[Transplantable sebaceous adenoma of the mouse with virus particles studied by electron microscopy (author's transl)].

Histology and ultrastructure of a sebaceous adenoma of the mouse -- derived from i.p.-injection of 0.1 ml DNA, isolated from cells of a transplantable mouse-leukemia -- were studied. The original tumour and all transplanted tumours display the same tissular differentiation. In all tumours virus particles were found. Structure and behaviour of the intracytoplasmic A-particles resemble those of the mouse mammary tumour virus while the C-particles found extracellular were identical with the leukemia viruses of the mouse. It might be suspected that mammary tumour virus and leukemia viruses are either non specific or that a new virus is present in the sebaceous adenoma.

Development of tumors in the glandular stomach of rats after oral administration of carcinogens. I. Histological findings.

Experimental carcinomas in the glandular stomach of rats were induced by oral administration of MNNG (M-methyl-N'-nitro-N-nitrosoguanidin) for 35 weeks or ENNG (N-ethyl-N'-nitro-N-nitrosoguanidin) for 20 weeks. Rats were killed at different times after beginning of carcinogen treatment and tissue specimens were prepared for histologic investigation. Particular interest was placed on the development of tumors and on pathological findings possibly contributing to early diagnosis of stomach cancer. During the development of tumors, several dysplastic reactions were observed in the antral mucosa. They could be classified into 4 groups: One was regenerative hyperplasia (1) that meant irregular glandular proliferations without cell atypism at the margin of erosions and ulcers. This lesion was mainly found 1-9 weeks after administration of MNNG. In glandular hyperplasia (2) either crypts or glands were extended and mucosal layers were thickened. No signs of cell atypism were observed. This lesion was mainly found 12-17 weeks after administration of MNNG. Dysplasia (3) was combined with considerable structural modifications and cellular atypism. However, this lesion was limited to the mucosal layer. Neoplastic changes (4) were characterized by marked cellular atypism and extension to tunica submucosa and tunica serosa. Some tumors showed the histological patterns of benign tumors, but most of them were adenocarcinomas. In some cases metastases into pancreas, liver and lymph nodes and in one case into the 12th rib were observed. No particular enzyme patterns were found by histochemistry.

Development of tumors in the glandular stomach of rats after oral administration of carcinogens. II. Different cell types in antral carcinoma as revealed by electron microscopy.

In the present study electron microscopic investigations of tumors of the glandular stomach of rats induced by oral administration of MNNG or ENNG are dealt with. The aim of the study was to describe the different cell types found in the carcinoma and to elucidate the possible relationship between the undifferentiated carcinoma cells and the more differentiated cell types found in the tumor. The results show that besides undifferentiaded carcinoma cells, several differentiated cell types such as goblet cells, endocrine cells, cells with lamellated inclusions in their cytoplasm, and squamous carcinoma cells can be observed. The most conspicuous findings were carcinomatous gland cells with well differentiated microvilli on their luminal surface and typical tonofilamentous structures in their cytoplasm. These cells exhibited signs of the squamous epithelium and of gland cells. Therefore they may be considered as intermediate variants between adenocarcinoma cells and squamous carcinoma cells. The occurrence of such intermediate steps points to the possibility of differentiation by cell division of adenocarcinoma cells into several metaplastic cell types in the experimental stomach carcinoma induced by MNNG or ENNG.

[Ultrastructure of an atrial myxoma (author's transl)].

Electron microscopic examination of a surgically removed atrial myxoma showed, besides fibrocytes and fibroblasts, mainly isolated smooth muscle cells arranged in solid cellstrands. They are responsible for collagen-and elastin-synthesis. They are presumed to arise from subendocardial smooth muscle cells and considered to be neoplastic.

[Ultrastructural aspects in the pathogenesis of desoxycholat--pancreatitis (author's transl)].

After intraperitoneal application of desoxycholate in cells of exocrine pancreas, ultrastructural alterations are observed. Initial changes are seen in mitochondria and focal parts of the endoplasmic reticulum. In later experimental stages there is sizable increase in the extent of the initially focal degradations, in such a way that great areas of the cytoplasm are involved. It is remarkable to note, that there are no indications about any disturbance in the synthesis and excretion of zymogen granules. The described morphological alterations will be discussed as conceivably being caused primarily by an irritation in the lipoid metabolism of the membrane.

[Colonization of the mouse liver by transplanted virogenic leukemia cells. Electron microscopic investigations (author's transl)].

Cells of a virogenic, immature myelogenic leukemia were injected in i.p. in one week old mice, strain NMRI, each animal receiving 10(5), 10(6) or 13(7) cells respectively in 0.2 ml Hanks' BSS. Mice were sacrificed at different times between 10 hrs and 21 days p.i. and liver specimens were prepared for electronmicroscopic studies. 10 hrs p.i. the leukemic cells are found in the sinusoids of the liver and after 30 hrs in the periportal fields. The leukemia cells migrate into the Disse spaces from the sinusoids. The leukemic cells penetrate with cytoplasmic digitations between connected endothelial cells. There is no lytic disintegration of the endothelial cells. The leukemic cells multiply by mitotic division of the Disse spaces, thereby compressing the liver cells. The cell membranes of both cell-types remain intact. Electronmicroscopically no evidence is found of a secretion of enzymes from the leukemia cells that destroy liver cells. Destruction of the liver cells is the consequence of the growth-pressure exerted by the dividing leukemic cells. Within the liver of the recipients the leukemic cells produce RNA-viruses. In the cytoplasm - frequently in the area of the Golgi-field - groups of immature A-particles are formed. In the virus fields the ribosomes disappear. The immature A-particles consist of two concentric electron dense shells. On the surface of the liver cells budding particles and immature C-particles develop. Extracellular mature C-particles with a homogeneous nucleoid and an irregular outer shell may be seen. The formation of viruses is found to be independent of the stage of the leukemia.

[Electron microscopic investigations in the origin of 'ghost bodies' in the aorta].

Electron microscopic studies on aortas of normal mongrel dogs revealed a special form of degeneration of vascular smooth muscle cells and the mode of origin of 'ghost bodies'. These are formed by budding off portions of cytoplasm of smooth muscle cells probably as a result of worsening of the metabolic situation of these cells with increasing age.

[Ultrastructural-morphometric analysis of liver biopsies in patients with intrahepatic cholestasis. I. Correlations between morphometry of bile canaliculi and so-called "marker enzymes of cholestasis" (author's transl)].

Reports on correlations between the activity of so-called "marker enzymes of cholestasis" in serum and the ultrastructural changes of the liver are rare. Therefore studies of ultrastructural changes were carried out in 40 patients with intrahepatic cholestasis. In the patients' serum activity of alkaline phosphatase, bile duct alkaline phosphatase, leucine-aminopeptidase (LAP), and 5'-nucleotidase (5'-Nu) as well as the concentration of bilirubin were determined. The results showed a significant correlation between the morphometry of the bile canaliculi and the serum activity of LAP and 5'-Nu. In patients with elevated LAP, an enlargement of the bile canaliculi could be proved. An increased serum activity of 5'-Nu correlated with a higher incidence of bile canaliculi in the ultrastructural picture. The results suggest an investigation of the ultrastructure of bile canaliculi and the determination of marker enzymes of cholestasis in the serum may both contribute to the assessment of cholestatic liver disease.

[Fine structure of megakaryocytes in aleukemic megakaryocytic meyelosis].

Megakaryocytes from a case of aleukemic megakaryocytic myelosis were examined by electron microscope. They were considerably increased in number and their ultrastructure was different from normal megakaryocytes in certain aspects. Myelinfigures became demonstrable in the cytoplasm in increased number. An effective development of the demarcation membran system could not be observed.

Electron microscopic study on permeability of coronary artery wall of normotensive rabbits using horseradish peroxidase as a tracer.

In order to study the pathways in the coronary artery vessel wall horseradish peroxidase (HRP) was used as a tracer. HRP was injected directly in the left ventricle of rabbit hearts. 30 seconds after intracardial injection of HRP in normotensive rabbits, peroxidase reaction products could be located in random areas of the coronary arterial system and could then be observed in all three layers of the coronary walls at these locations. HRP reaches the subendothelial space of the coronary arteries along two pathways: through intercellular junctions and by plasmalemmal vesicles. Through the fenestations of the internal elastic lamina HRP reaches the media and can be detected between the media smooth muscle cells. The influence of the methodology on the results is discussed.
