Effect of Nocardia rubra cell wall skeleton on murine interferon production in vitro.

Nocardia rubra cell wall skeleton (N-CWS) induced alpha- plus beta-interferons (IFN-alpha/beta) in resident peritoneal cells and bone marrow cells from normal mice. N-CWS also induced IFN-alpha/beta and IFN-gamma in the cultures of thioglycollate-elicited peritoneal cells. When induced with N-CWS, the mixed cultures of thioglycollate-elicited macrophages and spleen cells produced high titered IFN-gamma. Both macrophages and lymphocytes present in spleen cells (non-T, non-B, asialo-GM1-positive) were essential for the production. Immunofluorescence staining with anti-IFN-gamma antiserum showed the presence of IFN-gamma in lymphocytes but not in macrophages, suggesting that lymphocytes were producer cells and macrophages were accessory cells. Because IFN-gamma is an important lymphokine in immune response, the ability of N-CWS to stimulate IFN-gamma production may account for a significant portion of the antitumor activity of this agent.

[Chronology of recurrent gastric cancer].

The growth rate of 49 cases with recurrent gastric cancer was investigated with two tumor markers (AFP & CEA). The average doubling time of liver metastases (24.7 +/- 11.9 days) in 18 cases was significantly shorter from that of lymph node metastases (41.1 +/- 22.4 days) in 13 cases and of peritonitis carcinomatosa (42.2 +/- 19.6 days) in 18 cases. Latent period of recurrent cancer calculated by these doubling times was ranged from 1.0 to 3.5 years (mean 1.7 years) in liver metastases, from 1.0 to 5.0 years (mean 2.7 years) in lymph nodes metastases and from 1.5 to 6.0 years (mean 2.7 years) in peritonitis carcinomatosa. Only in liver metastases, positive correlation between the doubling time (X) and the duration of survival (Y) was observed by expressing the formula Y = 0.45 X-0.58 (R = 0.661, p less than 0.05). It is noteworthy that there is a significant correlation in spite of large differences in background subjects (systemic condition, size of metastatic lesion, etc.) and the growth rate of tumor is considered to play a very important role for determining the degree of biological malignancy of individual cancer patients in relation to survival.

[Combination of missile-induced hypertensive intra-aortic infusion chemotherapy in a patient with hepatocellular carcinoma].

A case of hepatocellular carcinoma was treated with anti-alpha-fetoprotein (AFP) antibody-adriamycin via the hepatic artery with induced hypertension by angiotensin II. A minor response was achieved (from 13.5 X 9.0 cm to 10.5 X 7.5 cm) and serum AFP decreased from 310,000 ng/ml to 26,000 ng/ml. The patient has been doing well during a follow-up period of one year. This result suggests that targeting chemotherapy used for each drug delivery system would be more effective if combined with two or more similar modalities.

[Clinicopathological study of minute stomach cancer].

During the last 10 years, we have experienced 41 cases of minute cancer, which is equivalent to 13.9% of all resected early stomach cancers. Out of 44 lesions in 41 cases, 17 lesions were diagnosed preoperatively, while 27 were unexpectedly detected by serial sections of the resected stomach. Most lesions were located along the lesser curvature of the body and antrum. There was no nodal involvement or vessel permeation. Histologically differentiated adenocarcinomas were encountered in 86.4%, associated with intensive intestinal mucosa in many cases. We have stressed that dye endoscopy using indigo carmine and methylene blue is very useful for the diagnosis of minute cancer.

Inhibitory effects of the analgesic neuropeptides kyotorphin and neo-kyotorphin on enkephalin-degrading enzymes from monkey brain.

The inhibitory effects of the analgesic neuropeptides kyotorphin (Tyr-Arg) and neo-kyotorphin (Thr-Ser-Lys-Tyr-Arg) on enkephalin-degrading enzymes were studied. The enzyme used were aminopeptidase (AP), dipeptidyl aminopeptidase (DPP), enkephalinase-A (ENK-A), and angiotensin-converting enzyme (ACE), which were prepared from the monkey brain membrane fraction. Kyotorphin inhibited only DPP (IC50, 18 microM), and the mode of inhibition was non-competitive (Ki, 6 microM). Neo-kyotorphin inhibited AP, DPP, and ACE with IC50 values of 131, 306, and 200 microM, respectively, but the inhibition of the enzyme activities were not effective. The selective inhibition by kyotorphin suggested that kyotorphin might protect the released Met-enkephalin against enzymatic degradation by DPP. Thus, kyotorphin may not only induce the release of Met-enkephalin but also stabilize the released neuropeptide.

[RI angiography with 99mTc and 201T1-chloride scintigraphy in determining the effectiveness of intra-arterial infusion in chemotherapy of locally advanced and recurrent breast cancer].

Eleven patients with local advanced and recurrent breast cancer were treated preoperatively with intraarterial infusion of chemotherapeutic agents. After the treatment, nine patients underwent radical mastectomy. RI angiography with 99mTc is a useful procedure for determining the correct position of the tips of an inserted cannula. The results of 201Tl-chloride scintigraphy have been found to be closely consistent with the results of the resected specimen histologically. Therefore, 201Tl-chloride scintigraphy is also a quite useful procedure for determining the effects of intraarterial infusion in cancer chemotherapy.

[Study of CEA doubling time in liver metastasis from colonic cancer and postoperative survival time].

We calculate the CEA doubling time of liver metastasis (13 cases) from colonic cancer and studied the factors influencing the growth rate. The doubling time of liver metastasis from colonic cancer ranged from 10 to 112 days (mean 57.8 +/- 35.4 days) was found significantly different among differentiations in histological type. Namely, the doubling time of poorly differentiated adenocarcinoma was shorter than that of well differentiated adenocarcinoma. Moreover the postoperative survival time (Y: months) was found to be significantly correlated with doubling time (X: days); Y = 0.155X + 4.452, r = 0.718, p less than 0.05. These results suggest that the growth rate calculated as CEA doubling time of cancer plays an important role in determining the degree of biological behavior in colonic cancer patients.

[A chronological study on gastric cancer originating from the fundus gland mucosa].

A chronological analysis of gastric cancer originating from fundic gland mucosa was made by measuring the tumor size with reference to the serosal invasion and peritoneal dissemination. As for serosal invasion by cancer cells there were significant differences in the group with tumors exceeding 4 cm in diameter, and peritoneal dissemination was liable to be seen in the group with those exceeding 5 cm. This suggests that gastric cancer originating from fundic gastric mucosa must be detected before it reaches 4 cm in diameter and that the intraperitoneal administration chemotherapeutic agents is recommendable, considering residual microcancer dissemination.

[Operated case of cerebral venous angioma with early venous filling on cerebral angiography--case report].

A 34-year-old woman had a generalized convulsion of sudden onset on April 30, 1985. The next day, she suffered a reattack of generalized convulsion. She was admitted to Kokura Memorial Hospital on May 13, 1985, and was found to have intracerebral hemorrhage on CT-scan then, underwent surgical procedure. Based on cerebral angiographic findings and histological examination, the diagnosis of cerebral venous angioma was done. Cerebral venous angioma is a relatively rare vascular malformation. Its characteristic angiographic findings are a normal arterial phase and a local network of small vein that converged centrally into a single large vein. However, this present case of venous angioma in the left frontal lobe with hematoma showed not only characteristically enlarged medurally vein, but also early venous filling. The literature of operated venous angioma with early venous filling of the brain are reviewed and discussed.

Effect of captopril on heavy proteinuria in azotemic diabetics.

We investigated whether captopril, an angiotensin-converting-enzyme inhibitor, would reduce proteinuria in patients with advanced diabetic nephropathy. Captopril (37.5 mg given in divided doses three times daily) was administered to 10 azotemic diabetics with heavy proteinuria. Urinary protein decreased promptly within two weeks (from 10.6 +/- 2.2 to 6.1 +/- 1.4 g per day [mean +/- S.E.M.]; P less than 0.01). The decrease in proteinuria did not coincide with a fall in systemic blood pressure or in the blood glucose concentration. Serum creatinine and potassium values did not change in any of the patients except one. We suggest that captopril caused a decrease in intrarenal hypertension, which contributed to the reduction of urinary protein excretion. The therapeutic value of this intervention remains to be established.

Immunocytochemical localization of factor VIII-related antigen in the human umbilical vein.

The immunocytochemical localization of factor VIII-related antigen in the human umbilical vein was investigated with the light microscopic immunoperoxidase and immunoelectron microscopical protein A-gold techniques. The light microscopic observation showed that peroxidase reactions were found exclusively in the endothelia of the vessel. By the protein A-gold method, immunoreactive gold particles were located in endothelial specific granules (Weibel-Palade bodies). These data suggest that endothelial specific granules are storage sites of factor VIII-related antigen.

Molecular cloning of the fibroin light chain complementary DNA and its use in the study of the expression of the light chain gene in the posterior silk gland of Bombyx mori.

Fibroin light chain (L-chain) mRNA (mol. wt 4.0 X 10(5) daltons) was purified from the posterior silk gland of the silkworm, Bombyx mori (J-131 strain). Double-stranded complementary DNA was synthesized and inserted into the PstI site of pBR322 employing the oligo(dC)-oligo(dG) tailing method. Several recombinant plasmids containing the inserts of about 800 base pairs were isolated. Hybridization-translation assay demonstrated that these clones hybridized specifically with the fibroin L-chain mRNA. One of these clones (pLA23) was used as a probe to investigate relative concentrations of the fibroin L-chain gene and mRNA in the posterior silk glands at different stages of late larval development.

Cellular vitamin A-binding proteins in liver tumors and adjacent tissues.

The levels of cellular retinol-binding protein (CRBP) and retinoic acid-binding protein (CRABP) were assessed in surgically resected ten human liver tumors and their adjacent tissues by sucrose density gradient centrifugation and Scatchard analysis. The tissues adjacent to hepatocellular carcinomas usually showed portal cirrhosis or hepatitis. There was no significant difference in the dissociation constant (Kd) values of binding proteins of CRBP or CRABP between tumors and adjacent tissues. Five of the ten liver tumors showed similar levels of CRBP as in their adjacent tissues whereas three of the ten liver tumors showed about 40% or less contents of CRBP in comparison with their adjacent tissues and two others had no CRBP activity. By contrast, CRABP activities were found in five liver tumors and in adjacent tissues of one case, whereas there were no detectable CRABP activities in the adjacent tissues except in one case.

Degradation of kyotorphin by a purified membrane-bound-aminopeptidase from monkey brain: potentiation of kyotorphin-induced analgesia by a highly effective inhibitor, bestatin.

Kyotorphin (Tyr-Arg) was rapidly degraded in rat brain homogenates and the Vmax and Km were 29.4 nmol/mg protein/min and 16.6 microM, respectively. This degradation was effectively inhibited by bestatin (IC 50; 0.08 microM) and p-chloromercuribenzoate (IC 50; 0.70 microM). Kyotorphin was also degraded by a membrane-bound aminopeptidase from monkey brains. The Vmax and Km of kyotorphin degradation by the aminopeptidase were 20.0 nmol/mg protein/min and 29.2 microM, respectively. The degradation of kyotorphin was also inhibited effectively by bestatin (KI; 0.4 microM). Co-administration with bestatin 50 micrograms (i.cist.) potentiated the analgesic effects of kyotorphin (i.cist.) by 4.8 times, and these effects were abolished by pretreatment with naloxone 0.5 mg/kg s.c. These results suggest that potentiation of analgesia by bestatin may be due to the protection against the degradation of kyotorphin and released enkephalin by a membrane-bound aminopeptidase.
