[The antipyrine test in occupational medicine research--evaluation of individual reproducibility and frequency distribution of half-life].

The antipyrine half-life is a reliable index of biotransformation capacity of the liver. The half-lives were estimated in 137 subjects; the result was a logarithmic normal distribution of these parameters. Therefore our patients were subdivided into groups of so-called fast, intermediate and slow metabolisers (fast: t50 less than 7.6 h and slow: t50 greater than 14.9 h). Under conditions given here the intraindividual coefficient of variation was about 11.5%. The antipyrine elimination curves were calculated by a computer programme and the values were analysed statistically.

Intertypic cross-neutralization of polioviruses by human monoclonal antibodies.

Human B cell lines producing monoclonal antibodies (mAbs) reactive with poliovirus type 1 were generated by transformation with Epstein-Barr virus (EBV) B 95-8 of tonsillar lymphocytes from several immune donors. EBV-transformed cells were cloned in semisolid agarose. Some neutralizing (Nt) human mAbs recognized and neutralized only poliovirus type 1, whereas other Nt mAbs neutralized either poliovirus type 1 and 2 or all three serotypes. mAbs reactive with poliovirus type 1 and 3 but not with type 2 were not detected. Immunoprecipitation of radiolabeled poliovirus type 1 with cross-reactive human Nt mAbs was inhibited competitively by preincubation of mAbs with cold poliovirus type 3 and/or 2.

Evidence for a complex structure of neutralization antigenic site I of poliovirus type 1 Mahoney.

We have selected neutralization escape mutants by using a monoclonal antibody (nt-MAb) against a sequential epitope between amino acids 93 through 104 (neutralization antigenic site I) of poliovirus type 1 Mahoney. The majority of mutants were also resistant against five strain-specific nt-MAbs which recognized conformation-dependent epitopes, suggesting that the neutralization antigenic site I must be involved in the formation of such epitopes. An analysis of all mutants by the binding of nt-MAbs and by isoelectric focusing of VP1 allowed discrimination of five classes of mutants. Sequence analysis of mutant RNAs revealed point mutations and deletions in the antibody-binding site.

In vitro stimulation of presensitized mouse spleen cells with poliovirus type 1, Mahoney, and enhancement of poliovirus-specific hybridomas.

In vivo immunization of BALB/c mice with poliovirus type 1, strain Mahoney, or with its purified polypeptides resulted in 0.2 to 0.5 antigen-specific hybridoma microcultures per 10(6) spleen cells. Stimulation of spleen cells from mice immunized with poliovirus or with its polypeptides in vitro with poliovirus 6 days prior to fusion with the myeloma cells led to a six- to 20-fold increase in the number of positive microcultures, i.e. after stimulation the yield of poliovirus-specific hybridomas was up to 3.8 antigen-specific microcultures per 10(6) spleen cells. The in vitro stimulation of spleen cells primed in vivo was demonstrated by the detection of poliovirus-specific antibody-producing cells 6 days after in vitro cultivation in the presence of poliovirus as antigen. Only spleen cells stimulated under these conditions in vitro gave rise to specific antibody-producing cells and yielded antigen-specific hybridomas after somatic hybridization.

Binding of neutralizing monoclonal antibodies to empty capsids of poliovirus can be blocked by monospecific antisera to structural polypeptides VP1 and VP2.

Binding of two neutralizing monoclonal antibodies (Nt-mAbs) to natural empty capsids (NEC) of poliovirus, type 1, was blocked to the extent of 83 per cent to 98 per cent by monospecific rabbit antisera directed against the structural polypeptides VP1 and VP2. Monospecific antisera against VP3 or VP4, however, did not show this blocking effect. It is therefore assumed that VP1 and VP2 are located close together at the antigenic sites for the two mAbs.

Self-reactive B lymphocytes detected in young adults, children and newborns after in vitro infection with Epstein-Barr virus.

B-lymphocytes from healthy children and young adults who were seronegative for autoantibodies and B lymphocytes from umbilical cord blood of newborns were induced to secrete a variety of autoantibodies upon infection with Epstein-Barr virus. Such autoantibody-secreting clones were obtained from different lymphoid tissues and occurred at frequencies of 1 in 10(6)-10(7) mononuclear cells. The autoantibodies were exclusively of the IgM class. They recognized normal cellular components, such as cytoplasmic, nuclear and cytoskeletal antigens. The fact that self-reactive clones were not efficiently eliminated during ontogeny suggests that suppressor mechanisms might be responsible for normal self tolerance.

Monoclonal antibodies reveal in blocking reactions the antigenicity and antibody response of poliovirus.

Neutralizing monoclonal antibodies against infectious poliovirus particles, type 1, strain Mahoney, were produced in hybridomas obtained from primed Balb/c mouse lymphocytes fused with the P3/NS-1/1-Ag4-1 mouse myeloma cell line (NS-1). From eleven neutralizing monoclonal antibodies, belonging to four groups differing in their reaction patterns, we chose for our experiments mNt-Abs reacting equally well with infectious poliovirus particles and naturally occurring empty capsids (NEC). We characterized the epitopes of NEC reacting with mNt-Abs by a quantitative absorption (blocking) ELISA with the help of monospecific polyvalent rabbit antisera directed against individual structural poliovirus polypeptides, obtained by two different separation procedures. We ascribe the "neutralizing antigenicity" of NEC to reside in or near polypeptides VP1 and VPO. Furthermore, we were able to demonstrate for the first time for poliovirus, that isolated polypeptides induced blocking antibodies, which were able to prevent binding of neutralizing antibodies to poliovirus NEC.

Evidence for several unrelated neutralization epitopes of poliovirus, type 1, strain Mahoney, provided by neutralization tests and quantitative enzyme-linked immunosorbent assay (ELISA).

With the aid of 11 neutralizing monoclonal antibodies which were investigated in four different neutralization tests, evidence was provided that several unrelated epitopes for neutralizing antibodies exist on the surface of poliovirus type 1. All monoclonal antibodies were able to neutralize poliovirus infectivity prior to and after virus adsorption to host cells. The quantitative enzyme-linked immunosorbent assay is introduced as a second independent system for the determination of antibody specificity. Antibodies could be divided into four groups according to their reaction patterns.

[Composition of the microorganism population in an unprotected fermentation process].

An accompanying flora of the production strains always develops in each unprotected continuous fermentation process. In the process of production of fodder biomass from gas oil the high specific growth rate provides sufficient selection advantages to the production strain Lodderomyces elongisporus "D" EH 15 against substrate competitors. The accompanying flora uses products of metabolism and lysis as carbon and energy sources. The concentration of these compounds in the medium and consequently the development of the accompanying flora depends on the process conditions. During a period of more than ten years the taxonomic-diagnostic and technological properties of the production strain Lodderomyces elongisporus "D" EH 15 did not change, caused by the permanent selection stress in the continuous fermentation process.

[Requirements and time factors in dental activities].

The exact determination of the demands on, and the allowed times for, the stomatological activity is an essential prerequisite of fixing measures for the increase in quality and efficiency of the stomatological care and for the control of their success. The results of a discriminating work analysis on the basis of an independent study on a representative sample of dentist-nurse collectives are compared with the findings of other authors. After resolution of the stomatological basic activities into partial activities and after calculations of the arithmetic means of the respective allowed times, the authors establish synthetic allowed times for the basic activities by summing up the adjustable, necessary and defined partial activities.

[Analysis of loss of working hours in a dental department].

The economical use of the societal working capacity, which is substantially influenced by the loss of working hours, requires a time study. The loss of working hours was analysed for three professional groups in a stomatological service in 1973.
