Constitutive expression of glutamic acid decarboxylase (GAD) by striatal cell lines immortalized using the tsA58 allele of the SV40 large T antigen.

Rodent striatal cells were immortalized using the A58 temperature-sensitive allele of the SV40 large T antigen. Seventy-eight clones and 10 mixed cultures were characterized at the nonpermissive and permissive temperatures. Based on morphology and expression of proteins, cells were classified into three primary types, with types b and c expressing some neuronal characteristics. Type a cells have an epithelial-like morphology with coarse cytoplasmic extensions and occasional fine processes. These cells express vimentin, do not grow well under serum-free conditions and, when confluent, form a uniform monolayer. Type b cells have a polygonal shape and usually extend multiple thin processes. These cells possess large nuclei with multiple nucleoli and do not express vimentin. Type c cells have a fibroblast-like appearance, are unipolar or multipolar, and their soma is smaller than that of type b cells. Type c cells do not express vimentin, and when confluent form a uniform monolayer. Some type b and c clones express N-CAM and MAP-2. Several type b and c cell lines were found to consistently express glutamic acid decarboxylase (GAD) immunoreactivity under several tissue culture conditions. Selected cell lines were transplanted into the intact adult rat brain in several locations. Cells survived well for 15 wk and did not form tumors. The proteins expressed in vivo were similar to those expressed in vitro.

Growth properties of neural cell lines immortalized with the tsA58 allele of SV40 large T antigen.

In vitro growth properties of three CNS-derived cell lines were compared under a variety of culture conditions. The M213-20 and J30a cell lines were each derived from embryonic CNS culture with the temperature-sensitive (ts) allele of SV40 large T antigen, tsA58, while the A7 cell line was immortalized using wild-type SV40 large T antigen. Cells immortalized with tsA58 SV40 large T proliferate at the permissive temperature, 33 degrees C, while growth is expected to be suppressed at the nonpermissive temperature, 39.5 degrees C. Both the M213-20 and J30a cell lines were capable of proliferating at 39.5 degrees C continuously for up to 6 mo. All three cell lines showed no appreciable differences in growth rates related to temperature over a 7-day period in either serum-containing or defined serum-free media. The percentage of cells in S-phase of the cell cycle did not decrease or was elevated at 39.5 degrees C for all three cell lines. After 3 wk at 39.5 degrees C, the three cell lines also showed positive immunostaining using two monoclonal antibodies reacting with different epitopes of SV40 large T antigen. Double strand DNA sequence analyses of a 300 base pair (bp) fragment of the large T gene from each cell line, which included the ts locus, revealed mutations in both the J30a and M213-20 cell lines. The J30a cell line ts mutation had reverted to wild type, and two additional loci with bp substitutions with predicted amino acid changes were also found. While the ts mutation of the M213-20 cells was retained, an additional bp substitution with a predicted amino acid change was found. The A7 cell line sequence was identical to the reference wild-type sequence. These findings suggest that (a) nucleic acid sequences in the temperature-sensitive region of the tsA58 allele of SV40 large T are not necessarily stable, and (b) temperature sensitivity of cell lines immortalized with tsA58 is not necessarily retained.

Correlation between clinical features and deletions of the gene for dystrophin in Duchenne muscular dystrophy.

The clinical course and prognosis of Duchenne muscular dystrophy (DMD) was compared in patients with deletions of the gene for dystrophin (cDMD) and those without such deletions. A total of 24 patients was followed for at least 2 yrs. At age 12 the rating of the activities of daily life (ADL) and disease stage were less favorable in those patients with deletions of the gene for cDMD. At age 14, no difference in ADL and disease stage was observed between the two groups. The percent vital capacity was lower in those patients with the cDMD deficit. When the prognosis was evaluated by multivariate analysis of the data obtained at age 12, the percent of patients predicted as dying before the age of 20 was 40% for those without the cDMD deficit but 76% for those who were cDMD defective. None of the cDMD defective patients lived longer than 20 yrs, whereas 5 of 14 patients without the cDND deficit survived longer than 20 yrs. Disorders such as cardiac and respiratory failure were also seen more frequently in the cDND defective patients. These results suggest that patients with Duchenne muscular dystrophy with defective cDMD have more severe disease than those without cDMD deficit.

Quantitative evaluation of pulmonary ventilation dynamics using MR imaging: comparison of smokers and non-smokers.

PURPOSE: This quantitative study was conducted to evaluate lobar ventilation dynamics in non-smokers and smokers using MR imaging. METHODS: Ten non-smoker males and ten smoker males underwent MR imaging in the supine position without breath-holding. The volume rates and fluctuation rates were calculated for each lobe from MR images. TVRCs were constructed and analyzed. RESULTS: In non-smokers, the mean volume rates for each lung and lobe arranged in order of diminishing values were RL (54.8%), LL (45.2%), RLL (25.9%), LUL (24.2%), LLL (21.0%), RUL (18.5%), and RML (10.3%). In smokers, the mean volume rates for each lung and lobe arranged in order of diminishing values were RL (55.7%), LL (44.3%), RLL (25.1%), LUL (24.0%), RUL (21.0%), LLL (19.9%), and RML (9.5%). In both groups, there was a tendency for the fluctuation rates of the lower lobes to be higher than those of the upper and middle lobes. Although TVRCs for the right and left lung were nearly parallel, with no time lag, the peaks of TVRCs for the upper and middle lobes appeared slightly earlier than those for the lower lobes. CONCLUSION: The evaluation of pulmonary ventilation dynamics using MR may be a useful non-invasive technique by which to assess lung lobar ventilation.

Perfusion, inhalation, and ventilation studies in patients with primary lung carcinoma.

To determine the relative roles of perfusion, inhalation, and ventilation lung scintigraphy, studies were done respectively in 48, 50, and 34 of 54 primary lung carcinoma patients (prior to thoracotomy and subsequent radical tumor resection). We observed the relationships between both the size of perfusion, inhalation, and ventilation defects observed scintigraphically, and the size of the mass lesion seen radiologically. We also estimated the correlation between those scintigraphic classifications and pTN. Although inhalation and perfusion images exhibited similar patterns, in hilar type lung carcinoma patients, the inhalation study showed superior mass lesion delineation. In peripheral type lung carcinoma, however, the incidence of negative inhalation defect (52%) differed significantly from that of negative perfusion defect (21%) (p less than 0.05). Xe-133 gas ventilation has limitations but is occasionally helpful in detecting obstructive airway disease. The coexistence ratio of COPD in lung carcinoma patients showed no significant difference from that of COPD in the controls. Perfusion and inhalation classification did not correlate with pTN factor.

Effects of prophylactic irradiation of acute lymphoblastic leukemia in the central nervous system of children.

The development of prophylactic therapy for acute lymphoblastic leukemia (ALL) in the central nervous system in children has resulted in an improvement of the prognosis and prolongation of patients' lives. On the other hand, late irradiation effects have increased with the improvement of the prognosis. Therefore, there is now an important need to lessen these late effects without deteriorating the prognosis. We have investigated the recurrence rate, the survival rate and the cause of death of ALL patients who were divided into a none prophylactic irradiation group and 15 Gy, 20 Gy and 24 Gy irradiation groups. The results show that the effect of prophylactic irradiation has been more effective in the 20 Gy group than that of the 24 Gy group. Furthermore, brain atrophy and leukoencephalopathy, which were investigated by X-ray CT in long-term survivors of post-prophylactic irradiation without recurrence, have been less in the 20 Gy group than in that of the 24 Gy group. This report presents the result that the most favorable dose for prophylactic irradiation for central nervous system leukemia of ALL in children is 20 Gy by a step-up method.

Transvenous embolization of high flow carotid cavernous fistula: a case report.

We treated a patient with a high flow traumatic carotid-cavernous fistula (CCF) by embolization using detachable balloons and metallic coils, by transarterial and transvenous approaches. The patient was a 20-year-old woman who had fractures in the skull base from a traffic accident. She was admitted to our hospital one month after the accident due to exophthalmos, chemosis, and periorbital bruit. Cerebral angiograms demonstrated left traumatic CCF and steal phenomenon of blood flow. Balloon embolization by transarterial approach performed three times was unsuccessful, probably because of balloon puncture due to bone fragments. Embolization using metalic coils via the superior ophthalmic vein route was then attempted. As a result of this approach, complete obliteration of CCF was obtained, and clinical symptoms subsided within a few days. Treatment of CCF by transvenous approach is one alternative when transarterial occlusion is difficult.

CT-pathologic correlation in the tumor margins of lung cancer.

A CT-pathologic correlative study concerning the nature of the tumor margin was performed in 22 lung cancer cases with solitary nodules in the peripheral lung field. Three of 14 adenocarcinoma cases showed an ill-defined margin and an adjacent subtle high attenuation area on CT, representing alveoli lined by tumor cells. In the other 11 cases, both ill- and well-defined margins were seen on CT. A well-defined margin of the mass represented alveoli filled entirely with mucinous material or tumor cells. In three of six squamous cell carcinomas an ill-defined margin in CT represented the infiltration of lymphatic cells surrounding the tumor and continuous spread of tumor cells from neighboring alveoli. A well-defined margin in the other cases and two small cell carcinoma cases represented alveoli filled entirely with tumor cells. CT was shown to be very useful in the assessment of the nature of the margin of the tumor.

Radiologic manifestation of spinal accessory neurinoma: a case report.

Spinal accessory nerve neurinoma is very rare; only 12 cases have been reported in the literature. We describe a 54-year-old woman who had a cisterna magna tumor expanding extracranially to the C1 cervical body. The tumor originated from the spinal root of the right accessory nerve. The radiological features including CT, MRI, and angiography are discussed. There have been reports on cranial MRI studies of spinal accessory neurinoma. MRI suggested an extra-axial tumor. The histological diagnosis was mixed Antoni A and B neurinoma. The neuroradiological findings of 12 cases of neurinoma of the accessory nerve reported in the literature are reviewed.

Rounded atelectasis formation following decrease of pleural effusion: a case report.

We present a case of rounded atelectasis (RA) observed continuously following decrease of pleural effusion. The collapsed right lower lobe adjacent to pleural effusion changed its shape from wedge-shaped consolidation with air-bronchogram to oval and flat formation at last. The vessels and bronchi curving into RA were detected by CT and we called it horizontal comettail sign. When a mass is detected following decrease of pleural effusion and vessels and bronchi are curving into the mass, the mass may be diagnosed as RA and no further examination may be necessary.

Effect of chemotherapy on Tl-201 tumor uptake: experimental study.

The purpose of this study was to investigate the correlation between histopathological changes and 201Tl accumulation in tumor following chemotherapy in the evaluation of chemotherapeutic effect. Walker 256 carcinoma implanted in the right thighs of Wistar rats was treated by the intravenous injection of adriamycin (ADR). 201Tl scintigraphy was performed before and 12, 24, 72, and 96 hours after chemotherapy. The count ratio of tumor to normal muscle (T/N ratio) was measured by gamma camera. Tumor growth rate was measured and histopathological examination of the tumor after 201Tl scintigraphy was performed. The tumors of all rats disappeared within 2 weeks after chemotherapy, but the T/N ratio did not change. Histopathological findings showed a decrement of tumor cells, small necrotic regions, and remarkable lymphocyte infiltration in the tumor bed in the healing stage. After 201Tl injection, lymphocytes isolated from tumor tissue showed noted 201Tl accumulation. These results suggest that the T/N ratio did not decrease after ADR chemotherapy because of the accumulation of 201Tl in lymphocytes. We concluded that the use of 201Tl may underestimate the therapeutic effect of ADR.
