Evaluation of cerebral collateral circulation by technetium-99m HM-PAO brain SPECT during Matas test: report of three cases.

Three cases with cerebral ischemic symptoms and an intracranial aneurysm are presented. Using [99mTc]hexamethyl-propyleneamine oxime (HM-PAO) single photon emission computed tomography imaging before and during the Matas test, a quantitative measurement method was developed for evaluating brain collateral circulation. The evaluation correlated well with findings of contrast carotid angiography. This noninvasive method seems to be useful for selection of patients for appropriate surgical treatment.

The hydrophobic cationic cyanine dye inhibits oxidative phosphorylation by inhibiting ADP transport, not by electrophoretic transfer, into mitochondria.

The effect of the divalent cationic cyanine dye tri-S-C4(5) on oxidative phosphorylation in rat liver mitochondria was examined. The dye at about 100 n mols per mg mitochondrial protein inhibited state 3 respiration and ATP synthesis almost completely. However, it had no effect on submitochondrial particles, like other hydrophobic cations. The dye inhibited the transport of ADP into mitochondria mediated by the adenine nucleotide translocator. Thus, the inhibition of oxidative phosphorylation by the cationic dye was concluded to be due to its action on the adenine nucleotide translocator, not to its electrophoretic transfer into the inner space of mitochondria according to the inside-negative electrochemical potential.

Stable phospholipid membrane supported on porous filter paper.

A novel method to construct a stable and uniform phospholipid membrane of large area and good manipulability is reported. Using the Langmuir-Blodgett (LB) technique, a monolayer of phospholipid can be transferred to filter paper. The electrical conductance across the pores of the lipid membrane is about 1.8 X 10(-9) S/cm2, corresponding to the conductance of 10(-7)-10(-10) S/cm2 reported for bilayer lipid membranes (BLM) of phospholipids. A scanning electron micrograph demonstrated that the phospholipid membrane on the filter paper was uniform.

Possible involvement of the 29 kDa protein in H+-ATPase in the action of cationic uncoupler of oxidative phosphorylation. Effect of the (o-phenanthroline)2-Cu2+ complex as a cationic uncoupler.

The divalent cation (o-phenanthroline)2-Cu2+ complex was found to uncouple oxidative phosphorylation in mitochondria. Its uncoupling activity depended on inorganic phosphate (Pi) in the incubation medium, and was inhibited by the SH-reagent N-ethylmaleimide, and retarded by ATP. The uncoupling by the (o-phenanthroline)2-Cu2+ complex was suggested to be due to its modification of sulfhydryl groups in the 29 kDa protein in H+-ATPase.

Why is Mg2+ necessary for specific cleavage of the terminal phosphoryl group of ATP?

The mechanism of specific cleavage of the terminal phosphoryl group in hydrolysis of ATP, and the role of Mg2+ in the hydrolysis were studied by ab initio molecular orbital calculations. The tetravalent anion of methyl triphosphate was used as a model of the ATP anion, and its electronic structures were determined as a function of the distance between Mg2+ and its beta-phosphoryl group. We found that the closer location of Mg2+ to the beta-phosphoryl group than to the alpha- or gamma-phosphoryl group was effective in weakening the P-O bond at which the cleavage of ATP catalyzed by most enzymes takes place. Moreover, the orbital coefficient of the frontier electron of P gamma, which is related to the nucleophilic reaction, was shown to increase greatly with increasing interaction between Mg2+ and the beta-phosphoryl group.

[Scintigraphy with 111In-labeled leukocytes: simplified procedure for labeling].

To utilize 111In leukocytes in a routine work, simplified procedure for sterile leukocytes preparation and labeling with water soluble oxine sulfate was performed. Viability and chemotaxis of leukocytes were maintained during separation and labeling. Chelated rate of 111In with oxine sulfate was 93.5%. Labeling efficiency of 111In leukocytes was 93.8%. Obvious blood pool images due to remained erythrocytes were not observed. 111In labeled leukocytes showed good migration into inflammatory focci.

Clinical and roentgenological studies on malalignment disorders of the patello-femoral joint. Part II: Relationship between predisposing factors and malalignment of the patello-femoral joint.

To clarify the relationship between the predisposing factors and malalignment of the patello-femoral joint, a study was conducted on the basis of roentgenograms of 621 cases in which a disorder of the dérangement was suspected in at least one knee. The degree of alignment was classified into four grade groups according to the dynamic skyline view arthrograms. Each predisposing factor was expressed by patellar tendon-patella ratio, patellar facet ratio, depth index, and femoro-tibial axis angle. As to each of the parameters each joint was categorized into three classes, on the basis of its mean and standard deviation. The frequency of occurrence of each of the three classes in each of the grade groups was then examined. For the patellar shape, the joints were categorized according to Baumgartl's taxonomy. The study indicates that there exists a causal relationship between the predisposing factors such as patella alta, narrow medial facet, shallow patellar groove of femur and unstable patellar shape, and the patello-femoral malalignment.

A new noninvasive quantitative assessment of cerebral blood flow using N-isopropyl-(iodine 123)p-iodoamphetamine.

The use of a nomogram is described as a new noninvasive quantitative assessment of cerebral blood flow using N-isopropyl-(123I)p-iodoamphetamine (IMP) and single-photon emission computed tomography. The nomogram was constructed from a computer analysis of the flow value relative to varying washout rate of brain activities between early and late tomographic images, and partition coefficient between brain tissue and blood. The validity of this method was evaluated in normal volunteers and in patients with cerebrovascular disorders by comparing the cortical flow values (Y) estimated from the IMP-nomogram with initial slope indices (X) by the 133Xe inhalation method. A significant correlation (N = 42, r = 0.88, P less than .001) was demonstrated between them, and the regression line was Y = 1.6X-7. A development and confirmation of this method made it possible to measure flow values noninvasively with the use of IMP.

An Escherichia coli mutant exhibiting temperature-sensitive ATP synthesis.

A mutant strain SM434 (ttr-3) of Escherichia coli that exhibits a temperature-sensitive Unc(succinate-nonutilizing) phenotype was characterized. The mutant allele ttr-3 was not linked to the ilvA gene, but was complemented by Fill carrying 81 min-91 min of the E. coli chromosome. The mutant strain SM434 exhibited resistance to N,N'-dicyclohexylcarbodiimide (DCCD) and a temperature-sensitive phenotype at the level of ATP synthesis, compatible with that of cell growth. These findings indicate that the mutant strain SM434 could carry a mutation (ttr-3) in an unknown gene responsible for the energy-transduction system.
