The volatile oil of Baizhu was included with beta-CD. The factors affecting the formation of the inclusion compounds were researched by L9(3(4)) orthogonal test. The ratio of the oil in inclusion compounds and recovery of inclusion compounds were used as criteria in the test. The best conditions for inclusion were A2B3C1, that is, the compounds should be formed in the aqueous solution containing beta-CD and oil with a raio of 8:1, aqua destillate and beta-CD with a ratio of 4:1 for two hours.
OBJECTIVE: To study the protective effects of nerve growth factor(NGF) on noise-induced hearing damage in guinea pigs. METHODS: NGF injected Guinea pigs were consecutively exposed to white noise of 115 dB(A) for 6 days continually (45 min.d-1). Auditory thresholds were measured using auditory cortex evoked response to tone bursts in different post-exposure intervals (1 h, 1 d, 2 d, 3 d and 6 d). The ultrastructural changes within hair cells were also observed by a transmission electron microscope(TEM). RESULTS: The auditory threshold shifts in test group A(NGF:1,000 U.kg-1.d-1, i.m.), B(NGF:2,000 U.kg-1.d-1, i.m.) and C(NGF:3,000 U.kg-1.d-1, i.m.) were significantly fewer than that in the control group(Saline: 1 ml.kg-1.d-1, i.m.). Threshold shifts almost recovered in test group B and C 3 days after the exposure; while a threshold shift of (16.43 +/- 6.91) dB was present 6 days after the exposure in the control group. TEM showed that all three rows of the outer hair cells(OHCs) of the basal turn in the control group displayed significant pathological changes. Depolymerization of actin filiaments within stereocilia, swelling of submembraneous cistern and the efferent nerve-ending and slight edema of hair cells were evident. In test group A, the hair cells display slight pathological changes, which are confined in the third row of OHCs in a local position of the basal turn. In group B and C hair cells have nearly normal appearance. CONCLUSION: NGF is able to reduce threshold shift, and promote the recovery of auditory threshold in acoustic trauma. This factor can, to some extent, protect against noise-induced hearing damage.
OBJECTIVE: To develop a valid behavioral animal model of tinnitus and to determine the animals whether have the perception of tinnitus and to evaluate effects of nimodipine and Chinese herbs on tinnitus using the behavioral model. METHODS: Adult male albino rats (n = 60) were fell evenly and randomly into ten groups. Animals in Group I and II were administrated sodium salicylate 350 mg/kg/d (i.p.). Instead, animals in control groups (Group III and IV) were saline injected. Pure tone (8 kHz, 65 dBSPL) was given to rats in Group V and VI instead of sodium salicylate. Nimodipine (1 mg/kg) were administrated to animals in Group VII and VIII. Gushenshuerfang, one kind of Chinese herbs were administrated to animals in Group IX and X. Medicine or pure tone were being given to all animals in Group I, III, V, VII, IX before conditioned reflex. Animals in Group II, IV, VI, VIII, X began to have medicine or pure tone after conditioned reflex. Thirsty animals (deprived of water for several days) were breeded in sound-proofed room and listened a persistent white noise (55 dB SPL) and trained daily with our instrument system, according to the procedure based on Pavlovian conditioned suppression paradigm. Subjects were permitted to lick water at an automatic dispenser during daily training sessions and the number of licks was recorded automatically. Noise offset (silence) was the conditioned stimulus and the electrical shock was the unconditioned stimulus. After two or three days training, subjects learnt rapidly to associate noise offset with shock and then established a behavioral conditioned reflex. Because of shock fear they decrease or even stop licking with the presentations of conditioned stimulus. However, subjects gradually forgot or ignored the fear without any shock present in extinction test sessions. RESULTS: Animals' licking recovered to their pre-training levels respectively at the following days: Group I--5 days, Group II--2 days, both Group III and IV--4 days, Group V--5 days, Group VI--2 days, Group VII--5 days, Group VIII--4 days, Group IX--5 days, Group X--4 days. There were much significant differences among all groups and all training days (F test, P < 0.001). Above results provided sufficient evidence for that tinnitus was induced by salicylate and tinnitus presented with noise offset. In group I, tinnitus became additional conditioned stimulus. Therefore, the fear of animals was much severe and the numbers of licking were much fewer. Extinction lasted longer. In group II, animals ignored the noise offset much easily and the extinction lasted much shorter. Moreover, results from group V and VI, pure tone instead of salicylate, indicated that sensation (tinnitus) sounded like pure tone. The groups VII, VIII, IX, X demonstrated that nimodipine and Gushenshuerfang, one kind of Chinese herbs can cure the tinnitus induced by salicylate. CONCLUSION: All behavioral evidence indicated that animals perceived tinnitus induced by salicylate and this animal model could be used in evaluating curative effects of some medicine on tinnitus.
OBJECTIVE: To map the tinnitus specific central activity and foci site in brain and to investigate the effects of hearing loss, tinnitus side, right or left handed on foci site. METHODS: Glucose metabolic activity of brain was studied with positron emission tomography (PET) using 18F-FDG as radiotracer. Seventeen patients with severe tinnitus and fifteen control subjects participated in this study. All subjects were fell into four groups according to their hearing levels. There were 13 tinnitus patients with hearing loss in Group one, 4 tinnitus patients without hearing loss in Group two, 2 control subjects with hearing loss in Group three and 13 control subjects without hearing loss in Group four. Statistical parameters mapping (SPM) was used to analyze the PET data and to determine the Brodmann area (BA) according to Talairach coordinate system. RESULTS: Increase of neuronal activity caused by tinnitus occurred predominately in the left hemisphere with significant foci in the transverse temporal gyrus (BA41), superior temporal gyrus (BA42, 22), anterior middle temporal gyrus (BA38) and hippocampus. Bilateral hemispheres were concerned with the neuronal activity caused by hearing loss while foci site were located at the posterior superior temporal gyrus(BA42,22), medial portion of middle temporal gyrus(BA21), combined auditory area(BA39). Left middle frontal gyrus (BA8, 9) as well as left inferior frontal gyrus(BA45) were also link up with hearing loss. CONCLUSION: As a new path in research, PET has provided an objective evidence for tinnitus and may be used as a potential tool in objectively measuring tinnitus. Tinnitus-related brain areas and their influence factors were discussed.
OBJECTIVE: To investigate the possible role of inducible nitric oxide synthase (iNOS) in esophageal carcinogenesis. METHODS: iNOS expressions in normal epithelial cells(NC), hyperplastic cells(HC), atypical hyperplastic cells(AHC) from grade I to III, carcinoma in situ(CIS), squamous cell carcinoma(SCC), and adenocarcinoma(ADC) were detected with immunohistochemistry. RESULTS: The positive rates of immunohistochemistry staining was 0 for NC, 2.5% for HC, 4.0%, 7.5% and 2.5% for AHC grade I to III respectively, 1.4% for CIS, 8.5% for SCC and 71.4% for ADC. CONCLUSIONS: There is a high expression of iNOS in human esophageal adenocarcinomas. Frequent expression of iNOS may be a potential event in ADC carcinogenesis. There is no correlation between iNOS and SCC carcinogenesis.
Condyloma acuminata(CA) is one of sexually transmitted diseases(STD) caused by human papillomavirus(HPV). We have done systemic study on the CA in recent years. Analyzing the morbidity of CA in our STD clinics showed that CA was in the first or second place of STD. Tissue specimens from CA were detected HPV DNA by dot blot hybridization and PCR. The result showed PCR was more sensitive than dot blot hybridization. HPV DNA were detected by PCR in tissue specimens of cervical squamous carcinoma, as well as cervical and vulvar CA. The result confirmed that HPV 16 was main type in cervical squamous carcinoma, and HPV 6 and 11 were main types in cervical and vulvar CA. To differentiate pseudocodyloma of vulva from CA, electron microscopy and HPV DNA detection were done. Certain ultrastructural changes were helpful, and detection of HPV DNA was better method in differentiating CA from pseudocondyloma. High purification of podophyllintoxin was studied and the effectiveness of podophyllintoxin was observed, compared to imported podophyllintoxin to treat CA, both cure rates were more than 80% after 3 week's therapy. Intralesional injection of different interferons for CA was effective.
OBJECTIVE: To observe preventive and therapeutic effect of Chinese herbs Naofeikang on hypoxic pulmonary artery hypertension of elastase-induced emphysema hamsters and investigate its mechanism. METHODS: Hamsters for 30 days after intratracheally-instilled elastase, were kept in hypoxia environment under normal atmospheric pressure for 15 days, 50 hamsters were divided into prevention group (Prv), treatment group (T), emphysema + hypoxia (EH), and control group(N). Mean pulmonary artery pressure (MPAP) was measured before hamsters were killed. And then right ventricle hypertrophy index (RVHI) was measured, as well as circulating endothelial cells (CEC) and cells recoveries from bronchoalveolar lavage fluid (BALF) were counted. Meanwhile, pulmonary tissue changes were studied under light microscope with morphometric analysis. RESULTS: Compared with EH group, MPAP, CEC, and cells recoveries of BALF of Prv and T groups were significantly decreased (P < 0.05 or P < 0.01); Mean linear intercept and percentage of arterial media area of Prv and T groups had totally a significant difference (P < 0.01) in comparison with EH group. CONCLUSIONS: Chinese herbs Naofeikang could lower pulmonary hypertension, preserve vessel endothelial cells and lessen the inflammatory reaction in pulmonary tissue. Thereby, it could hinder the further development of emphysema and inhibit the remodeling of pulmonary small artery.
OBJECTIVE: To understand the characteristic of steroid diabetes by the analysis for clinical data of 162 cases with Cushing syndrome (CS) proved by pathological report. METHODS: Retrospective analysis for the data of CS about age of CS onset, duration of CS, body mass index (BMI), 24 hours urine free cortisol (24 h UFC), pathological report, fasting blood glucose (FBG) pre- and post-operation, family history of diabetes, treatment of glucose dysbolism (GD) pre- and post-operation. RESULTS: The onset age and duration of CS in GD group was obviously earlier and longer than normal glucose (NG) group. BMI and 24 h UFC were no difference between two groups. Besides adrenal adenocarcinoma, no difference was found in types of pathology. FBG of diabetes pre-operation was obviously lower than post-operation. After operation, most diabetes need not on drug treatment. Only one secondary diabetes has family history. CONCLUSIONS: Steroid diabetes easily occurs in patients who is senility and suffer longer course of CS. No relation to diabetic family history, concentric obesity, the level of UFC and benign types of pathology. Therapeutical emphasis of steroid diabetes is radical cure of CS.

RESULTS: Recurrent rate of oral furtulon therapy for bladder tumors (8.8%) and instillation of mitomycin C for preventing the recurrence of bladder tumors (12.5%) as compared to recurrent rate of control group(40%), which was statistically significant(P < 0.01). Overall, 4 patients (33.3%) had an effective response with furtulon. CONCLUSIONS: Furtulon has more effective for treatment and prophylaxis of bladder tumors.
OBJECTIVE: Patients with Graves' disease (GD) have marked lymphocytic infiltration in their thyroid glands. We examined the gene for the variable regions of the alpha-chain of the Chinese T-cell receptor (V(alpha) gene) in intrathyroidal T cells to determine the role of T cells in the pathogenesis of GD and offer potential for the development of immunotherapeutic remedies for GD. METHODS: We used the reverse transcription and polymerase chain reaction (RT-PCR) to amplify complementary DNA(cDNA) for the 18 known families of the V(alpha) gene in intrathyroidal T cells from 5 patients with Graves' disease. The findings were compared with the results of peripheral blood T cells in the same patients as well as those in normal subjects. RESULTS: We found that marked restriction in the expression of T cell receptor V(alpha) genes by T cells from the thyroid tissue of Chinese patients with GD(P < 0.001). An average of only 4.6 +/- 1.52 of the 18 V(alpha) genes were expressed in such samples, as compared with 10.4 +/- 2.30V(alpha) genes expressed in peripheral blood T cells from the same patients. The pattern of expressed V(alpha) genes differed from patient to patient with no clear predominance. CONCLUSIONS: Expression of intrathyroidal T cell receptor V(alpha) genes in GD is highly restricted suggesting the primacy of T cells in causing the disorders.

In this paper, the absorption and fluorescence spectra of 7-Hydroxyquinoline (7HQ) in the copolymer of methyl methacrylate (MMA) and methacrylic acid (MAA) monomers were observed. The influence of different matrices on the species of 7HQ and the excited state proton transfer effect were investigated. The results showed that, as the number of carboxylic group in the matrix was increased, the relative contents of different species of 7HQ should be changed and the excited state proton transfer effect of 7HQ was enhanced.
In this paper, the fluoride-oxide glass doped with Er3+ for upconversion from IR to green was report. The component of sample in experiment is 60TeO2-8PbF2-10AlF3-10BaF2-10NaF-2ErO3/2, and the prepared method is obtained. The absorption spectrum was measured at room temperature. Intense green upconversion emissions from Er3+ doped the sample were observed under 808 nm and 970 nm excitation, and the upconversion emissions spectra under 808 nm and 970 nm LD excitation were measured at room temperature. The upconversion mechanics was discussed. The relationship between working currents of LD and intensity of upconversion emissions has been studied. The results was improved that needs of two photons for upconversion process under 808 nm and 970 nm excitation respectively.
A new method of second order derivative fluorimetry for the simultaneous determination of tryptophan and tyrosine is described. The measurement of emission spectra is carried out with excitation at 221 nm in a KH2PO4-NaOH buffer solution (pH = 7.4). The second-derivative peak is at 318 nm and 283 nm for tryptophan and tyrosine respectively. The two amino acids can be determined directly from corresponding second order derivative signals. Linear regression equation of the calibration graph is c = 0.000 7H-0.004, with a correlation coefficient of linear regression of 0.9964 for tryptophan and c = 0.001 2H-0.0040, with a correlation coefficient of linear regressin of 0.9971 for tyrosine. The linear ranges of determination of tryptophan and tyrosine are from 0.004 to 0.200 microgram.mL-1 and from 0.002 to 0.025 microgram.mL-1, respectively. Effects of pH, temperature and foreign ions on the determination of tryptophan and tyrosine have been examined. Recovery is from 92.0% to 104% for tryptophan and 98.7% to 102.0% for tyrosine, and relative standard deviation of 3.5% and 2.8%, respectively.

Although the important roles of RANK/RANKL in osteoclastogenesis have been established, their roles in the regulation of mature osteoclasts remain uncertain. Microisolation has been used to obtain pure populations of rat and human osteoclasts for RT-PCR analysis. RANK and calcitonin receptor mRNA was detected in all the samples whereas OPG and ALP mRNA was not present in any. RANKL mRNA was detected in two of eight rat and one of four human samples. Treatment of osteoclasts with soluble RANKL resulted in translocation of NF-kappaB to the nucleus and elevation of cytosolic and nuclear calcium levels. We have shown that RANK is highly expressed in mature osteoclasts and that its stimulation by RANKL results in activation of NF-kappaB and calcium signalling.
Methylenetetrahydrofolate reductase (MTHFR) is the least understood enzyme of folate-mediated one-carbon metabolism in plants. Genomics-based approaches were used to identify one maize and two Arabidopsis cDNAs specifying proteins homologous to MTHFRs from other organisms. These cDNAs encode functional MTHFRs, as evidenced by their ability to complement a yeast met12 met13 mutant, and by the presence of MTHFR activity in extracts of complemented yeast cells. Deduced sequence analysis shows that the plant MTHFR polypeptides are of similar size (66 kDa) and domain structure to other eukaryotic MTHFRs, and lack obvious targeting sequences. Southern analyses and genomic evidence indicate that Arabidopsis has two MTHFR genes and that maize has at least two. A carboxyl-terminal polyhistidine tag was added to one Arabidopsis MTHFR, and used to purify the enzyme 640-fold to apparent homogeneity. Size exclusion chromatography and denaturing gel electrophoresis of the recombinant enzyme indicate that it exists as a dimer of approximately 66-kDa subunits. Unlike mammalian MTHFR, the plant enzymes strongly prefer NADH to NADPH, and are not inhibited by S-adenosylmethionine. An NADH-dependent MTHFR reaction could be reversible in plant cytosol, where the NADH/NAD ratio is 10(-3). Consistent with this, leaf tissues metabolized [methyl-(14)C]methyltetrahydrofolate to serine, sugars, and starch. A reversible MTHFR reaction would obviate the need for inhibition by S-adenosylmethionine to prevent excessive conversion of methylene- to methyltetrahydrofolate.
