[Lonazolac-Ca-a non-steroidal antirheumatic agent. Long-term clinical study in chronic polyarthritis].

During a controlled open long-term study, Lonazolac-Ca, a new nonsteroidal antiphlogistic substance, has been tested in 35 patients with rheumatoid arthritis (stage I-III, according to Steinbrocker). The patients took 600 mg Lonazolac-Ca daily in three equal doses. The observation period was one year. Clinical and laboratory controls were carried out in monthly intervals. In more than 50% of the patients the tested parameters pain, swelling and active mobility improved significantly. Because of acute deterioration of the basic disease which necessitated the application of steroids, therapy with Lonazolac-Ca was discontinued in some cases. Lonazolac-Ca is well tolerated. But also as with other non-steroidal antiphlogistics, administration of the substance may be restricted in individual cases due to gastric symptoms. No adverse effect on blood count, liver and kidney function was seen.

[Age-related changes in peripheral thyroid parameters, it hypophyseal regulation and relation to fat metabolism].

In 50 women and 57 men aged 20 to 80 years serum levels of T3, T4 and rT3 as well as TSH levels before and after intravenous application of TRH and serum triglycerid- and cholesterol-levels were determined. Regressions and multiple regressions were calculated with Broca-Index, and age. Serum-T3 are significantly lower in old age than in younger people. In men serum T4-levels remain unchanged in old age, in women a significant rise can be observed. With rising age TBK rises in men and falls in women. Women older than 50 years have significantly higher Serum-triglycerid- and cholesterrol-levels, Broca-Index being unchanged, than younger women. In men no remarkable change of these parameters could be detected. Calculating multiple regressions we found significant relations parameters could be detected. Calculating multiple regressions we found significant relations of T3-serum levels to either age and TBK or age and T4, as well as relations of rT3 serum levels to age and T4 levels in men and women. Only in men TSH-secretion following intravenous TRH application is related to age and serum-T3, or age and T4-serum levels or age and rT3-levels in serum. Broca-Index in men is related to age and T3-levels in serum. In both sexes serum-triglycerid-levels are significantly related to Broca-Index and rT3-levels or Broca-Index and T4-concentration in serum. Serum-cholesterol or triglyceride-levels correlate in women with age and T3-serum-concentrations. With increasing age in men and women hypothyreodism seems to be increasing.

[Gold treatment of progressive chronic polyarthritis--a clinical study].

In an intraindividual series of investigation consisting of 30 patients with rheumatoid arthritis the effect of gold therapy (Aurothioglucose) has been proved controlling biochemical and clinical parameters. Controls of the function-index by Steinbrocker et al. have shown a significant improvement in comparison with the stage before gold treatment. Futhermore a significant decrease of erythrocyte sedimentation rate (Westergren) as well as a significant increase of albumins and a significant decrease of gamma-globulins in proteinelectrophoresis provoked by gold treatment could be demonstrated. Side effects during gold treatment were observed in 5 patients; therapy was stopped in 2 patients.

Sex chromosome determination in extragonadal teratomas by interphase cytogenetics: clues to histogenesis.

Teratomas are neoplasms that are composed of tissues from all three germinal layers. The exact histogenetic origin of teratomas, however, is still controversial. In order to gain more insight into histogenesis of extragonadal teratomas (EGTs), the gonosomal status in 13 congenital EGTs was studied by means of interphase cytogenetics using nonradioactive in situ hybridization (NISH) with centromere-specific DNA probes. By use of this technique a direct correlation of cytogenetic results with morphology was possible. In all EGTs analyzed the gonosomal status in tissues derived from the different germinal layers was identical to that of the nontumorous fetal and placental tissue. This was true irrespective of localization, age, histological type, and classification of the EGT. Our results strongly suggest that EGTs arise from pluripotent diploid precursor cells, for example, either premeiotic germ cells that have not yet undergone the first meiotic division or pluripotent ectopic embryonal or extraembryonal cells. Our data do not support the theory of parthenogenetic EGT development, at least in males.
