Strongyloides venezuelensis infections in mice.

The course and intensity of Strongyloides venezuelensis infection as compared with S. ratti infection were investigated in BALB/c mice. The mice were found to be much more susceptible to infection with S. venezuelensis than S. ratti. The majority of worms inoculated were recovered from the lungs and subsequently from the small intestines, suggesting that their migratory route via the lungs to the small intestine was comparable to that of S. stercoralis in humans. Spontaneous expulsion of worms occurred by about 10 days after infection, which was the same as that of S. ratti. Different infectivities, as assessed by faecal egg excretion, age, sex and strain of mouse were observed in mice infected with S. venezuelensis, as well as in those infected with S. ratti. A striking immunity was acquired following a primary exposure to S. venezuelensis. Mice infected with S. venezuelensis are considered to provide as useful a model as those infected with S. ratti for the study of human strongyloidiasis.

Effects of spleen cells and serum on transfer of immunity to Strongyloides venezuelensis infection in hypothymic (nude) mice.

The course of Strongyloides venezuelensis infection in congenitally hypothymic (nu/nu) mice and their heterozygous thymus-bearing littermates (nu/+) was followed. Unlike the infected nu/+ mice, the nu/nu mice were unable to expel the worms until the end of the observation period (98 days post-infection). In addition, about three times as many eggs were counted at the peak level of infection in faeces of the infected nu/nu mice in comparison with the nu/+ mice. No acquired resistance to rechallenge was observed among the nu/nu mice. Auto-reinfection within the infected nu/nu mice could not be supposed in the present study. The worm expulsion mechanism was generated by nu/nu mice which had been given syngeneic spleen cells from intact +/+ mice. The expulsion of adult worms, as well as the protection against migrating larvae, occurred anamnestically when spleen cells from immune +/+ mice were transferred. The serum transfer, however, only caused a retardation of larval migration. The results support the hypothesis that direct worm immunity and worm expulsion are a T cell-dependent phenomenon.

[5-Fluorouracil (5-FU) concentration in prostatic tissue of rats and in prostatic cancer patients after oral administration of 5-FU].

5-FU has been widely used in the treatment of urogenital cancers. The concentration of orally administered 5-FU was investigated in both the serum and prostate of rats and prostatic cancer patients. In the experimental studies rats were divided into two groups. In rats in group 1 (single administration group), after oral administration of 20 mg/kg/day 5-FU, the 5-FU concentration in serum was 0 at 0 min., 0.916 +/- 0.694 at 10 min., 2.000 +/- 1.159 at 20 min., 1.029 +/- 0.570 at 30 min., 0.119 +/- 0.033 at 60 min., 0.020 +/- 0.020 at 2 hr., 0 at 4 hr. and 0 micrograms/ml at 24 hr.; the 5-FU concentration in prostatic tissue as 0 at 0 min., 0.324 +/- 0.190 at 10 min., 0.843 +/- 0.544 at 20 min., 0.469 +/- 0.252 at 30 min., 0.132 +/- 0.027 at 60 min., 0.094 +/- 0.024 at 2 hr., 0.057 +/- 0.020 at 4 hr. and 0 micrograms/g at 24 hr. In rats in group 2 (daily administration group), 20 mg/kg/day 5-FU was orally administered for seven days. On the seventh day, the 5-FU concentration in serum was 0 at 0 min., 1.877 +/- 0.957 at 10 min., 4.091 +/- 2.184 at 20 min., 1.692 +/- 1.033 at 30 min., 0.345 +/- 0.084 at 60 min., 0.036 +/- 0.019 at 2 hr., 0.005 +/- 0.011 at 4 hr. and 0 micrograms/ml at 24 hr.(ABSTRACT TRUNCATED AT 250 WORDS)

[Postoperative pyuria after TUR-P: the study of postoperative pyuria by using NFLX].

The postoperative duration of pyuria was studied in 35 patients who underwent transurethral resection of the prostate (TUR-P). The average postoperative duration of pyuria was 58.0 +/- 23.6 days. The age over 70 years, preoperative indwelling of urethral catheter and the preoperative urinary tract infection did not make the duration of pyuria longer. The volume of resected prostatic tissue over 20 g and the existence of diabetes mellitus make it significantly longer. It is effective and safe to use a low-dose antibacterial agent such as NFLX which has a broad spectrum and hardly develops bacterial resistance after TUR-P. It is suggested unnecessary to change the anti-bacterial agent even when pyuria continues.

Effect of endocrine treatment on prostatic blood flow in patients with prostatic adenocarcinoma.

Prostatic blood flow in 19 patients with prostatic adenocarcinoma was measured by the hydrogen gas clearance method before and during endocrine treatment. Prostatic volume was reduced to 70 per cent of the pre-treatment volume by 3 months after the beginning of treatment. Prostatic blood flow was remarkably depressed in patients who had never had any treatment of the prostatic carcinoma (22.2 +/- 8.3 ml. per minute per 100 gm.), while prostatic blood flow increased significantly after endocrine treatment (56.3 +/- 21.8 ml. per minute per 100 gm.). It was likely that prostatic blood flow increased as the prostate volume decreased. Final histology of serial prostatic biopsy specimens after endocrine treatment, revealed distinct deterioration of tumor cells and slight stromal hyperplasia compared to the initial pre-treatment biopsy. The stromal-epithelial ratio, which was calculated by computer-assisted image analysis, was markedly increased after endocrine treatment. Our study indicated that endocrine treatment caused a growth-inhibitory effect that was accompanied by increased blood flow in the prostatic carcinoma.

Cyclosporine-treated kidney transplants from living-related donors--a single center trial.

Analysis of 369 CsA-treated recipients transplanted with living-related donor grafts was presented along with the results of a control group of 160 recipients treated with AZ. 1. The 5-year graft survival was 78.2% in the CsA group as opposed to 58.5% in the control group. However, serum creatinine levels in the CsA group were higher than those in the CT group. 2. DST was carried out in recipients showing response to MLR. The fact that no significant difference was found between the DST and non-DST groups despite the poor tissue matching, may favor the effect of DST in CsA-treated recipients. 3. The graft survival rate in patients with a transient positive crossmatch was lower than that obtained in patients with no history of a positive crossmatch. 4. Incidence of acute rejection was highest in T-cell antibody positive cases, while the incidence of chronic rejection was highest in cold B-cell positive cases. 5. The combined use of AZ and local graft irradiation successfully suppressed the formation of antibodies to blood group antigens. 6. Detailed analysis and follow-up studies will be necessary to establish the optimum regimen of CsA administration for the long-term maintenance of renal function in CsA-treated recipients.
