For the development of 99mTc labeled tumor seeking agents, 273 drugs and chemicals were evaluated. Agents labeled by SnCl2-HCl method were injected to rats bearing Yoshida sarcoma. Scintigrams of the rats 2 hours after injection and the excised tumor and organs placed on a plate were taken and evaluated their affinity to the tumor. Isoniazid (INH) was found to be the most useful agent as a result of this screening study. Intensive study on 99mTc-INH was subsequently performed. Another labeling method, FeSO4-H2SO4 method, was evaluated and shown to be superior in both absolute tumor uptake and tumor to muscle ratio. Uptake of 99mTc-INH to Yoshida sarcoma occurred early after injection and maintained its level until later time. On the other hand, radioactivity in the blood and muscle decreased rapidly; that is, tumor to blood and muscle ratio showed rapid increase. Other animal tumor models, mouse bearing Ehrlich tumor or Sarcoma 180, were also used to evaluate the usefulness of 99mTc-INH. The labeled agent showed good localization in these tumors too. In conclusion 99mTc-INH might be a good tumor seeking agent and further clinical trials would be warrant.
To get the better therapeutic ratio with low LET radiation therapy alone, the improvement of the physical dose distribution and the mode of dose fractionation are required. Since 1972, uneven fractionation irradiation with electron beam has been used for head and neck tumors. The 2 year control rates for primary tongue cancer T1, T2 and T3 by this method were approximately comparable to that of the small source implantation. In the treatment results for the cervical lymph node metastasis, uneven fractionation technique is superior to conventional one. Furthermore, this results was reviewed by means of multivariate analysis.

The present study was undertaken to investigate changes in the muscle fiber when treated with calcium ionophore. Muscles treated with ionophore showed disruption of the plasma membrane of the muscle fiber, delta lesions, marked contraction of the myofibrills, and dissolution of Z lines and I bands. Black granules of calcium pyroantimonate were observed inside the plasma membrane in ionophore-treated muscle fibers without alteration of the other muscle organelles. The density of the intramembranous particles was less in muscle treated with calcium ionophore than in the control muscle. These results support the previous hypothesis that the increased concentration of intracellular calcium activates calcium-activated neutral protease and induces necrosis of the myofiber. The mechanism for the decrease in the density of intramembranous particles is unsolved. However, the disruption of the plasma membrane may not be a direct effect of calcium ionophore on it, but a secondary phenomenon which occurs after the calcium-induced necrosis of the muscle fibers.
We examined the effect of local x-ray irradiation on the changes in the lipid peroxide level in submandibular gland, liver, and blood plasma. Rats five weeks of age received a single low dose of 3 Gy x-ray irradiation to their neck regions. The lipid peroxide level in the submandibular gland was significantly enhanced at seven days after irradiation, as was the level in the blood plasma at two hours, seven and 14 days after irradiation. The lipid peroxide level in the liver decreased, as compared with levels in the controls. There was a slight tendency for acinar cells of the submandibular gland to show pyknosis and anomalous nuclei within three days after irradiation. These results suggest that radiation injury results in an elevation of the lipid peroxides in the submandibular gland, and an increased level in the blood.

The kinetics of the hydrogen-deuterium exchange reactions of poly(dA).poly(rU) and poly(rA).poly(dT) has been examined, at pH 7.0 and at various temperatures in the 15-35 degrees C range, by stopped-flow ultraviolet spectrophotometry. For comparison, the deuteration kinetics of poly[d(A-T)].poly[d(A-T)] and poly(rA).poly(rU) has been reexamined. At 20 degrees C, the imino deuteration (NH----ND) rates of the two hybrid duplexes were found to be 1.5 and 1.8 s-1, respectively. These are nearly equal to the imino deuteration rates of poly[d(A-T)].poly[d(A-T)] (1.1 s-1) and poly(rA).poly(rU) (1.5 s-1) but appreciably higher than that of poly(dA).poly(dT) (0.35 s-1). It has been suggested that a DNA.RNA hybrid, an RNA duplex, and the AT-alternating DNA duplex have in general higher base-pair-opening reaction rates than the ordinary DNA duplex. The amino deuteration (NH2----ND2) rates, on the other hand, have been found to be 0.25, 0.28, and 0.33 s-1, respectively, for poly(dA).poly(rU), poly(rA).poly(dT), and poly[d(A-T)].poly[d(A-T)], at 20 degrees C. These are appreciably higher than that for poly(rA).poly(rU) (0.10 s-1). In general, the equilibrium constants (K) of the base-pair opening are considered to be greatest for the DNA.RNA hybrid duplex (0.05 at 20 degrees C), second greatest for the RNA duplex (0.02 at 20 degrees C), and smallest for the DNA duplex (0.005 at 20 degrees C), although the AT-alternating DNA duplex has an exceptionally great K (0.07 at 20 degrees C). From the temperature effect on the K value, the enthalpy of the base-pair opening was estimated to be 3.0 kcal/mol for the DNA.RNA hybrid duplex.
The gene expression of myelin basic proteins (MBPs) in shiverer mutant mice was investigated by the Northern and Southern hybridization techniques. In the control mice RNA molecules from the brains which were about 2,300 nucleotides in length were hybridized to cDNA of 1.8 kb encoding for a mouse MBP, but RNA from the brains of 3-week-old shiverer mutant mice contained no detectable amount of MBP transcripts hybridizing to this probe. Moreover the shiverer mutant mice lost several restriction fragments that hybridized to the same probe in the control mice when each of the five restriction enzymes, i.e., HindIII, PstI, PvuII, AccI, and StuI, was used. These data suggest that the shiverer mutation may correspond to the deletion of a large portion of MBP exon(s) in the gene, and this deletion causes inefficient transcription leading to the depletion of MBPs in the myelin and the dysmyelination observed in these mice.
Concomitant with frequent occurrence of disorders of the neck, arm, hand and low back among assembly-line female workers in confectionery work, there is an increased number of patients with occupational cervicobrachial disorders and/or low back pain. In suspicion of the close correlation between the working conditions and development of these local disorders, a field study was undertaken. The following are the results obtained. More than 90% of these assembly-line female workers consisted of inexperienced part-time employees, mostly of middle to old age. They were engaged in decorating conveyor-carried cakes with cream and chocolate. The work necessitated repetitive movements of the upper limbs and concentration of visuosensory and nervous attention in a half-sitting slouching posture. This was considered to exert excess load on the local muscles and nervous and sensory systems. The causes that intensified the local symptoms as pointed out by the workers consisted of (1) repetitive use of the arms and hands, (2) static posture during the work and (3) sustained standing position. Complaints of low back pain were conspicuous from the unsuitable height of a conveyor-belt. Thirty five percent of the female workers needed medical treatment for cervicobrachial and/or low back pain. The forced adaptation to the belt height, sustained unnatural working posture and the imposition of forced movements seemed to be the main factors in the onset of cervicobrachial and/or low back pain in interrelation with working hours. On the basis of these results, the work load on a machine-paced assembly-line was analyzed and the necessity of improvement of working conditions was discussed.
