Histopathology of cutaneous changes in non-drug-induced coma.

To compare the histological features of non-drug-induced and drug-induced coma blister, we performed histopathological and immunopathological studies of four biopsy specimens from three patients with non-drug-induced coma. These results were compared with the previously well-documented histology of drug-induced coma. The findings of the present study of non-drug-induced coma included (a) a variable degree of epidermal cell degeneration, including vacuolation of basal cells, intraepidermal blister formation with pale cytoplasm, and extensive coagulation necrosis with pale nuclei; (b) alteration of the outer root sheath of telogen follicles, ranging from focal necrosis to total coagulation necrosis, and degeneration of sebaceous gland with disappearance of the germinative cell layer; (c) secretory eccrine cells with pyknotic nuclei, vacuolation of the cytoplasm, and intercellular edema, resulting in poorly defined cytoplasm, although the nuclei of the outer basal layer were partially preserved; (d) from slight edema of the vessel wall of the venules to fibrinoid, thrombosis and/or fibrinoid necrotic degeneration of arterioles and venules; and (e) deposits of immunoglobulins or complement as detected by direct immunofluorescent technique in all the three cases. One significant difference between non-drug-induced and drug-induced coma blister was the presence of fibrinoid thrombi in the lumina of non-drug-induced coma blisters. Since one of the three cases of non-drug-induced coma studied in the present report did not show thrombi in the lumina, this feature may not always be available for the differential diagnosis of these two conditions. However, fibrinoid thrombi may be a good marker for the differentiation of these two conditions, when the depth and duration of non-drug-induced coma are severe enough to induce these lesions.

Does coinfection of Bartonella henselae and FIV induce clinical disorders in cats?

It was found that Bartonella henselae (B. henselae) may induce clinical disorders in cats in natural conditions from a comparison of the serological status for B. henselae with the serostatus for feline immunodeficiency virus (FIV) and several clinical characteristics in 170 domestic cats. Seropositivity for B. henselae was not significantly different between FIV antibody-positive and -negative cats (18.4% vs 16.0%). The incidence of clinical characteristics were compared among four cat groups distinguished by the reactivity of sera against B. henselae and FIV. The incidence of lymph node swelling was lower in only FIV antibody-positive cats (3.0%), but higher in B. henselae antibody-positive cats (13.6%) and significantly higher in both B. henselae and FIV antibody-positive cats (42.9%) compared with the incidence of lymph node swelling in cats which were negative for both antibodies (5.5%). The same relation was also observed for the incidence of gingivitis among the 4 cat groups, suggesting that coinfection of B. henselae and FIV may be associated with gingivitis and lymphadenopathy in cats.

Serological survey of spotted fever group rickettsia in wild rats in Thailand in the 1970s.

In Thailand, the first human cases of spotted fever group rickettsiosis were reported in 1994, but no serosurveys on wild rats have yet been conducted. We investigated the seroepidemiology in wild rats collected in the 1970s from two regions in Thailand, and found a 62.2% positive rate of antibodies for spotted fever group rickettsia (SFGR) by the indirect immunofluorescence antibody test. Of the antibody-positive rats, 82.2% had higher titers of antibodies against TT-118 than those against Rickettsia japonica, which suggests that Thailand is infested mainly with the TT-118 strain or its antigenically related organisms. The prevalence of antibodies in Bandicota indica was significantly higher than that in other species, which suggests that B. indica is important as a reservoir of SFGR in Thailand.

The prevalence of Listeria monocytogenes in the environment of dairy farms.

The prevalence of Listeria monocytogenes in the environment of dairy farms was surveyed from December 1993 to June 1994 in one city of Hokkaido. L. monocytogenes was isolated from 3 out of 5 farms investigated. Serovar 4b organism was isolated from the brain stem of a cow from one farm which was clinically diagnosed as having listeriosis. The same serovar of L. monocytogenes was also isolated from the rectal contents of a healthy cow, straw on the floor, straw in the barn, and silage scattered around the silo from the same farm. At another farm, with no reported cases of bovine listeriosis, serovar 1/2 organism was isolated from the same types of samples as the above mentioned farm except from straw on the floor. The difference in the isolation rates of the organism from straw on the floor between the two farms (22%:5/23 vs 0%:0/24) is considered to be caused by the different feeding methods of silage between the two farms.

Seroepidemiological survey of lymphocytic choriomeningitis virus in wild house mice in China with particular reference to their subspecies.

Serum samples from 337 wild house mice (Mus musculus) from 35 sites in China, collected in 1992 and 1993, were examined for antibodies against lymphocytic choriomeningitis virus (LCMV). Ten samples from eight sites were found to contain such antibodies. Six of the eight positive sites were located in the territory of M. m. gansuensis. One of the other two sites was located in the territory of M. m. castaneus in southern China and the other site was in a habitat of M. m. castaneus which had invaded into the western end of the territory of M. m. homourus. It seems likely that LCMV is distributed in the territories of M. m. gansuensis and M. m. castaneus in China. This is the first report of detection of these antibodies in wild house mice in China and specifically in the territories of M. m. gansuensis and M. m. castaneus.

ATP-sensitive K+ channel opener pinacidil augments beta 1-adrenoceptor-induced coronary vasodilation in dogs.

The opening of ATP-sensitive K+ (K+ATP) channels contributes to the mechanism of metabolic coronary vasodilation. The aim of the present study was to determine whether K+ATP channel opener pinacidil augments coronary vasodilation induced by beta-adrenoceptor stimulation. In anesthetized dogs, coronary vasodilation in response to intracoronary infusion of a beta 1-adrenoceptor agonist denopamine, selective beta 2-adrenoceptor stimulation with isoproterenol after bisoprolol or nitroglycerin was studied before and during simultaneous intracoronary infusion of pinacidil at a dose of 1 microgram/min, which had no effect on basal hemodynamics. Pinacidil augmented the denopamine-induced increase in coronary blood flow (CBF) from 38 +/- 9 to 66 +/- 16% (P < 0.05) but did not affect the denopamine-induced by isoproterenol or nitroglycerin. Thus pinacidil selectively augmented beta 1-adrenoceptor-mediated coronary vasodilation. These observations suggest that the K+ATP channel opener pinacidil may increase myocardial perfusion during metabolic stress associated with beta 1-adrenoceptor stimulation.

Adenovirus-mediated gene transfer into injured arteries: quantitative analysis of repeated administration.

We evaluated quantitatively the efficiency and safety of, and influence of either prior arterial injury or immunity against adenovirus on adenovirus-mediated gene transfer into injured arteries of a relatively large animal model, the dog. A replication-defective adenovirus expressing bacterial lacZ was introduced percutaneously into balloon-injured femoral arteries through a double balloon catheter. After a single dose of adenovirus, up to 90% of surface and smooth muscle cells in multiple layers of the media showed transgene expression evaluated by X-Gal histostaining without extralocal expression, as assessed by the polymerase chain reaction. High level expression measured as beta-galactosidase activity peaked at 7 days after transfer and was transient, though retained for a month. Gene transfer performed just after balloon injury induced submaximal gene expression. A second administration of the same adenovirus to the same arterial site enhanced lacZ expression significantly despite the presence of an immune response. Expression of lacZ was also detected in preimmunized dogs, although the expression levels correlated inversely to the titer of neutralizing antibodies in their serum. These data may provide a scientific foundation for the use of adenovirus-mediated arterial gene transfer in future clinical practice.

MR imaging of the lacrimal gland. Age-related and gender-dependent changes in size and structure.

PURPOSE: The subject of this study was to define age-related and gender-dependent changes in MR features of the lacrimal gland. MATERIAL AND METHODS: MR images were retrospectively analyzed in 104 normal subjects aged 2-79 years to measure thickness and area of the lacrimal gland, its MR signal intensity ratio, and SD of the MR signal intensity profile. RESULTS: Thickness and area of the lacrimal gland decreased with age in women (p < 0.001), but not in men. Furthermore, MR signal intensity ratio of the lacrimal gland showed an age-related increase in women (p < 0.001), but not in men. On the other hand, SD of the MR signal intensity increased with age in both women and men (p < 0.001). CONCLUSION: These findings demonstrate that gender has a significant influence on lacrimal gland structure during development and aging.

Primary pericardial mesothelioma demonstrated by magnetic resonance imaging.

A 24-year-old man was hospitalized for an investigation of cardiac enlargement. Serum hyaluronic acid was increased to 238 ng/ml. Echocardiogram showed pericardial effusion with an area of increased density adjacent to the anterolateral wall of the left ventricle. A soft-tissue mass within the pericardial space was enhanced with gadolinium DTPA on magnetic resonance imaging. Open biopsy confirmed the presence of a malignant mesothelioma. Magnetic resonance imaging provided useful information regarding the tumor.

S-(1,2- dicarboxyethyl)glutathione and glutathione in lens and liver of napthalene-treated rabbits.

The determination of S-(1,2-dicarboxyethyl)glutathione and reduced glutathione (GSH) in the rabbit lens and liver was developed using an isotachophoretic analyser. The recovery of S-(1, 2-dicarboxyethyl)GSH from the rabbit liver after ion-exchange treatment was 96.8 +/- 11.3% (n=3). The contents of S-(1,2-dicarboxyethyl)GSH in the rabbit lens and liver were 219.9 +/- 29.1 (n=5) and 44.0 +/- 13.5 (n = 8) nmol/g, respectively. The contents of S-(1, 2-dicarboxyethyl)GSH in the lens and GSH in the lens and liver of naphthalene-treated rabbits was also determined by this method 24 hours after naphthalene administration, at which time the axial opacity "spichen" was observed at the equatorial region of the lens. The content of S-(1,2-dicarboxyethyl)GSH in the lens decreased in proportion to the content of GSH. During the further development of true lens opacity after naphthalene administration, the S-(l, 2-dicarboxyethyl)GSH content further compared with that in the spichen stage, but the S-(1, 2-dicarboxyethyl)GSH content of the lens that did not develop true opacity after naphthalene administration returned to the normal level. The change of S-(1, 2-dicarboxyethyl)GSH content of the lens in the spichen and true opacity stages coincided with that of GSH content. On the other hand, the content of GSH of the liver decreased markedly until 24 hours after naphthalene administration, then returned to normal, irrespective of whether true opacity did or did not subsequently develop.

Effects of the functional elimination of sarcoplasmic reticulum on the manganese-dependent norepinephrine-induced contractions of the guinea pig vas deferens.

We investigated the effects of inhibitors of the sarcoplasmic reticulum (SR) functions on the tonic contractions induced by norepinephrine (NE) in the Ca(2+)-depleted Mn(2+)-loaded vas deferens of the guinea pig in the absence of both Ca2+ and Mn2+ (Mn(2+)-dependent NE-contraction). In control preparations without Ca(2+)-depletion and Mn(2+)-loading, either cyclopiazonic acid (CPA, 10 microM) or ryanodine (RYA, 3 microM) inhibited the initial phasic and tonic components but not the large phasic component of NE-induced contraction in normal medium containing 2.2 mM Ca2+. In contrast, CPA did not affect the Mn(2+)-dependent NE-contractions. The inhibitory effect of RYA slowly developed with each repetition of the Mn(2+)-dependent NE-contraction and the magnitude of the inhibition was slight. A23187 (10 microM) inhibited the NE-induced contractions of the control preparations in the same manner as CPA and RYA. Although A23187 did not induce contractions in the Mn(2+)-loaded preparations, A23187 augmented the Mn(2+)-dependent NE-contractions. The augmented tonic contractions returned to the resting level by washing NE and A23187. The augmentation remained for 3 successive contractions in the absence of A23187. However, the 2nd application of A23187 did not augment the contraction. These results suggest that neither Mn(2+)-release from SR nor Mn(2+)-influx from the extracellular space contributes to the Mn(2+)-dependent NE-contractions. We concluded that NE induces Mn(2+)-dependent contractions by increasing Mn2+ sensitivity of contractile processes but not by increasing intracellular Mn2+ concentration.

Human obese gene: molecular screening in Japanese and Asian Indian NIDDM patients associated with obesity.

The mouse obese (ob) gene has recently been isolated through the positional cloning technique and has been proved to result in the obese and NIDDM phenotype in mice when mutated (Nature 372:425-432, 1994). More recently, it has been demonstrated, by experiments with recombinant ob protein, that ob gene product can cause mice, including ob/ob mice, diet-induced obesity mice, and normal mice, to lower their food intake and body weight (Science 269:540-549, 1995). To investigate the genetic and/or environmental influences underlying the development of NIDDM associated with obesity, we isolated and partially sequenced the human obese (OB) gene. The human OB gene isolated in this study encoded 167 amino acids and its open reading frame was revealed to be divided into two parts with an intermediate intron of approximately 2.4 kb. Using the single-strand conformation polymorphism (SSCP) technique, we screened Japanese and Asian Indian subjects for mutations in the protein coding regions of the OB gene. A total of 75 NIDDM patients with obesity (54 Japanese and 21 Asian Indians), 40 NIDDM patients without obesity (34 Japanese and 6 Asian Indians), and 34 Japanese patients with simple obesity showed no abnormal SSCP patterns in either component of the coding sequences. These results suggested that mutations in the coding regions of the OB gene are not likely to be commonly identifiable and that there would likely be a kind of obesity-associated NIDDM not caused by mutations of the OB gene.

Epidemiological survey of small rodents for spotted fever rickettsial antibody in Hokkaido, Japan.

The prevalence of antibody against spotted fever group rickettsia (SFGR) was investigated with Rickettsia japonica antigen in small rodents captured in an area in Hokkaido, where no human cases of SFGR infection had been reported. Antibody against SFGR was found in Apodemus speciosus and Clethrionomys rufocanus but not in Apodemus argenteus. None of these rodents had antibody against R. typhi. This is the first report of the antibody against SFGR in rodents in Hokkaido.

[Dielectric behavior of rat lens and changes due to cold cataract].

To correlate the formation of cold cataract with changes in the passive electrical properties of the lens, we measured impedances of isolated rat lenses in the frequency range from 100 Hz to 500 MHz. Temperature-dependent, reversible changes were confirmed in the dielectric behavior as well as in the histological characteristics. The impedance of lenses showed two separate peaks (i.e. P1 and P2) when expressed in terms of loss tangent. At a low temperature of 4 degrees C, cytoplasmic "aggregates" characteristic of the cold cataract were formed inside the fiber cells of young rats with concomitant decreases in P2 (the peak value at higher frequencies), whereas no remarkable changes occurred in the control group. These results indicate that the lowering of P2 may reflect functional changes in the lens fiber membrane, an augmented heterogeneity in cytoplasm and/or decreases of protein-bound water. We conclude that the P2 is a useful dielectric index for the assessment of the nuclear opacity. This is the first report on the application of dielectric techniques to lenses with nuclear cataract. We suggest that dielectric spectroscopy may be applicable to a quantitative evaluation of cataract, not only of the cortical type but also of the nuclear type.
