Roentgenologic appearance of a thorotrast-induced small cholangiocarcinoma in a case of thorotrastosis. An autopsy case of massive gastrointestinal bleeding of esophageal varices.

A small cholangiocarcinoma was detected at autopsy in a patient with thorotrastosis who died from the rupture of esophageal varices at the esophagogastric junction. Prior to the advent of recent diagnostic imaging technique, a correct antemortem diagnosis could only be obtained from tumor markers. However, the tendency for the opacity of the liver to decrease slowly with time and develop uneven trabeculation suggests that small tumors may be difficult to detect against such a non-homogeneous background.

Leukocytosis in mice following therapy with a novel antitumor agent, RA-700.

Nine daily intravenous (iv) injections of RA-700 (an antitumor cyclic hexapeptide) at doses of 2 to 6 mg/kg/day caused increases of WBC counts at 4-6 days after treatment in normal C57BL/6 X DBA/2 (BDF1) mice. The percentages of neutrophils and lymphocytes were modified. There was a decrease of colony-forming units in culture (CFUc) in bone marrow to 40% of the control value on day 1 but CFUc rapidly returned to normal values on day 3. The colony-forming units in spleen (CFUs) in bone marrow decreased during treatment. On the other hand, CFUc and CFUs in spleen were increased from the initiation of treatment to the time prior to the increase of WBC count. Spleen weight increased after treatment, and histologically, increases of immature and also mature granulocytes and megakaryocytes were observed. However, RA-700 did not stimulate the progress of hematoprogenitors in vitro. The results indicated that RA-700 stimulates the progress of hematoprogenitors in the spleen, but this effect is probably indirect.

Effects of neonatal ovarian X-irradiation in the Chinese hamster. I. Correlation between the age of irradiation and the fertility span.

The fertility span between the first and the last estrus was compared among female Chinese hamsters exposed to 1 Gy X-rays on neonatal day 0, 2, 4, 6, 8, 10 and 14. The fertility span changed dramatically according to the age at the time of irradiation. The females irradiated on day 4, 6 and 8 showed a definite delay in sexual maturation and a very early cessation of the estrous cycles, while those irradiated on day 10, 2 and 14 (in this order) showed a much longer, nevertheless significantly shortened, fertility span. Reduction in the number of juvenile oocytes by acute killing was responsible for this shortened reproductive life. The present study indicated again that the sensitivity of the oocytes to the lethal effects of X-rays is unquestionably meiotic stage-dependent.

Effects of neonatal ovarian X-irradiation in the Chinese hamster. II. Absence of chromosomal and developmental damages in surviving oocytes irradiated at the pachytene and resting dictyate stages.

Ovaries of Chinese hamsters were irradiated with 1 Gy of X-rays on day 0, 4, 8 and 14 after birth, and the late effects on pachytene and resting dictyate oocytes, which had been proven to be radioresistant and to survive acute oocyte-killing, were studied in the progeny of the irradiated females. Thirty hours after mating, fertilized ova were collected and female pronuclear chromosomes were karyotyped. On day 18.5 of gestation, the number of corpora lutea and implantation sites were counted and the live and dead fetuses were examined phenotypically. There was no increase of chromosome aberrations in female pronuclei of one-cell embryos of the irradiated groups. There was no significant difference in the incidences of pre- and postimplantation deaths and of congenital malformations between the irradiated groups and the non-irradiated controls. These findings indicated clearly that oocytes at the pachytene and resting dictyate stages which had survived the acute oocyte-killing effects of X-rays were also free from late deleterious effects on genetic and developmental abilities. The radiosensitivity of oocytes of the Chinese hamster was compared with that of the mouse with respect to chromosome aberrations, lethal mutations and congenital anomalies, and the difference between both species was discussed.

Repair of salivary fistula after reconstruction of pharyngoesophagus.

We describe three surgical methods used to repair salivary fistulas in different situations that are seen after reconstruction of the pharyngoesophagus in patients with hypopharyngeal cancer. The principle of these methods is the same; a cervical skin flap next to the fistula is used as the internal lining and an anterior chest skin flap is rotated as an external lining to cover it. For the primary closure of heavily irradiated fistulas, this combination may not always be effective, and we emphasize that a combined use of a pedicled pectoralis major muscle flap with these two kinds of pedicled skin flap is extremely reliable.

A chromosomal method to distinguish between X- and Y-bearing spermatozoa of the bull in zona-free hamster ova.

The interspecific in-vitro fertilization system using zona-free hamster ova was adopted to distinguish chromosomally between X- and Y-bearing bull spermatozoa. Frozen semen samples were thawed and washed with modified BWW medium including 0.3% BSA (pH 7.2) to remove cryoprotective medium. Motile spermatozoa were recovered from the semen by the 'swim-up' method. These spermatozoa were treated with ionophore A23187 to facilitate capacitation. Adequate capacitation of spermatozoa was found by their movement patterns and the insemination was performed at the optimum time. The fertilized ova were cultured in Medium TC199 with 10% FBS including podophyllotoxin and vinblastine until they reached first cleavage metaphase. Chromosome slides were prepared by our gradual fixation-air drying method. The success rate of the method was 56% of the number corresponding to the zona-free ova used. Altogether 1116 spermatozoa from 4 different bulls were successfully analysed and the ratio of X- and Y-bearing spermatozoa was 542:574 (P greater than 0.3).

Absence of late effects on survival and developmental abilities of pachytene oocytes X-irradiated during neonatal stages in the Chinese hamster.

In neonatal Chinese hamsters, all oocytes on day 0 after birth are at pachytene and ovarian X-irradiation with 1 Gy causes no acute oocyte killing. The same treatment on day 4, however, kills all diplotene-dictyate oocytes within 48 h, and consequently only a small number of pachytene oocytes survives. We investigated late effects of X-rays on the pachytene oocytes in relation to the reproductive capacity of irradiated females. There was no difference between the 0-day irradiated females and the controls with respect to the age of sexual maturation, fertility span and the rate of age-dependent reduction in the number of oocytes due to follicular atresia. On the other hand, the 4-day irradiated females showed a marked delay in sexual maturation and a very precocious cessation of oestrous cycles. However, the degeneration of the surviving oocytes was not accelerated. Fertility tests on day 18.5 of gestation revealed that there was no difference between both irradiated groups and the controls with respect to the mean number of corpora lutea and to the occurrence of dead and abnormal conceptuses. These results indicate clearly that pachytene oocytes were not affected at all by either acute or late effects of X-rays although the dose was high enough to kill diplotene-dictyate oocytes completely.

[Histologic extension of lobular carcinoma--study of a case of infiltrating lobular carcinoma accompanied by various patterns of lobular proliferation].

Histologic examination using serial sections of a mastectomy specimen from an ILC patient with LCIS and unusual lobular proliferation showed the following: 1) Cancer cells and their infiltration patterns varied and were not typical of ILC, so that this case was categorized as ILC partially possessing some histologic features of IDC. 2) Unusual lesions of proliferation, which were initially regarded as ALH, consisted of both pure ALH and ILC invading the ALH and normal lobules. For the correct diagnosis of ALH in biopsy specimens, therefore, we must bear in mind the (possible) concomitant presence of ILC. 3) The histologic criteria for ALH and ILC must be defined more clearly.
