[Study of the distribution and migration laws of nitroglycerin in energy increasing propellant by size exclusion chromatography].

It is very important to increase the energy of high-nitrogen-content single-base propellant by absorbing nitroglycerin in the case of keeping the advantage of the original propellant. In this paper, the distribution laws and migration laws in the aging of nitroglycerin have been studied by size exclusion chromatography (SEC) after high-nitrogen-content single-base propellant absorbing nitroglycerin. The results show that the distribution of nitroglycerin in high-nitrogen-content sigle-base propellant follows the Gauss distribution. In aging, this distribution changes towards the big variance Gauss distribution. The relationship between migration mass and aging time is y = k1e-k2/t, for external layer k1 = 0.1208 and k2 = 17.229, for inner hole k1 = 0.0348 and k2 = 10.97. The migration velocity constant, K = A0e-E/RT, of nitroglycerin is also given in this article, where A0 = 0.4086 and E0 = 1.6702 kJ/mol for inner hole, A0 = 31.2775 and E0 = 3.5428 kJ/mol for external layer.

[Distinction of yeast flocculent phenotypes and studies of the physiological and biochemical characteristics of yeast flocculation].

More than 400 yeast strains were examined for their flocculation, and five of them displayed strong flocculation. The 5 strains were divided into two groups which are Flo 1 and NewFlo phenotypes on the basis of their response to sugar inhibition. Related physiological and biochemical characteristics of the two phenotypes' strains were studied. The results showed that the flocculation of Flo 1 phenotype was only inhibited by mannose; it was sensitive to high temperature (70 degrees C), pronase E, trypsin, whereas tolerant to pronase K, chymotrypsin, Ca2+, pH; the flocculation of NewFlo was inhibited by many sugars, such as glucose, maltose, sucrose, mannose; it was sensitive to high temperature (70 degrees C), pronase, Ca2+, pH. The Calcium concentration and pH value of the optimum flocculaton of the Flo1 and NewFlo phenotypes strains were respectively 10 mmol/L-1 mol/L; 3.0-4.5.

[Effect of promoter-PTH4 on Streptomyces sporulation].

233 bp of promoter--PTH4 was subcloned into Streptomyces plasmid pIJ4083. pIJ4470 was obtained after recombinant plasmid was introduced into Streptomyces coelicolor J1501. Both Streptomyces coelicolor J1501/pIJ4083 and J1501/pIJ4470 were grown on minimal medium (MM) agar containing mannitol and 10 micrograms/mL thiostrepton for 6 days, and then ultrathin sections of colonies were prepared and stained with silver proteinate to observe glycogen biosynthesis in cell by the electron microscope. Result showed that the glycogen was produced by J1501/pIJ4083, whereas little glycogen was produced by J1501/pIJ4471. The dark color spores were not observed in J1501/pIJ4470 with lead stain in contrast with dark color spores which were produced by J1501/pIJ4083, indicating that promoter PTH4 may play an important role in physiological and morphological differentiation of Streptomyces.

[Study on the inkblot of carbon pen fields by Raman micro-spectroscopy].

The four carbon-pen inkblots in market were studied by Raman micro-spectroscopy. The Raman spectra obtained in the experiments indicated that four brands of carbon-pen have not only similarity but also differentiation. Raman spectra from the same brand but different batch demonstrated difference too. Hence one can use the Raman technique for characterization and identification of inkblots from market.

Seroprevalence of enzootic bovine leukosis in Trakya district (Marmara region) in Turkey.

In this study, 481 cattle belonging to 77 farms from nine localities in Trakya district in the Marmara region of Turkey were blood sampled and serologically tested for enzootic bovine leukosis (EBL). Antibodies to bovine leukosis virus (BLV) were detected in 51 cattle sera (11%) belonging to nine farms in five localities. Cattle tested were mostly female Holstein or Brown Swiss of ages ranging between 18 months and 10 years. Analysis of the relationships between age, breed or sex and seropositivity to EBL in seropositive herds indicated no significant associations (p > 0.05). The relationship between seropositivity and haematological changes was also studied, and seropositive cattle had higher lymphocyte percentage and lower neutrophil percentage than seronegative cattle (p < 0.001).

Automation and integration of multiplexed on-line sample preparation with capillary electrophoresis for high-throughput DNA sequencing.

An integrated and multiplexed on-line instrument starting from DNA templates to their primary sequences has been demonstrated based on multiplexed microfluidics and capillary array electrophoresis. The instrument automatically processes eight templates through reaction, purification, denaturation, preconcentration, injection, separation, and detection in a parallel fashion. A multiplexed freeze/thaw switching principle and a distribution network were utilized to manage flow and sample transportation. Dye-labeled terminator cycle-sequencing reactions are performed in an eight-capillary array in a hot-air thermal cycler. Subsequently, the sequencing ladders are directly loaded into separate size exclusion chromatographic columns operated at approximately 60 degrees C for purification. On-line denaturation and stacking injection for capillary electrophoresis is simultaneously accomplished at a cross assembly set at approximately 70 degrees C. Not only the separation capillary array but also the reaction capillary array and purification columns can be regenerated after every run. The raw data allow base calling up to 460 bp with an accuracy of 98%. The system is scalable to a 96-capillary array and will benefit not only high-speed, high-throughput DNA sequencing but also genetic typing.

Disruption of primary mesenchyme cell patterning by misregulated ectodermal expression of SpMsx in sea urchin embryos.

The patterning of the mesoderm of the sea urchin embryo is a classical paradigm of epithelial mesenchymal interactions in organogenesis, yet little is known of its molecular basis. Here we address the role of the homeobox gene, SpMsx, a member of the highly conserved Msx gene family, in this process. Msx genes have been shown to function in the dorsoventral patterning of the central nervous system in Drosophila and in a variety epithelial-mesenchymal interactions in vertebrates. We showed previously that the SpMsx gene is expressed during embryogenesis in a complex and dynamic pattern consistent with roles in the development of subpopulations of endoderm, mesoderm, and oral ectoderm. To perturb this pattern of expression and thus probe the function of SpMsx, we injected SpMsx mRNA into single-cell zygotes and monitored development morphologically and with a series of territory-specific molecular markers. RT-PCR analysis revealed that injected SpMsx transcripts persisted at least until the gastrula stage in amounts comparable to endogenous levels. Injected embryos exhibited deficiencies in the organization of primary and secondary mesenchyme cells within the blastocoelic cavity, as well as abnormalities in spicule number and shape. Defects in the endoderm were also common, including reduced or absent archenterons. Micromere transplantation experiments revealed that the defects in skeletogenic mesenchyme patterning were non-cell autonomous, consistent with findings that cell-cell interactions between ectoderm and the progenitors of the skeletogenic mesenchyme, the primary mesenchyme cells (PMCs), are important both for PMC guidance and spicule morphogenesis. Our data, taken together with observations in other organisms on the role of Msx genes in embryonic signaling processes, particularly involving the BMP pathway, suggest that SpMsx may be a part of the mechanism by which the ectoderm influences both the arrangement of primary mesenchyme cells within the blastocoel and the shapes of the skeletal rods.

The Streptomyces coelicolor sporulation-specific sigma WhiG form of RNA polymerase transcribes a gene encoding a ProX-like protein that is dispensable for sporulation.

In the non-motile mycelial organism Streptomyces coelicolor A3(2), the sporulation gene whiG encodes a protein that closely resembles RNA polymerase sigma factors such as sigma D of Bacillus subtilis, which mainly control motility and chemotaxis genes. Here, we show that the whiG gene product, purified from an Escherichia coli strain carrying an expression construct, could activate E. coli core RNA polymerase in vitro to transcribe a sigma D-dependent motility-related promoter from B. subtilis. Such RNA polymerase holoenzyme preparations could also transcribe from an S. coelicolor promoter, PTH4, previously shown to require an intact whiG gene for in-vivo transcription. The in-vivo dependence on whiG was therefore shown to be direct. Unusually, the initiation of PTH4 transcription in vitro depended on the provision of appropriate dinucleotides. The whiG-dependent PTH4 transcription unit consisted of a single gene, orfTH4. Sequence comparisons suggested that the gene product was a member of a small group of proteins that include the B. subtilis and E. coli ProX proteins. Though none of these proteins shared more than about 30% of extended primary sequence identity, they had similar size and hydropathy profiles, and could be aligned end to end to reveal a mosaic of similarities. The ProX proteins of B. subtilis and E. coli are implicated in glycine betaine transport in response to hyperosmotic stress. However, disruption of orfTH4 did not cause any obvious phenotypic changes in growth or development on media of varying osmotic strengths.

Atmospheric mercury deposition on Fanjing Mountain Nature Reserve, Guizhou, China.

Fanjing Mountain Nature Reserve (FMNR) is surrounded with several Hg emission sources within distances of 100-200 km. At the two sites studied, Tongren and Danzai, Hg emission and deposition fluxes, Hg concentration in the air, soil and other samples are all several hundred times higher than at other relatively clean areas. Hg accumulation in soil and moss at FMNR varies with the sampling heights. Total Hg deposition to this area has been estimated to be 115 micrograms m-2 y-1 using moss bag technique. Dry deposition was determined to be about 5.2 micrograms m-2 month-1 during March to June, corresponding to more than 50% of the total deposition.

Tuberculosis of the nasopharynx following radiotherapy.

We report the case history of a patient who was treated with radiotherapy for nasopharyngeal carcinoma. During follow-up, she showed signs, symptoms and radiological evidence of disease recurrence and progression. However, repeated biopsies of the posterior nasal space (PNS) failed to demonstrate malignancy. A diagnosis of nasopharyngeal tuberculosis was finally made when tissue from a PNS biopsy stained positive for acid-fast bacillus. The patient responded to antituberculous chemotherapy.

A rapid method for determination of in vitro susceptibility to antibiotics with a bulk acoustic wave bacterial growth biosensor.

A novel bulk acoustic wave (BAW) bacterial growth biosensor was developed to study in vitro susceptibility by continuous monitoring of disturbances of bacterial growth at low antibiotic concentrations, followed by the accurate and rapid estimation of growth kinetic parameters and minimum inhibitory concentrations (MICs). The susceptibilities of bacteria, e.g. Escherichia coli, Staphylococcus aureus, Proteus vulgaris, Pr. morganii and Pr. mirabilis, to various antibiotics, e.g. penicillin, streptomycin, gentamicin and cefotaxime, were investigated, respectively, and the MICs were rapidly determined with a higher reproducibility than the conventional broth micro-dilution technique (BMDT). The effects of cell constant of conductivity electrode, pH and temperature on bacterial growth and biosensor signals were discussed in detail. The proposed method offers an effective alternative to the conventional methods.

Reversed-phase high-performance liquid chromatographic measurement of the P-aminobenzoic acid synthesized by Streptococcus sanguis.

OBJECTIVE: This study investigated the biosynthesis of P-aminobenzoic acid by Streptococcus sanguis and the role of P-aminobenzoic acid in the interaction between Streptococcus sanguis and Streptococcus mutans. METHODS: A reversed-phase high-performance liquid chromatographic technique was used to analyze the P-aminobenzoic acid synthesized by Streptococcus sanguis in an anaerobic atmosphere. RESULTS: The results showed, that (1) the chromatographic method established in this study had good linear relation and the average recovery of P-aminobenzoic acid was 80%, and (2) Streptococcus sanguis did synthesize P-aminobenzoic acid, and the mean concentration of P-aminobenzoic acid was 1.23 micrograms/mL. CONCLUSIONS: The results of this study will help further studies of the factors that may have effects on P-aminobenzoic acid synthesis by Streptococcus sanguis and on the role of P-aminobenzoic acid in the microbial homeostasis of dental plaque.

[The second phase clinical observation of anti-radiation effect by superoxide dismutase].

Multiple center randomized controlled double blind clinical trait was conducted to evaluate the anti-radiotherapy effect by SOD (produced by Hunan Biochemical Work) in 159 patients. Injection of 4000U SOD immediately after receiving radiotherapy significantly reduced the occurrence rate of skin, oral mucosal, pelvic visceral and systematic adverse reaction, only the reduction of leukopenia did not reach the statistical significant level. No adverse effect of SOD injection was observed. The results suggest that SOD is a safe and effective agent to attenuate the radiotherapy reactions.

[A multiple variable analysis about the risk factors of breast cancer in women].

A comprehensive investigation to the risk factors of breast cancer was carried out in 146 pairs of women subjects, with the method of 1:1 matched case-control study, and of logistic regression analysis and principal component analysis. As a result, a regression equation which includes 5 independent variables has been established by logistic regression analysis; Ten principal components were extracted from 22 risk factors of breast cancer through principal component analysis, and the cumulative contribution of variances of the 10 principal components is 64.8%. The result combining the mono-factor analysis and the multiple variables analysis indicated that the principal risk factors of breast cancer are cadre occupation, the age of primiparity and late marriage, more intake of salted meat and sausage, passive smoking, having the history of family tumor, of breast benign tumor, and of the mastadenitis; but the more number of giving births, longer period of time of lactation, more vegetable intake, and taking more vitamin may have preventive effect for breast cancer.

[Spinal segment distribution of neural innervation related to houhai acupoint and compared with zusanli and dazhui acupoints in domestic chicken].

OBJECTIVE: The aim of this study was to demonstrate the spinal segmental innervation of Houhai, Dazhui and Zusanli acupoints in domestic chicken. METHODS: The horseradish peroxidase conjugated cholera toxin B subunit (CB-HRP) was injected into the regions of the three acupoints namely, Houhai Dazhui and Zusanli respectively in separate groups of chicken. The frozen sections of spinal cord and dorsal ganglia were prepare for labelling using TMB as a chromogen. RESULTS: (1) In Houhai group the anterograde transganlionic labelling terminals were densely distributed in the dorsal horn and dorsal commissure nucleus. The moto-neurons in ventral horn labelled retrogradely appeared fine arborization of dendrites. There were labelled cell bodies in the dorsal region of the ventral horn (lamina VII) or dorsal commissure nucleus. (2) In Dazhui group, the bundles of labelled fibers presented from laminea I and II to IX in the caudal cervical enlargements and two upper thoracic segments. The labelled motoneurons were shown in medial and lateral groups. The most numourous of labelled ganglionic cells appeared in this group. (3) The labelling sensory ganglionic neurons and their terminals in spinal cord related Zusanli acupoint were overlaped with those related Houhai acupoint. The largest numbers of the labelled ventral motoneurons occured in the Zusanli group. CONCLUSIONS: The results demontrated the patterns of segmental innervation corresponding respectively to Houhai, Dazhui, and Zusanli acupoints. The anterograde afferent labelling terminals and interrelationship of the dendritic arborization of retrograde labelling motoneurons (somatic or visceral) provided neuroanatomical evidences for the innervation of the acupoints.

[Augmented effect of Composite Radix Salviae Miltiorrhizae Injection on radiosensitivity of leukemia cells].

OBJECTIVE: To explore the effect of Composite Radix Salviae Miltiorrhizae Injection (CSMI) on radiosensitivity of leukemia cells. METHODS: Semisolid agar culture and flow cytometry assay were performed for studying the change in radiosensitivity of HL60 cell line and fresh human leukemia cells after exposing to CSMI. RESULTS: The D0(the inverse of the slope of the survival curve) and SF2(survival fraction at 2 Gy) of HL60 cell line were decreased from 1.53, 0.34 to 0.93, 0.12, respectively and apoptosis rates after radiation were raised significantly by CSMI. Furthermore, the concentration of CSMI and the time of mixed culture with CSMI before irradiation had positive relation to the effects mentioned above. Compared with control group, CSMI could increase the radiation-induced apoptosis of fresh leukemia cells (5.89 +/- 2.91% vs 12.05 +/- 3.06%). CONCLUSION: CSMI could obviously enhance the radiosensitivity of leukemia cells.

[Biological responses of a Streptomyces strain producing-Nikkomycin to space flight].

In order to see biological responses to the production of Nikkomycins in general and Nikkomycin X and Z in particular by space conditions, Streptomyces ansochromogenus, a Nikkomycins-producing strain, was carried onboard a satellite for 15 d in 1996. Several strains were isolated from the treated sample and found that the productivity of Nikkomycins in all was increased by 13-18 percent, and the proportion of Nikkomycin X and Z increased correspondingly. Besides, some biological properties of the isolated strains varied markedly.

[Studies on the functional anatomy of Golgi-phobic dendrites of spinal motoneurons in rats--different dendroarchitectures of the motoneurons to slow muscle and fast one].

OBJECTIVE: The functional anatomy of Golgi-phobic dendrites (GBD) of spinal motoneurons (Mn) in rats was studied from their postnatal development. METHODS: Cholera toxin B subunit conjugated with horseradish peroxidase (CB-HRP) retrograde labelling method was mainly used. RESULTS: In the extensor digitorum longus Mn pool (EDL-Mn) GBD gradually decrease during development and are only a few in adults. The related value of the surface density of the white matter dendrites, most of them are GBD, of the soleus Mn pool (SOL-Mn) is nearly double than that of EDL-Mn. CONCLUSIONS: GBD of motoneurons might be related to their action patterns, especially the tonic action of slow muscle, such as the soleus. The hypotheis of dual circuits in motor control, the paralateral circuit on GBD regulates control of tonic action and the core circuit on Golgi-philic dendrites (GLD) and somata regulates phasic action, such as on EDL-Mn, has been further enhanced.
