56 Bacteroides fragilis strains, 19 Bacteroides thetaiota-omicron strains and two Bacteroides splanchnicus strains. The difference seemed less pronounced with five Bacteroides vulgatus and four Bacteroides variabilis cultures. Three Bacteroides melaninogenicus and six Bacteroides oralis strains displayed similar sensitivity to both cephalosporins. At concentrations of 32 microng/ml or less, cefoxitin inhibited 91% and cephalothin 33% of the Bacteroides strains. Two strains of Sphaerophorus varius were likewise much more sensitive to cefoxitin than to cephalothin (MIC values 8 micron8/ml of cefoxitin and 256 microng/ml of cephalothin). Three other Sphaerophorus strains (the species necrophorus and freundii/mortiferum) and two of Fusobacterium fusiforme (Fusobacterium nucleatum) strains were very similar in their susceptibility to both cephalosporins. Statistical analysis of the relationship between MIC and zone size diameter showed relatively high correlation coefficients for cefoxitin and cephalothin (r=-0.741 and r=-0.703 respectively).

[In vitro activity of mezlocillin, azlocillin and carbenicillin against bacteroidaceae with particular reference to bacteroides fragilis (author's transl)].

The in vitro susceptibility of approximately 90 Bacteroidaceae strains to the the new broad-spectrum penicillins mezlocillin and azlocillin was determined by tube dilution tests and compared to susceptibility to carbenicillin. Bacteroides fragilis and Bacteroides thetaiotaomicron predominated among the strains tested. These strains are particularly important as pathogens and are known to produce beta-lactamase. At concentrations of 16 microng/ml or less, mezlocilin inhibited 81% of the Bacteroides cultures, and at 32 microng/ml 96% of the cultures. The amount of Bacteroides strains inhibited by 16 and 32 microng/ml of azlocillin was 67% and 93% respectively. Similar results were obtained with carbenicillin, concentrations of 32, 64 and 256 microng/ml inhibiting 55%, 87% and 94% of the strains respectively. Mezlocillin and azlocillin did not appear to be any more effective than carbenicillin against Sphaerophorus strains. Statistical analysis of the relationship between minimal inhibiting concentration and inhibition zone diameter showed that the susceptibility of Bacteroidaceae to mezlocillin, azlocillin and carbenicillin cannot be determined with sufficient accuracy by a standardized agar diffusion test.

A comparison of the kinetics of the macrophage electrophoretic mobility (MEM) and the tanned sheep erythrocyte electrophoretic mobility (TEEM) tests.

In this study the macrophage electrophoretic mobility (MEM) test was modified by using tanned sheep erythrocytes in place of guinea pig peritoneal macrophages as the indicator cells of lymphocyte sensitization to antigens. This modification is named the tanned sheep erythrocyte electrophoretic mobility (TEEM) test, and a comparison of the kinetics of the two systems allowed the following conclusions to be made: 1) Treatment of freshly drawn sheep red blood cells with a concentration of 1/40,000 tannic acid produced optimum results in the TEEM test. 2) Lymphocyte-antigen and lymphocyte-number response curves show similarity in the two test systems. 3) A plateau response with slowing factor is achieved at a lower dilution in the TEEM test than in the MEM test. 4) Whilst similarity in the first stage reaction was found in the two systems, in the second stage of the test (at 37 degrees C) tanned sheep red cells gave a plateau response after 45 min instead of the 60 min found in the MEM test. 5) The two slowing factors showed similar gel filtration patterns with molecular weights between 13,000 and 15,000 daltons, and had equivalent activity in both test systems. 6) The disadvantages of the guinea pig macrophage as an indicator cell are discussed. 7) The TEEM test seems simpler to perform than the MEM test and may be widely applicable in clinical immunology for the estimation of lymphocyte sensitization.

Lymphokines of T and B cells. I. Demonstration that T and B cells of tumor antigensensitised mice produce different lymphokines changing surface charge of target cells.

Lymphocytes from MC sarcoma-bearing mice were exposed to a soluble tumor antigen. The lymphokine-containing supernatant thus obtained was fractionated on Sephadex G-75. Fractions were tested for their activity to reduce the surface charge of indicator macrophages by cell electrophoresis technique. When unseparated spleen or lymph node cells were used the charge-reducing activity of the supernatant was found in three regions according to molecular weights of about 100,000, 60,000 - 35,000 and 13,000 Daltons. The appearance of activity was found to be time-dependent. As indicated by tests with separated lymphocytes both B and T cells appear to release charge-reducing activity.

[Growth of non-sporing anaerobes in an oxygen-free blood culture system (author's transl)].

The efficacy of the commercially available Vacutainer blood culture system to support the growth of non-sporing anaerobes was compared with two laboratory-prepared blood culture media (supplemented thioglycollate medium and brain heart infusion). The media were inoculated with 10, 100 and 1000 organisms of the species tested, and the number of colony-forming units was determined at intervals of 8-10 hrs. Analogous experiments were performed with batches of the media to which 10% vol/vol of human blood had been added. From the results obtained with strains of Bacteroides fragilis, Bacteroides thetaiotaomicron, Bacteroides splanchnicus, Sphaerophorus necrophorus, Peptostreptococcus anaerobius and Propionibacterium acnes it became obvious that growth of non-sporing anaerobes was significantly enhanced in the Vacutainer culture tubes, whereas Enterobacteriaceae and Streptococcus spp. grew equally well in all the media tested.
