[Factors affecting the setting time of zinc oxide-eugenol impression materials--the influence of humidity and temperature to the reaction velocity].

Zinc oxide-eugenol paste are widely used in clinical dentistry, principally for impression of non-undercut edentulous ridges, or bite taking materials. However it has been realized by those who use these paste that different products, direction for mixing, or different conditions for mixing, have different characteristics. An experiment have been made to observe the influence of different conditions for mixing to the setting time. Zinc oxide-eugenol impression pastes were mixed in accordance with the manufacturer's instruction when supplied. The condition for mixing was at temperature of 15 +/- 1, 20 +/- 1, 25 +/- 1, 30 +/- 1 degrees C and at humidity of 40 +/- 5, 70 +/- 5, 90 +/- 5%. The setting time (a initial setting time, a final setting time) was defined in the A.D.A. specification No. 16. The following results were obtained. 1. A higher temperature and humidity shortened the initial setting time of all paste. 2. A higher temperature and humidity shortened slightly the time interval between a initial and a final setting time of all paste. 3. In clinical use, sample C is better than other ones, so that a setting time of the sample is property for impression making.

[Experimental studies on platelet preservation with prostaglandin I2 and its derivative (OP-41483) during cardiopulmonary bypass].

Platelet number and its function, specially of aggregability, the changes of intrinsic vasoactive prostanoids were studied in experimental cardiopulmonary bypass (CPB) with PGI2 and its derivative, OP-41483 (OP). Twenty mongrel dogs were divided into the following 4 groups; control group (noCPB, C), heparin group (heparin 2 mg/Kg, H), PGI2 group (heparin 2 mg/Kg + PGI2(1) ng/Kg/min, PG) OP-41483 group (heparin 2 mg/Kg + OP-41483 1 ng/Kg/min, OP). CPB was carried out for 2 hours by roller pump through a membrane oxygenator. The following results were obtained. 1. The drop of mean arterial pressure during CPB with OP was not shown at this dosage. 2. Platelet counts during CPB were well maintained by both PGI2 and OP infusions. 3. The recovery of platelet aggregability examined by 2 microM ADP and whole blood aggregometer induced by collagen with OP were superior to that with PGI2. 4. In spite of PGI2 or OP infusion, no remarkable improvements of intrinsic PGI2 production and TXA2 inhibition were observed. In conclusion, the PGI2 and OP infusion, especially OP during CPB, could well preserve platelet functions and this experimental study might indicate the possibility of the clinical application of these drugs, for the prevention of postoperative hemorrhage.

Signet cell carcinoma of the stomach in a patient with acquired immunodeficiency syndrome: a case report.

A case of signet cell carcinoma associated with AIDS is presented. A 50-year old Japanese man with hemophilia A was suffering from human immunodeficiency virus (HIV) infection, the result of multiple injections of clotting factor concentrates. A diagnosis of signet cell carcinoma of the stomach was reached upon endoscopic and histological examinations. Opportunistic infections of esophageal candidiasis and candida septicemia occurred. The patient died of repeated gastrointestinal bleeding and cachexia. Although there is a possibility of the patient having a coincidential carcinoma along with AIDS, the HIV infection, perhaps, had a role in causing signet cell cercinoma.

[Effect of adriamycin on cell cycle by bromodeoxyuridine-labeling method].

We studied the effect of adriamycin on DNA synthesizing cells from the view point of cell cycle. DNA synthesizing cells were marked by bromodeoxyuridine, which is a known marker for such cells, before administration of adriamycin. Then, the level of this marker was sequentially measured by flow cytometry. At the low concentration (0.01 micrograms/ml), adriamycin caused delay of the shifting time to the S phase 16 hours after administration. The marked cells were accumulated at the G2M phase at a moderate concentration (0.1 micrograms/ml), and blocked at the S phase at the higher concentration (1.0 micrograms/ml).

Estradiol receptors in the cytosol of peripheral blood mononuclear cells in hepatitis virus carriers.

Sex hormones are known to affect the immune system. Asymptomatic hepatitis B virus (HBV) carriers are thought to be immunologically tolerant to HBV, but in some HBV carriers, the immune response to HBV is induced, especially in females during their sexually-mature periods. This may be caused by the effects of estrogen on the immune system. In this study, we found that the level of estradiol receptors in the cytosol of peripheral blood mononuclear cells was significantly lower in asymptomatic HBV carriers and patients with chronic hepatitis (B type and non A non B type) compared to healthy controls. This suggests that one cause of the deficiency in virus elimination in hepatitis virus carriers may be the unresponsiveness of the immune system to sex hormones, as a result of the low level of cytosol estradiol receptors.

[An immunohistochemical study of endogenous estradiol and testosterone in human colo-rectal cancer tissues].

An immunohistochemical method (peroxidase antiperoxidase method) was carried out to examine the distribution of endogenous estradiol (E2) and testosterone (Te). One hundred and twenty four (48.2%) of 257 colo-rectal cancer cases, and 36.7% of male cases and 58.4% of female cases revealed E2-immunostain. This immunohistochemical reaction for E2 was found more frequently in female patients than in male patients, while Te-immunostain was found in 15 (17.6%) of 85 colo-rectal cancer cases. E2 and Te were demonstrated mainly in the cytoplasms and occasionally in the nuclei of cancer cells. Since E2 or Te-positive cancer cells were mainly distributed in the deeper or invasive portion of tissue sections, it might deserve further investigation to elucidate a certain relationship between cancer growth and endogenous sex-hormones.

[Clinical study on surgical treatment of aortic arch aneurysm using selective cerebral perfusion or hypothermic circulatory arrest].

To improve the surgical results of aneurysms of the transverse aortic arch, it is essential to select the optimal support technique to protect the cerebral ischemia during the aortic arch occlusion. In the four year period between 1983 and 1987, 21 consecutive patients had surgical correction of aneurysms of the transverse aortic arch at our institution. The causes of aneurysms were dissection (type A) in 16 patients and arteriosclerosis in 5 patients. Seven patients had emergency operation for frank or impending rupture. Two method for cerebral protection were employed during the period of arch exclusion. In Group I, 11 patients underwent selective cerebral perfusion both to innominate and left common carotid arteries via one roller pump at a rate of 600 ml/min (25 degrees C). The average cerebral perfusion time was 70.4 +/- 20.5 minutes. In Group II, 10 patients underwent deep hypothermia (15 degrees C to 20 degrees C) and total circulatory arrest to allow repair of the transverse aortic arch. The concomitant AVR was performed in two patients and CABG in one patient. The average cerebral arrest time was 35.2 +/- 3.4 minutes. Two out of 10 patients had additional cerebral perfusion because cerebral ischemic time exceeded over 45 minutes. There were three early deaths (14.3%) in this series. The causes of early death were bleeding in two patients and renal failure in one patient. There were no cerebral complications in both groups. The duration of extracorporeal bypass necessary for cooling and rewarming phase in Group II was longer than that in Group I.(ABSTRACT TRUNCATED AT 250 WORDS)

[Synthesis and evaluation of the adaptive control system for the ventricular assist device by using the circulatory system simulator].

An on-line digital simulator using microcomputer system was developed to mimic the hemodynamic behavior of the human circulatory system under ventricular assist device (VAD) pumping. This simulator could calculate the response to the variation of the cardiac function or the driving mode of VAD in the real-time fashion. This simulator was used as the mock controlled object to evaluate and improve the algorithm of an adaptive controller of the drive unit for VAD. The adaptive one-step ahead controller was introduced as the precompensator for the PI-controller, which decides the outflow volume from VAD in order to follow up the reference flow value by changing the systolic duration. It was confirmed that the proposed adaptive control system improved the response speed of the VAD driving system automatically according to the variation of the controlled object.

[Expanded indication of percutaneous transluminal coronary angioplasty to the infarcted zone--using thallium-201 exercise scintigram after rehabilitation].

In some cases of old myocardial infarction (OMI), left ventricular wall motion was improved after revascularization, though viability of the infarcted myocardium was not detected by left ventriculography (LVG) and exercise thallium-201 myocardial scintigraphy (EX-Tl). So to obtain more correct information of the viability, EX-Tl was performed again after ergometer rehabilitation for an OMI patient whose myocardial viability of the infarcted zone could not detected by LVG and EX-Tl. Incomplete fill in was showed in the EX-Tl after rehabilitation. So percutaneous transluminal coronary angioplasty (PTCA) was performed. Left ventricular wall motion was improved after three months. EX-Tl after rehabilitation is useful to evaluate the viability that could not detected by LVG and EX-Tl.

[An adaptive control system of the ventricular assist device with the functions of measurement and analysis].

A personal computer based digital control system for driving the ventricular assist device (VAD) has been developed. Adaptive control, monitoring and analyzing functions are integrated into a software of the personal computer in this system. These functions can operate in parallel with one another because of the multi-interrupt processing used in the software. The system enables the operator to analyze the hemodynamic behavior of the circulatory system simultaneously with VAD pumping and to make on-line use of the result of the analysis in order to improve the driving condition of VAD.

A role for Ca2+ in mediating hormone-induced biphasic pepsinogen secretion from the chief cell determined by luminescent and fluorescent probes and X-ray microprobe.

In isolated chief cells from the guinea pig, cholecystokinin (10 nM) and a high concentration of ionomycin each caused a biphasic pattern of pepsinogen secretion. The initial fast response to cholecystokinin was not dependent on medium Ca2+ ans was mimicked by low concentration of ionomycin (100 nM). Inositol 1,4,5-trisphosphate caused a similar fast release from permeabilized cells. The slow component of release was dependent on medium Ca2+, however, and was mimicked by the phorbol ester 12-O-tetradecanoylphorbol 13-acetate (TPA) (100 nM) or the diacylglycerol analogue 1-oleoyl-2-acetylglycerol (OAG) (100 microM). Ionomycin (100 nM) and TPA (and/or OAG), when applied together, reproduced the biphasic pattern of pepsinogen secretion, suggesting that the signalling pathways utilized by both types of agonist contribute to the response evoked by cholecystokinin-hormone stimulation. Both fura-2 and aequorin were used to monitor changes of intracellular Ca2+. Three pathways were found to contribute to the Ca2+ transient. A rapid release of Ca2+ from intracellular store(s), a rapid Ca2+ entry from the extracellular space, and a more sustained Ca2+ entry from the extracellular space. Cholecystokinin induced a rapid increase in cytoplasmic Ca2+ ([Ca2+]i) as estimated with fura-2 and aequorin. This rise was reduced but not abolished upon removal of extracellular Ca2+, suggesting that both Ca2+ entry from the extracellular space and Ca2+ mobilization from the intracellular store(s) contribute to the initial, fast component of the Ca2+ transient. A second, more sustained component of the Ca2+ transient induced by cholecystokinin was abolished by lanthanum. TPA and OAG induced a biphasic Ca2+ transient that could be detected only with aequorin. The late, sustained component of this response was again abolished by lanthanum as well as by removal of extracellular Ca2+. It appears that the late component of the Ca2+ transient is dependent on Ca2+ influx from the extracellular space and is too localized to be detected by fura-2. Prestimulation of cells with TPA or OAG prevented the aequorin transient caused by cholecystokinin and vice versa, suggesting that TPA, OAG and cholecystokinin activate the same pathways of Ca2+ entry into the cytosol from the intracellular store(s) or the extracellular space. The stimulation-sensitive Ca2+ pool was examined with electron probe X-ray microanalysis. It appears to be restricted to an area enriched in secretory granules or peripheral endoplasmic reticulum just beneath the apical plasma membrane and in close association with the microtubular-microfilamentous system.(ABSTRACT TRUNCATED AT 400 WORDS)

Initial and sustained calcium mobilizations in the parietal cell during stimulations with gastrin, inositol trisphosphate, phorbol ester and exogenous diacylglycerol.

Electron probe X-ray microanalysis revealed that cytoplasmic Ca2+ concentration increased in the restricted apical cytoplasm during stimulation of isolated guinea pig parietal cells with gastrin. Furthermore, this study, using 45Ca2+, aequorin and fura-2, revealed the mechanism involved in intracellular Ca2+ shifts caused by gastrin and the involvements of inositol 1,4,5-trisphosphate (IP3) and diacylglycerol in producing those shifts. The gastrin-mediated and IP3-sensitive Ca2+ pool was located in the smooth-surfaced membrane-enriched areas and released Ca2+ in the initial phase. Gastrin-mediated Ca2+ mobilization was also evoked by diacylglycerol, comprising an intracellular Ca2+ mobilization followed by a late, sustained and more localised Ca2+ entry from the extracellular space.
